| 2 | Expansion and differentiation of human hepatocyte-derived liver progenitor-like cells and their use for the study of hepatotropic pathogens显示文摘The study of pathophysiological mecha nisms in huma n liver disease has bee n constrained by the in ability to expa nd primary hepatocytes in vitro while maintaining proliferative capacity and metabolic function. We and others have previously shown that mouse mature hepatocytes can be con verted to liver progenitor-like cells in vitro with defined chemical factors. Here we describe a protocol achieving efficient conversion of human primary hepatocytes into liver progenitor-like cells (HepLPCs) through delivery of developmentally relevant cues, including NAD +-dependent deacetylase SIRT1 signaling. These HepLPCs could be expanded sign ifica ntly duri ng in vitro passage. The expa nded cells can readily be con verted back into metabolically functional hepatocytes in vitro and upon tran splantati on in vivo. Under three-dime nsional culture conditions, differentiated cells gen erated from HepLPCs regai ned the ability to support infect io n or reactivatio n of hepatitis B virus (HBV). Our work dem on strates the utility of the con versi on betwee n hepatocyte and liver proge nitor-like cells for studying HBV biology and an tiviral therapies. | 3 | Guiding-controlling technology of coal seam hydraulic fracturing fractures extension显示文摘Aiming at the uncontrollable problem of extension direction of coal seam hydraulic fracturing,this study analyzed the course of fractures variation around the boreholes in process of hydraulic fracturing,and carried out the numerical simulations to investigate the effect of artificial predetermined fractures on stress distribution around fractured holes.The simulation results show that partial coal mass occurs relatively strong shear failure and forms weak surfaces,and then fractures extended along the desired direction while predetermined fractures changed stress distribution.Directional fracturing makes the fractures link up and the pressure on coal mass is relieved within fractured regions.Combining deep hole controlling blasting with hydraulic fracturing was proposed to realize the extension guiding-controlling technology of coal seam fractures.Industrial experiments prove that this technology can avoid local stress concentration and dramatically widen the pressure relief scope of deep hole controlling blasting.The permeability of fractured coal seam increased significantly,and gas extraction was greatly improved.Besides,regional pressure relief and permeability increase was achieved in this study. | Zhai Cheng Li Min Sun Chen Zhang Jianguo Yang Wei Li Quangui | 2012 | International Journal of Mining Science and Technology2012,22,6: | 12 |
| 4 | Warming effects on plant biomass allocation and correlations with the soil environment in an alpine meadow,China显示文摘Alpine meadow ecosystem is fragile and highly sensitive to climate change.An understanding of the allocation of above-and below-ground plant biomass and correlations with environmental factors in alpine meadow ecosystem can result in better protection and effective utilization of alpine meadow vegetation.We chose an alpine meadow in the Qinghai-Tibetan Plateau of China as the study area and designed experimental warming plots using a randomized block experimental design.We used single-tube infrared radiators as warming devices,established the warming treatments,and measured plant above- (AGB) and below-ground biomass (BGB) during the growing seasons (May to September) in 2012 and 2013.We determined the allocation of biomass and the relationship between biomass and soil environment under the warming treatment.Biomass indices including above-ground biomass,below-ground biomass and the ratio of root to shoot (R/S) ,and soil factors including soil moisture and soil temperature at different depths were measured.The results showed that (1) BGB of the alpine meadow had the most significant allometric correlation with its AGB (y=298.7x~ (0.44) ,P<0.001) ,but the relationship decreased under warming treatment and the determination coefficient of the functional equation was 0.102 which was less than that of 0.188 of the unwarming treatment (control) ; (2) BGB increased,especially in the deeper soil layers under warming treatment (P>0.05) .At 0–10 cm soil depth,the percentages of BGB under warming treatment were smaller than those of the control treatment with the decreases being 8.52% and 8.23% in 2012 and 