| 1 | The Chrysanthemum nankingense Genome Provides Insights into the Evolution and Diversification of Chrysanthemum Flowers and Medicinal Traits显示文摘The Asteraceae (Compositae),a large plant family of approximately 24 000-35 000 species,accounts for^10% of all angiosperm species and contributes a lot to plant diversity.The most representative members of the Asteraceae are the economically important chrysanthemums (Chrysanthemum L.)that diversified through reticulate evolution.Biodiversity is typically created by multiple evolutionary mechanisms such as wholegenome duplication 0NGD)or polyploidization and locally repetitive genome expansion.However,the lack of genomic data from chrysanthemum species has prevented an in-depth analysis of the evolutionary mechanisms involved in their diversification.Here,we used Oxford Nanopore long-read technologyto sequence the diploid Chrysanthemum nankingense genome,which represents one of the progenitor genomes of domesticated chrysanthemums.Our analysis revealed that the evolution of the C.nankingense genome was driven by bursts of repetitive element expansion and WGD events including a recentWGD that distinguishes chrysanthemum from sunflower,which diverged from chrysanthemum approximately 38.8 million years ago.Variations of ornamental and medicinal traits in chrysanthemums are linked to the expansion of candidate gene families by duplication events including paralogous gene duplication.Collectively,our study of the assembled reference genome offers new knowledge and resources to dissect the history and pattern of evolution and diversification of chrysanthemum plants,and also to accelerate their breeding and improvement. | Chi Song Yifei Liu Aiping Song Gangqiang Dong Hongbo Zhao Wei Sun Shyam Ramakrishnan Ying Wang Shuaibin Wang Tingzhao Li Yan Niu Jiafu Jiang Bin Dong Ye Xia Sumei Chen Zhigang Hu Fadi Chen Shilin Chen | 2018 | Molecular Plant2018,11,12: | 24 |

| 4 | Dynamics of Soil Moisture and Salt Content After Infiltration of Saline Ice Meltwater in Saline-Sodic Soil Columns显示文摘Saline ice meltwater can be used for irrigation and leaching of salts in salt-affected soil regions.A laboratory experiment was conducted using soil columns to investigate the redistribution of soil moisture, salt and sodium adsorption ratio(SAR) in saline-sodic soil under the infiltration of saline ice meltwater.Soils were treated using saline water of three irrigation volumes(1 600, 2 400 and 3 200 mL) at four salinity levels.These four salinity levels included salt free(0 g L^(-1)), low salinity level(1.4 g L^(-1)), moderate salinity level(2.7 g L^(-1)) and high salinity level(4.1 g L^(-1)).The prepared saline water was frozen into ice, and then the ice was put on the surface of soil columns.After 96 h, the infiltration rate and soil moisture content of saline ice treatments were greater than those of salt-free ice treatments, increasing with the increase of ice salinity.Infiltration of saline ice meltwater increased soil moisture content in the upper layers for all treatments.Both salt contents and SAR values in the upper soil layers decreased in all saline ice treatments and were lower than those in salt-free ice treatment.However, this trend was reversed in the deeper(below 20 cm) soil layers.The highest desalting rate and lowest SAR were observed in high-salinity treatment under three irrigation volumes in the 0–15 cm soil layer,especially under irrigation volume of 2 400 mL.These results indicate that saline ice(0–20 cm) meltwater irrigation is beneficial to saline-sodic soil reclamation, and the best improvement effect would be achieved when using high-salinity ice under optimal irrigation volume. | XIAO Ye HUANG Zhigang YANG Fan WANG Zhichun ZHOU Xia LIU Zhijun | 2017 | Pedosphere2017,27,6: | 8 |
