| 2 | Weaning stress and gastrointestinal barrier development:Implications for lifelong gut health in pigs显示文摘The gastrointestinal(GI) barrier serves a critical role in survival and overall health of animals and humans. Several layers of barrier defense mechanisms are provided by the epithelial, immune and enteric nervous systems. Together they act in concert to control normal gut functions(e.g., digestion,absorption, secretion, immunity, etc.) whereas at the same time provide a barrier from the hostile conditions in the luminal environment. Breakdown of these critical GI functions is a central pathophysiological mechanism in the most serious GI disorders in pigs. | 3 | Effect of myosin heavy chain composition of muscles on meat quality in Laiwu pigs and Duroc显示文摘In order to explain the mechanism of high meat quality in Laiwu pigs and investigate the relation between myosin heavy chains (MyHC) composition and meat quality, meat quality analysis was conducted and mRNA expression of MyHC I, IIa, IIx, IIb was quantified by real-time fluorescence PCR in longissimus muscle (LM) and semimembranous muscle of Laiwu pigs and Duroc. The result indicated that, compared with Duroc, mRNA expression of MyHC IIa, IIx in LM and semimembranous muscle of Laiwu pigs was significantly increased, mRNA expression of MyHC IIb was dramatically decreased. However, the expression of MyHC I was not significantly affected by breeds. The correlation between mRNA expression of MyHC I, IIa, IIx in LM and meat color, pH value, marbling, intramuscular fat content was positive, but shear value of LM was negative. The relation between MyHC IIb mRNA expression and marbling, intramuscular fat content was dramatically negative, whereas shear value was strikingly positive, as well as fiber diameter, but without reaching statistical significance. | 6 | Hermetia illucens larvae as a potential dietary protein source altered the microbiota and modulated mucosal immune status in the colon of finishing pigs显示文摘Background: Insects, such as Hermetia illucens larvae, are rich in chitin and proteins, and represent a suitable feed ingredient replacement for animals. However, little is known about the effect of administering H. illucens larvae on intestinal microbiota, bacterial metabolite profiles, and mucosal immune status in animals. This study aimed to investigate the effects of administering H. illucens larvae on colonic microbiota and bacterial metabolites production in finishing pigs. Seventy-two crossbred(Duroc × Landrace × Large White) female pigs(initial body weight, 76.0 ± 0.52 kg) were randomly allocated to three different dietary treatments: a control diet(Control group) and two diets corresponding to 4%(H1 group) and 8%(H2 group) H. illucens larvae inclusion levels, respectively. Each treatment consisted of eight pens(replicates), with three pigs per pen. After 46 days of feeding, eight pigs per treatment(n =8) were slaughtered, and the colonic digesta and mucosa were collected for microbial composition and microbial fermentation products, and genes expression analyses.Results: The results showed that the H1 diet significantly increased the abundance of Lactobacillus,Pseudobutyrivibrio, Roseburia, and Faecalibacterium compared with those in the control group(P < 0.05), with a decrease in the abundance of Streptococcus. The numbers of Lactobacillus, Roseburia, and Clostridium cluster XIVa were significantly greater in the H1 group than in the control group(P < 0.05). Meanwhile, H2 diet increased the number of Clostridium cluster XIVa compared with the control group(P < 0.05). For colonic metabolites, total short chain fatty acids, butyrate, and isobutyrate concentrations were significantly higher in the H1 group than those in the control group(P < 0.05);the H1 treatment caused a striking decrease in protein fermentation compared with the control group, as the concentrations of total amines, cadaverine, tryptamine, phenol, p-cresol, and skatole were significantly lower(P < 0.05). Additionally, H2 diet also increased butyrate concentration compared with control group(P < 0.05), while decreased the concentrations of phenol, p-cresol, and skatole(P < 0.05). Pigs in the H1 group down-regulated the expression of TLR-4 and pro-inflammatory cytokines(IFN-γ) compared with pigs in the control group(P < 0.05), and up-regulated anti-inflammatory cytokine(IL-10) and intestinal barrier genes(ZO-1, occludin, and mucin-1). H2 diet up-regulated the expression of ZO-1 compared with control group(P < 0.05). Furthermore, the changes in the colonic mucosal gene expression were associated with changes in the bacterial composition and their metabolites.Conclusions: Collectively, dietary inclusion of Hermetia illucens larvae may enhance mucosal immune homeostasis of pigs via altering bacterial composition and their metabolites. | 10 | Alfalfa-containing diets alter luminal microbiota structure and short chain fatty acid sensing in the caecal mucosa of pigs显示文摘Background: Pork produced by outdoor-reared pigs raised