4High-throughput experiments facilitate materials innovation:A review显示文摘Since the Material Genome Initiative(MGI) was proposed, high-throughput based technology has been widely employed in various fields of materials science. As a theoretical guide, material informatics has been introduced based on machine learning and data mining and high-throughput computation has been employed for large scale search, narrowing down the scope of the experiment trials. High-throughput materials experiments including synthesis, processing, and characterization technologies have become valuable research tools to pin down the prediction experimentally, enabling the discovery-to-deployment of advances materials more efficiently at a fraction of cost. This review aims to summarize the recent advances of high-throughput materials experiments and introduce briefly the development of materials design based on material genome concept. By selecting representative and classic works in the past years, various high-throughput preparation methods are introduced for different types of material gradient libraries, including metallic, inorganic materials, and polymers. Furthermore, high-throughput characterization approaches are comprehensively discussed, including both their advantages and limitations. 5Effects of Starving and Re-feeding Strategies on the Growth Performance and Physiological Characteristics of the Juvenile Tongue Sole(Cynoglossus semilaevis)显示文摘Starving and re-feeding trials were conducted to evaluate the effects of starvation duration and recovery times on the growth performance and physiological characteristics of juvenile tongue sole Cynoglossus semilaevis, which included growth, body composition, intestinal morphology and digestive enzymes activities. The fish starved for one(D1), two(D2) and four(D4) days, respectively, were recovered for three(R3, R6, R12), seven(R7, R14, R28) and eleven(R11, R22, R44) folds of starvation days while the control fish were fed constantly. The experiment was conducted for 96 days. The fish individuals in D1R11, D2R14 and D2R22 caught up in weight with the control, indicating that complete compensatory growth existed in these fish individuals. The highest body weight gain and total specific growth rates were observed in D2R22. Food intake increased significantly in most treatments in comparison with that of the control except for D1R7 and D2R22(P < 0.05). Food conversion efficiency and apparent digest coefficient in D2R22 were significantly promoted, being higher than that of the control(P < 0.05). The activities of trypsin and lipase were found to be closely related to the growth performance. The activities of liver and hindgut trypsin, also midgut and hindgut lipase in D2R22 were significantly higher than those of the control(P < 0.05). The fold height(HF) of foregut and midgut increased significantly in D2R22, and HF decreased significantly in D1R3, D2R6 and D4R12(P < 0.05). On our findings, we may conclude that the optimum starving and re-feeding strategy is starving for 2 days and re-feeding for 22 days. 6A biomimetic and bioactive scaffold with intelligently pulsatile teriparatide delivery for local and systemic osteoporosis regeneration显示文摘Osteoporosis is one of the most disabling consequences of aging,osteoporotic fractures and higher risk of the subsequent fractures leading to substantial disability and deaths,indicating both local fractures healing and the early anti-osteoporosis therapy are of great significance.Teriparatide is strong bone formation promoter effective in treating osteoporosis,while side effects limit clinical applications.Traditional drug delivery is lack of sensitive and short-term release,finding a new non-invasive and easily controllable drug delivery to not only repair the local fractures but also improve total bone mass has remained a great challenge.Thus,bioinspired by the natural bone components,we develop appropriate interactions between inorganic biological scaffolds and organic drug molecules,achieving both loaded with the teriparatide in the scaffold and