1A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 2Surface wear evolution of traction motor bearings in vibration environment of a locomotive during operation显示文摘Wear is a major cause of bearing failure,however,compared with other impact failures,smooth wear is often neglected because of its slow gradualness.In a locomotive,under the effect of the intensified wheel-rail interaction and gear mesh,the surface wear can increase the radial clearances of motor bearings,trigger abnormal vibration and noise,and adversely affect the efficiency and safety of the traction motor.In this study,considering the mechanical structure of the traction motor,a locomotive-track spatially coupled dynamics model with traction power transmissions(LTMM)is adopted to provide accurate dynamic loads and obtain the dynamic responses of the system.From the perspective of contact stress and relative skidding at the roller-race interface,the causes and evolution of motor bearings surface wear are analyzed.The results indicate that the wear of inner races of motor bearings is uniform.The outer races of the driving/non-driving end bearings(DB or NDB)are worn in the loaded/main loadbearing regions.The increasing surface wear can increase the internal dynamic forces(e.g.,the centrifugal force of rotor and unbalanced magnetic pull)induced by the instantaneous eccentricity of the rotor,intensify the interactions between the roller and races,and deteriorate the vibrations of the traction motor and its neighboring components in the locomotive.In addition,the effects of the friction coefficient at the roller-race interface and track random irregularity are analyzed.This study offers a theoretical basis in service life prediction and design of the motor bearings.LIU YuQing CHEN ZaiGang WANG KaiYun ZHAI WanMing 2022Science China(Technological Sciences)2022,65,4:2

5Target Extension-Activated DNA Walker on Nanoparticles for Digital Counting-Based Analysis of MicroRNA显示文摘MicroRNAs(miRNAs),especially exosomal miRNAs,are promising noninvasive biomarkers in early-stage cancer diagnosis and disease treatment monitoring.However,their precise and sensitive quantification remains challenging due to their small size and low abundance.Herein,we have developed a nanoparticle-confined DNA walker strategy for the specific detection of miRNA.In the existence of the target miRNA;the on-particle DNA walking reaction will be initiated,providing a fluorescence-positive nanoparticle.Otherwise,the nanoparticle would be fluorescence-negative.Utilizing the total internal reflection fluorescent microscope(TIRFM)to digitally count the fluorescence-positive nanoparticles,the proposed method possesses a detection limit of 0.2 pmol/L miRNA and can accurately distinguish the single-base mismatched target.This design combines the merits of the DNA walker for signal amplification and the TIRFM for highly sensitive detection,paving a new way for the digital counting-based analysis of exosomal miRNAs.Qinya Feng Yuqing Zhai Wei Ren Chenghui Liu 2021Chinese Journal of Chemistry2021,39,6:1

7Improved Dynamics Model of Locomotive Traction Motor with Elasticity of Rotor Shaft and Supporting Bearings显示文摘The locomotive traction motor is described as a rotor-bearing system coupling the kinetic equations of the traction shaft and its support bearings with the determination of their elastic deformations in this study.Under the effect of excitations induced by the dynamic rotor eccentric distance and time-varying mesh stiffness,the elastic structure deformations of the shaft and support bearings are formulated in the vibration environment of the locomotive.In addition,the nonlinear contact forces between the components of the rolling bearing,the lubricating oil film,and radial clearance are comprehensively considered in this study.The results indicate that the elastic deformations of the shaft and bearings can change the dynamic responses of the traction motor and its support bearings.There are large differences between the ranges of the rotor motion calculated by the rigid and the flexible traction motor models when the intensified wheel-rail interaction is considered.With the increase of the rotor eccentricity,the results underscore the role of the elasticity of traction shaft and support bearings in dynamic researches of the traction motor.The critical value of the initial eccentric distance for the rub-impact phenomenon decreases from 1.23 mm to 1.15 mm considering the flexible effect of the shaft and bearings.This dynamics model of the traction motor can provide more accurate and reasonable simulation results for correlational dynamic researches.Yuqing Liu Zaigang Chen Jieyu Ning Kaiyun Wang Wanming Zhai 2022Chinese Journal of Mechanical Engineering2022,35,5:0
8Changes in intracellular and extracellular proteins after ERGIC3 knockdown in lung cancer:Proteins interacting with ERGIC3,HORN,and FLNA显示文摘Objective:Endoplasmic reticulum-Golgi intermediate compartment 3(ERGIC3)promotes cell proliferation and metastasis in lung cancer,but its molecular mechanism is unclear.Methods:The GLC-82 cells were randomly divided into the ERGIC3i group and the negative control group.The cells were transfected with ERGIC3 siRNA or control siRNA in the groups,respectively.The ERGIC3-interacting proteins expressed in cells or extracellularly were isolated by the immunoprecipitation method and detected by isobaric tags for relative and absolute quantitation and liquid chromatography-tandem mass spectrometry.The differentially expressed proteins were determined by bioinformatic methods.Results:After ERGIC3 knockdown,88 extracellular differentially expressed proteins,41 upregulated and 47 down-regulated,were detected in the supernatant of cultured cells.Among 52 intracellular differentially expressed proteins,33 were up-regulated and 19 down-regulated.Cluster analysis showed that the extracellular differential proteins are mainly involved in Ca2+binding and transport and I-kappa B kinase/NF-kappa B signal transduction.The upregulated proteins are mainly involved in the biological process of H3-K27 and H3-K4 methylation in cells.Co-immunoprecipitation assay showed that proteins interacting with ERGIC3 were rich in cytoskeleton construction and RHO GTPases activated p21-activated kinases.The intersection of these two research methods shows that ERGIC3 interacts with HORN and filamin A(FLNA).Conclusion:Proteomic analyses reveal that ERGIC3 acts as a vesicle transmembrane protein on the distribution of various extracellular and intracellular proteins and regulates the extracellular and intracellular biological processes by specifically binding hornin(HORN)and FLNA proteins.These findings maybe provide new methods and ideas for ERGIC3 as a therapeutic target for lung cancer.WEI ZHAI XIANG ZHENG MENGYUAN LIU QIURONG ZHAO YUSHU ZHANG YUQING LUO SHANSHAN FENG JINJING WANG XUEYING LI MINGSONG WU 2023BIOCELL2023,47,8:0

