| 1 | Copper ions influence the toxicity of β-amyloid(1-42) in a concen-tration-dependent manner in a Caenorhabditis elegans model of Alzheimer’s disease显示文摘β-amyloid (Aβ) and copper play important roles in the pathogenesis of Alzheimer’s disease (AD).However,the behavioral correlativity and molecular mechanisms of Aβ and copper toxicity have been investigated less often.In the present study,we investigated the interaction and toxicity of Aβ1-42 and copper in the Aβ1-42 transgenic Caenorhabditis elegans worm model CL2006.Our data show that the paralysis behavior of CL2006 worms significantly deteriorated after exposure to 10-3 mol L-1 copper ions.However,the paralysis behavior was dramatically attenuated with exposure to 10-4 mol L-1 copper ions.The exogenous copper treatment also partially changed the homeostatic balance of zinc,manganese,and iron.Our data suggest that the accumulation of reactive oxygen species (ROS) was responsible for the paralysis induced by Aβ and copper in CL2006.The ROS generation induced by Aβ and copper appear to be through sod-1,prdx-2,skn-1,hsp-60 and hsp-16.2 genes. | LUO YunFeng ZHANG Jie LIU NianQing LUO Yuan ZHAO BaoLu | 2011 | Science China(Life Sciences)2011,54,6: | 26 |
| 2 | Antioxidant effects of green tea polyphenols显示文摘The unprecedented interest in the antioxidant activity of green tea polyphenols (GTP) is due to the appar-ent health benefits of tea drinking and the experimental re-sults with the polyphenols. The results suggest that the gal-loylated catechins show stronger antioxidant effect than that of nongalloylated catechins and the double bond in C ring also plays an important role in this effect. There are synergic effects between different catechins and the antioxidant effect of (+)-epimers is stronger than that of (-)-epimers. The active site to scavenge free radicals mainly locates in p-hydroxyl of phenol in gallic acid and hydroxyl of phenol in the pyrane also shows activity. The large p bond in chromane and benzene ring of catechin is the structure base for their anti-oxidant effects. | 3 | Green tea catechins prevent obesity through modulation of peroxisome proliferator-activated receptors显示文摘Epidemiological evidence and experimental studies suggest that drinking green tea is associated with a lower risk of obesity and related diseases. However, the mechanisms of these effects are not clear. In the present study, we investigated the anti-obesity mechanisms of green tea catechins (GTCs) through modulation of peroxisome proliferator activated-receptor (PPAR) pathways in high-fat diet-induced obesity in rats. GTC supplementation significantly attenuated the increased body and liver weights and the elevated serum and liver triglyceride levels. Meanwhile, GTCs increased the PPARγ levels in subcutaneous white adipose tissue (SWAT) and decreased the PPAR levels in visceral white adipose tissue (VWAT). In addition, GTC treatment up-regulated the levels of PPARδ in SWAT, VWAT, and brown adipose tissue and increased the expression of genes involved in fatty acid oxidation in brown adipose tissue. | 4 | Effects of r-HuEPO on the biophysical characteristics of erythrocyte membrane in patients with anemia of chronic renal failure显示文摘Using electron spin resonance (ESR) spin labeling technique, we have studied the conformation of sulfhydryl groups(-SH) binding sites in membrane proteins and membrane fluidity of red blood cells (RBCs) from two groups of patients with anemia of chronic renal failure (ACRF). One of the groups is composed of patients who were untreated with recombinant human erythropoietin(r-HuEPO), and the other is composed of patients who were treated with r-HuEPO. The results indicated: 1) the conformation of SH group binding site in RBC membrane proteins from former group was different from those of health people. 2)the fluidity in the region near the surface of RBC membrane from former group was lower than those of healthy people. 3) However, the above biophysical properties of RBC membrane from later group were normal. | 5 | L-theanine inhibits nicotine-induced dependence via regulation of the nicotine acetylcholine receptor-dopamine reward pathway显示文摘In this study, the inhibitory effect of L-theanine, an amino acid derivative of tea, on the rewarding effects of nicotine and its underlying mechanisms of action were studied. We found that L-theanine inhibited the rewarding effects of nicotine in a con- ditioned place preference (CPP) model of the mouse and reduced the excitatory status induced by nicotine in SH-SY5Y cells to the same extent as the nicotine receptor inhibitor dihydro-beta-erythroidine (DH E). Further studies using high performance liquid chromatography, western blotting and immunofluorescence staining analyses showed that L-theanine significantly inhibited nicotine-induced tyrosine hydroxylase (TH) expression and dopamine production in the midbrain of mice. L-theanine treatment also reduced the upregulation of the 4, 2 and 7 nicotine acetylcholine receptor (nAChR) subunits induced by nico- tine in mouse brain regions that related to the dopamine reward pathway, thus decreasing the number of cells that could react to nicotine. In addition, L-theanine treatment inhibited nicotine-induced c-Fos expression in the reward circuit related areas of the mouse brain. Knockdown of c-Fos by siRNA inhibited the excitatory status of cells but not the upregulation of TH induced by nicotine in SH-SY5Y cells. Overall, the present study showed that L-theanine reduced the nicotine-induced reward effects via inhibition of the nAChR-dopamine reward pathway. In its first phase, JUNA aims at the direct measurements of^(25)Mg(p,γ)^(26)Al,^(19)F(p,α)^(16)O,^(13)C(α,n)^(16)O and ^(12)C(α,γ)^(16)O reactions. | 8 | Geodynamic processes of the southeastern Neo-Tethys Ocean and the formation mechanism of the curved subduction system in Southeast Asia显示文摘Southeast Asia is located at the intersection of the Tethys and Pacific domains. The superimposed effects of the two tectonic domains have resulted in complicated deep structure, surface magma responses, and dynamic processes of Southeast Asia. Based on the latest long-term passive seismic experiment and numerical modeling, this study reconstructs the dynamic processes of the closure of the Neo-Tethys Ocean and the formation of the curved subduction system in Southeast Asia since the Late Mesozoic. P-wave velocity structure shows a remnant of the Neo-Tethys subducted slab in the lower mantle beneath Southeast Asia at a depth of approximately 1500 km. On the Java-East Timor subduction zone, the remnant slab is coupled with the Indo-Australian subducting slab in the upper mantle with the same direction, while on the Sumatra subduction zone, the remnant slab is decoupled from the Indo-Australian subducting slab in different directions. The formation of the curved subduction system in Southeast Asia is resulted from the northward subdcutions of previous Neo-Tethys and current IndoAustralian Plate, and the westward subduction of the Pacific Plate since Mesozoic. The former is characterized by continuous subduction and subsequent continental block collision, forming the current continental lithosphere in Southeast Asia and the curve-shaped Sumatra-Java subduction zone;the latter is characterized by subduction retreat and back-arc spreading, forming the eastern Philippine subduction zone and a series of marginal sea basins. The results showed that nitric oxide donor S-nitroso-N-acetyl-penicillamine (SNAP; 500 (imol/L) could induce apoptosis in immature cultures of cerebellar granule cells. Flow cytometry and HPLC analyses revealed that after treatment with SNAP, the mitochondrial transmembrane potential and the cellular ATP content decreased significantly. Nitric oxide scavenger hemoglobin could effectively prevent the neuronal mitochondria from dysfunction and attenuate apoptosis.