capable of releasing on demand.Herein,biomimetic bone microstructure of mesoporous bioglass,a near-infrared ray triggered switch,thermosensitive liposomes based on a valve,and polydopamine coated as a heater is developed rationally for osteoporotic bone regeneration.Teriparatide is pulsatile released from intelligent delivery,not only rejuvenating osteoporotic bone defect,but also presenting strong systemic anti-osteoporosis therapy.This biomimetic bone carrying novel drug delivery platform is well worth expecting to be a new promising strategy and clinically commercialized to help patients survive from the osteoporotic fracture.Lingbin Che Ying Wang Dongyong Sha Guangyi Li Ziheng Wei Changsheng Liu Yuan Yuan Dianwen Song 2023Bioactive Materials2023,,1:3

9ContractGuard:面向以太坊区块链智能合约的入侵检测系统显示文摘以太坊智能合约本质上是一种在网络上由相互间没有信任关系的节点共同执行的已被双方认证程序。目前,大量的智能合约被用于管理数字资产,使智能合约成为黑客的重要攻击对象。常见的攻击方法是通过利用智能合约的漏洞来实现特定操作的入侵攻击。ContractGuard是首次提出面向以太坊区块链智能合约的入侵检测系统,它能检测智能合约的潜在攻击行为。ContractGuard的入侵检测主要依赖检测潜在攻击可能引发的异常控制流来实现。由于智能合约运行在去中心化的环境以及在高度受限的环境中运行,现有的IDS技术或者工具等以外部拦截形式的部署架构不适合于以太坊智能合约。为了解决这些问题,通过设计一个嵌入式的架构,实现了把ContractGuard直接嵌入智能合约的执行代码中,作为智能合约的一部分。在运行时刻,ContractGuard通过相应的context-tagged无环路径来实现入侵检测,从而保护智能合约。由于嵌入了额外的代码,ContractGuard一定程度上会增加智能合约的部署开销与运行开销,为了降低这两方面的开销,基于以太坊智能合约的特性对ContractGuard进行优化。实验结果显示,可有效地检测83%的异常行为,其部署开销仅增加了36.14%,运行开销仅增加了28.17%。赵淦森 谢智健 王欣明 何嘉浩 张成志 林成创 Ziheng Zhou 陈冰川 Chunming Rong 2020网络与信息安全学报2020,6,2:2

11Comparison of algorithms for monitoring wheat powdery mildew using multi-angular remote sensing data显示文摘Powdery mildew is a disease that threatens wheat production and causes severe economic losses worldwide. Its timely diagnosis is imperative for preventing and controlling its spread. In this study, the multiangle canopy spectra and disease severity of wheat were investigated at several developmental stages and degrees of disease severity. Four wavelength variable-selected algorithms: successive projection(SPA), competitive adaptive reweighted sampling(CARS), feature selection learning(Relief-F), and genetic algorithm(GA), were used to identify bands sensitive to powdery mildew. The wavelength variables selected were used as input variables for partial least squares(PLS), extreme learning machine(ELM), random forest(RF), and support vector machine(SVM) algorithms, to construct a suitable prediction model for powdery mildew. Spectral reflectance and conventional vegetation indices(VIs) displayed angle effects under several disease severity indices(DIs). The CARS method selected relatively few wavelength variables and showed a relatively homogeneous distribution across the 13 viewing zenith angles.Overall accuracies of the four modeling algorithms were ranked as follows: ELM(0.70–0.82) > PLS(0.63–0.79) > SVM(0.49–0.69) > RF(0.43–0.69). Combinations of features and algorithms generated varied accuracies, with coefficients of determination(R^(2)) single-peaked at different observation angles. The constructed CARS-ELM model extracted a predictable bivariate relationship between the multi-angle canopy spectrum and disease severity, yielding an R^(2)> 0.8 at each measured angle. Especially for larger angles,monitoring accuracies were increased relative to the optimal VI model(40% at-60°, 33% at +60°), indicating that the CARS-ELM model is suitable for extreme angles of-60° and +60°. 13Synchronous conjugation of i-motif DNA and therapeutic siRNA on the vertexes of tetrahedral DNA nanocages for efficient gene silence显示文摘The functionality of DNA biomacromolecules has been widely excavated,as therapeutic drugs,carriers,and functionalized modification derivatives.In this study,we developed a series of DNA tetrahedron nanocages(Td),via synchronous conjugating different numbers of i-(X)and therapeutic siRNA on four vertexes of tetrahedral DNA nanocage(aX-Td@bsiRNA,a+b=4).This i-motif-conjugated Td exhibited good endosomal escape behaviours in A549 tumor cells,and the escape efficiency was affected by the number of i-motif.Furthermore,the downregulating mRNA and protein expression level of epidermal growth factor receptor(EGFR)caused by this siRNA embedded Td were verified in A549 cells.The tumor growth inhibition efficiency of the 2X-Td@2siRNA treated group in tumorbearing mice was significantly higher than that of non-i-motif-conjugated Td@2siRNA(3.14-fold)and free siRNA(3.63-fold).These results demonstrate a general strategy for endowing DNA nanostructures with endosomal escape behaviours to achieve effective in vivo gene delivery and therapy.Xiu Han Xiang Xu Ziheng Wu Zhenghong Wu Xiaole Qi 2021Acta Pharmaceutica Sinica B2021,11,10:2

