1SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality.Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li 2019Horticulture Research2019,6,1:39
2Biofunctional magnesium coated Ti6Al4V scaffold enhances osteogenesis and angiogenesis in vitro and in vivo for orthopedic application显示文摘The insufficient osteogenesis and osseointegration of porous titanium based scaffold limit its further application.Early angiogenesis is important for scaffold survival.It is necessary to develop a multifunctional surface on titanium scaffold with both osteogenic and angiogenic properties.In this study,a biofunctional magnesium coating is deposited on porous Ti6Al4V scaffold.For osseointegration and osteogenesis analysis,in vitro studies reveal that magnesium-coated Ti6Al4V co-culture with MC3T3-E1 cells can improve cell proliferation,adhesion,extracellular matrix(ECM)mineralization and ALP activity compared with bare Ti6Al4V cocultivation.Additionally,MC3T3-E1 cells cultured with magnesium-coated Ti6Al4V show significantly higher osteogenesisrelated genes expression.In vivo studies including fluorochrome labeling,micro-computerized tomography and histological examination of magnesium-coated Ti6Al4V scaffold reveal that new bone regeneration is significantly increased in rabbits after implantation.For angiogenesis studies,magnesium-coated Ti6Al4V improve HUVECs proliferation,adhesion,tube formation,wound-healing and Transwell abilities.HUVECs cultured with magnesium-coated Ti6Al4V display significantly higher angiogenesis-related genes(HIF-1αand VEGF)expression.Microangiography analysis reveal that magnesium-coated Ti6Al4V scaffold can significantly enhance the blood vessel formation.This study enlarges the application scope of magnesium and provides an optional choice to the conventional porous Ti6Al4V scaffold with enhanced osteogenesis and angiogenesis for further orthopedic applications.Peng Gao Bo Fan Xiaoming Yu Wenwen Liu Jie Wu Lei Shi Di Yang Lili Tan Peng Wan Yulin Hao Shujun Li Wentao Hou Ke Yang Xiaokang Li Zheng Guo 2020Bioactive Materials2020,5,3:15
3Cotton bollworm resistance to Bt transgenic cotton: A case analysis显示文摘Cotton bollworm (Helicoverpa armigera) is one of the most serious insect pests of cotton. Transgenic cotton expressing Cry toxins derived from a soil bacterium, Bacillus thuringiensis (Bt), has been produced to target this pest. Bt cotton has been widely planted around the world, and this has resulted in efficient control of bollworm populations with reduced use of synthetic insecticides. However, evolution of resistance by this pest threatens the continued success of Bt cotton. To date, no field populations of bollworm have evolved significant levels of resistance; however, several laboratory-selected Cry-resistant strains of H. armigera have been obtained, which suggests that bollworm has the capacity to evolve resistance to Bt. The development of resistance to Bt is of great concern, and there is a vast body of research in this area aimed at ensuring the continued success of Bt cotton. Here, we review studies on the evolution of Bt resistance in H. armigera, focusing on the biochemical and molecular basis of Bt resistance. 4Genome-wide Dissection of Co-selected UV-B Responsive Pathways in the UV-B Adaptation of Qingke显示文摘Qingke (Tibetan hulless barley) has long been cultivated and exposed to long-term and strong UV-B radiation on the Tibetan Plateau, which renders it an ideal species for elucidating novel UV-B responsive mechanisms in plants. Here we report a comprehensive metabolite profiling and metabolite-based genome-wide association study (mGWAS) using 196 diverse qingke and barley accessions. Our results demonstrated both constitutive and induced accumulation, and common genetic regulation, of metabolites from different branches of the phenylpropanoid pathway that are involved in UV-B protection. A total of 90 significant mGWAS loci for these metabolites were identified in barley-qingke differentiation regions, and a number of high-level metabolite trait alleles were found to be significantly enriched in qingke, suggesting co-selection of various phenylpropanoids. Upon dissecting the entire phenylpropanoid pathway, we identified some key determinants controlling natural variation of phenylpropanoid content, including three novel proteins, a flavone C-pentosyltransferase, a tyramine hydroxycinnamoyl acyltransferase, and a MYB transcription factor. 