| 1 | Recombinant human PDCD5 protein enhances chemosensitivities of hematologic malignancies显示文摘PDCD5 is a novel apoptosis-related gene. Overexpression of PDCD5 facilitates apoptosis in various tumor cells. Here, we investigated the roles of recombinant human PDCD5 (rhPDCD5) protein in chemosensitivities of hematologic malignancies. The rhPDCD5 increased the in vitro cytotoxicity of daunorubicin (DNR), adriamycin (ADM) and As2O3 in three hematologic tumor cell lines. In vivo studies showed that DNR combined with rhPDCD5 significantly suppressed tumor growth of U937 xenograft nude mice compared with DNR alone. | 3 | Supplementation with yak (Bos grunniens) bone collagen hydrolysate altered the structure of gut microbiota and elevated short-chain fatty acid production in mice显示文摘In this study, yak bone collagen hydrolysate(YBCH)was produced by mixed proteases and provided to standard-diet mice at a different dose(low dose(LD), medium dose(MD), and high dose(HD))to investigate its effects on the composition of gut microbiota and short-chain fatty acids(SCFA)production. It was found that YBCH was mainly composed of small molecular peptides whose molecular weight below 2 000 Da. Notably, supplementation with different doses of YBCH could significantly downregulate the ratio of Firmicutes to Bacteroidetes in the fecal microbiota. At the family level, the Lachnospiraceae abundance was significantly reduced in the YBCH gavage groups(mean reduction ratio 41.7 %, 35.2%, and 36.4% for LD, MD, and HD group, respectively). The predicted functions of gut microbes in the MD group were significantly increased at “lipid metabolism” and “glycan biosynthesis and metabolism”. Moreover, the SCFA production in the YBCH groups was elevated. Especially, the concentration of acetic acid, propionic acid, and butyric acid in the MD group was separately increased 79.7%, 89.2%, and 78.8% than that in the NC group. | 4 | The alteration of gut microbiota by bioactive peptides:a review显示文摘Evidences suggest that the homeostasis of gut microbiota is among the most important factors for maintaining the physical and mental health of the host.Among the multiple factors affecting the homeostasis of gut microbiota,diet is one of the decisive factors.Bioactive peptides derived from protein hydrolyzed by protease or fermented by microorganism have many physiological activities that their parent proteins do not have.Currently,bioactive peptides attract more and more attention due to their bidirectional interaction with gut microbes.It has been reported that some bioactive peptides could alter the composition of gut microbiota by influencing the intestinal microenvironment.Meanwhile,quite a few bioactive peptides that are released by gut microbes or intestinal cells could resist the pathogenic bacteria to sustain the homeostasis of gut microbiota.In this review,some exogenous bioactive peptides derived from food and some endogenous bioactive peptides released from intestinal cells or microbes were discussed to summary their effects on the modulation of gut microbiota.This review is expected to provide new ideas for related research,and as well to promote the application of bioactive peptides in the fields of food and medicine. | Zitao Guo Dalong Yi Bo Hu Yi Shi Yu Xin Zhenghua Gu Huaigao Liu Liang Zhang | 2021 | Systems Microbiology and Biomanufacturing2021,1,4: | 0 |
| 5 | Probabilistic analysis of secant piles with random geometric imperfections显示文摘The failure to achieve minimum design overlap between secant piles compromises the ability of a structure to perform as designed,resulting in water leakage or even ground collapse.To establish a more realistic simulation and provide guidelines for designing a safe and cost-effective secant-pile wall,a three-dimensional model of a secant pile,considering the geometric imperfections of the diameter and direction of the borehole,is introduced.An ultrasonic cross-hole test was performed during the construction of secant piles in a launching shaft in Beijing,China.Based on the test results,the statistical characteristics of the pile diameters and orientation parameters were obtained.By taking the pile diameter D,inclination angleβ,and azimuth angleαas random variables,Monte Carlo simulations were performed to discuss the influence of different design parameters on the probability density functions of the overlap of secant piles.The obtained results show that the randomness of the inclination angle and pile diameter can be well described by a normal distribution,whereas the azimuth angle is more consistent with a uniform distribution.The integrity of the secant-pile wall can be overestimated without considering the uncertainty of geometric imperfections.The failure of the secant-pile wall increases substantially with increasing spatial variability in drilling inclination and diameter.A design flowchart for pile spacing under the target safety level is proposed to help engineers design a safe and economical pile wall. | Yi YANG Dalong JIN Xinggao LI Weilin SU Xuyang WANG | 2021 | Frontiers of Structural and Civil Engineering2021,15,3: | 0 |
