| 5 | Supplementation with yak (Bos grunniens) bone collagen hydrolysate altered the structure of gut microbiota and elevated short-chain fatty acid production in mice显示文摘In this study, yak bone collagen hydrolysate(YBCH)was produced by mixed proteases and provided to standard-diet mice at a different dose(low dose(LD), medium dose(MD), and high dose(HD))to investigate its effects on the composition of gut microbiota and short-chain fatty acids(SCFA)production. It was found that YBCH was mainly composed of small molecular peptides whose molecular weight below 2 000 Da. Notably, supplementation with different doses of YBCH could significantly downregulate the ratio of Firmicutes to Bacteroidetes in the fecal microbiota. At the family level, the Lachnospiraceae abundance was significantly reduced in the YBCH gavage groups(mean reduction ratio 41.7 %, 35.2%, and 36.4% for LD, MD, and HD group, respectively). The predicted functions of gut microbes in the MD group were significantly increased at “lipid metabolism” and “glycan biosynthesis and metabolism”. Moreover, the SCFA production in the YBCH groups was elevated. Especially, the concentration of acetic acid, propionic acid, and butyric acid in the MD group was separately increased 79.7%, 89.2%, and 78.8% than that in the NC group. | 9 | CcpA mutants infuence selective carbon source utilization by changing interactions with target genes in Bacillus licheniformis显示文摘The gram-positive bacterium Bacillus licheniformis exhibits obvious selective utilization on carbon sources.This process is mainly governed by the global regulator catabolite control protein A(CcpA),which can recognize and bind to multiple target genes that are widely distributed in metabolic pathways.Although the DNA-binding domain of CcpA has been predicted,the infuence of key amino acids on target gene recognition and binding has yet to be uncovered.In this study,the impact of Lys31,Ile42 and Leu56 on in vitro protein–DNA interactions and in vivo carbon source selective utilization was investigated.The results showed that alanine substitution of Lys31 and Ile42,located within the 3rd helices of the DNAbinding domain,signifcantly weakened the binding strength between CcpA and target genes.These mutations also lead to alleviated repression of xylose utilization in the presence of glucose.On the other hand,the Leu56Arg mutant in the 4th helices exhibited enhanced binding afnity compared with that of the wild-type one.When this mutant was used to replace the native one in B.licheniformis cells,the selective utilization of glucose over xylose increased.This study provides a new strategy for understanding the relationship between the function and structure of regulatory proteins.This study also used a new strategy was used to regulate carbon source utilization beyond CCR engineering. | Yupeng Zhang Youran Li Fengxu Xiao Hanrong Wang Liang Zhang Zhongyang Ding Sha Xu Zhenghua Gu Guiyang Shi | 2022 | Systems Microbiology and Biomanufacturing2022,2,1: | 0 |
| 10 | Heterologous expression,refolding,and characterization of a calcium-independent phospholipase A1 from Streptomyces albidoflavus显示文摘Phospholipase A1(PLA_(1))is a member of the hydrolase family with applications in various fields,especially in the food industry.A calcium-independent PLA_(1) from Streptomyces albidoflavus was expressed in E.coli BL21(DE3)in this study.The results indicated that the soluble expression of PLA_(1) was at low level,which was possibly due to the toxicity of PLA_(1) to the host.In contrast,the expression of the enzyme as inclusion bodies exhibited a high-level expression and 0.3 mg inclusion bodies protein could be derived from 1 mL culture medium.Furthermore,the renaturation of PLA_(1) was achieved through a direct dilution method,yielding 29.6 U/mL PLA_(1) activity after 16 h of renaturation at 4℃.For improving the efficiency of the dilution refolding process,a continuous refolding strategy was established,and 155 U/mL PLA_(1) activity was derived from the continuous refolding process.With soybean lecithin as the substrate,the specific activity of purified renatured PLA_(1) was 1380 U/mg and the optimal temperature and pH was found to be 60℃ and 6.5.In addition,the renatured PLA_(1) was observed with better activity towards phosphatidyl inositol,whilst lipase activity was detected when the catalyzing temperature was below 55℃.Overall,this study provides a possible solution to obtain calcium-independent PLA_(1) with high yield by heterologous expression in E.coli and hence to promote its further application in the field of food industry. | Shi Cheng Zitao Guo Chaojuan Liang Yi Shi Peng Geng Yu Xin Zhenghua Gu Liang Zhang | 2022 | Systems Microbiology and Biomanufacturing2022,2,3: | 0 |
