| 1 | Exosome-guided bone targeted delivery of Antagomir-188 as an anabolic therapy for bone loss显示文摘The differentiation shift from osteogenesis to adipogenesis of bone marrow mesenchymal stem cells(BMSCs)characterizes many pathological bone loss conditions.Stromal cell-derived factor-1(SDF1)is highly enriched in the bone marrow for C-X-C motif chemokine receptor 4(CXCR4)-positive hematopoietic stem cell(HSC)homing and tumor bone metastasis.In this study,we displayed CXCR4 on the surface of exosomes derived from genetically engineered NIH-3T3 cells.CXCR4+exosomes selectively accumulated in the bone marrow.Then,we fused CXCR4+exosomes with liposomes carrying antagomir-188 to produce hybrid nanoparticles(NPs).The hybrid NPs specifically gathered in the bone marrow and released antagomir-188,which promoted osteogenesis and inhibited adipogenesis of BMSCs and thereby reversed age-related trabecular bone loss and decreased cortical bone porosity in mice.Taken together,this study presents a novel way to obtain bone-targeted exosomes via surface display of CXCR4 and a promising anabolic therapeutic approach for age-related bone loss. | Yan Hu Xiaoqun Li Qin Zhang Zhengrong Gu Ying Luo Jiawei Guo Xiuhui Wang Yingying Jing Xiao Chen Jiacan Su | 2021 | Bioactive Materials2021,6,9: | 12 |

In both hMSCs and hNSCs, RAP1 not only negatively regulated telomere length but also acted as a transcriptional regulator of RELN by tuning the methylation status of its gene promoter. | 5 | Famitinib in combination with concurrent chemoradiotherapy in patients with locoregionally advanced nasopharyngeal carcinoma: a phase 1, open-label, dose-escalation Study显示文摘Background:Famitinib is a tyrosine kinase inhibitor against multiple targets,including vascular endothelial growth factor receptor 2/3,platelet-derived growth factor receptor,and stem cell factor receptor(c-kit).Previous studies have demonstrated anti-tumour activities of famitinib against a wide variety of advanced-stage solid cancers.We aimed to determine the safety and efficacy of famitinib with concurrent chemoradiotherapy(CCRT)in patients with locoregionally advanced nasopharyngeal carcinoma(NPC).We also evaluated the feasibility of contrast-enhanced ultrasound(D-CEUS)as a predictor of early tumour response to famitinib and to correlate functional parameters with clinical efficacy.Methods:The trial was conducted in subjects with stage III or IVa-b NPC using a 3+3 design of escalating fami-tinib doses.Briefly,subjects received 2 weeks of famitinib monotherapy followed by 7 weeks of famitinib plus CCRT.D-CEUS of the neck lymph nodes was performed at day 0,8 and 15 after famitinib was administered before starting concurrent chemoradiotherapy.End points included safety,tolerability and anti-tumour activity.Results:Twenty patients were enrolled(six each for 12.5,16.5 and 20 mg and two for 25 mg).Two patients in the 25 mg cohort developed dose-limiting toxicities,including grade 4 thrombocytopenia and grade 3 hypertension.The most common grade 3/4 adverse events were leukopenia,neutropenia and radiation mucositis.D-CEUS tests showed that more than 60%of patients achieved a perfusion parameter response after 2 weeks taking famitinib alone,and the parameter response was associated with disease improvement.In the famitinib monotherapy stage,three patients(15%)showed partial responses.The complete response rate was 65%at the completion of treatment and 95%3 months after the treatment ended.After a median follow-up of 44 months,the 3-year progression-free survival(PFS)and distant metastasis-free survival were 70%and 75%,respectively.Subjects with a decrease of perfusion parameter response,such as peak intensity decreased at least 30%after 1 week of famitinib treatment,had higher 3-year PFS(90.9%vs.44.4%,95%CI 73.7%-100%vs.11.9%-76.9%,P<0.001)than those with an increase or a reduction of less than 30%.Conclusions:The recommended famitinib dose for phase II trial is 20 mg with CCRT for patients with local advanced NPC.D-CEUS is a reliable and early measure of efficacy for famitinib therapies.Further investigation is required to confirm the effects of famitinib plus chemoradiotherapy. | Qiuyan Chen Linquan Tang Na Liu Feng Han Ling Guo Shanshan Guo Jianwei Wang Huai Liu Yanfang Ye Lu Zhang Liting Liu Pan Wang Yingqin Li Qingmei He Xiaoqun Yang Qingnan Tang Yang Li YuJing Liang XueSong Sun Chuanmiao Xie Yunxian Mo Ying Guo Rui Sun Haoyuan Mo Kajia Cao Xiang Guo Musheng Zeng Haiqiang Mai Jun Ma | 2018 | Cancer Communications2018,38,1: | 6 |
