1Role of PLC-PIP2 and cAMP-PKA Signal Pathways in Radiation-induced Immune-suppressing Effect显示文摘Objective The purpose of the present study was to observe the changes in CD4+CD25+Nrp1+Treg cells after irradiation with different doses and explore the possible molecular mechanisms involved. Methods ICR mice and mouse lymphoma cell line(EL-4 cells) was used. The expressions of CD4,CD25,Nrp1,calcineurin and PKC-α were detected by flow cytometry. The expressions of TGF-β1,IL-10,PKA and cAMP were estimated with ELISA. Results At 12 h after irradiation,the expression of Nrp1 increased significantly in 4.0 Gy group,compared with sham-irradiation group(P<0.05) in the spleen and thymus,respectively,when ICR mice received whole-body irradiation(WBI). Meanwhile the synthesis of Interleukin 10(IL-10) and transforming growth factor-β1(TGF-β1) increased significantly after high dose irradiation(HDR)(> or = 1.0 Gy). In addition,the expression of cAMP and PKA protein increased,while PKC-α,calcineurin decreased at 12h in thymus cells after 4.0 Gy X-irradiation. While TGF-β1 was clearly inhibited when the PLC-PIP2 signal pathway was stimulated or the cAMP-PKA signal pathway was blocked after 4.0 Gy X-irradiation,this did not limit the up-regulation of CD4+CD25+Nrp1+Treg cells after ionizing radiation. 2A novel synthetic route for LiFePO_4/C cathode materials by addition of starch for lithium-ion batteries显示文摘LiFePO4/Carbon composite cathode material was prepared using starch as carbon source by spray-pelleting and subsequent pyrolysis in N2. The samples were characterized by XRD, SEM, Raman, and their electrochemical performance was investigated in terms of cycling behavior. There has a special micro-morphology via the process, which is favorable to electrochemical properties. The discharge capacity of the LiFePO4.C composite was 170 mAh g-1, equal to the theoretical specific capacity at 0.1 C rate. 3Exposure Effects of Terahertz Waves on Primary Neurons and Neuron-like Cells Under Nonthermal Conditions显示文摘Objective This study aimed to explore the potential effects of terahertz(THz) waves on primary cultured neurons from 4 rat brain regions(hippocampus, cerebral cortex, cerebellum, and brainstem)and 3 kinds of neuron-like cells(MN9 D, PC12, and HT22 cells) under nonthermal conditions.Methods THz waves with an output power of 50(0.16 THz) and 10(0.17 THz) m W with exposure times of 6 and 60 min were used in this study. Analysis of temperature change, neurite growth, cell membrane roughness, micromorphology, neurotransmitters and synaptic-related proteins(SYN and PSD95) was used to evaluate the potential effects.Results Temperature increase caused by the THz wave was negligible. THz waves induced significant neurotransmitter changes in primary hippocampal, cerebellar, and brainstem neurons and in MN9 D and PC12 cells. THz wave downregulated SYN expression in primary hippocampal neurons and downregulated PSD95 expression in primary cortical neurons.Conclusion Different types of cells responded differently after THz wave exposure, and primary hippocampal and cortical neurons and MN9 D cells were relatively sensitive to the THz waves. 4A novel approach for designing efficient broadband photodetectors expanding from deep ultraviolet to near infrared显示文摘Broadband photodetection(PD)covering the deep ultraviolet to near-infrared(200–1000 nm)range is significant and desirable for various optoelectronic designs.Herein,we employ ultraviolet(UV)luminescent concentrators(LC),iodinebased perovskite quantum dots(PQDs),and organic bulk heterojunction(BHJ)as the UV,visible,and near-infrared(NIR)photosensitive layers,respectively,to construct a broadband heterojunction PD.Firstly,experimental and theoretical results reveal that optoelectronic properties and stability of CsPbI3 PQDs are significantly improved through Er3+doping,owing to the reduced defect density,improved charge mobility,increased formation