| 1 | Chemical screen identifies a geroprotective role of quercetin in premature aging显示文摘Aging increases the risk of various diseases. The main goal of aging research is to find therapies that attenuate aging and alleviate aging-related diseases. In this study, we screened a natural product library for geroprotective compounds using Werner syndrome (WS) human mesenchymal stem cells (hMSCs), a premature aging model that we recently established. Ten candidate compounds were identified and quercetin was investigated in detail due to its leading effects. Mechanistic studies revealed that quercetin alleviated senescence via the enhancement of cell proliferation and restoration of heterochromatin architecture in WS hMSCs. RNA-sequencing analysis revealed the transcriptional commonalities and differences in the geroprotective effects by quercetin and Vitamin C. Besides WS hMSCs, quercetin also attenuated cellular senescence in Hutchinson-Gilford progeria syndrome (HGPS) and physiological-aging hMSCs. | 2 | SIRT7 antagonizes human stem cell aging as a heterochromatin stabilizer显示文摘SIRT7,a sirtuin family member implicated in aging and disease,is a regulator of metabolism and stress responses.It remains elusive how human somatic stem cell populations might be impacted by SIRT7.Here,we found that SIRT7 expression declines during human mesenchymal stem cell(hMSC)aging and that SIRT7 deficiency accelerates senescence.Mechanistically,SIRT7 forms a complex with nuclear lamina proteins and heterochromatin proteins,thus maintaining the repressive state of heterochromatin at nuclear periphery.Accordingly,deficiency of SIRT7 results in loss of heterochromatin,derepression of the LINE1 retrotransposon(LINE1),and activation of innate immune signaling via the cGAS-STING pathway.These agingassociated cellular defects were reversed by overexpression of heterochromatin proteins or treatment with a LINE1 targeted reverse-transcriptase inhibitor.Together,these findings highlight how SIRT7 safeguards chromatin architecture to control innate immune regulation and ensure geroprotection during stem cell aging. | Shijia Bi Zunpeng Liu Zeming Wu Zehua Wang Xiaoqian Liu Si Wang Jie Ren Yan Yao Weiqi Zhang Moshi Song Guang-Hui Liu Jing Qu | 2020 | Protein & Cell2020,11,7: | 10 |

| 5 | High yield production of 1,4-cyclohexanediol from lignin derived2,6-dimethoxybenzoquinone via Raney Ni Mn catalyst in hydrogen free conditions显示文摘Transformation of lignin to valuable chemicals via sustainable pathways is recognized as one of the most efficient ways to explore its value and replace the nonrenewable petroleum resource. In this work, an environmental-friendly transfer hydrogenation process has been developed to convert lignin derived2,6-dimethoxybenzoquinone to 1,4-cyclohexanediol. Compared with previous work under hydrogen pressure(30 bar), this process uses isopropanol as both solvent and hydrogen donor, which significantly simply the operation process. The core of this study is the design and preparation of Mn modified Raney Ni catalysts by ball milling process. A series of Raney Ni Mn catalysts with different ball milling time and Mn content were prepared and investigated. Characterizations by X-ray diffraction(XRD), X-ray photoelectron spectroscopy(XPS), scanning electron microscope(SEM) and transmission electron microscope(TEM) etc. showed that Ni Mn Al alloy was formed during the ball milling process and then transformed to Ni Mn alloy after treatment by aqueous Na OH. | 6 | An Improved Calibration Method of Grating Projection Measurement System显示文摘In the traditional fringe projection profilometry system,the projector and the camera light center are both spatially virtual points.The spatial position relationships specified in the model are not easy to obtain,leading to inaccurate system parameters and affectingmeasurement accuracy.This paper proposes a method for solving the system parameters of the fringe projection profilometry system,and the spatial position of the camera and projector can be adjusted in accordance with the obtained calibration parameters.The steps are as follows:First,in accordance with the conversion relationship of the coordinate system in the calibration process,the calculation formula of the vertical distance from the camera light center to the reference plane and the calculation formula of the distance between the projector and the camera light center are given respectively.Secondly,according to the projector calibration principle,the position of the projector light axis perpendicular to the reference plane is gained by comparing the parallel relationship between the reference plane coordinate system and the projector coordinate system’s Z-axis.Then,in order to fulfill the position restriction that the line between the projector light center and the camera light center must be parallel to the reference plane,the camera’s spatial location is adjusted so that the vertical distance between it and the reference plane tends to that between the projector light center and the reference plane.And finally,the three-dimensional(3D)reconstruction of the target object can be finished using the phase height model’s system parameters once the aforementioned position limitations are put into practice.Experimental results demonstrate that the method improves the measurement accuracy,and verifies that it is effective and available in 3D shape measurement. | Qiucheng Sun Weiyu Dai Mingyu Sun Zeming Ren Mingze Wang | 2023 | Computers, Materials & Continua2023,,5: | 0 |
