2Arginine bi-directional translocation and breakdown into ornithine along the arbuscular mycorrhizal mycelium显示文摘Bi-directional translocation and degradation of Arginine (Arg) along the arbuscular mycorrhizal (AM) fungal mycelium were testified through 15N and/or 13C isotopic labeling. In vitro mycorrhizas of Glomus intraradices and Ri T-DNA-transformed carrot roots were grown in dual compartment Petri dishes. [15N- and/or13C]Arg was supplied to either the fungal compartment or the mycorrhizal compartment or separate dishes containing the uncolonized roots. The levels and labeling of free amino acids (AAs) in the mycorrhizal roots and in the extraradical mycelia(ERM) were measured by gas chromatogra- phy/mass spectrometry (GC-MS) and high-performance liquid chromatography (HPLC). The ERM of AM fungi exposed in either NH4+ or urea as sole external nitrogen source had much higher 15N enrichment of Arg, compared with those in nitrate or exogenous Arg; however, glycerol supplied as an external car- bon source to the ERM had no significant effect on the level of Arg in the ERM. Meanwhile, Arg bio- synthesized in the ERM could be translocated intact to the mycorrhizal roots and thereby the level of Arg in the mycorrhizal roots increased to about 20% after culture of ERM in 4 mmol/L NH4+ for 6 weeks. Also Arg was found to be bi-directionally transported along the AM fungal mycelium through [U-13C]Arg labeling either in the mycorrhizal compartment or in the fungal compartment. Once Arg was translo- cated to the potential N-limited sites, it would be further degraded into ornithine (Orn) and urea since either [U-13C] or [U-15N/U-13C]Orn was apparently shown up in the mycorrhizal root tissues when [U-13C] or [U-15N/U-13C]Arg was labeled in the fungal compartment, respectively. 3Arbuscular mycorrhizal fungi ameliorate the chemical properties and enzyme activities of rhizosphere soil in reclaimed mining subsidence in northwestern China显示文摘In semi-arid region of northwestern China, underground mining subsidence often results in decreased vegetation coverage, impoverishment of soil fertility and water stress. In addition, the physical-chemical and biological properties of soil also change, resulting in more susceptible to degradation. In particular, subsidence causes disturbance of the symbioses of plant and microbe that can play a beneficial role in the establishment of vegetation communities in degraded ecosystems. The objective of this study was to evaluate the effects of revegetation with exotic arbuscular mycorrhizal fungi(AMF) inoculum on the chemical and biological properties of soil over time in mining subsidence areas. Soils were sampled at a depth up to 30 cm in the adjacent rhizosphere of Amorpha fruticose Linn. from five reclaimed vegetation communities in northwestern China. In August 2015, a field trial was set up with five historical revegetation experiments established in 2008(7-year), 2011(4-year), 2012(3-year), 2013(2-year) and 2014(1-year), respectively. Each reclamation experiment included two treatments, i.e., revegetation with exotic AMF inoculum(AMF) and non-AMF inoculum(the control). Root mycorrhizal colonization, glomalin-related soil protein(GRSP), soil organic carbon(SOC), soil nutrients, and enzyme activities were also assessed. The results showed that mycorrhizal colonization of inoculated plants increased by 33.3%–163.0% compared to that of non-inoculated plants(P<0.05). Revegetation with exotic AMF inoculum also significantly improved total GRSR(T-GRSP) and easily extracted GRSP(EE-GRSP) concentrations compared to control, besides the T-GRSP in 1-year experiment and the EE-GRSP in 2-year experiment. A significant increase in SOC content was only observed in 7-year AMF reclaimed soils compared to non-AMF reclaimed soils. Soil total N(TN), Olsen phosphorus(P) and available potassium(K) were significantly higher in inoculated soil after 1–7 years of reclamation(except for individual cases), and increased with reclamation time(besides soil Olsen P). The exotic AMF inoculum markedly increased the average soil invertase, catalase, urease and alkaline phosphatase by 