energy,tolerance factor,etc.The narrow bandgap of CsPbI3:Er3+PQDs serves as a visible photosensitive layer of PD.Secondly,considering the matchable energy bandgap,the BHJ(BTP-4Cl:PBDB-TF)is selected as to NIR absorption layer to fabricate the hybrid structure with CsPbI3:Er3+PQDs.Thirdly,UV LC converts the UV light(200–400 nm)to visible light(400–700 nm),which is further absorbed by CsPbI3:Er3+PQDs.In contrast with other perovskites PDs and commercial Si PDs,our PD presents a relatively wide response range and high detectivity especially in UV and NIR regions(two orders of magnitude increase that of commercial Si PDs).Furthermore,the PD also demonstrates significantly enhanced air-and UV-stability,and the photocurrent of the device maintains 81.5%of the original one after 5000 cycles.This work highlights a new attempt for designing broadband PDs,which has application potential in optoelectronic devices.Nan Ding Yanjie Wu Wen Xu Jiekai Lyu Yue Wang Lu Zi Long Shao Rui Sun Nan Wang Sen Liu Donglei Zhou Xue Bai Ji Zhou Hongwei Song 2022Light(Science & Applications)2022,11,5:5
5Interferon-induced Transmembrane Protein 3 Prevents Acute Influenza Pathogenesis in Mice显示文摘Objective Interferon-induced transmembrane protein 3(IFITM3)is an important member of the IFITM family.However,the molecular mechanisms underlying its antiviral action have not been completely elucidated.Recent studies on IFITM3,particularly those focused on innate antiviral defense mechanisms,have shown that IFITM3 affects the body’s adaptive immune response.The aim of this study was to determine the contribution of IFITM3 proteins to immune control of influenza infection in vivo.Methods We performed proteomics,flow cytometry,and immunohistochemistry analysis and used bioinformatics tools to systematically compare and analyze the differences in natural killer(NK)cell numbers,their activation,and their immune function in the lungs of Ifitm3-/-and wild-type mice.Results Ifitm3-/-mice developed more severe inflammation and apoptotic responses compared to wild-type mice.Moreover,the NK cell activation was higher in the lungs of Ifitm3-/-mice during acute influenza infection.Conclusions Based on our results,we speculate that the NK cells are more readily activated in the absence of IFITM3,increasing mortality in Ifitm3-/-mice.SUN Qiang LEI Na LU Jian GAO Rong Bao LI Zi LIU Li Qi SUN Ying GUO Jun Feng WANG Da Yan SHU Yue Long 2020Biomedical and Environmental Sciences2020,33,5:4

9An Investigation on the Molecular Characteristics and Intracellular Growth Ability among Environmental and Clinical Isolates of Legionella pneumophila in Sichuan Province, China显示文摘Objective To investigate the molecular characteristics and intracellular growth ability of Legionella pneumophila(L. pneumophila) strains from 1989 to 2016 in Sichuan Province, China. Methods Seventy-nine isolates of L. pneumophila were collected from environmental and clinical sources, including cooling towers, hot springs, bath water, fountains, and patients, and identified with 16 S rRNA gene analysis and serum agglutination assay. The isolates were then typed by Sequence-Based Typing(SBT), and Genotyping of forty-two LP1 strains were analyzed by means of multiple-locus VNTR analysis with 8 loci(MLVA-8). All strains were further analyzed for two virulence genes: Legionella vir homologue(lvh) and repeats in structural toxin(rtx A). The intracellular growth ability of 33 selected isolates was determined by examining their interaction with J774 cells. Results All isolates were identified to L. pneumophila including 11 serogroups, among which the main serogroup were LP1, accounting for 54.43%. Thirty-three different sequence types(STs) from five main clonal groups and five singletons were identified, along with 8 different MLVA patterns. Both the lvh and rtx A loci were found in all 79 strains. Thirty isolates showed high intracellular growth ability in J774 cells.