mostly on alfalfa pastures attracts increasing population of consumer from most of the world. In China, pigs were raised with alfalfa-containing diets to seek for good quality pork.However, the influence of dietary alfalfa involving high level of insoluble dietary fiber(IDF) on pig intestinal luminal microbiota composition remains unclear. The objective of this study was to investigate the effects of alfalfa on luminal microbiota and short chain fatty acids(SCFA) production, and gene expressions involved in SCFA sensing, transporting and absorbing in pig caecal mucosa.Results: Twenty-four growing pigs were randomly allotted to four diets containing 0%, 5%, 10% and 15% alfalfa meal for a 28-d experiment. Ingestion of alfalfa meal-contained diets significantly increased the ratio of body weight gain to feed consumption. Illumina MiS eq sequencing of the V3 region of the 16 S r RNA genes showed that alfalfa-containing diet significantly decreased the relative abundance of genera Turicibacter, Acidiphilium, Paracoccus, Propionibacterium,Corynebacterium, Pseudomonas, Acinetobacter, and Staphylococcus, and increased the relative abundance of genera Lachnospira, Marvinbryantia, and Desulfovibrio in the caecal digesta. Butyrate concentration was significantly increased in the hindgut by the supplementation of alfalfa meal in diets. | 11 | Alginate oligosaccharide-induced intestinal morphology, barrier function and epithelium apoptosis modifications have beneficial effects on the growth performance of weaned pigs显示文摘Background: Alginate oligosaccharide(AOS), produced from alginate by alginate lyase-mediated depolymerisation, is a potential substitute for antibiotics and possesses growth-enhancing effects. Nevertheless, the mechanisms by which AOS regulates porcine growth remain to be elucidated. Therefore, we investigated the AOS-mediated changes in the growth performance of weaned pigs by determining the intestinal morphology, barrier function,as well as epithelium apoptosis.Methods: Twenty-four weaned pigs were distributed into two groups(n = 12) and received either a basal diet(control group) or the same diet supplemented with 100 mg/kg AOS. On d 15, D-xylose(0.1 g/kg body weight)was orally administrated to eight randomly selected pigs per treatment, and their serum and intestinal mucosa samples were collected 1 h later.Results: Our results showed that inclusion of AOS in the diet for 2 wk increased(P < 0.05) the average daily body weight gain in weaned pigs. Notably, AOS supplementation ameliorated the intestinal morphology and barrier function, as suggested by the enhanced(P < 0.05) intestinal villus height, secretory immunoglobulin A content and goblet cell counts. Compared to the control group, AOS ingestion both decreased(P < 0.05) the total apoptotic percentage and increased(P < 0.05) the proportion of S phase in the intestinal epithelial cells. Dietary fatty acids not only play critical roles in energy homeostasis and cel ular membrane composition, but also exert potent effects on intestinal development, immune function, and inflammatory response. Recent studies support potential therapeutic roles for specific fatty acids(short chain and medium chain fatty acids and long chain polyunsaturated fatty acids) in intestinal inflammation of pigs. | 14 | Effect of dietary supplementation of xylanase on apparent ileal digestibility of nutrients,viscosity of digesta,and intestinal morphology of growing pigs fed corn and soybean meal based diet显示文摘This study was to determine apparent ileal digestibility of acid detergent fiber(ADF), neutral detergent fiber(NDF), dry matter(DM), energy, organic matter(OM), crude ash, digesta viscosity, and gut morphology in nursery pigs fed diets containing xylanase(Lohmann Animal Nutrition GmbH, Cuxhaven, Germany). The diet(61% corn, 35% soybean meal, 1% poultry fat, and 3% minerals and vitamins) was mixed with 3 levels of xylanase(0,700, and 1400 LXU/kg). Thirty-six barrows(17.6 ± 3.3 kg) received one of 3 treatment diets based on a randomized complete block design with the initial body weight(BW) as a block. Pigs were individually housed and received experimental diets twice daily(0700 and 1700 h) at a fixed amount based on BW of pigs(0.09 x BW^(0.75) kg). Pigs were fed diets for 10 d, and chromium oxide(0.3%) was added to the diets from d 6 as an indigestible external marker. Pigs were euthanized at the end of d 10 for the collection of digesta and tissues. Jejunal digesta were centrifuged to measure viscosity using a viscometer(Brookfield Engineering Laboratories, Stoughton, MA). Diets and freeze-dried ileal digesta were used to measure ADF, NDF, and chromium to calculate apparent ileal digestibility of ADFand NDF. Villus height and crypt depth of jejunum were measured using a microscope(Fisher Scientific, Hampton, NH). Data were analyzed using polynomial contrasts in the MIXED procedure of SAS version 9.3(SAS Inc., Cary, NC, USA). Morphological measurements and ileal ADF digestibility were not affected by increasing