10Performance of novel overhead crane fume-collecting hood for pollutant removal显示文摘In industrial buildings,the presence of overhead cranes severely affects roof exhaust ventilation systems when capturing and discharging fumes,resulting in severe deterioration of the indoor plant environment.In this study,an overhead crane-based ventilation auxiliary device,called overhead crane fume-collecting hood(CFCH),is proposed to guide pollutants blocked by the overhead crane back to the roof exhaust hood.The airflow characteristics and pollutant distribution under the three modes of no overhead crane,overhead crane,and overhead crane+CFCH were compared using numerical simulations.Subsequently,the effects of the CFCH length(a),width(b),and height(h)on the pollutant capture performance were determined through orthogonal experiments and computational fluid dynamics.Finally,the pollutant capture efficiency(PCE)of the optimal CFCH was investigated considering different exhaust airflow rates.The results showed that the pollutants captured by the CFCH can be classified into directly and secondary captured pollutants,with the directly captured pollutants dominating.In addition,with the introduction of different sizes of CFCH around the overhead crane girders,the PCE significantly improved by 49.9%–74.6%.The length,width,and height of the CFCH on the PCE were statistically significant,and the priority of the three factors was as follows:h>b>a.The PCE decreased with increasing a,initially increased and then decreased with increasing b,and increased with h.Subsequently,when the optimal CFCH was used,the excessive exhaust air rate had no evident PCE improvement.This provides a new concept for the control of pollutants in industrial buildings and provides a theoretical basis for the design of CFCHs.Zhixiang Cao Pan Xiao Yi Wang Yuqing Bai Chen Zhang Tongtong Zhao Chao Zhai Minghao Zhang Songheng Wu 2023Building Simulation2023,16,7:0
11Antioxidant and anti-HepG2 cell activities of a novel bioactive peptide from cowhide collagen in vitro显示文摘In the present study,the antioxidant and anti-human liver cancer(HepG2)cells effects of bioactive peptides from cowhide collagen(BPCC)were evaluated.BPCC exhibited significant scavenging effect on l,1-diphenyl-2-picrylhydrazyl(DPPH)radicals((60.09±3.51)%),2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)radicals((77.40±3.10)%)and hydroxyl radicals((56.00±2.30)%)as well as strong reducing power(0.320士0.025).Meanwhile,BPCC effectively protected biomacromolecules including proteins,lipids and DNA from oxidative damage induced by Cu^(2+)H_(2)O_(2)and 2,2'-azobis(2-methylpropionamidine)dihydrochloride(AAPH).Moreover,BPCC significantly inhibited cell viability of HepG2 cells in a dose-dependent manner with an estimated IC_(50)of 7.61 mg/mL.The results of 4',6-diamidino-2-phenylindole(DAPI)and acridine:orange/ethidium bromide(AO/EB)staining demonstrated the apoptotic morphological changes and cell mediated death in BPCC treated HepG2 cells.In addition,BPCC induced decrease of mitochondrial membrane potential(MMP)in HepG2 cells.Therefore,the present finding proved that BPCC encompasses significant antioxidant activity and anticancer property on HepG2 cells and can be used as alternative food antioxidants for cancer prevention benefits.Zhike Xie Yuhan Zhai Yuqing Zhang Ming He Xuguang Wang Shaoxuan Yu Haifang Xiao Yuanda Song 2024Journal of Future Foods2024,4,3:0
