| 2 | Xinglou Chengqi Decoction improves neurological function in experimental stroke mice as evidenced by gut microbiota analysis and network pharmacology显示文摘The current study was designed to explore the brain protection mechanism of Xinglou Chengqi Decoction(XCD)based on gut microbiota analysis and network pharmacology. A transient middle cerebral artery occlusion(MCAO) model of mice was established, followed by behavioral evaluation, TTC and TUNEL staining. Additionally, to investigate the effects of gut microbiota on neurological function after stroke, C57BL/6 mice were treated with anti-biotic cocktails 14 days prior to ischemic stroke(IS) to deplete the gut microbiota. High-throughput 16S rDNA gene sequencing, metabonomics technique, and flow multifactor technology were used to analyze bacterial communities, SCFAs and inflammatory cytokines respectively. Finally, as a supplement, network pharmacology and molecular docking were applied to fully explore the multicomponent-multitarget-multichannel mechanism of XCD in treating IS, implicated in ADME screening, target identification, network analysis, functional annotation, and pathway enrichment analysis.We found that XCD effectively improved neurological function, relieved cerebral infarction and decreased the neuronal apoptosis.Moreover, XCD promoted the release of anti-inflammatory factor like IL-10, while down-regulating pro-inflammatory factors such as TNF-α, IL-17A, and IL-22. Furthermore, XCD significantly increased the levels of short chain fatty acids(SCFAs), especially butyric acid. The mechanism might be related to the regulation of SCFAs-producing bacteria like Verrucomicrobia and Akkermansia, and bacteria that regulate inflammation like Paraprevotella, Roseburia, Streptophyta and Enterococcu. Finally, in the network pharmacological analysis, 51 active compounds in XCD and 44 intersection targets of IS and XCD were selected. As a validation, components in XCD docked well with key targets. It was obviously that biological processes were mainly involved in the regulation of apoptotic process, inflammatory response, response to fatty acid, and regulation of establishment of endothelial barrier in GO enrichment. XCD can improve neurological function in experimental stroke mice, partly due to the regulation of gut microbiota. | 9 | Robust UV/moisture dual curable PDMS-microcapsule-silica functional material for self-healing,antifouling,and antibacterial applications显示文摘Polydimethylsiloxane containing methacryloyloxy and methoxy silane groups(MAPDMS)-microcapsule-SiO_(2)(MPMS)functional materials were prepared by constructing micro-nano hierarchical structures on the surface of MAPDMS matrix.Herein,MAPDMS@1,1-stilbene-modified hydrolyzed polyglycidyl methacrylate/graphene oxide/dimethyloctadecyl[3-(trimethoxysilyl)propyl]ammonium chloride(MAPDMS@PGMA_(m)/GO/QC18)self-healing microcapsules with compact multi-shell structure were synthesized and combined with nano-SiO_(2)to construct the hierarchical structures.Furthermore,ultraviolet(UV)/moisture dual curing mode was introduced into deep curing reaction and efficient self-healing reaction of the MPMS.The results show that the introduction of UV/moisture dual curing mode and micro-nano hierarchical structure gives MPMS functional materials excellent mechanical properties,antifouling properties,self-healing properties,antibacterial properties.The shear strength and tensile strength of MPMS increase from 3.32 and 4.26 MPa of MAPDMS to 3.81 and 5.06 MPa,respectively.Its static contact angle increases from 115.9°of MAPDMS to 156.5°,its slide angle decreases from 68.5°of MAPDMS to 7.8°,respectively.The antifouling performance of MPMS against seawater,soy sauce,juice,coffee,protein,other contaminants is effectively improved compared with MAPDMS matrix.At the same time,the tensile strength and elongation at break of MPMS after healing reach 98.22%and 96.57%of those in original state,respectively.In addition,the antibacterial rates of MPMS against Escherichia coli and Staphylococcus aureus reach 99.85%and 100%,respectively.The MPMS prepared in this paper is expected to be widely used in marine antifouling,pipeline network,anti-icing,microfluidics,wearable devices,medical devices,electrochemical biosensors,other fields. | Nan Zheng Jie Liu Guoqing Wang Pan Yao Lihong Dang Ze Liu Jiufu Lu Wenge Li | 2023 | Nano Research2023,16,5: | 2 |
| 10 | Salmonella-mediated blood-brain barrier penetration,tumor homing and tumor microenvironment regulation for enhanced chemo/bacterial glioma therapy显示文摘Chemotherapy is an important adjuvant treatment of glioma,while the efficacy is far from satisfactory,due not only to the biological barriers of blood-brain barrier(BBB)and blood-tumor barrier(BTB)but also to the intrinsic resistance of glioma cells via multiple survival mechanisms such as upregulation of P-glycoprotein(P-gp).To address these limitations,we report a bacteria-based drug delivery strategy for BBB/BTB transportation,glioma targeting,and chemo-sensitization.Bacteria selectively colonized into hypoxic tumor region and modulated tumor microenvironment,including macrophages repolarization and neutrophils infiltration.Specifically,tumor migration of neutrophils was employed as hitchhiking delivery of doxorubicin(DOX)-loaded bacterial outer membrane vesicles(OMVs/DOX).By virtue of the surface pathogen-associated molecular patterns derived from native bacteria,OMVs/DOX could be selectively recognized by neutrophils,thus facilitating glioma targeted delivery of drug with significantly enhanced tumor accumulation by 18-fold as compared to the classical passive targeting effect.Moreover,the P-gp expression on tumor cells was silenced by bacteria typeⅢsecretion effector to sensitize the efficacy of DOX,resulting in complete tumor eradication with 100%survival of all treated mice.In addition,the colonized bacteria were finally cleared by anti-bacterial activity of DOX to minimize the potential infection risk,and cardiotoxicity of DOX was also avoided,achieving excellent compatibility.This work provides an efficient trans-BBB/BTB drug delivery strategy via cell hitchhiking for enhanced glioma therapy. | Ze Mi Qing Yao Yan Qi Jinhai Zheng Jiahao Liu Zhenguo Liu Hongpei Tan Xiaoqian Ma Wenhu Zhou Pengfei Rong | 2023 | Acta Pharmaceutica Sinica B2023,13,2: | 1 |

These complexes also represent major antigenic targets for anti-viral immune responses. The gC II complex consists of two glycoproteins, gM and gN. In the current study, DNA vaccines expressing the murine cytomegalovirus (MCMV) homologs of the gM and gN proteins were evaluated for protection against lethal MCMV infection in a mouse model. Humoral and cellular immune responses, spleen viral titers, and mice survival and body-weight changes were examined. The results showed that immunization with gM or gN DNA vaccine alone was not able to offer good protection, whereas co-immunization with both gM and gN induced an effective neutralizing antibody response and cellular immune response, and provided mice with complete protection against a lethal MCMV challenge.
