1Development of a Haploid-Inducer Mediated Genome Editing System for Accelerating Maize Breeding显示文摘Crop breeding aims to generate pure in bred lines with multiple desired traits. Doubled haploid (DH) and genome editing using CRISPR/Cas9 are two powerful game-changing technologies in crop breeding. However, both of them still fall short for rapid generation of pure elite lines with integrated favorable traits. Here, we report the development of a Haploid-Inducer Mediated Genome Editing (IMGE) approach, which utilizes a maize haploid inducer line carrying a CRISPR/Cas9 cassette targeting for a desired agronomic trait to pollinate an elite maize in bred line and to generate genome-edited haploids in the elite maize background. 2FHY3 and FAR1 Integrate Light Signals with the miR156-SPL Module-Mediated Aging Pathway to Regulate Arabidopsis Flowering显示文摘In response to competition for light from their neighbors,shade-intolerant plants flower precociously to ensure reproductive success and survival.However,the molecular mechanisms underlying this key developmental switch are not well understood.Here,we show that a pair of Arabidopsis transcription factors essential for phytochrome A signaling,FAR-RED ELONGATED HYPOCOTYL3(FHY3)and FAR-RED IMPAIRED RESPONSE1(FAR1),regulate flowering time by integrating environmental light signals with the miR156-SPL module-mediated aging pathway.We found that FHY3 and FAR1 directly interact with three flowering-promoting SQUAMOSA-PROMOTER BINDING PROTEIN-LIKE(SPL)transcription factors,SPL3,SPL4,and SPL5,and inhibit their binding to the promoters of several key flowering regulatory genes,including FRUITFUL(FUL),LEAFY(LFY),APETALA1(AP1),and MIR172C,thus downregulating their transcript levels and delaying flowering.Under simulated shade conditions,levels of SPL3/4/5 proteins increase,whereas levels of FHY3 and FAR1 proteins decline,thus releasing SPL3/4/5 from FHY3/FAR1 inhibition to allow activation of FUL,LFY,AP1,and MIR172C and,consequently,early flowering.Taken together,these results unravel a novel mechanism whereby plants regulate flowering time by integrating environmental cues(such as light conditions)and an internal developmental program(the miR156-SPL module-mediated aging pathway).Yurong Xie Qin Zhou Yongping Zhao Quanquan Li Yang Liu Mengdi Ma Baobao Wang Rongxin Shen Zhigang Zheng Haiyang Wang 2020Molecular Plant2020,13,3:8

8Research on the Longitudinal Transmission Mechanism of Prices for Urban Vegetable Supply Chain-Based on the Empirical Research in Chongqing显示文摘This paper mainly introduces the longitudinal transmission mechanism of urban vegetable prices in Chongqing area. On the basis of the supply chain theory,the ' farm price- wholesale price- retail price' of vegetables in Chongqing has been identified as the research object. Through statistical analysis,it is found that the above three types of price show a modest annual fluctuation of 7- 12% from 2008 to2013. Later,the paper focuses on the empirical research on the weekly price data in 2013 applying the co-integration theory,which reveals that the farm price of vegetables is largely influenced by themselves; the wholesale price of vegetables is mainly affected by their own price,the production price and the retail price of the earlier stage without the current influence of the production price; and the retail price of vegetables is also affected by their own price,but is not affected by the their current local price and the wholesale price. 9Cathepsin B S-nitrosylation promotes ADAR1-mediated editing of its own mRNA transcript via an ADD1/MATR3 regulatory axis显示文摘Genetic information is generally transferred from RNA to protein according to the classic“Central Dogma”.Here,we made a striking discovery that post-translational modification of a protein specifically regulates the editing of its own mRNA.We show that S-nitrosylation of cathepsin B(CTSB)exclusively alters the adenosine-to-inosine(A-to-I)editing of its own mRNA.Mechanistically,CTSB S-nitrosylation promotes the dephosphorylation and nuclear translocation of ADD1,leading to the recruitment of MATR3 and ADAR1 to CTSB mRNA.ADAR1-mediated A-to-I RNA editing enables the binding of HuR to CTSB mRNA,resulting in increased CTSB mRNA stability and subsequently higher steady-state levels of CTSB protein.Together,we uncovered a unique feedforward mechanism of protein expression regulation mediated by the ADD1/MATR3/ADAR1 regulatory axis.Our study demonstrates a novel reverse flow of information from the post-translational modification of a protein back to the post-transcriptional regulation of its own mRNA precursor.We coined this process as“Protein-directed EDiting of its Own mRNA by ADAR1(PEDORA)”and suggest that this constitutes an additional layer of protein expression control.“PEDORA”could represent a currently hidden mechanism in eukaryotic gene expression regulation.Zhe Lin Shuang Zhao Xuesong Li Zian Miao Jiawei Cao Yurong Chen Zhiguang Shi Jia Zhang Dongjin Wang Shaoliang Chen Liansheng Wang Aihua Gu Feng Chen Tao Yang Kangyun Sun Yi Han Liping Xie Hongshan Chen Yong Ji 2023Cell Research2023,33,7:0
