2High-speed image reconstruction for optically sectioned,super-resolution structured illumination microscopy显示文摘Super-resolution structured illumination microscopy(SR-SIM)is an outstanding method for visualizing the subcellular dynamics in living cells.To date,by using elaborately designed systems and algorithms,SR-SIM can achieve rapid,optically sectioned,SR observation with hundreds to thousands of time points.However,real-time observation is still out of reach for most SIM setups as conventional algorithms for image reconstruction involve a heavy computing burden.To address this limitation,an accelerated reconstruction algorithm was developed by implementing a simplified workflow for SR-SIM,termed joint space and frequency reconstruction.This algorithm results in an 80-fold improvement in reconstruction speed relative to the widely used Wiener-SIM.Critically,the increased processing speed does not come at the expense of spatial resolution or sectioning capability,as demonstrated by live imaging of microtubule dynamics and mitochondrial tubulation.Zhaojun Wang Tianyu Zhao Huiwen Hao Yanan Cai Kun Feng Xue Yun Yansheng Liang Shaowei Wang Yujie Sun Piero RBianco Kwangsung Oh Ming Lei 2022Advanced Photonics2022,4,2:9
3Sodium Stress in the Halophyte Thellungiella halophila and Transcriptional Changes in a thsos1-RNA Interference Line显示文摘The plasma membrane Na^+/H^+-antiporter salt overly sensitive1(SOS1)from the halophytic Arabidopsis-relative Thel-lungiella halophila(ThSOS1)shows conserved sequence and domain structure with the orthologous genes fromArabidopsis thaliana and other plants.When expression of ThSOS1 was reduced by RNA interference(RNAi),pronouncedcharacteristics of salt-sensitivity were observed.We were interested in monitoring altered transcriptional responsesbetween Thellungiella wild type and thsosl-4,a representative RNAi line with particular emphasis on root responses to saltstress at 350 mmol/L NaCl,a concentration that is only moderately stressful for mature wild type plants.Transcript profilingrevealed several functional categories of genes that were differently affected in wild-type and RNAi plants.Down-regulationof SOS1 resulted in different gene expression even in the absence of stress.The pattern of gene induction in the RNAi plantunder salt stress was similar to that of glycophytic Arabidopsis rather than that of wild type Thellungiella.The RNAi plantsfailed to down-regulate functions that are normally reduced in wild type Thellungiella upon stress and did not up-regulatefunctions that characterize the Thellungiella salt stress response.Metabolite changes observed in wild type Thellungiellaafter salt stress were less pronounced or absent in RNAi plants.Transcript and metabolite behavior suggested SOS1functions including but also extending its established function as a sodium transporter.The down-regulation of ThSOS1converted the halophyte Thellungiella into a salt-sensitive plant.Dong-Ha Oh Qingqiu Gong Alex Ulanov Quan Zhang Youzhi Li Wenying Ma Dae-Jin Yun Ray A. 4液态与固态酵素理化成分及抗氧化活性的研究显示文摘为了对比分析液态酵素与固态酵素这2类市场上常见的酵素产品的差异,以3种液态酵素和3种固态酵素为原料,对其理化成分、生物活性物质及其体外抗氧化活性进行了分析与比较。结果显示,液态酵素产品的总酸、还原糖、可溶性固形物、总酚、总黄酮、花青素含量均显著高于固态酵素,液态酵素超氧化物歧化酶(superoxide dismutase,SOD)活力较高,但差异很大,在193.23~1214.46 U/mL,显著高于固态酵素;2类酵素的蛋白酶、脂肪酶、α-淀粉酶活力均较低,液态酵素蛋白酶、脂肪酶活力显著高于固态酵素,而液态酵素几乎没有α-淀粉酶活力。不同酵素产品具有较强的抗氧化能力且差异较大,液态酵素1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基清除能力的IC 50值在5.65~21.08μL/mL,而固态酵素最小的IC50值为29.77μL/mL,液态酵素的抗氧化能力显著强于固态酵素。该研究为帮助人们了解不同状态的酵素产品品质,正确看待酵素产品提供了参考,对于促进酵素行业健康发展具有指导意义。余思洁 OH YOUNG JOO 李洪军 KIM TAE SUK 贺稚非 LEE SANG YUN 李少博 李敏涵 2020食品与发酵工业2020,46,13:6

