| 1 | Alginate oligosaccharide-induced intestinal morphology, barrier function and epithelium apoptosis modifications have beneficial effects on the growth performance of weaned pigs显示文摘Background: Alginate oligosaccharide(AOS), produced from alginate by alginate lyase-mediated depolymerisation, is a potential substitute for antibiotics and possesses growth-enhancing effects. Nevertheless, the mechanisms by which AOS regulates porcine growth remain to be elucidated. Therefore, we investigated the AOS-mediated changes in the growth performance of weaned pigs by determining the intestinal morphology, barrier function,as well as epithelium apoptosis.Methods: Twenty-four weaned pigs were distributed into two groups(n = 12) and received either a basal diet(control group) or the same diet supplemented with 100 mg/kg AOS. On d 15, D-xylose(0.1 g/kg body weight)was orally administrated to eight randomly selected pigs per treatment, and their serum and intestinal mucosa samples were collected 1 h later.Results: Our results showed that inclusion of AOS in the diet for 2 wk increased(P < 0.05) the average daily body weight gain in weaned pigs. Notably, AOS supplementation ameliorated the intestinal morphology and barrier function, as suggested by the enhanced(P < 0.05) intestinal villus height, secretory immunoglobulin A content and goblet cell counts. Compared to the control group, AOS ingestion both decreased(P < 0.05) the total apoptotic percentage and increased(P < 0.05) the proportion of S phase in the intestinal epithelial cells. | 3 | Tannic acid extracted from gallnut prevents post-weaning diarrhea and improves intestinal health of weaned piglets显示文摘This experiment was conducted to evaluate the effects of different levels of tannic acid(TA)on growth performance,diarrhea rate,nutrient digestibility and intestinal health in weaned piglets.A total of 180 weaned piglets(Duroc×Landrace×Yorkshire,24 d of age,initial average BW=7.77±0.17 kg)were allotted to 5 groups(6 pigs/pen and 6 replicates/group)in a randomized complete block design according to their gender and body weight.Piglets were fed a basal diet,or the basal diet supplemented with 0.05%,0.1%,0.2%or 0.4%TA for 28 d.The supplementary levels of TA in the diets were obtained by adding tannalbin containing 51%TA and 40.17%protein.The results showed that,compared with the CON group,dietary TA did not affect ADFI,ADG or F:G,and linearly reduced(P<0.01)the diarrhea rate and diarrhea index of piglets.There were no significant effects on apparent total tract digestibility(ATTD)in the 0.05%,0.1%and 0.2%TA groups,while negative effects(P<0.05)on apparent digestibility of crude protein and gross energy were observed in the 0.4%TA group.In addition,the nutrient digestibility of dry matter,crude protein and gross energy linearly decreased(P<0.01)with the increase of TA dosage.Supplementation of TA increased(P<0.05)the villus height of the duodenum and jejunum,as well as increased(P<0.05)catalase(CAT)activity in serum.Dietary TA improved(P<0.05)the Bacillus counts in cecal digesta.Further,TA significantly improved(P<0.05)Bacillus counts and reduced(P<0.05)the Escherichia coli counts in colonic digesta.The concentration of acetic acid,pro pionic acid,butyric acid and isovaleric acid in cecal digesta were significantly increased(P<0.05).The mRNA expression level of zonula occludens-1(Z0-1),zonula occludens-2(ZI-2),and claudin-2(CLDN-2)in the jejunum were greater(P<0.05)in TA supplemented groups.The study showed that,compared to the control,TA prevented post-weaning diarrhea and improved intestinal health of weaned piglets,and the appropriate level of TA supplementation would be from 0.1%to 0.2%. | Yanyan Song Yong Luo Bing Yu Jun He Ping Zheng Xiangbing Mao Zhiqing Huang Junqiu Luo Yuheng Luo Hui Yan Quyuan Wang Huifen Wang Daiwen Chen Jie Yu | 2021 | Animal Nutrition2021,,4: | 5 |
