| 1 | Microvessel changes in the gerbil hippocampus after cerebral ischemia and reperfusion by Buyang Huanwu decoction pretreatment显示文摘Buyang Huanwu decoction(BYHWD) is a classic recipe for the prevention and treatment of ischemic cerebrovascular disease.Gerbils were pretreated with BYHWD,and then subjected to cerebral ischemia and reperfusion.Microvascular changes were determined with laser Doppler monitoring,tannic acid-ferric chloride mordant,and electron microscopy.Results showed that BYHWD pretreatment could enhance the function of hippocampal microvessels,prevent injury,and increase microvasular density and microvasular area density.Thus,these results suggest that BYHWD pretreatment could prevent microvascular occlusion,enhance the capacity of microvascular reperfusion,increase cerebral blood flow,and inhibit neuronal damage,and may be an effective therapy against brain ischemic injury. | Xiaoguang Wu Yixue Li Haixia Liu Yuhua Yin Shumin Zhao Yuanyuan Guo | 2011 | Neural Regeneration Research2011,6,9: | 11 |
| 2 | The molecular determinants governing the immunogenicity of Japanese encephalitis live attenuated vaccines显示文摘In the course of isolating the attenuated Japanese encephalitis vaccine SA14-14-2,two attenuated strains SA14-9-7 and SA14-5-3 were also obtained that elicited low antibody responses in humans(o10%and 62%,respectively)and exerted much weaker immune protection in animal challenge experiments.However,the reason for these differences remains unknown.In order to understand why SA14-14-2 is superior to SA14-9-7 and SA14-5-3,we employed a reverse genetics method to identify the key mutations in the virus genome that determine the immunogenicity of live attenuated Japanese encephalitis viruses.We first sequenced the full genomic sequences of SA14-9-7 and SA14-5-3 and found mutations that changed four amino-acid base pairs when compared to the envelope gene of SA14-14-2.We mutated the genome of SA14-14-2 to generate these mutations both singly(E-177,E-264,E-279 and E-315)and in combination(E-177/264,E-279/315 and E-177/264/279/315)and tested these mutants along with parental strains SA14-14-2,SA14-9-7 and SA14-5-3 for their immunogenicity in vivo.When mice were immunized with SA14-9-7 and SA14-5-3,lower levels of neutralizing antibodies were generated compared with the immune response to SA14-14-2.Furthermore,SA14-5-3 was more immunogenic than SA14-9-7,which replicated the results previously seen in humans.Point mutations E-177,E-264,E-279 and E-315 diminished the immunogenicity of SA14-14-2 with E-264 and E-315,contributing the most to this phenotype.The mutant rJEV(E-177/E-264/E-279/E-315)containing all four point mutations exhibited the lowest immunogenicity with a seroconversion rate of 0 at an inoculation dose of 103 PFU(plaque-forming unit).We have identified the key amino acids in the envelope protein that account for the superior immunogenicity of SA14-14-2. | Yuhua Li Yin Fu Xinyu Liu Huiqiang Yang Yongxin Yu Lili Jia Xuguang Li Aaron Farnsworth Junzhi Wang | 2017 | Signal Transduction and Targeted Therapy2017,2,1: | 3 |
| 3 | Two-stage degradation and novel functional endothelium characteristics of a 3-D printed bioresorbable scaffold显示文摘Bioresorbable scaffolds have emerged as a new generation of vascular implants for the treatment of atherosclerosis,and designed to provide a temporary scaffold that is subsequently absorbed by blood vessels over time.Presently,there is insufficient data on the biological and mechanical responses of blood vessels accompanied by bioresorbable scaffolds(BRS)degradation.Therefore,it is necessary to investigate the inflexion point of degradation,the response of blood vessels,and the pathophysiological process of vascular,as results of such studies will be of great value for the design of next generation of BRS.In this study,abdominal aortas of SD rats were received 3-D printed poly-l-actide vascular scaffolds(PLS)for various durations up to 12 months.The response of PLS implanted aorta went through two distinct processes:(1)the neointima with desirable barrier function was obtained in 1 month,accompanied with slow degradation,inflammation,and intimal hyperplasia;(2)significant degradation occurred from 6 months,accompanied with decreasing inflammation and intimal hyperplasia,while the extracellular matrix recovered to normal vessels which indicate the positive remodeling.These in vivo results indicate that 6 months is a key turning point.This“two-stage degradation and vascular characteristics”is proposed to elucidate the long-term effects of PLS on vascular repair and demonstrated the potential of PLS in promoting endothelium function and positive remodeling,which highlights the benefits of PLS and shed some light in the future researches,such as drug combination coatings design. | Tieying Yin Ruolin Du Yang Wang Junyang Huang Shuang Ge Yuhua Huang Youhua Tan Qing Liu Zhong Chen Hanqing Feng Jie Du Yazhou Wang Guixue Wang | 2022 | Bioactive Materials2022,7,4: | 2 |

We analyzed the relationship between the “wrap around” effect and the distribution of FRAT (Finite Radon Transform) coefficients first, and then based on study of some properties of the columnwisely FRAT reconstruction procedure, we proposed an energy-based adaptive orthogonal FRIT scheme (EFRIT). Experiments using nonlinear approximation show its superiority in energy concentration over both Discrete Wavelet Transform (DWT) and Finite Ridgelet Transform (FRIT). Furthermore, we have modeled the denoising problem and proposed a novel threshold selecting method. | 13 | The synthesis of PROTAC molecule and new target KAT6A identification of CDK9 inhibitor i CDK9显示文摘Cyclin-dependent kinases(CDKs) have become potential targets for treating various diseases, especially cancer. Compound i CDK9 is an excellent and selective CDK9 inhibitor, but its major limitation is the potential toxicity and poor understanding of the underlying mechanism. The PROTAC(proteolysis targeting chimera) degraders of bioactive molecules can significantly induce in vitro and in vivo degradation of their target protein with high selectivity and effectively reduce the dose-limiting toxicity of small molecule drugs. Therefore, we designed and synthesized the bifunctional PROTAC molecules of i CDK9, being used for identifying its previously unknown target and revealing the underlying pharmacological mechanism.The PROTAC bifunctional molecule CD-5 could selectively and significantly degrade CDK9 with low cell toxicity. Therefore, we selected CD-5 as a chemical prober in the SILAC quantitative proteomic analysis, which disclosed that CD-5 could enormously lessen the lysine acetyltransferase KAT6A. Furthermore,KAT6A degradation induced by CD-5 repressed the levels of H3K14Ac and H3K23Ac. Lastly, the streptavidin immunoprecipitation(IP) assay confirmed a direct interaction between KAT6A and i CDK9. Collectively, our results uncover that KAT6A is a potential non-kinase target of i CDK9.