5Perchlorate removal using granular activated carbon supported iron compounds: Synthesis, characterization and reactivity显示文摘Synthesis and use of the iron compounds supported on granular activated carbon (ICs/GAC) have shown significant environmental implications for perchlorate (ClO 4 ) removal. ICs/GAC was synthesized via hydrolyzing FeSO 4 ·7H 2 O on GAC, reduced by NaBH 4 solution in polyethylene glycol 6000 and ethanol solution, dried in vacuum condition and exposed to air. Synthesized ICs/GAC was characterized using transmission electron micrograph (TEM), Brunauer-Emmett-Teller, X-ray photoelectron spectroscopy (XPS). ICs/GAC was determined to be containing a large amount of FeOHSO 4 , Fe 2 O 3 and a small amount of zero-valent iron (ZVI) nanoparticles according to TEM and XPS measurements. Batch static kinetic tests showed that 97% of ClO 4 was removed within 10 hr at 90°C and 86% of ClO 4 was removed within 12 hr at 25°C, at ICs/GAC dosage of 20 g/L. The experimental results also showed that FeOHSO 4 and Fe 2 O 3 nanoparticles have the function of perchlorate adsorption and play important roles in ClO 4 removal. 6Characteristics and model studies for fluoride and arsenic adsorption on goethite显示文摘Fluoride and arsenic are major anionic elements of concern in drinking water treatment. The effects of contact time, pH, surface loading and ionic strength on adsorption of fluoride and As(V) were investigated using batch methods. Adsorption of fluoride and As(V) onto goethite obeyed a pseudo second-order rate law. Through experimental data and adsorption kinetic analysis, the affinity of As(V) onto goethite was stronger than fluoride. Fluoride and As(V) uptake by goethite all decreased with pH increasing at the same surface loading; however, ionic strength had slight influence on their adsorption. A surface sites-species model was used to quantify the adsorption of fluoride and As(V) onto goethite as function of pH and surface loading. 7High-Energy Batteries:Beyond Lithium-Ion and Their Long Road to Commercialisation显示文摘Rechargeable batteries of high energy density and overall performance are becoming a critically important technology in the rapidly changing society of the twenty-first century.While lithium-ion batteries have so far been the dominant choice,numerous emerging applications call for higher capacity,better safety and lower costs while maintaining sufficient cyclability.The design space for potentially better alternatives is extremely large,with numerous new chemistries and architectures being simultaneously explored.These include other insertion ions(e.g.sodium and numerous multivalent ions),conversion electrode materials(e.g.silicon,metallic anodes,halides and chalcogens)and aqueous and solid electrolytes.However,each of these potential“beyond lithium-ion”alternatives faces numerous challenges that often lead to very poor cyclability,especially at the commercial cell level,while lithium-ion batteries continue to improve in performance and decrease in cost.This review examines fundamental principles to rationalise these numerous developments,and in each case,a brief overview is given on the advantages,advances,remaining challenges preventing cell-level implementation and the state-of-the-art of the solutions to these challenges.Finally,research and development results obtained in academia are compared to emerging commercial examples,as a commentary on the current and near-future viability of these“beyond lithium-ion”alternatives.Yulin Gao Zhenghui Pan Jianguo Sun Zhaolin Liu John Wang 2022Nano-Micro Letters2022,14,6:5
8Facile one-step synthesis of graphitic carbon nitride-modified biochar for the removal of reactive red 120 through adsorption and photocatalytic degradation显示文摘Converting waste biomass into value-added biochar has been considered as a green and sustainable strategy for resource management and pollution control.In this study,graphitic carbon nitride(g-C_(3)N_(4))modified biochars(BCs)were produced through one-pot pyrolysis of urea and hickory chips in differential ratios at 520℃.The resulting BC/g-C_(3)N_(4)composites were evaluated in laboratory for their physicochemical,adsorptive,and photocatalytic properties.The characterization tests showed the successful synthesis of the BC/g-C_(3)N_(4)composites that introduced g-C_(3)N_(4)structure,N-containing surface func-tional groups,reduced surface area,and better thermal stability to the biochar.After modification,the BC/g-C_(3)N_(4)composites showed better adsorption ability to reactive red 120(RR120)than the pristine BC,due to the strong electrostatic attrition between N-containing functional groups of g-C_(3)N_(4)on biochar surface and anionic RR120.The BC/g-C_(3)N_(4)composites also inherited g-C_(3)N_(4)’s photocatalytic activity,which is visible light responsive to generate free radicals for RR120 degradation.In addition,the composites with higher urea modification ratios were more effective in the degradation of RR120.Overall,this study demonstrates the feasibility and promising potential of combining biochar and photocatalyst for the removal of aqueous dye.Because of the synergistic adsorption and photodegradation ability,BC/g-C_(3)N_(4)composites present a novel and cost-effective solution for the removal of aqueous dye and other photodegradable contaminants under natural conditions.Yulin Zheng Yichen Yang Yue Zhang Weixin Zou Yidan Luo Lin Dong Bin Gao 2019Biochar2019,1,1:5