23.8%, 21.3%, 18.8% and 8.6%, respectively(P<0.01), compared with the control. Root mycorrhizal colonization was positively correlated with soil parameters(SOC, TN and soil available K) and soil enzyme activities(soil invertase, catalase, urease and alkaline phosphatase) in both AMF and non-AMF reclaimed soils(P<0.05), excluding availableK in non-AMF reclaimed soils. T-GRSP(P<0.01) and EE-GRSP(P<0.05) were significantly correlated with the majority of edaphic factors, except for soil Olsen P. The positive correlation between root mycorrhizal colonization and available K was observed in AMF reclaimed soils, indicating that the AMF reclaimed soil with a high root mycorrhizal colonization could potentially accumulate available K in soils. 4Arbuscular mycorrhizal fungi combined with exogenous calcium improves the growth of peanut(Arachis hypogaea L.)seedlings under continuous cropping显示文摘The growth and yield of peanut are negatively affected by continuous cropping.Arbuscular mycorrhizal fungi(AMF)and calcium ions(Ca^(2+))have been used to improve stress resistance in other plants,but little is known about their roles in peanut seedling growth under continuous cropping.This study investigated the possible roles of the AMF Glomus mosseae combined with exogenous Ca^(2+)in improving the physiological responses of peanut seedlings under continuous cropping.G.mosseae combined with exogenous Ca^(2+)can enhance plant biomass,Ca^(2+)level,and total chlorophyll content.Under exogenous Ca^(2+)application,the F_v/F_m in arbuscular mycorrhizal(AM)plant leaves was higher than that in the control plants when they were exposed to high irradiance levels.The peroxidase,superoxide dismutase,and catalase activities in AM plant leaves also reached their maximums,and accordingly,the malondialdehyde content was the lowest compared to other treatments.Additionally,root activity,and content of total phenolics and flavonoids were significantly increased in AM plant roots treated by Ca^(2+)compared to either G.mosseae inoculation or Ca^(2+)treatment alone.Transcription levels of AhCaM,AhCDPK,AhRAM1,and AhRAM2 were significantly improved in AM plant roots under exogenous Ca^(2+)treatment.This implied that exogenous Ca^(2+)might be involved in the regulation of G.mosseae colonization of peanut plants,and in turn,AM symbiosis might activate the Ca^(2+)signal transduction pathway.The combination of AMF and Ca^(2+)benefitted plant growth and development under continuous cropping,suggesting that it is a promising method to cope with the stress caused by continuous cropping.CUI Li GUO Feng ZHANG Jia-lei YANG Sha MENG Jing-jing GENG Yun WANG Quan LI Xin-guo WAN Shu-bo 2019Journal of Integrative Agriculture2019,18,2:9
5Metal concentrations and mycorrhizal status of plants colonizing copper mine tailings: potential for revegetation显示文摘A field survey of metal concentrations and mycorrhizal status of plants growing on copper mine tailings was conducted in Anhui Province, China. Available phosphorus and organic matter in the tailings were very low. High concentrations of Pb, Zn, As and Cd as well as Cu were observed on some sites. The dominant plants growing on mine tailings belonged to the families Gramineae and Compositae, and the most widely distributed plant species were Imperata cylindrica, Cynodon dactylon and Paspalum distichum. Coreopsis drummondii also grew well on the arid sites but not on wet sites. Very low or zero arbuscular mycorrhizal (AM) fungal colonization was observed in most of the plants, but extensive mycorrhizal colonization was recorded in the roots of C. drummondii and C. dactylon. Metal concentrations in plant tissues indicated that l.cylindrica and P. distichum utilized avoidance mechanisms to survive at high metal concentrations. The investigation suggests that remediation and revegetation of heavy metal contaminated sites might be facilitated by selection of tolerant plant species. 