xylanase. However, increasing xylanase supplementation from 0 to 1400 LXU/kg enhanced ileal digestibility of NDF(P < 0.042, linear) from 27.9 to 40.3%,DM(P < 0.006, linear) from 55.4 to 64.6%, OM(P < 0.006, linear) from 59.2 to 67.7%, and energy(P < 0.003,linear) from 58.8 to 68.0%. | 15 | Perivascular adipose tissue dysfunction aggravates adventitial remodeling in obese mini pigs via NLRP3 inflammasome/IL-1 signaling pathway显示文摘Perivascular adipose tissue (PVAT), a special type of adipose tissue, closely surrounds vascular adventitia and produces numerous bioactive substances to maintain vascular homeostasis. PVAT dysfunction has a crucial role in regulating vascular remodeling, but the exact mechanisms remain unclear. In this study, we investigated whether and how obesity-induced PVAT dysfunction affected adventitia remodeling in early vascular injury stages. Mini pigs were fed a high sugar and fat diet for 6 months to induce metabolic syndrome and obesity. In the mini pigs, left carotid vascular injury was then generated using balloon dilation. Compared with normal mini pigs, obese mini pigs displayed significantly enhanced vascular injury-induced adventitial responses, evidenced by adventitia fibroblast (AF) proliferation and differentiation, and adventitia fibrosis, as well as exacerbated PVAT dysfunction characterized by increased accumulation of resident macrophages, particularly the M1 pro-infiammatory phenotype, increased expression of leptin and decreased expression of adiponectin, and production of pro-infiammatory cytokines interleukin (IL)-1β and IL-18. Primary AFs cultured in PVAT-conditioned medium from obese mini pigs also showed significantly increased proliferation and differentiation. We further revealed that activated nod-like receptor protein 3 (NLRP3) infiammasome and its downstream products, i.e., IL-1 family members such as IL-1β and IL-18 were upregulated in the PVAT of obese mini pigs;PVAT dysfunction was also demonstrated in preadipocytes treated with palmitic acid. Finally, we showed that pretreatment with IL-1 receptor (IL-1R) antagonist or IL-1R knockdown blocked AF proliferation and differentiation in AFs cultured in PVAT-conditioned medium. | 16 | Overall assessment of antibiotic substitutes for pigs:a set of meta-analyses显示文摘Background:Antibiotic growth promoters are widely used to improve weight gain.However,the abuse of antibiotics can have many negative effects on people.Developing alternatives to antibiotics is an urgent need in livestock production.We aimed to perform a meta-analysis and network meta-analysis(NMA)to investigate the effects of feed additives as potential antibiotic substitutes(ASs)on bacteriostasis,growth performance,intestinal morphology and immunity.Furthermore,the primary,secondary,and tertiary ASs were defined by comparing their results with the results of antibiotics.Results:Among 16,309 identified studies,37 were summarized to study the bacteriostasis effects of feed additives,and 89 were included in the meta-analysis and NMA(10,228 pigs).We summarized 268 associations of 57 interventions with 32 bacteria.The order of bacteriostasis effects was as follows:antimicrobial peptides(AMPs)≈antibiotics>organic acids>plant extracts>oligosaccharides.We detected associations of 11 feed additives and 11 outcomes.Compared with a basal diet,plant extract,AMPs,probiotics,microelements,organic acids,bacteriophages,lysozyme,zymin,and oligosaccharides significantly improved growth performance(P<0.05);organic acids,probiotics,microelements,lysozyme,and AMPs remarkably increased the villus height:crypt depth ratio(V/C)(P<0.05);and plant extracts,zymin,microelements,probiotics,and organic acids notably improved immunity(P<0.05).The optimal AMP,bacteriophage,lysozyme,microelements,oligosaccharides,organic acids,plants,plant extracts,probiotics,and zymin doses were 0.100%,0.150%,0.012%,0.010%,0.050%,0.750%,0.20%,0.040%,0.180%,and 0.100%,respectively.Compared with antibiotics,all investigated feed additives exhibited no significant difference in effects on growth performance,IgG,and diarrhoea index/rate(P>0.05);AMPs and microelements significantly increased V/C(P<0.05);and zymin significantly improved lymphocyte levels(P<0.05).Furthermore,linear weighting sum models were used to comprehensively estimate the overall impact of each feed additive on pig growth and health.Conclusions:Our findings suggest that AMPs and plant extracts can be used as primary ASs for weaned piglets and growing pigs,respectively.Bacteriophages,zymin,plants,probiotics,oligosaccharides,lysozyme,and microelements can be regarded as secondary ASs.Nucleotides and organic acids can be considered as tertiary ASs.Future studies should further assess the alternative effects of combinational feed additives. | Bocheng Xu Jie Fu Luoyi Zhu Zhi Li Mingliang Jin Yizhen Wang | 2021 | Journal of Animal Science and Biotechnology2021,12,2: | 9 |