9SSHT Process -a Low Cost Solution for Low Sulfur and Low Aromatics Diesel显示文摘The need for cleaner fuels has resulted in a continuing worldwide trend to reduce diesel sulfur and aromatics. There are many approaches to reducing sulfur and aromatics in diesel. Most of them have a common drawback of high cost because of adopting two stages of hydrotreating and using noble-metal catalyst, especially for reducing aromatics. The attempt to resolve this issue has led to the recent development of the Single Stage Hydrotreating (SSHT) process by Research Institute of Petroleum Processing (RIPP), SINOPEC.The SSHT process is a single-stage hydrotreating technology for producing low sulfur and low aromatics diesel. The process uses one or two non-noble-metal catalysts system and operates at moderate pressure. When revamping an existing unit to meet low aromatics diesel specification, the only thing to do is to add a reactor or replace the existing reactor, In pilot plant tests, the SSHT technology has successfully treated SRGO (Straight Run Gas Oil), LCO (Light Cycle Oil) or the blend of them. It is shown that by using the SSHT process diesel with sulfur of 30 ppm and aromatics of 15 m% can be produced from Middle-East SRGO and diesel with aromatics content of 25 m% can be produced from cracked feed, such as FCC-LCO. High diesel yield and cetane number gain (from cracked feed stocks) give the SSHT technology a performance advantage compared to conventional hydrocracking and hydrotreating processes.The lower investment and operating cost is another advantage. 10Mutant p53 increases exosome-mediated transfer of mi R-21-3p and mi R-769-3p to promote pulmonary metastasis显示文摘Objective: Tumor metastasis is a complex, multistep process that depends on tumor cells and their communication with the tumor microenvironment. A p53 gain-of-function mutant has been shown to enhance the tumorigenesis, invasion, and metastasis abilities of tumor cells. This study aimed to investigate the roles of p53 R273 H mutation in the tumor microenvironment.Methods: The in vitro and in vivo effects of the p53 R273 H mutant on the invasion and metastasis of HCT116 cells were investigated. Exosomes from wild-type and HCT116-TP53(R273 H) cells were cocultured with mouse embryonic fibroblasts(MEFs). The roles of differentially expressed exosomal micro RNAs identified by microarray analysis were investigated. The functions of the p53 R273 H mutant in tumor cells were also investigated via gene expression microarray and quantitative polymerase chain reaction(q PCR) analyses.Results: Introducing p53 R273 H mutant into HCT116 cells significantly potentiated pulmonary metastasis in vivo. In the presence of exosomes derived from HCT116-TP53(R273 H) cells, the exosomes were taken up by MEFs and became activated. Microarray analysis showed that the p53 R273 H mutation increased the exosomal levels of mi R-21-3 p and mi R-769-3 p. Intriguingly, in clinical samples, mi R-21-3 p and mi R-769-3 p levels were significantly higher in patients with a p53 mutation than in those without this mutation. Furthermore, both mi R-21-3 p and mi R-769-3 p activated fibroblasts and exerted a synergistic effect via their target genes on the transforming growth factor-β(TGF-β)/Smad signaling pathway. The activated fibroblasts excreted cytokine TGF-β and may have reciprocally induced cancer cells to undergo epithelial-mesenchymal transition(EMT). Indeed, HCT116-TP53(R273 H) cells showed increased expression of ZEB1 and SNAI2 and decreased transcription of several cell adhesion molecules.Conclusions: The mutant p53-exosomal mi R-21-3 p/mi R-769-3 p-fibroblast-cytokine circuit appears to be responsible for communication between tumor and stromal cells, with exosomal mi RNAs acting as a bridge. mi R-21-3 p and mi R-769-3 p are potential predictive markers of pulmonary metastasis and candidate targets for therapeutic interventions.Qiang Ju Lina Zhao Jiajia Gao Lanping Zhou Yang Xu Yulin Sun Xiaohang Zhao 2019Chinese Journal of Cancer Research2019,31,3:4

13Effect of fluoride on the corrosion behavior of nanostructured Ti-24Nb-4Zr-8Sn alloy in acidulated artificial saliva显示文摘The surface of titanium dental implants is highly susceptible to aggressive fluoride ions in the oral environment. Nanotechnology has proven an effective approach to improve the stability and corrosion resistance of titanium by applying a passive film. In this study, we investigated the effects of fluoride on the corrosion behavior of nanostructured(NS) Ti-24 Nb-4 Zr-8 Sn(Ti2448) alloy in acidulated artificial saliva(AAS)at 37 ℃, and then conducted comparisons with its coarse grained(CG) counterpart. Electrochemical techniques, such as potentiodynamic polarization and electrochemical impedance spectroscopy(EIS), as well as surface analysis including X-ray photoelectron spectroscopy(XPS) with argon ion sputtering, and scanning electronic microscopy(SEM) were employed to evaluate the effects of fluoride on sensitivity to pitting and the tolerance of Ti2448 to