6Gardenia jasminoides protects against cerulein-induced acute pancreatitis显示文摘AIM: To investigate the effect of Gardenia jasminoides (GJ) on cerulein-induced acute pancreatitis (AP) in mice.METHODS: C57BL/6 mice weighing 18-20 g were divided into three groups.(1) Normal saline-treated group,(2) treatment with GJ at a dose of 0.1 g/kg,(3) treatment with GJ at a dose of 1 g/kg.GJ was administered orally (n = 6 per group) for 1 wk.Three hours later,the mice were given an intraperitoneal injection of cerulein (50 μg/kg),a stable cholecystokinin (CCK) analogue,every hour for a total of 6 h as described previously.The mice were sacrificed at 6 h after completion of cerulein injections.Blood samples were obtained to determine serum amylase,lipase and cytokine levels.The pancreas was rapidly removed for morphologic examination and scoring.A portion of pancreas was stored at -70℃ and prepared for the measurement of tissue myeloperoxidase (MPO) activity,an indicator of neutrophil sequestration,and for reverse-transcriptase PCR (RT-PCR) and real-time PCR measurements.RESULTS: Treatment with GJ decreased significantly the severity of pancreatitis and pancreatitis-associated lung injury.Treatment with GJ attenuated the severity of AP compared with saline-treated mice,as shown by reduction in pancreatic edema,neutrophil infiltration,serum amylase and lipase levels,serum cytokine levels,and mRNA expression of multiple inflammatory mediators.CONCLUSION: These results suggest that GJ attenuated the severity of AP as well as pancreatitis-associated lung injury.Won-Seok Jung Young-Seok Chae Do-Yun Kim Sang-Wan Seo Hee-Je Park Gi-Sang Bae Tae-Hyeon Kim Hyo-Jeong Oh Ki-Jung Yun Rae-Kil Park Jong-Suk Kim Eun-Cheol Kim Sung-Yeon Hwang Sung-Joo Park Ho-Joon Song 2008World Journal of Gastroenterology2008,14,40:4
7Analysis on sports and recreation activity-related eye injuries presenting to the Emergency Department显示文摘AIM:To investigate the incidence and general characteristics of sports-related eye injuries in patients visiting the Emergency Department. METHODS: A cross-sectional,multi-center,observational study.Patients with an injured eye who visited the Emergency Department at one of nine hospitals in Korea were enrolled.All data were prospectively collected between March and September 2010 using a questionnaire.Eye injuries that occurred during risky sports were examined by gender and age.Additionally,the rate of open globe injuries that occurred with and without protective eyewear was examined for each activity.Continuous variables were compared using Student's t-test and categorical variables were compared using Chi-square test.RESULTS: A total of 446 patients had sports-related eye injuries.Teenagers(10-19 years old) and young adults(20-29 years old) had the most eye injuries.Eye injuries accounted for 0.2% of Emergency Department patients.Baseball was the most common cause of sports-related eye injuries,followed by soccer and hiking.Protective gear was worn by 9.4% of all patients.Patients that were 30-39 years of age had the highest rate of protective gear use,followed by patients that were 40-49 years of age.The proportion of sports-related eye injuries that were open-globe injuries was highest for soccer and hiking.CONCLUSION: Although injuries were most common in patients below the age of 10 years,these patients had the lowest rate of protective eyewear use.Injuries in adults over 40 years of age most commonly occurred during hiking,but the rate of protective eyewear use was low.Young athletes should be educated on and provided with protective eyewear and policies protective gear use should be established.For older adults,eye protection should be encouraged,especially during hiking.Sungbae Moon Hyun Wook Ryoo Jae Yun Ahn Jung Bae Park Kang Suk Seo Sang Do Shin Kyoung Jun Song Kang Hyun Lee In Sool Yoo Jin Seong Cho Hyun Ho Ryu Tae Oh Jeong Seok Ran Yeom Young Taek Kim Sung Ok Hong 2016International Journal of Ophthalmology(English edition)2016,9,10:3
8Effects of Gastrodiae rhizoma on proliferation and differentiation of human embryonic neural stem cells显示文摘Objective:To investigate the effects of Gastrodiae rhizoma,a dried root of Gastrodia elata Blume,on proliferation and differentiation of human NSCs derived from embryonic stem cells.Methods:A 70%ethanol extract of Gastrodiae rhizoma(EEGR) was estimated with4-hydroxybenzyl alcohol as a representative constituent by HPLC.Results:MTT assay showed that the treatment with EEGR increased the viability of NSCs in growth media.Compared to contro1,EEGR increased the number of dendrites and denritic spines extended from a differentiated NSC.Whereas EEGR decreased the mRNA expression of Nestin,it increased that of Tuj1 and MAP2 in NSCs grown in differentiation media.Immunocytochemical analysis using confocal microscopy also revealed the increased expression of MAP2 in dendrites of EEGR-treated NSCs.Furthermore,EEGR decreased mRNA expression of Sox2 in NSCs grown even in growth media.Conclusions:In conclusion,our study demonstrates for the first time that EEGR induced proliferation and neuronal differentiation of NSCs,suggesting its potential benefits on NSC-based therapies and neuroregeneration in various neurodegenerative diseases and brain Injuries.Samrat Baral Ramesh Pariyar Chi-Su Yoon Dong-Cheol Kim Jong-Min Yun Seok O Jang Sung Yeon Kim Hyuncheol Oh Youn-Chul Kim Jungwon Seo 2015Asian Pacific Journal of Tropical Medicine2015,8,10:2