| 5 | DFT study of difference caused by catalyst supports in Pt and Pd catalysis of oxygen reduction reaction显示文摘Based on an experimental phenomenon that catalytic activity of Pt and Pd for oxygen reduction reaction (ORR) changes with catalyst supports from C to TiO2, density function theory (DFT) was used to elucidate the cause behind the difference in catalysis caused by catalyst supports. First, factors closely associated with the first electron transfer of the ORR were assessed in the light of quantum chemistry. Then intermediate (atomic oxygen, O) adsorption strength on the catalyst surface was calculated. The results show that, in terms of minimum energy difference, the best orbital symmetry match, and the maximum orbital overlap, TiO2 does bring about a very positive effect on catalysts Pd/TiO2 for the first electron transfer of the ORR. Especially, TiO2 remarkably expands the space size of Pd/TiO2 HOMO orbital and improves orbital overlap of Pd/TiO2 HOMO and O2 LUMO. The analysis of deformation density and partial density of state shows that the strong interaction between Pt and Ti leads to a strong adsorption of intermediate O on Pt/TiO2, but the strong interaction between Pd and surface O causes positive net charge of Pd and a weak adsorption of intermediate O on Pd/TiO2. Thus, the ORR can proceed more smoothly on Pd/TiO2 than Pt/TiO2 in every respect of maximum orbital overlap and rate delay by intermediate O. The research also discloses that several factors lead to less activity of TiO2-supported Pt and Pd catalysts than the C-supported ones for the ORR. | 6 | Ripple-effect analysis for operational architecture of air defense systems with supernetwork modeling显示文摘In order to solve the problem that the ripple-effect analysis for the operational architecture of air defense systems(OAADS)is hardly described in quantity with previous modeling approaches,a supernetwork modeling approach for the OAADS is put forward by extending granular computing. Based on that operational units and links are equal to different information granularities, the supernetwork framework of the OAADS is constructed as a 'fournetwork within two-layer' structure by forming dynamic operating coalitions, and measuring indexes of the ripple-effect analysis for the OAADS are given combining with Laplace spectral radius. In this framework, via analyzing multidimensional attributes which inherit relations between operational units in different granular scales, an extended granular computing is put forward integrating with a topological structure. Then the operation process within the supernetwork framework, including transformation relations between two layers in the vertical view and mapping relations among functional networks in the horizontal view, is studied in quantity. | 7 | Expression of reconstructive hF Ⅷ in the hrDNA by using hrDNA targeting vector显示文摘Our lab has constructed a new nonviral vec-tor—hrDNA targeting vector(pHrneo). pHrneo is a human derived vector that can target gene into human ribosomal DNA(hrDNA) locus. In this study, we inserted expression cassette of reconstructive hF Ⅷ (hFVIII-BDDAK39) to pHrneo to construct targeting vector: pHrneo-BDDAK39. Through electroporation of pHrneo-BDDAK39 into HT1080 cells, we identified the homologous recombinants by PCR and Southen blotting, and tested the expression of hFVIII- BDDAK39 in the hrDNA locus. The hFⅧ-BDDAK39 was successfully targeted into hrDNA locus of HT-1080 by pHrneo-BDDAK39, and the efficiency of site-specific inte-gration was 2.0×10?5. hFⅧ-BDDAK39 in hrDNA locus of HT-1080 is found to be able to express efficiently (32±5 ng·106 cells?1·24 h?1). | 8 | Facile microwave-assisted ionic liquid synthesis of sphere-like BiOBr hollow and porous nanostructures with enhanced photocatalytic performance显示文摘In this work, two kinds of self-assembled hierarchical BiOBr microcrystals were rapidly synthesized through a simple microwave-assisted route in the presence of reactable ionic liquid 1-hexadecyl-3-methylimidazolium bromide([C_(16)mim]Br). These porous and hollow BiOBr microspheres were obtained via a facile solvothermal method with or without polyvinyl pyrrolidone(PVP), respectively. During the synthetic process, ionic liquid [C_(16)mim]Br played as solvent, reactant and template at the same time. Moreover, the BiOBr hollow and porous microspheres exhibited outstanding photocatalytic activities for the degradation of rhodamine B(RhB) under visible light irradiation. A possible photocatalytic mechanism was also discussed in detail. | 10 | Progressive deterioration of the upper