This study aimed to evaluate the redox system in tissues of finishing pigs by supplementation with high dose of daidzein(640 mg/kg).Results: The supplementation of high dose of daidzein for 64 d increased the activity of superoxide dismutase and total antioxidant capacity in longissimus muscle but down-regulated the expression of reactive oxygen species(ROS)-producing enzyme NADPH oxidase-2 and cyclooxygenase-2. In contrast, high-level supplementation with daidzein exerted pro-oxidant changes in back fat, abdominal fat, liver, and plasma, as reflected by increased contents of malondialdehyde, a lipid peroxidation product, in these tissues. Furthermore, daidzein supplementation resulted in higher expression of ROS-producing enzymes, including NADPH oxidase-1 and cyclooxygenase-1in liver, 5-lipoxygenase(5-LOX) in backfat and NADPH oxidase-2 both in abdominal fat and backfat. | 19 | Preliminary attempt to distinguish the domesticated pigs from wild boars by the methods of carbon and nitrogen stable isotope analysis显示文摘Despite great achievements in the origins of domestic pigs made by the methods of zooarchaeology and molecular biology,how to scientifically distinguish the domesticated pigs from wild boars during the early stage of pig domestication is still poorly understood.Compared to wild boar's diets which come from the natural environment,the diets of domestic pigs are more easily influenced by human feeding activities.Therefore,in principle,exploration of the dietary differences among pigs and understanding the impact on pig diets fed by humans can have great potential to differentiate between wild boars and domesticated pigs.To reveal dietary differences among pigs and distinguish the domesticated pigs from wild boars based on comparison with the diets of humans and other animals,we analyzed the carbon and nitrogen stable isotopes of human bones from Xiaojingshan Site and animal bones from Yuezhuang Site,both of which belong to Houli Culture in Shandong Province and date to about 8500―7500 years ago.The mean δ 13C value((-17.8 ± 0.3)‰) and δ 15N value((9.0±0.6)‰) in human collagen indicate that although millet agriculture began it was not the main subsistence strategy as millets are typical of C4 plants and that humans made a living mainly by gathering,hunting or raising some domesticated animals.The δ 13C value(-16.1‰) and δ 15N value(6.9‰) in the bovine suggest that C3 plants were dominant in its diet with some C4 plants complemented.The fish has lower δ 13C value(-24.9‰) and higher δ 15N value(8.8‰) than the bovine,which is the characteristic of the isotopic values from Eurasian freshwater fish.Based on the differences in carbon and nitrogen isotope values,the pigs can be divided into three groups.A group,composed of two pigs,has low δ 13C values(-18.1‰,-20.0‰) and low δ 15N values(4.7‰,6.0‰).B group,only one pig,has the highest δ 13C value(-10.6‰) and mediate δ 15N value(6.4‰).As for the C group,also only one pig,low δ 13C value(-19.0‰) and the highest δ 15N value(9.1‰) are observed.Previous studies on the stable isotopes from modern or ancient wild boars' bones have suggested that C3 plants are predominated in their diets and that their δ 15N values are close to those in herbivores and far from those in carnivores.Based on the comparison with the isotope values from humans,the wild boars and the domestic pigs from Xipo Site in Henan 6000―5500 years ago and Kangjia Site in Shaanxi 4500―4000 years ago,we conclude that A pig group belongs to wild boars while B and C groups can be attributed to domesticated pigs. | Michael P.RICHARDS | 2009 | Science China Earth Sciences2009,52,1: | 7 |
| 20 | Effects of a two-meal daily feeding pattern with varied crude protein levels on growth performance and antioxidant indexes in pigs显示文摘The present study aimed to evaluate the effects of daily feeding pattern on growth performance, blood biochemistry, and antioxidant indexes in pigs. One hundred and eighty female Duroc x Landrace x Yorkshire(DLY) pigs with similar body weight(11.00 ± 0.12 kg) were randomly assigned to 3 groups: the control group(fed 17.01% CP diet, twice daily); high-low group(H-L group, fed18.33% CP diet in the morning, followed by 15.70% CP diet in the afternoon); and low-high group(L-H group, fed 15.70% CP diet in the morning, followed by 18.33% CP diet in the afternoon)(n = 6). Comparable amounts of their respective diets were given at 05:30 and 15:00 throughout the experimental periods to make all the treatments consumed the same type of food and the same amount of calories on a daily basis. On day 30, one pig was randomly selected per litter for blood samples. Compared with the control group, ADG in the H-L and L-H groups increased by 8.11% and 16.23%, but not significant(P > 0.05); and blood urea nitrogen(BUN) in the H-L and L-H groups decreased by 26.76% and 41.04%(P < 0.05), respectively. The H-L group feeding pattern could significantly improve levels of serum superoxide dismutase(SOD), when compared with the control group. These findings suggest that the twomeal daily feeding pattern with varied levels of CP affects serum levels of BUN and SOD.