14Attenuated FOLFIRINOX in the salvage treatment of gemcitabine-refractory advanced pancreatic cancer: a phase II study显示文摘Background:Combination therapy with oxaliplatin,irinotecan,fluorouracil,and leucovorin(FOLFIRINOX)chemotherapy drastically improves survival of advanced pancreatic cancer patients.However,the efficacy of FOLFIRINOX as a second-line treatment after gemcitabine failure has not been tested prospectively.We investigated the feasibility and safety of attenuated FOLFIRINOX in patients with gemcitabine-refractory advanced pancreatic cancer.Methods:A multicenter phase II prospective open-label,single-arm study was conducted at 14 hospitals.Patients with histologically proven invasive ductal pancreatic adenocarcinoma,a measurable or evaluable lesion,Eastern Cooperative Oncology Group performance status 0 or 1,adequate organ function,and aged 19 years or older were eligible.Attenuated FOLFIRINOX consisted of oxaliplatin 65 mg/m2,irinotecan 135 mg/m2,and leucovorin 400 mg/m2 injected intravenously on day 1 and 5-fluorouracil 2000 mg/m2 continuously infused intravenously over 46 h on days 1-2,repeated every 2 weeks.The primary endpoint was progression-free survival from the initiation of FOLFIRINOX.Secondary endpoints were the objective response rate,disease control rate,overall survival,safety,and tolerability.We estimated overall survival and progression-free survival using the Kaplan-Meier methods.Results:We enrolled 39 patients from 14 institutions.The objective response rate was 10.3%,while the disease control rate was 64.1%.The 6-month and 1-year overall survival rates were 59.0%and 15.4%,respectively.Median progression-free survival and overall survival were 3.8 months(95%confidence interval[CI]1.5-6.0 months)and 8.5 months(95%CI 5.6-11.4 months),respectively.Grade 3 or 4 adverse events were neutropenia(41.0%),nausea(10.3%),anorexia(10.3%),anemia(7.7%),mucositis(7.7%),pneumonia/pleural effusion(5.1%),and fatigue(5.1%).One treatment-related death attributable to septic shock occurred.Conclusion:Attenuated FOLFIRINOX may be promising as a second-line therapy for gemcitabine-refractory pancre-atic cancer.Jung Hoon Kim Sang-Cheol Lee Sung Yong Oh Seo-Young Song Namsu Lee Eun Mi Nam Soonil Lee In Gyu Hwang Hyo Rak Lee Kyu Taek Lee Sang-Byung Bae Han Jo Kim Joung Soon Jang Do Hyoung Lim Hyun Woo Lee Seok Yun Kang Jung Hun Kang 2018Cancer Communications2018,38,1:2

16Bisphosphonate use and gastrointestinal tract cancer risk:Meta-analysis of observational studies显示文摘AIM:To perform a meta-analysis of observational studies to further elucidate the relationship between oral bisphosphonate use and gastrointestinal cancer risk.METHODS:Systematic literature search was conducted in MEDLINE,EMBASE,and the Cochrane Library to identify studies through January 2011.Search terms were 'bisphosphonates' or trade names of the drugs,and 'observational studies' or 'cohort studies' or 'case-control studies'.Two evaluators reviewed and selected articles on the basis of predetermined selection criteria as followed:(1) observational studies(casecontrol or cohort studies) on bisphosphonate use;(2) with at least 2 years of follow-up;and(3) reported data on the incidence of cancer diagnosis.The DerSimonian and Laird random effects model were used to calculate the pooled relative risk(RR) with 95% confidence interval(CI).Two-by-two contingency table was used to calculate the outcomes not suitable for meta-analysis.Subgroup meta-analyses were conducted for the type of cancer(esophageal,gastric and colorectal cancers).Sensitivity analyses were performed to examine the effect sizes when only studies with