1A de novo Genome of a Chinese Radish Cultivar显示文摘Here, we report a high-quality draft genome of a Chinese radish(Raphanus sativus) cultivar. This draft contains 387.73 Mb of assembled scaffolds, 83.93% of the scaffolds were anchored onto nine pseudochromosomes and 95.09% of 43 240 protein-coding genes were functionally annotated. 184.75 Mb(47.65%) of repeat sequences was identified in the assembled genome. By comparative analyses of the radish genome against 10 other plant genomes, 2 275 genes in 780 gene families were found unique to R. sativus. 2Magnetic performance and corrosion resistance of electroless plating CoWP film显示文摘The CoWP film with good magnetic performance and corrosion resistance was electrolessly plated from alkaline solution. The technical parameters of the electroless plating system were optimized. When the pH value of electroless plating solution was 11.0 and the reducing agent (NaH2PO2) content was 0.4 mol L 1, the target chemical reactions proceeded in the electroless plating solution smoothly with negligible interference and side effects. CoWP film prepared under optimal deposition condition contained more hexagonal close-packed (hcp) cobalt (ε-Co) of [110] and crystallographic orientation of Co [002]. VSM analysis reveals that the saturation magnetization of the CoWP film is 100~220 Am2 kg 1 and coercivity is (2.87~4.38)×104 A·cm 1. The corrosion behavior of CoWP film in 3.5% NaCl aqueous solution was studied by electrochemical experiments. The results prove that the corrosion resistance of the CoWP film deposited under the optimal depo-sition condition exhibites relatively lower corrosion current of (3.054~3.162)×10 6 A·cm 2. 3Hydrothermal exfoliated molybdenum disulfide nanosheets as anode material for lithium ion batteries显示文摘Ultrathin MoS2nanosheets were prepared in high yield using a facile and effective hydrothermal intercalation and exfoliation route. The products were characterized in detail using X-ray diffraction, scanning electron microscopy, transmission electron microscopy and Raman spectroscopy. The results show that the high yield of MoS2nanosheets with good quality was successfully achieved and the dimensions of the immense nanosheets reached 1 μm–2 μm. As anode material for Li-ion batteries, the as-prepared MoS2nanosheets electrodes exhibited a good initial capacity of 1190 mAh g-1and excellent cyclic stability at constant current density of 50 mA g-1. 5Derivation of androgenetic embryonic stem cells from m-carboxycinnamic acid bishydroxamide (CBHA) treated androgenetic embryos显示文摘The androgenetic embyronic stem (aES) cells are useful models in studying the effects of imprinted genes on pluripotency maintaining and embryo development. The expression patterns of imprinted genes are significantly different between uniparental derived aES cells and zygote-derived embryonic stem (ES) cells, therefore, the imprinting related cell pluripotency needs further exploitation. Several approaches have been applied in generation of androgenetic embryos and derivation of aES cell lines. Here, we describe a method to generate androgenetic embryos by injecting two mature sperms into one enucleated oocyte. Then these androgenetic embryos were treated with a histone deacetylase inhibitor: m-carboxycinnamic acid bishydroxamide (CBHA). Further, aES cell lines were successfully derived from these treated androgenetic embryos at blastocyst stage. The CBHA could improve not only the quality of androgenetic embryos, but also the efficiencies of aES (CaES) cells derivation and chimeric mice generation. 6Preparation of CoP films by ultrasonic electroless deposition at low initial temperature显示文摘Electroless deposition technology has been considered as a kind of common ways to obtain cobalt alloy films. However, in order to get cobalt alloy films, high temperature(353 K) is necessary during the electroless deposition process which will increase costs and energy consumption. Ultrasonic was introduced during electroless plating process to obtain cobalt alloy films at lower initial temperature. It was found that the cobalt thin films could be prepared at lower initial temperature(323 K) with the introduction of ultrasonic. Therefore, different powers of ultrasonic were applied during the electroless