respiratory tract and the gut microbiomes in children during the early infection stages of COVID-19显示文摘Children are less susceptible to coronavirus disease 2019 (COVID-19), and they have manifested lower morbidity and mortality after infection, for which a multitude of mechanisms may be considered. Whether the normal development of the gut-airway microbiome in children is affected by COVID-19 has not been evaluated. Here, we demonstrate that severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2)infection alters the upper respiratory tract and the gut microbiomes in nine children. The alteration of the microbiome is dominated by the genus Pseudomonas, and it sustains for up to 25e58 days in different individuals. Moreover, the patterns of alternation are different between the upper respiratory tract and the gut. Longitudinal investigation shows that the upper respiratory tract and the gut microbiomes are extremely variable among children during the course of COVID-19. The dysbiosis of microbiome persists in7 of 8 children for at least 19e24 days after discharge from the hospital. | 11 | Oscillation of Surface PM2.5 Concentration Resulting from an Alternation of Easterly and Southerly Winds in Beijing: Mechanisms and Implications显示文摘We used simultaneous measurements of surface PM_(2.5) concentration and vertical profiles of aerosol concentration,temperature, and humidity, together with regional air quality model simulations, to study an episode of aerosol pollution in Beijing from 15 to 19 November 2016. The potential effects of easterly and southerly winds on the surface concentrations and vertical profiles of the PM_(2.5) pollution were investigated. Favorable easterly winds produced strong upward motion and were able to transport the PM_(2.5) pollution at the surface to the upper levels of the atmosphere. The amount of surface PM_(2.5) pollution transported by the easterly winds was determined by the strength and height of the upward motion produced by the easterly winds and the initial height of the upward wind. A greater amount of PM_(2.5) pollution was transported to upper levels of the atmosphere by upward winds with a lower initial height. The pollutants were diluted by easterly winds from clean ocean air masses. The inversion layer was destroyed by the easterly winds and the surface pollutants and warm air masses were then lifted to the upper levels of the atmosphere, where they re-established a multi-layer inversion. This region of inversion was strengthened by the southerly winds, increasing the severity of pollution. A vortex was produced by southerly winds that led to the convergence of air along the Taihang Mountains. Pollutants were transported from southern–central Hebei Province to Beijing in the boundary layer. Warm advection associated with the southerly winds intensified the inversion produced by the easterly winds and a more stable boundary layer was formed. The layer with high PM_(2.5) concentration became dee-per with persistent southerly winds of a certain depth. | 13 | Investigation of hrDNA targeting vector-mediated tumor-specific suicide gene therapy for hepatocellular carcinoma显示文摘Human ribosomal DNA (hrDNA) target- ing vector (pHrn) is one of the human derived vectors, which was devised by our lab and has got patent authority. To investigate its effect on gene therapy for hepatocellular carcinoma, a double suicide fusion gene expression cassette, CDUPRT/GFP controlled by a synthetic CMV enhancer-enhanced hTERT promoter (CeTp) which was determined by luciferase assays was constructed in pHrn backbone, creating an expression vector pHr-CeTpCDUPRT/GFP. After transfer of plasmid to hepatocellular carcinoma cell line Bel7402 in vitro, the transfection efficiency reached 30%―50% by using flow cytometer. The expression of CDUPRT/GFP was detected by RT-PCR and Western Blotting. After the administra- tion of 5-FC, high performance liquid chromatography (HPLC) was applied to examine the level of 5-FU in supernatant, resulting in a concentration of 60.15 μg/mL. The Methylthiazolyl tetrazolium (MTT) assay was then utilized to investigate the antitumor effect of pHr-CeTpCDUPRT/GFP in Bel7402 cells, and rela- tive