fluoride in AAS solution. The results demonstrate that corrosion current density increased with F-concentration. In all respects, the NS Ti2448 alloy presented corrosion resistance superior to that of its coarse grained(CG) counterpart at low F-concentrations(<0.1%).Furthermore, a high content of F-(1%) was shown to promote the active dissolution of both alloys by increasing the rate of corrosion. Following immersion in the fluoridated AAS solution for 60 days, a tissuefriendly compound, Ca_3(PO_4)_2, was detected on the surface of the NS when F-= 0.01% and Na_2 TiF_6 was identified as the main component in the corrosion products of the CG as well as NS Ti2448 alloys when F-= 1%. High concentrations of F-produced pitting corrosion on the CG alloy, whereas NS Ti2448 alloy presented general corrosion in the form of lamellar separation under the same conditions. 14Gold nanoparticle-directed autophagy intervention for antitumor immunotherapy via inhibiting tumor-associated macrophage M2 polarization显示文摘Tumor-associated macrophages(TAMs),one of the dominating constituents of tumor microenvironment,are important contributors to cancer progression and treatment resistance.Therefore,regulation of TAMs polarization from M2 phenotype towards M1 phenotype has emerged as a new strategy for tumor immunotherapy.Herein,we successfully initiated antitumor immunotherapy by inhibiting TAMs M2 polarization via autophagy intervention with polyethylene glycol-conjugated gold nanoparticles(PEG-Au NPs).PEG-Au NPs suppressed TAMs M2 polarization in both in vitro and in vivo models,elicited antitumor immunotherapy and inhibited subcutaneous tumor growth in mice.As demonstrated by the m RFP-GFP-LC3 assay and analyzing the autophagy-related proteins(LC3,beclin1 and P62),PEGAu NPs induced autophagic flux inhibition in TAMs,which is attributed to the PEG-Au NPs induced lysosome alkalization and membrane permeabilization.Besides,TAMs were prone to polarize towards M2phenotype following autophagy activation,whereas inhibition of autophagic flux could reduce the M2polarization of TAMs.Our results revealed a mechanism underlying PEG-Au NPs induced antitumor immunotherapy,where PEG-Au NPs reduce TAMs M2 polarization via induction of lysosome dysfunction and autophagic flux inhibition.This study elucidated the biological effects of nanomaterials on TAMs polarization and provided insight into harnessing the intrinsic immunomodulation capacity of nanomaterials for effective cancer treatment.Siyue Zhang Fangyuan Xie Kaichun Li He Zhang You Yin Yuan Yu Guangzhao Lu Shihao Zhang Yan Wei Ke Xu Yan Wu Hong Jin Lan Xiao Leilei Bao Can Xu Yulin Li Ying Lu Jie Gao 2022Acta Pharmaceutica Sinica B2022,12,7:2

16Bottom-up lithium growth guided by Ag concentration gradient in 3D PVDF framework towards stable lithium metal anode显示文摘Three-dimensional(3 D)frameworks have received much attention as an effective modification strategy for next-generation high-energy-density lithium metal batteries.However,the top-growth mode of lithium(Li)on the 3 D framework remains a tough challenge.To achieve a uniform bottom-up Li growth,a scheme involving Ag concentration gradient in 3 D PVDF framework(C-Ag/PVDF)is proposed.Ag nanoparticles with a concentration gradient induce an interface activity gradient in the 3 D framework,and this gradient feature is still maintained during the cycle.As a result,the C-Ag/PVDF framework delivers a long lifespan over 1800 h at a current density of 1 mA cm^(-2) with a capacity of 1 mAh cm^(-2),and shows an ultra-long life(>1300 h)even at a high current density of 4 mA cm^(-2) with a capacity of 4 mAh cm^(-2).The advantage of concentration gradient provides further insights into the optimal design of the 3 D framework for stable Li metal anode.Yulin Zhao Liping Wang Jian Zou Qiwen Ran Li Li Pengyu Chen Hailong Yu Jian Gao Xiaobin Niu 2022Journal of Energy Chemistry2022,31,2:2
17Effects of surface states over core-shell Ni@SiO_2 catalysts on catalytic partial oxidation of methane to synthesis gas显示文摘In the present work, core-shell Ni@Si O2 catalysts were investigated in order to evaluate the relevance of catalytic activity and surface states of Ni core as well as Ni nanoparticles size to catalytic partial oxidation of methane(POM). The catalysts were characterized by N2 adsorption,H2-TPR, XRD, TEM and XPS techniques. The catalytic performance of the core-shell catalysts was found to be dependent on the surface states of catalyst, which influenced the formation of products. It was considered that carbon dioxide formed on the oxidized nickel sites(Ni O)and carbon monoxide produced on the reduced sites(Ni). The surface states of active metal in the dynamic were influenced both by the size of Ni core and the porosity of silica shell. However, the catalytic activity would be debased when the size of Ni core was under a certain extent,which can be ascribed to the fact the carbon deposition increased with the increasing content of Ni O.