deposition process to prepare CoP thin films on copper substrates from an alkaline bath in this investigation. The effects of different powers of ultrasonic on deposition rate, surface morphology, anticorrosion performance and magnetic property of films were studied. It was found that the deposition rate increased gradually with the rise in ultrasonic powers due to cavitation phenomenon. All the CoP films presented the typical spherical nodular structures with the impact of ultrasonic. Smaller and regular shaped structures could be observed when the films were deposited with higher power of ultrasonic which contributed directly to enhancement of anticorrosion performance. Saturation magnetization and coercivity of thin films increased gradually with the rise in ultrasonic powers during the electroless deposition process due to the higher amounts of cobalt.& 2014 Chinese Materials Research Society. 8Downstream signaling of reactive oxygen species,protein kinase C epsilon translocation and delayed neuroprotection in sevoflurane preconditioned rats following cerebral ischemia/reperfusion显示文摘BACKGROUND:Brief exposure to the anesthetic sevoflurane results in delayed neuroprotection. However,few studies have addressed delayed neuroprotection after preconditioning with a single administration of sevoflurane. OBJECTIVE:To explore the relationship between a single preconditioning administration of sevoflurane and reactive oxygen species production and protein kinase C-epsilon(PKC-ε) translocation. DESIGN,TIME,AND SETTING:The randomized,controlled,animal experiment was conducted at the Central Laboratory,Xiangya Hospital,Central South University,China from November 2007 to April 2008. MATERIALS:A total of 120 healthy,male,Sprague Dawley rats were equally and randomly assigned into five groups:sham operation,ischemia/reperfusion,sevoflurane,2-mercaptopropionylglycine (2-MPG,a selective reactive oxygen species scavenger) + sevoflurane(MPG + sevoflurane),and MPG.Sevoflurane(Baxter,USA) and MPG(Sigma,USA) were used in this study. METHODS:Intervention consisted of three procedures.(1) MPG injection:a selective reactive oxygen species scavenger,MPG(20 mg/kg),was infused into the rat caudal vein in the MPG and MPG + sevoflurane groups.(2) Sevoflurane preconditioning:30 minutes following MPG injection, rats in the sevoflurane and MPG + sevoflurane groups breathed a mixed gas of 2.4%sevoflurane and 97.6%oxygen for 60 minutes.Rats in the sham operation,ischemia/reperfusion,and MPG groups breathed 100%pure oxygen for 60 minutes.(3) Ischemia/reperfusion:24 hours after sevoflurane or pure oxygen preconditioning,middle cerebral artery occlusion models were established in the ischemia/reperfusion,sevoflurane,MPG + sevoflurane,and MPG groups. Following 2 hours ischemia/6 hours and 24 hours reperfusion,the carotid artery was separated,but the middle cerebral artery was not occluded,in the sham operation group. MAIN OUTCOME MEASURES:In the ischemic hemisphere,PKC-εtranslocation in the rat parietal cortex was measured by Western blot analysis.Infarct volume was calculated using the TTC assay.Neurological deficits were evaluated in rats using a scoring system of 8 points. RESULTS:After 6 hours reperfusion,the ratio of PKC-εin membrane/(cytosol + membrane) was significantly less in the sham operation group than in the ischemia/reperfusion,sevoflurane,MPG + sevoflurane),and MPG groups(P<0.05).The ratio of PKC-εin membrane/(cytosol + membrane) was significantly greater in the sevoflurane group than in the sham operation,ischemia/reperfusion, MPG + sevoflurane,and MPG groups(P<0.05).No significant differences were observed in the ischemia/reperfusion,MPG + sevoflurane,and MPG groups(P>0.05).Following 24 hours reperfusion, the ratio of PKC-εin membrane/(cytosol + membrane) was significantly less in the sham operation group than in the ischemia/reperfusion,sevoflurane,MPG + sevoflurane,and MPG groups(P<0.05). No significant differences were detected in the ischemia/reperfusion,sevoflurane,MPG + sevoflurane, and MPG groups(P>0.05).Compared with the ischemia/reperfusion,MPG + sevoflurane,and MPG groups,infarct volume was significantly smaller,and neurological deficits were significantly improved, in the sevoflurane group(P<0.05).No significant differences in infarct volume and neurological deficits were observed among the ischemia/reperfusion,MPG + sevoflurane,and MPG groups(P>0.05).Infarcts or neurological deficits were not detected in the sham operation group. 