6A LysM Receptor Heteromer Mediates Perception of Arbuscular Mycorrhizal Symbiotic Signal in Rice显示文摘Symbiotic microorganisms improve nutrient uptake by plants.To initiate mutualistic symbiosis with arbus-cular mycorrhizal(AM)fungi,plants perceive Myc factors,including lipochitooligosaccharides(LCOs)and short-chain chitooligosaccharides(CO4/CO5),secreted by AM fungi.However,the molecular mechanism of Myc factor perception remains elusive.In this study,we identified a heteromer of LysM receptor-like kinases consisting of OsMYR1/OsLYK2 and OsCERK1 that mediates the perception of AM fungi in rice.CO4 directly binds to OsMYR1,promoting the dimerization and phosphorylation of this receptor complex.Compared with control plants,Osmyr1 and Oscerk1 mutant rice plants are less sensitive to Myc factors and show decreased AM colonization.We engineered transgenic rice by expressing chimeric receptors that respectively replaced the ectodomains of OsMYR1 and OsCERK1 with those from the homologous Nod factor receptors MtNFP and MtL YK3 of Medicago truncatula.Transgenic plants displayed increased cal-cium oscillations in response to Nod factors compared with control rice.Our study provides significant mechanistic insights into AM symbiotic signal perception in rice.Expression of chimeric Nod/Myc recep-tors achieves a potentially important step toward generating cereals that host nitrogen-fixing bacteria.Jiangman He Chi Zhang Huiling Dai Huan Liu Xiaowei Zhang Jun Yang Xi Chen Yayun Zhu Dapeng Wang Xiaofeng Qi Weichao Li Zhihui Wang Guoyong An Nan YU Zuhua He Yong-Fei Wang Youli Xiao Peng Zhang Ertao Wang 2019Molecular Plant2019,12,12:9
7Rapid assessment of acid phosphatase activity in the mycorrhizosphere and in arbuscular mycorrhizal fungal hyphae显示文摘A pot experiment has been carried out under controlled conditions to study the possibility of applying the technique of in vivo staining for acid phosphatase activity on the roots of mycorrhizal plants and arbuscular mycorrhizal hyphae. The pots had 5 compartments. The central root compartment was separated from the two adjacent hyphal compartments using nylon nets of 30 μm mesh, and the two hyphal compartments were separated from the two outermost compartments with 0.45 μm membranes. Red clover was grown in the root compartment and was either inoculated with the arbuscular mycorrhizal fungus (AMF) Glomus mosseae or uninoculated. Sodium phytate was applied to all compartments. The results show that AMF can increase acid phosphatase activity of clover roots. The plant roots acquired deep red 'mycorrhizal prints'. 8Coinoculation of bioinoculants improve Acacia auriculiformis seedling growth and quality in a tropical Alfisol soil显示文摘We conducted a study to find out if arbuscular mycorrhizal(AM) fungi(Acaulospora scrobiculata,Scutellospora calospora) and phosphate solubilizing bacteria(PSB, Paenibacillus polymyxa) inoculation either individually or in combinations can improve Acacia auriculiformis seedling growth, uptake of nutrients and quality in a phosphorus deficient tropical Alfisol. The seedlings were assessed for various growth and nutrient uptake parameters after 60 days of treatment. Inoculation with P.polymyxa stimulated mycorrhizal formation. Seedling height, stem girth, taproot length, number of leaves and leaf area, plant dry matter production, nodulation, and nodular dry weight were significantly higher for seedlings that were either dual inoculated or triple inoculated compared to individual inoculation of AM fungi or PSB, and uninoculated seedlings. Dual and triple application of AM fungi and PSB also significantly improved the nutrient contents of shoots and roots and nutrient uptake efficiencies. The calculated seedling quality indexes of the AM fungi and PSB inoculated seedling were 25–208% higher than uninoculated seedlings. These findings show that A.auriculiformis seedlings when dual inoculated or triple inoculated performed better than seedlings inoculated with the microbes individually and compared with uninoculated control seedlings. 