| 1 | Inhibition of hepatitis B virus replication by quercetin in human hepatoma cell lines显示文摘Hepatitis B virus(HBV) infection is one of the most serious and prevalent viral diseases in the world. Although several anti-HBV drugs have been used clinically, their side and adverse effects limit treatment efficacy. Therefore, it is necessary to identify novel potential anti-HBV agents. The flavonol quercetin has shown activity against some retroviruses, but its effect on HBV remains unclear. In the present study, quercetin was incubated with Hep G2.2.15 cells, as well as Hu H-7 cells transfected with an HBV plasmid. Quercetin was shown to significantly reduce Hepatitis B surface antigen(HBs Ag) and Hepatitis B e antigen(HBe Ag), secretion and HBV genomic DNA levels in both cell lines. In addition, co-incubation with lamivudine(3TC), entecavir(ETV), or adefovir(Ade) further enhanced the quercetin-induced inhibition of HBV replication. This inhibition was partially associated with decreased heat shock proteins and HBV transcription levels. | 5 | The deep structure of the Duanqiao hydrothermal field at the Southwest Indian Ridge显示文摘Polymetalic sulfide is the main product of sea-floor hydrothermal venting, and has become an important sea-floor mineral resources for its rich in many kinds of precious metal elements. Since 2007, a number of investigations have been carried out by the China Ocean Mineral Resources Research and Development Association(COMRA)cruises(CCCs) along the Southwest Indian Ridge(SWIR). In 2011, the COMRA signed an exploration contract of sea-floor polymetallic sulfides of 10 000 km2 on the SWIR with the International Seabed Authority. Based on the multibeam data and shipborne gravity data obtained in 2010 by the R/V Dayang Yihao during the leg 6 of CCCs21, together with the global satellite surveys, the characteristics of gravity anomalies are analyzed in the Duanqiao hydrothermal field(37°39′S, 50°24′E). The 'subarea calibration' terrain-correcting method is employed to calculate the Bouguer gravity anomaly, and the ocean bottom seismometer(OBS) profile is used to constrain the two-dimensional gravity anomaly simulation. The absent Moho in a previous seismic model is also calculated.The results show that the crustal thickness varies between 3 and 10 km along the profile, and the maximum crustal thickness reaches up to 10 km in the Duanqiao hydrothermal field with an average of 7.5 km. It is by far the most thicker crust discovered along the SWIR. | 6 | GCN5 Acetyltransferase Inhibits PGC1α-induced Hepatitis B Virus Biosynthesis显示文摘Hepatitis B virus (HBV) biosynthesis is primarily restricted to hepatocytes due to the governing of liver-enriched nuclear receptors (NRs) on viral RNA synthesis. The liver-enriched NR hepatocyte nuclear factor 4α (HNF4α), the key regulator of genes implicated in hepatic glucose metabolism, is also a primary determinant of HBV pregenomic RNA synthesis and HBV replication. Peroxisome proliferator-activated receptor-γ coactivator 1α (PGC1α) coactivates and further enhances the effect of HNF4α on HBV biosynthesis. Here, we showed that the acetyltransferase General Control Non-repressed Protein 5 (GCN5) acetylated PGC1α, leading to alteration of PGC1α from a transcriptionally active state into an inactive state. As a result, the coactivation activity of PGC1α on HBV transcription and replication was suppressed. Apparently, an acetylation site mutant of PGC1α (PGC1αR13) still had coactivation activity as GCN5 could not suppress the coactivation activity of the mutant. Moreover, a catalytically inactive acetyltransferase mutant GCN5m, due to the loss of acetylation activity, failed to inhibit the coactivation function of PGC1α in HBV biosynthesis. | 20 | Effects of slurry viscosity and particle additive size on filter cake formation in highly permeable sand显示文摘Filter cake is critical to maintaining the stability of the excavation face of an underwater shield tunnel in a high-permeability stratum.In a high-permeability formation,generating an effective filter cake on the excavation face is difficult with a pure bentonite slurry,which penetrates the ground and may not achieve the required suspension pressure.Determining how to efficiently and quickly form a thin and low-permeability filter cake on the tunnel working face has become a key engineering problem in the construction of slurry shield tunnels in high-permeability strata.In this study,the relationship between slurry viscosity and the slurry pressure gradient of pure bentonite was established by performing slurry permeability experiments.The influence of slurry viscosity on the formation of the filter cake in a high-permeability formation was studied under different pressure gradients.In addition,the effect of additive particle size on the slurry filter cake formation was analyzed by introducing additives with different particle sizes to pure bentonite slurries with different viscosities.The test results indicate that(1)for the pure bentonite slurry,the critical initial pressure gradient can be used as a rough indicator of the formation of the filter cake,and the relationship between the critical maximum pore diameter and the average pore diameter of the for-mation can be compared to establish and analyze the formation law of the slurry filter cake;(2)adding particles to the slurry can enhance the effect of the pure bentonite slurry;and(3)adding coarse-grained materials can effectively improve the film-forming effect.The slurry is more compact when the particle size is close to the average pore size of the formation. | Zhikui Wang Yuan Wang Di Feng Jinhui Zhang Sijin Liu | 2022 | Underground Space2022,7,2: | 1 |