cell survival of 60%-35% was observed after 5-FC treatment. In vivo experiments, the nude mouse model of hepatocellular carcinoma was constructed and in situ gene therapy was performed. The results indicated the tumor growth of treatment group was obviously suppressed, and some even shrank, whenthe vectors and prodrugs were injected continuously. The expression of CDUPRT in tumor tissues was also identified by RT-PCR, and the concentration of 5-FU was 7.694 μg/mL in blood serum using HPLC detection. Then the pathological section of tumor tissues revealed significant tumor cell necrosis. | 18 | The direct and gut microbiota-mediated effects of dietary bile acids on the improvement of gut barriers in largemouth bass(Micropterus salmoides)显示文摘Fish gut barrier damage under intensive culture model is a significant concern for aquaculture industry.This study aimed to investigate the effects of bile acids(BAs)on gut barriers in Micropterus salmoides.A germ-free(GF)zebrafish model was employed to elucidate the effects of the direct stimulation of BAs and the indirect regulations mediated by the gut microbiota on gut barrier functions.Four diets were formulated with BAs supplemented at 0,150,300 and 450 mg/kg,and these 4 diets were defined as control,BA150,BA300 and BA450,respectively.After 5 weeks of feeding experiment,the survival rate of fish fed with BA300 diet was increased(P<0.05).Histological analysis revealed an improvement of gut structural integrity in the BA150 and BA300 groups.Compared with the control group,the expression of genes related to chemical barrier(mucin,lysozyme and complement 1)and physical barrier(occludin and claudin-4)was increased in the BA150 and BA300 groups(P<0.05),and the expression of genes related to immunological barrier(interleukin[IL]-6,tumor growth factorβ,IL-10,macrophage galactosetype lectin and immunoglobulin M[Ig M])was significantly increased in the BA300 group(P<0.05),but the expression of genes related to chemical barrier(hepcidin)and immunological barrier(IL-1β,tumor necrosis factor-a,IL-6 and arginase)was significantly decreased in the BA450 group(P<0.05).Gut microbiota composition analysis revealed that the abundance of Firmicutes was augmented prominently in the BA150 and BA300 groups(P<0.05),while that of Actinobacteriota and Proteobacteria showed a downward trend in the BA150 and BA300 groups(P>0.05).The results of the gut microbiota transferring experiment demonstrated an upregulation of gut barrier-related genes,including immunoglobulin Z/T(Ig Z/T),IL-6,IL-1βand IL-10,by the gut microbiota transferred from the BA300 group compared with the control(P<0.05).Feeding the BA300 diet directly to GF zebrafish resulted in enhanced expression of Ig M,Ig Z/T,lysozyme,occludin-2,IL-6 and IL-10(P<0.05).In conclusion,BAs can improve the gut barriers of fish through both direct and indirect effects mediated by the gut microbiota. | Rui Xia Qingshuang Zhang Dongmei Xia Qiang Hao Qianwen Ding Chao Ran Yalin Yang Aizhi Cao Zhen Zhang Zhigang Zhou | 2023 | Animal Nutrition2023,,3: | 2 |

| 20 | Silica nanoparticle is a possible safe carrier for gene therapy显示文摘In order to develop a safe and effective gene therapy carrier, some toxicological and biodynamical ex- periments were carried out on silica nanoparticles (SiNPs). First we prepared SiNPs with appropriate portions of cyclo- hexane, deionized water and ethyl silicate, and then trans- fected the modified SiNPs and GFP plasmid DNA complex into the HT1080 cells to test the effectiveness of transfection for gene therapy. At the same time, we injected the SiNPs into a number of mice through tail vein. Then we made the mice crossed to evaluate the acute, long-term and reproduc- tive toxicity. In vivo distribution analysis and pathological examination were made on both adult mice and their off- spring. SiNPs were uniform and had an average diameter of 40 nm, and the modified SiNPs carried exogenous DNA molecules into target cells and the transferred GFP fusion gene was effectively expressed in the cells. The SiNPs injected via tail vein were widely distributed in almost all of tissues, and the injected mice had the ability to reproduce normally.