9Natural Colonization of Rice by Arbuscular Mycorrhizal Fungi in Different Production Areas显示文摘Interactions between plants and soil microorganisms can influence the other interactions in which plants participate, including interactions with herbivores. Many fungi, including arbuscular mycorrhizal fungi(AMF), form symbiotic relationships with the roots they inhabit, and potentially alter defense against pests. The objective of this study was to document the extent of root colonization by AMF on non-flooded rice plants grown under conditions typical of commercial fields. We hypothesized that AMF naturally colonized rice plants in different rice producing field locations. Rice plant samples were collected from areas across the southern United States, including Texas, Mississippi, Arkansas and two research stations in Louisiana. We quantified the amount of AMF colonization in insecticide-free rice plants over three consecutive years(2014–2016). The results revealed natural colonization of AMF in all rice producing areas. In all the three years of survey, rice-AMF associations were the greatest in Arkansas followed by Mississippi and Texas. 10Nested multiplex PCR——A feasible technique to study partial community of arbuscular mycorrhizal fungi in field-growing plant root显示文摘Plant can be infected by different arbuscular mycorrhizal fungi, but little is known about the interaction between them within root tissues mainly because different species cannot be distin-guished on the basis of fungal structure. Accurate species identification of Arbuscular mycorrhizal fungi (AMF) colonized in plant roots is the cornerstone of mycorrhizal study, yet this fundamental step is impossible through its morphological character alone. For accurate, rapid and inexpensive detec-tion of partial mycorrhizal fungal community in plant roots, a nested multiplex polymerase chain reac-tion (PCR) was developed in this study. Five discriminating primers designed based on the variable region of the 5′ end of the large ribosomal subunit were used in the experiment for testing their speci-ficity and the sensitivity in nested PCR by using spores from Glomus mosseae (BEG12), Glomus intraradices (BEG141), Scutellospora castaneae (BEG1) and two unidentified Glomus sp. HAUO3 and HAUO4. The feasibility assay of nested multiplex PCR was conducted by use of spore mixture, Astragalus sinicum roots co-inoculated with 4 species of arbuscular mycorrhizal fungi from pot cul-tures and 15 different field-growing plant roots respectively after analyses of the compatibility of primers. The result indicated that the sensitivity was in the same range as that of the corresponding single PCR reaction. Overall accuracy was 95%. 11Humus-Rich Compost Increases Lettuce Growth, Nutrient Uptake, Mycorrhizal Colonisation, and Soil Fertility显示文摘Sandy soils, typical of Australia's west, either have little or no habitat protection for microbes including arbuscular mycorrhizal(AM) fungi, which are essential for nutrient cycling. To minimize this problem, the application of organic matter, such as humus-rich composts, is necessary during vegetable crop production. This study aimed at determining the effects of humus-rich composts on either indigenous or inoculated AM fungal colonisation in roots, lettuce(Lactuca sativa L. var. Quechua) growth, and soil fertility improvement. Four different humus-rich composts with varying humus contents were applied at the same standard rate to lettuce grown under glasshouse conditions for 10 weeks after sowing and compared with two low-humus composts and non-amended soil(control). Humus-rich composts significantly increased lettuce shoot growth, root growth, and AM fungal colonisation in roots. Humus contents in the composts were also correlated with lettuce shoot and root growth. Soil dissolved organic carbon, microbial biomass carbon, and fertility were increased with the application of humus-rich composts. These humus-rich composts, especially the compost of higher humic acid with and without AM inoculation, might have a significant role in sustainable vegetable production, for example lettuce growth. 