| 6 | Improvement of the Compressive Strength of Carbon Fiber/Epoxy Composites via Microwave Curing显示文摘Microwave processing was used to cure the carbon fiber/epoxy composites and designed for improving the compressive strength of the materials. By controlling the power of microwave heating, vacuum bagged laminates were fabricated under one atmosphere pressure without arcing. The physical and mechanical properties of composites produced through vacuum bagging using microwave and thermal curing were compared and the multistep(2-step or 3-step) microwave curing process for improved compressive properties was established. The results indicated that microwave cured samples had somewhat differentiated molecular structure and showed slightly higher glass transition temperature. The 2-step process was found to be more conducive to the enhancement of the compressive strength than the 3-step process. A 39% cure cycle time reduction and a 22% compressive strength increment were achieved for the composites manufactured with microwave radiation. | 10 | Identification of late-stage pollen-specific promoters for construction of pollen-inactivation system in rice显示文摘Large-scale production of male sterile seeds can be achieved by introducing a fertility-restoration gene linked with a pollen-killer gene into a recessive male sterile mutant.We attempted to construct this system in rice by using a late-stage pollen-specific(LSP)promoter driving the expression of maizeα-amylase gene ZM-AA1.To obtain such promoters in rice,we conducted comparative RNA-seq analysis of mature pollen with meiosis anther,and compared this with the transcriptomic data of various tissues in the Rice Expression Database,resulting in 269 candidate LSP genes.Initial test of nine LSP genes showed that only the most active OsLSP3 promoter could drive ZM-AA1 to disrupt pollen.We then analyzed an additional 22 LSP genes and found 12 genes stronger than OsLSP3 in late-stage anthers.The promoters of OsLSP5 and OsLSP6 showing higher expression than OsLSP3 at stages 11 and 12 could drive ZM-AA1 to inactivate pollen,while the promoter of OsLSP4 showing higher expression at stage 12 only could not drive ZM-AA1 to disrupt pollen,suggesting that strong promoter activity at stage 11 was critical for pollen inactivation.The strong pollen-specific promoters identified in this study provided valuable tools for genetic engineering of rice male sterile system for hybrid rice production. | Menglong Wang Wei Yan Xiaoqun Peng Zhufeng Chen Chunjue Xu Jianxin Wu Xing Wang Deng Xiaoyan Tang | 2020 | Journal of Integrative Plant Biology2020,62,8: | 4 |

| 13 | Comparative study of carbonic anhydrase activity in waters among different geological eco-environments of Yangtze River basin and its ecological significance显示文摘This study provides the presence of carbonic anhydrase(CA) activity in waters of the Yangtze River basin, China, as well as the correlation of CA activity with HCO-3 concentration and CO_2 sink flux. Different degrees of CA activity could be detected in almost all of the water samples from different geological eco-environments in all four seasons. The CA activity of water samples from karst areas was significantly higher than from non-karst areas(PP3-concentration(r = 0.672, P2 sink flux(r = 0.602, P = 0.076) in karst areas. This suggests that CA in waters might have a promoting effect on carbon sinks for atmospheric CO_2 in karst river basins. In conditions of similar geological type, higher CA activity was generally detected in water samples taken from areas that exhibited better eco-environments, implying that the CA activity index of waters could be used as an indicator for monitoring ecological environments and protection of river basins. | 20 | Loss of the centrosomal protein Cenpj leads to dysfunction of the hypothalamus and obesity in mice显示文摘Cenpj is a centrosomal protein located at the centrosomes and the base of cilia,it plays essential roles in regulating neurogenesis and cerebral cortex development.Although centrosomal and cilium dysfunction are one of the causes of obesity,insulin resistance,and type 2 diabetes,the role that Cenpj plays in the regulation of body weight remains unclear.Here,we deleted Cenpj by crossing Cenpjflox/flox mice with Nkx2.1-Cre mice.Loss of the centrosomal protein Cenpj in Nkx2.1-expressing cells causes morbid obesity in mice at approximately 4 months of age with expended brain ventricles but no change of brain size.We found that hypothalamic cells exhibited reduced proliferation and increased apoptosis upon Cenpj depletion at the embryonic stages,resulting in a dramatic decrease in the number of Proopiomelanocortin(POMC)neurons and electrophysiological dysfunction of NPY neurons in the arcuate nucleus(ARC)in adults.Furthermore,depletion of Cenpj also reduced the neuronal projection from the ARC to the paraventricular nucleus(PVN),with decreased melanocortin-4 receptors(MC4R)expression in PVN neurons.The study defines the roles that Cenpj plays in regulating hypothalamus development and body weight,providing a foundation for further understanding of the pathological mechanisms of related diseases. | Wenyu Ding Changjiang Zhang Baisong Wang Xin Zhou Le Sun Suijuan Zhong Jing Liu Junjing Zhang Xiaoqun Wang Qian Wu | 2021 | Science China(Life Sciences)2021,64,3: | 1 |