long-term follow-up(mean 5 years;subgroup 3 years) were included.RESULTS:Of 740 screened articles,3 cohort studies and 3 case-control studies were included in the analyses.At first,4 cohort studies and 3 case-control studies were selected for the analyses but one cohort study was excluded because the cancer outcomes were not categorized by type of gastrointestinal cancer.More than 124 686 subjects participated in the 3 cohort studies.The mean follow-up time in all of the cohort studies combined was approximately 3.88 years.The 3 casecontrol studies reported 3070 esophageal cancer cases and 15 417 controls,2018 gastric cancer cases and 10 007 controls,and 11 574 colorectal cancer cases and 53 955 controls.The percentage of study participants who used bisphosphonate was 2.8% among the cases and 2.9% among the controls.The meta-analysis of all the studies found no significant association between bisphosphonate use and gastrointestinal cancer.Also no statistically significant association was found in a meta-analysis of long-term follow-up studies.There was no negative association between bisphosphonate use and the incidence of esophageal cancer in the overall analysis(RR 0.96,95% CI:0.65-1.42,I 2 = 52.8%,P = 0.076) and no statistically significant association with long-term follow-up(RR 1.74,95% CI:0.97-3.10,I 2 = 58.8%,P = 0.119).No negative association was found in the studies reporting the risk of gastric cancer(RR 0.89,95% CI:0.71-1.13,I 2 = 0.0%,P = 0.472).In case of colorectal cancer,there was no association between colorectal cancer and bisphosphonate use(RR 0.62,95% CI:0.30-1.29,I 2 = 88.0%,P = 0.004) and also in the analysis with long-term follow-up(RR 0.61,95% CI:0.28-1.35,I 2 = 84.6%,P = 0.011).CONCLUSION:Oral bisphosphonate use had no significant effect on gastrointestinal cancer risk.However,this finding should be validated in randomized controlled trials with long-term follow-up.Yun Hwan Oh Chan Yoon Sang Min Park 2012World Journal of Gastroenterology2012,18,40:2
17Comparative effectiveness of adipose-derived mesenchymal stromal cells in the management of knee osteoarthritis:A meta-analysis显示文摘BACKGROUND Osteoarthritis(OA)is the most common joint disorder,is associated with an increasing socioeconomic impact owing to the ageing population.AIM To analyze and compare the efficacy and safety of bone-marrow-derived mesenchymal stromal cells(BM-MSCs)and adipose tissue-derived MSCs(AD-MSCs)in knee OA management from published randomized controlled trials(RCTs).METHODS Independent and duplicate electronic database searches were performed,including PubMed,EMBASE,Web of Science,and Cochrane Library,until August 2021 for RCTs that analyzed the efficacy and safety of AD-MSCs and BM-MSCs in the management of knee OA.The visual analog scale(VAS)score for pain,Western Ontario McMaster Universities Osteoarthritis Index(WOMAC),Lysholm score,Tegner score,magnetic resonance observation of cartilage repair tissue score,knee osteoarthritis outcome score(KOOS),and adverse events were analyzed.Analysis was performed on the R-platform using OpenMeta(Analyst)software.Twenty-one studies,involving 936 patients,were included.Only one study compared the two MSC sources without patient randomization;hence,the results of all included studies from both sources were pooled,and a comparative critical analysis was performed.RESULTS At six months,both AD-MSCs and BM-MSCs showed significant VAS improvement(P=0.015,P=0.012);this was inconsistent at 1 year for BM-MSCs(P<0.001,P=0.539),and AD-MSCs outperformed BM-MSCs compared to controls in measures such as WOMAC(P<0.001,P=0.541),Lysholm scores(P=0.006;P=0.933),and KOOS(P=0.002;P=0.012).BM-MSC-related procedures caused significant adverse events(P=0.003)compared to AD-MSCs(P=0.673).CONCLUSION Adipose tissue is superior to bone marrow because of its safety and consistent efficacy in improving pain and functional outcomes.Future trials are urgently warranted to validate our findings and reach a consensus on the ideal source of MSCs for managing knee OA.Sathish Muthu Sandesh C Patil Naveen Jeyaraman Madhan Jeyaraman Prakash Gangadaran Ramya Lakshmi Rajendran Eun Jung Oh Manish Khanna Ho Yun Chung Byeong-Cheol Ahn 2023World Journal of Orthopedics2023,14,1:2