2013,respectively.However,the BGB increased 2.13% and 2.06% in 2012 and 2013,respectively,at 10–50 cm soil depths; (3) BGB had significant positive correlations with soil moisture at 100 cm depth and with soil temperature at 20–100 cm depths (P<0.05) ,but the mean correlation coefficient of soil temperature was 0.354,greater than the 0.245 of soil moisture.R/S ratio had a significant negative correlation with soil temperature at 20 cm depth (P<0.05) .The warmer soil temperatures in shallow layers increased the biomass allocation to above-ground plant parts,which leading to the increase in AGB;whereas the enhanced thawing of frozen soil in deep layers causing by warming treatment produced more moisture that affected plant biomass allocation. | XU Manhou LIU Min XUE Xian ZHAI Datong | 2016 | Journal of Arid Land2016,8,5: | 12 |

| 6 | Effects of electroacupuncture at Guanyuan(CV 4)or Sanyinjiao(SP 6)on hypothalamus-pituitary-ovary axis and spatial learning and memory in female SAMP8 mice显示文摘OBJECTIVE: To investigate the effects of electroacupuncture(EA) at the Guanyuan(CV 4) or Sanyinjiao(SP 6) acupoints on the hypothalamus-pituitary-ovary(HPO) axis and spatial learning and memory in female mice.METHODS: Nine-month-old female mice with senescence-accelerated mouse prone 8(SAMP8)were divided into three groups: the disease model,EA-Guanyuan and EA-Sanyinjiao groups. Concurrently, 9-month old female mice with senescence-accelerated mouse resistance 1(SAMR1)were set as the control model group. The two treatment groups were given the same pattern of EA stimulation. Gonadotropin-releasing hormone, luteinizing hormone, follicle-stimulating hormone(FSH) and Serum estradiol levels in the Hypothalamus-pituitary-ovary axis were assessed by enzyme-linked immunosorbent assay to determinethe HPO axis function level. Spatial learning and memory were assessed by the Morris Water Maze(MWM) test.RESULTS:(a) HPO axis: compared with the control model group, the disease model group displayed a decrease in E2 levels(P < 0.01), and an increase in Gn RH, LH and FSH levels(P < 0.01). E2 levels were increased in EA treatment groups compared with the disease model group(P < 0.05). In contrast,Gn RH and LH and FSH levels were reduced(P <0.05). EA-Sanyinjiao group was superior than EA-Guanyuan group on increasing E2 and declining Gn RH levels(P < 0.01).(b) The MWM test demonstrated that the response latency in the EA-Sanyinjiao treatment group declined from day 2 to day5 compared with the disease model group(P <0.05), whereas the EA-Guanyuan treatment group showed no significant difference.CONCLUSION: EA can regulate hormone(E2, FSH,LH, Gn RH) levels in the HPO axis and the spatial learning and memory ability in female SAMP8 mice. Moreover, this effect may have been more pronounced in the EA-Sanyinjiao group than the EA-Guanyuan group. However, the existing technical guides in China are behind the application requirements. This study analyzed the most important indicators of air-sea gravimetry, including the density of survey lines, gravimetry accuracy and space resolution, stability and reliability of the air-sea gravimeter, and proposed a gravimetry accuracy assessment system consisting of gravity RMS of error, systematic error and mean error, and an assessment system for the gravimeter stability consisting of the relative accuracy of the scale value, monthly zero-drift, RMS of the monthly nonlinear zero-drift variation and the threshold of the monthly nonlinear zero-drift variation. The mathematic models for the measurement point determination in shipborne gravimetry, E tv s correction for airborne gravimetry, platform tilt correction and evaluation air-sea gravimetry were also analyzed and modified. | 11 | Serum Gamma-glutamyl Transferase Levels Predict Functional Outcomes after Aneurysmal Subarachnoid Hemorrhage显示文摘Objective We aim to explore the potential association between serum gamma-glutamyl transferase levels and functional outcome after aneurysmal subarachnoid hemorrhage in a Chinese population. Methods A total of 386 aneurysmal subarachnoid hemorrhage patients were included in the study from September 2007 to February 2015. Baseline serum gamma-glutamyl transferase levels and 6-month follow-up functional outcomes were determined. A poor outcome was defined as a modified ranking scale score of ≥ 3. The multivariable logistic model was used to analyze the relationship between serum gamma-glutamyl transferase and clinical outcomes after aneurysmal