12Biodiversity of arbuscular mycorrhizal fungi in Amygdalus scoparia Spach plantations and a natural stand显示文摘The biodiversity of arbuscular mycorrhizal fungi(AMF) was surveyed in the Kolm region of Iran in three adjacent sites, a natural stand, a 10-year-old and a 15-yearold plantation of Amygdalus scoparia. To date, there have been few studies of AMF biodiversity in Iran, especially in the western forests of the country. For this study, soil and root samples were taken from A. scoparia rhizosphere soil in spring and autumn. Almost half of the root length was colonized by AMF. We identified 13 AMF species belonging to Glomeraceae, Claroideoglomeraceae or Diversisporaceae. The three plantations differed in terms of soil electrical conductivity, organic C and P. Spore density was significant correlated with P concentration. Root length colonization was correlated only with soil Ca.Species diversity and richness were significantly correlated with soil N, P, organic C and spore density. AMF diversity in 15-year-old plantations was more similar to that in the natural stand than in the 10-year-old plantation. We confirmed that a 15-year-old plantation is not similar in terms of AMF colonization to natural stands. 13Trait complementarity between fine roots of Stipa purpurea and their associated arbuscular mycorrhizal fungi along a precipitation gradient in Tibetan alpine steppe显示文摘Development of fine roots and formation of symbiosis with arbuscular mycorrhizal(AM) fungi represent two strategies for plants to acquire nutrient and water from soil. Here, we elucidated how fine root development and symbolized mycorrhizal fungi with Stipa purpurea responded to the precipitation change in Tibetan alpine steppe ecosystem across a precipitation gradient from 50 mm to 400 mm. As precipitation increased, the proportion of thinner fine roots(diameter < 0.4 mm) in total roots increased significantly; while the mycorrhizal colonization percentage, either associated with thinner or thicker roots, decreased. This phenomenon indicated that fine root development and symbolized mycorrhizal fungi are likely alternative, and plant preferred to develop fine root rather than build a symbiotic relationship with mycorrhizal fungi in more benign niches with higher precipitation. Also, root diameter was negatively correlated with specific root length(SRL), but positively correlated with AM fungal colonization percentage, indicating thicker-root species rely more on mycorrhizal fungi in alpine steppe. 14Response of arbuscular mycorrhizal fungi and phosphorus solubilizing bacteria to remediation abandoned solid waste of coal mine显示文摘Coal is the vital resource of energy in China,but abandoned coal ash and gangue lead to the degradation of vegetation cover and reduce soil quality.Both arbuscular mycorrhizal fungi (AMF) and phosphate solubilizing bacteria (PSB) play a key role in biogeochemical cycle such as soil organic matter decomposition,nutrition release,and energy flow.To improve and reclamation the soil quality and ecological efficiency of the coal mining waste,we investigated the effects of an AMF strain (Glomus mosseae) and a PSB strain (Pantoesstewarti) on phytate mineralization and subsequent transfer to the host plant (Medicago sativa L.) using a two-compartment microcosm with a central 30 mm nylon mesh barrier.The results showed that significantly higher available P (AP),above ground biomass (AGB) and underground biomass (UGB) were in combined inoculation of AMF-PSB than other treatments in root and hyphae compartment.The microbial inoculum of the AMF or PSB had a significant influence on soil acid phosphatase activities (ACP).AMF-PSB enhanced phytate mineralization,improved plant biomass.AP and ACP positively influenced the AGB and UGB.AMF-PSB could be used as bioinoculant to enhance sustainable production of the plant in abandoned solid waste of coal mine.Yinli Bi Li Xiao Rongrong Liu 2019International Journal of Coal Science & Technology2019,6,4:2