15Profiles of immune status and related pathways in sepsis: evidencebased on GEO and bioinformatics显示文摘Sepsis,characterized as life-threatening sequential organ failure,is caused by a dysregulated host immune response to a pathogen.Conventional practice for sepsis is to control the inflammation source and administer highgrade antibiotics.However,the mortality rate of sepsis varies from 25–30%and can reach 50%if a septic shock occurs.In our current study,we used bioinformatics technology to detect immune status profiles in sepsis at the genomic level.We downloaded and analyzed gene expression profiles of GSE28750 from the Gene Expression Omnibus(GEO)database to determine differential gene expression and immune status between sepsis and normal samples.Next,we used the CIBERSORT method to quantify the proportions of immune cells in the sepsis samples.Then we explored the differentially expressed genes(DEGs)related to sepsis.Furthermore,gene ontology(GO)function and the Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis were used to present potential signaling pathways in sepsis.We found that in the sepsis samples,the CD8+T cell fraction was consistently lower,based on the CIBERSORT method,whereas the neutrophil fraction was significantly higher in the sepsis samples.The GO function and KEGG pathway enrichment analysis identified 1573 DEGs that were significantly associated with neutrophil activation,neutrophil degranulation,neutrophil activation involved in the immune response,neutrophil-mediated immunity,and T cell activation in the biological processes group.In our study,we provided a first glance of associations between immune status and sepsis.Furthermore,our data regarding the reciprocal interaction between immune cells(neutrophils and CD8+T cells)could improve our understanding of immune status profiles in sepsis.However,additional investigations should be performed to verify their clinical value.HANYU SHEN SHIQI REN WEI WANG CHENLIN ZHANG HAIYAN HAO QIUYAN SHEN YINONG DUAN ZIHENG WANG WENLIANG GE 2020BIOCELL2020,44,4:1
16Increasing the heterologous production of spinosad in Streptomyces albus J1074 by regulating biosynthesis of its polyketide skeleton显示文摘Spinosyns are natural broad-spectrum biological insecticides with a double glycosylated polyketide structure that are produced by aerobic fermentation of the actinomycete,Saccharopolyspora spinosa.However,their large-scale overproduction is hindered by poorly understood bottlenecks in optimizing the original strain,and poor adaptability of the heterologous strain to the production of spinosyn.In this study,we genetically engineered heterologous spinosyn-producer Streptomyces albus J1074 and optimized the fermentation to improve the production of spinosad(spinosyn A and spinosyn D)based on our previous work.We systematically investigated the result of overexpressing polyketide synthase genes(spnA,B,C,D,E)using a constitutive promoter on the spinosad titer in S.albus J1074.The supply of polyketide synthase precursors was then increased to further improve spinosad production.Finally,increasing or replacing the carbon source of the culture medium resulted in a final spinosad titer of~70 mg/L,which is the highest titer of spinosad achieved in heterologous Streptomyces species.This research provides useful strategies for efficient heterologous production of natural products.Ziheng An Hui Tao Yong Wang Bingqing Xia Yang Zou Shuai Fu Fang Fang Xiao Sun Renqiong Huang Yao Xia Zixin Deng Ran Liu Tiangang Liu 2021Synthetic and Systems Biotechnology2021,6,4:1