subarachnoid hemorrhage. Results The adjusted poor outcome rates of patients with gamma-glutamyl transferase levels of < 30 U/L, 30-50 U/L and ≥ 50 U/L were 16.7%, 19.6%, and 34.4%, respectively(P < 0.01). The age-sex and multivariable adjusted odds ratios(95% confidence intervals) of poor prognosis comparing the top group(≥ 50 U/L) with the lowest group(< 30 U/L) were 5.76(2.74-12.13), 6.64(2.05-21.52), and 6.36(1.92-21.02). A significant linear trend existed between gamma-glutamyl transferase level and aneurysmal subarachnoid hemorrhage prognosis. This association was also observed among nondrinkers. Conclusion Patients with higher gamma-glutamyl transferase levels were more likely to have a poor prognosis. | 13 | Microscopic properties and sealing performance of new gas drainage drilling sealing material显示文摘The geological condition of Chinese coal mines are complex and high gassy,which account for ffty percent to seventy percent.Because of the abundant pores and cracks around the drainage drilling hole,the gas concentration attenuates rapidly,and the effective gas drainage period is short.The traditional sealing materials of yellow mud and cement-sand grout will readily shrink after the drilling hole is sealed,the sealing length is short and the sealing quality is not satisfactory.Currently widely used polyurethane material will shrink when it comes into contact with water,and the price is also very high.In this study,taking cement as a base material,a novel composite sealing material mixed by expansion admixture,additive,and fbrin and coupling agent was developed and the sealing performance and expansion property of the material were also studied and analyzed.The FEI Quanta TM 250 environmental scanning electron microscope was used to investigate the microstructure of material.The results revealed that the new composite sealing material had a desirable expansion performance and a defnite fluidity convenient for grouting.The solidifed material,combining closely with the drilling wall,possessed an adequate strength and was not easy to shrink.Compared to the conventional polyurethane,the gas drainage concentration by drilling sealing exceeded 40 percent,and the sealing capacity improves5 times,the sealing effect increases signifcantly. | Zhai Cheng Yu Xu Ni Guanhua Li Min Hao Zhiyong | 2013 | International Journal of Mining Science and Technology2013,23,4: | 4 |

The cell constants, obtained by least-squares calculation from direct θ -value’s measurements on the diffractometer are: a =1.158 8(5), b =1.174 4(4), c =0.395 5(2) nm; orthorhombic; V =0.538 23 nm+3; Z =4. The space group is Pbnm. X-ray single crystal data, using Mo K α radiation, were measured on a RIGAKU RASA-5RP automated diffractometer and refined to a final R index of 0.048 1. Sb-2Se-3 is isostructural with Sb-2S-3 and Bi-2S-3. Each Sb(1) atom is six-coordinated by 3 Se(1), 1 Se(2) and 2 Se(3) atoms at distances 0.266 0-0.323 6 nm. Each Sb(2) atom is seven-coordinated by 2 Se(1), 2 Se(2) and 3 Se(3) atoms at distances of 0.258 1- 0.346 7 nm. The crystal structure consists of chains parallel to c or needle axis. The strongest bonds (shortest separations) are within the chains. | 19 | Qualitative Analysis and Quality Evaluation of Cnidium monnieri Using UHPLC-ESI-Q-TOF/MS显示文摘Objective To identify the coumarins constituents in Cnidium monnieri and classify ten samples into three groups and this helpful chemical information could be used for the further pharmacological and clinical study on C. monnieri. Methods Qualitative analysis of coumarins in C. monnieri was detected by UHPLC-ESI-Q-TOF/MS. Quadrupole TOF/MS in either full scan mode or extracted ion mode was used for the qualitative analysis of the constituents. Relative peak area of each component was used for the hierarchical cluster analysis. Results According to UHPLC-ESI-QTOF-MS data, chemical structures of 28 coumarins in the fruits of C. monnieri were identified, including 19 simple coumarins, seven linear coumarins, and two angular coumarins. Among these constituents, ten coumarins were firstly identified in C. monnieri. In addition, hierarchical cluster analysis suggested that C. monnieri from different regions could be classified into four groups, and this clustering was correlated to the distribution significantly, Xuzhou could be regarded as the genuine producing area. Conclusion UHPLC-ESI-Q-TOF-MS is a viable method for qualitative analysis and quality evaluation of coumarins from the fruit of C. monnieri.