10Characteristics of urban heat island effect in Lhasa City显示文摘This paper analyzes the Urban Heat Island(UHI) effect in Lhasa City of Tibet using meteorological observations,the Normalized Difference Vegetation Index(NDVI),Land Surface Temperature(LST) data obtained from Earth Observing System/Moderate resolution Imaging Spectroradiometer(EOS/MODIS) information,and correlation and composite analyses.The results show:(1) Areas with high temperature are primarily located in the center of the city or nearby counties,while low temperature areas are in the suburbs of counties.The area with high temperature has expanded in recent years and some high-temperature centers have even migrated to certain other regions.(2) The UHI intensity tends to be stronger both in annual and seasonal variations,especially in winter.Also,LST is somewhat positively related to mean air temperature.(3) A negative correlation exists between the changes of LST and NDVI with the increase of vegetation from urban to rural areas in different seasons.(4) The UHI intensity is nega-tively correlated with precipitation while positively correlated with wind speed,and the relation between the UHI intensity and evaporation varies with the seasons,namely,the intensity is positively correlated with summer evaporation but negatively corre-lated with winter evaporation.(5) UHI intensity might be enhanced by intensified urbanization,wherein built-up areas expand,there is increased heat from human activity,and there is more artificial heat input to the atmosphere.Zhuo Ga YunDan NiMa Jian Jun PuBu CiRen 2011Research in Cold and Arid Regions2011,3,1:1

14High throughput monoclonal antibody generation by immunizing multiple antigens显示文摘Recognizing proteins via the production of highly specific monoclonal antibodies(mAbs)is crucial to identifying proteins for proteomic research.However,traditional mAb generation is time-consuming with low efficiency.In this study,we assessed the high throughput method of producing mAbs by immunizing mice with multiple antigens in order to obtain hybridomas against these multiple antigens in one cell fusion.We selected eight proteins that play important roles in human physiological or pathological processes.These proteins were mixed and simultaneously administered to one mouse.We observed the immunizing period for 10 d,and determined the effect of liquid medium and semi-solid medium in hybridoma generation.As a result,all eight immunogens induced antibodies in the immunized mouse in one cell fusion,we obtained hybridomas specific to all eight proteins by enzyme-linked immuno sorbent assay(ELISA)screening,hybridomas against five out of eight showed specific positive in Western-blotting assays.This indicates that we generated mAbs specific to eight proteins in one cell fusion,greatly increasing the efficiency of mAb generation.Furthermore,we observed that hybridomas selected from the liquid medium and semi-solid medium showed different reactivity to antigens.Our study established high-throughput and time-saving methods for production of mAbs.These results provide alternative approaches for increasing the efficacy of mAb generation.LIU Ying WANG YunDan LIU Jing ZUO Wei HAO Lu ZHANG LiLi ZHEN Bei 2014Science China(Life Sciences)2014,57,7:0

17Effect of magnetic fields on pulse plating of cobalt films显示文摘Magnetic fields parallel to the electrodes were introduced during a pulse plating process to obtain cobalt thin films from alkaline baths. Ef-fects of different magnetic intensities on the composition, microstructure, and magnetic properties of cobalt thin films were investigated. It was found that the deposition speed increased gradually with the increase of magnetic intensity. Almost all of the deposited films were crys-talline and showed Co(002), Co(100) peaks. With the rise on the magnetic intensity, the intensity of Co (002) peak raised gradually. Mag-netic fields would induce cobalt growing along (002) orientation. The films were densely covered with typical nodular structure. Films of smaller grain size and smooth surface could be formed under high magnetic intensity (1 T) as a result of magnetic force and MHD effects. Moreover, higher magnetic intensity induced larger saturation magnetization and lower coercivity. 