15The diversity of arbuscular mycorrhizal fungi of Rosa acicularis ’Luhe’ in saline areas显示文摘Arbuscular mycorrhizal fungi(AMF)in the soil encompass a diversity of species and play an important role in maintaining ecosystem balance.However,little research has focused on the AMF diversity of Rosa acicularis Lindl.’Luhe’,which has been cultivated in northeastern saline areas,because of its strong salt tolerance and cold resistance.In the present study the AMF in the rhizosphere of R.acicularis Luhe were identified and AMF diversity and community composition were assessed using morphological and molecular techniques.Vesicles,hyphae,and arbuscular structures were observed in seedlings of different ages.Result shows that AMF established a good symbiotic relationship with R.acicularis Luhe.The colonization rate and spore density tended to increase over time.The AMF diversity in the rhizosphere of R.acicularis Luhe was low and four species of AMF were validated:Rhizophagus irregularis,Glomus aggregatum,Septoglomus furcatum,and Funneliformis mosseae.Of these,R.irregularis and G.aggregatum were dominant and have high salt tolerance.Determining the AMF diversity in the rhizosphere of R.acicularis Luhe will aid in screening AMF species with strong resistance and using mycorrhizas for plant establishment in breeding programs.Yanrong Zu Yuan Ping Liqiang Mu Tiantian Yang 2019Journal of Forestry Research2019,30,4:1
16Transcriptome analysis of the symbiosis-related genes between Funneliformis mosseae and Amorpha fruticosa显示文摘Arbuscular mycorrhizal fungi(AMF) can colonize and form associations with the roots of Amorpha fruticosa L.(desert false indigo). Various genes are induced during the symbiotic process. In this study, de novo transcriptome sequencing using RNA-seq was conducted for the first time for a comprehensive analysis of AMF-A. fruticosa symbionts at the transcript level. We obtained 12 G of raw data from illumina sequencing and recovered 115,786 unigenes with an average length of547 bp, among them 41,848 of significance. A total of2460 diffexpression genes were identified, including 1579 down-regulated and 881 up-regulated genes. A threshold for false discovery rate of \ 0.001 and fold change of [ 1 determined significant differences in gene expression.Using these criteria, we screened 285 significant differentially expressed genes, of which 82 were up-regulated and203 down-regulated. The 82 up-regulated genes were classified according to their functions and assigned into seven categories: stress and defense, metabolism, signaling transduction, protein folding and degradation, energy,protein synthesis, and transcription. The 203 down-regulated genes were screened according to fold change [ 2,and 50 highly significant down-regulated genes were obtained related to stress and defense. 17Effect of Arbuscular Mycorrhizal Fungal Inoculation on Sorghum bicolor Growth at Different Phosphate Levels: A Greenhouse Study显示文摘In sub-Saharan Africa, Sorghum (Sorghum bicolor) is an important cereal for both human being and animals. Unfortunately, its production is confronted to soils with deficiency of phosphorus. Traditional use of mineral phosphate on this culture fertilization is expensive and may cause contamination. It is thus necessary to seek more efficient and economic reasonable techniques to improve sorghum growth. Arbuscular mycorrhizal fungi (AMF) constitute a reference for phosphorus improvement and plant nutrition. This study aimed to investigate the effects of AMF strains (Rhizophagus irregulare, Glomus aggregatum, G. mosseae) on growth of sorghum cultivated in greenhouse on Sangalkam soil (Senegal) sterilized with or without Tilemsi natural phosphate (PNT). The phosphorus can represent until 0.2% of the dry weight of the plant. Two fertilizers were used separately and together to doses of 20 g by strain, 100 mg and 200 mg of PNT. The experiment lasted for 120 d. Results showed that mycorrhizal colonization intensity varied between 40% and 80% for all treatments. AMF inoculation increased sorghum plant height and biomass, regardless of PNT amendment. 19丛枝菌根真菌对旱稻根际Pb形态分布的影响(英文)显示文摘[目的]为了研究丛枝菌根真菌对旱稻根际土壤中重金属铅(Pb)化学赋存形态的影响。[方法]采用盆栽实验的方法,研究了不同Pb处理水平(0、300、600mg/kg)下,接种丛枝菌根真菌(AMF)Glomus mosseae对旱稻(OryzalsativaL.)根际Pb形态分布的影响。[结果]接种处理下,旱稻根系侵染率以及根外菌丝量随着土壤Pb含量增加而显著降低(P<0.05)。与未接种处理相比,接种处理显著提高Pb处理下根际pH;300mg/kgPb处理下,接种显著增加土壤球囊霉素含量,600mg/kgPb处理下,接种处理显著降低土壤球囊霉素含量(P<0.05);在Pb处理下,与未接种处理相比,接种处理显著提高可交换态Pb含量,显著降低了碳酸盐结合态以及铁锰氧化物结合态Pb含量,却显著增加了土壤中有机质结合态Pb含量(P<0.05)。[结论]这说明接种丛枝菌根真菌可以改变旱稻根际土壤中Pb的形态分布。张旭红 林爱军 张莘 郭兰萍 2012Agricultural Science & Technology2012,13,7:0