18Carbon matrix/SiNWs heterogeneous block as improved reversible anodes material for lithium ion batteries显示文摘A novel carbon matrix/silicon nanowires(SiNWs) heterogeneous block was successfully produced by dispersing SiNWs into templated carbon matrix via a modified evaporation induced self-assembly method. The heterogeneous block was determined by X-ray diffraction, Raman spectra and scanning electron microscopy. As an anode material for lithium batteries, the block was investigated by cyclic voltammograms(CV), charge/discharge tests, galvanostatic cycling performance and A. C. impedance spectroscopy. We show that the SiNWs disperse into the framework, and are nicely wrapped by the carbon matrix. The heterogeneous block exhibits superior electrochemical reversibility with a high specific capacity of 529.3 mAh/g in comparison with bare SiNWs anode with merely about 52.6 mAh/g capacity retention. The block presents excellent cycle stability and capacity retention which can be attributed to the improvement of conductivity by the existence of carbon matrix and the enhancement of ability to relieve the large volume expansion of SiNWs during the lithium insertion/extraction cycle. 19Anshen Babu Ointment Improves Sleep Function of Insomnia Mice and Its Mechanism显示文摘[Objectives]This study aimed to study the sleep-improving effect of Anshen Babu ointment on insomnia mice and its mechanism of action.[Methods]Mouse models of insomnia were established by p-chlorophenylalanine(PCPA)injection.They were given high,middle and low doses of Anshen Babu ointment,for 7 consecutive days,and then,the spontaneous activity of insomnia mice and the effect on hypnotic time and subthreshold hypnotic dose of pentobarbital sodium were observed,and the levels of 5-HT,IL-6 and TNF-α in the hippocampus of mice were determined.[Results]High and middle-dose Anshen Babu ointment significantly reduced the spontaneous activity of insomnia mice and shorten the hypnotic time of pentobarbital sodium;high and middle-dose Anshen Babu ointment increased the 5-HT content and inhibited the IL-6 and TNF-α expression in the hippocampus of mice.[Conclusions]Anshen Babu ointment has hypnotic and sedative effect on insomnia mice.The mechanism may be achieved by increasing 5-HT and reducing IL-6 and TNF-α levels.Qin GONG Xiaoxue GONG Sen LIU Yundan LI Xi WANG 2020Medicinal Plant2020,11,6:0
20Localization method of subsynchronous oscillation source based on high-resolution time-frequency distribution image and CNN显示文摘The penetration of new energy sources such as wind power is increasing,which consequently increases the occurrence rate of subsynchronous oscillation events.However,existing subsynchronous oscillation source-identification methods primarily analyze fixed-mode oscillations and rarely consider time-varying features,such as frequency drift,caused by the random volatility of wind farms when oscillations occur.This paper proposes a subsynchronous oscillation sourcelocalization method that involves an enhanced short-time Fourier transform and a convolutional neural network(CNN).First,an enhanced STFT is performed to secure high-resolution time-frequency distribution(TFD)images from the measured data of the generation unit ports.Next,these TFD images are amalgamated to form a subsynchronous oscillation feature map that serves as input to the CNN to train the localization model.Ultimately,the trained CNN model realizes the online localization of subsynchronous oscillation sources.The effectiveness and accuracy of the proposed method are validated via multimachine system models simulating forced and natural oscillation events using the Power Systems Computer Aided Design platform.Test results show that the proposed method can localize subsynchronous oscillation sources online while considering unpredictable fluctuations in wind farms,thus providing a foundation for oscillation suppression in practical engineering scenarios.Hui Liu Yundan Cheng Yanhui Xu Guanqun Sun Rusi Chen Xiaodong Yu 2024Global Energy Interconnection2024,7,1:0