| 4 | Differential expression, molecular cloning,and characterization of porcine beta defensin 114显示文摘Background:β-defensins have attracted considerable research interest because of their roles in protecting hosts from various pathogens.This study was conducted to investigate the expression profiles of the porcineβ-defensin114(PBD114)in different breeds and in response to infections.Moreover,the function of PBD114 protein was partially investigated.Methods:Six Tibetan pigs(TP)and six DLY(Duroc×Landrace×Yorkshire)pigs were slaughtered to explore the expression profiles of PBD114 in different breeds and tissues.For infection models,sixteen DLY pigs were divided into two groups and challenged either with sterile saline or E.coli K88.The recombinant protein PBD114(r PBD114)was obtained by using a heterologous expression system in E.coli.Results:PBD114 gene was highly expressed in tissues such as the intestine,liver,spleen,and thymus.Interestingly,the expression level of PBD114 gene was higher in the TP pigs than in the DLY pigs(P<0.05),and was significantly elevated upon E.coli K88 challenge(P<0.05).The nucleotide sequences of PBD114 from Tibetan and DLY pigs was identical,and both showed a 210-bp open reading frame encoding a 69-amino acid mature peptide.To explaore the function of PBD114 protein,PBD114 gene was successfully expressed in E.coli Origami B(DE3)and the molecular weight of the r PBD114 was estimated by SDS-PAGE to be 25 k Da.The r PBD114 was purified and mass spectrometry verified the protein as PBD114.Importantly,r PBD114 showed antimicrobial activities against E.coli DH5αand E.coli K88,and the minimal inhibitory concentrations(MICs)were 64 and 128μg/m L,respectively.Hemolytic and cytotoxicity assays showed that r PBD114 did not affect cell viability under physiological concentrations.Conclusions:PBD114 is an infection response gene that is differentially-expressed between different porcine breeds and tissues.The antimicrobial activity of PBD114 protein,against pathogens such as the E.coli K88,suggested that it may serve as a candidate for the substitution of conventionally used antibiotics. | Guoqi Su Kunhong Xie Daiwen Chen Bing Yu Zhiqing Huang Yuheng Luo Xiangbing Mao Ping Zheng Jie Yu Junqiu Luo Jun He | 2019 | Journal of Animal Science and Biotechnology2019,10,4: | 4 |

It captures considerable interest in redesign of its function for either the mechanism study or the pharmacological development as an antioxidant. In order to de- velop a general strategy for specifically targeting and operating selenium in active sites of enzymes, the catalytically essential residue selenocysteine (Sec) was first successfully bioincorporated into the catalytic center of subtilisin by using an auxotrophic expression system. The studies of the catalytic activity and the steady-state kinetics demonstrated that selenosubtilisin is an excellent GPx-like bio- catalyst. In comparison with the chemically modified method, biosynthesis exhibits obvious advan- tages: Sec could be site-directly incorporated into active sites of enzymes to overcome the non-speci- ficity generated by chemical modification. | 8 | Leucine modulates the IPEC-J2 cell proteome associated with cell proliferation, metabolism and phagocytosis显示文摘Leucine can affect intestinal protein expressions, and improve mucosal immune function. However, little study has been conducted to determine the change of protein component by leucine treatment in intestine epithelial cells. The present study was to cover the key proteins and cell pathways that could be regulated by leucine treatment in porcine intestinal epithelial cell line(IPEC-J2) cells with the approach of proteome analysis. A total number of 3,211 proteins were identified in our approach by searching the database of Uniprot sus scrofa. Among identified proteins, there were 101 proteins expressed differently between control group and leucine group. Compared with the control group, there were 50 up-regulated proteins and 51 down-regulated proteins in leucine group. In these proteins, leucine treatment decreased the expression of some proteins including pyruvate kinase, glyceraldehyde-3-phosphate dehydrogenase,E3 ubiquitin ligase, cathepsin D, caspase 3 and caspase 6, and increased the levels of some proteins, such as some eukaryotic translation initiation factors, ubiquitin carboxyl-terminal hydrolase, DNA-related RNA polymerase II, urokinase plasminogen activator, cyclin-dependent kinase inhibitor 2 b, Mut L homolog 1,5-methylcytosine binding domain 4, polymerase d, a-tubulin, syntaxin 18, Ras homolog D, actin related protein 2/3 complex and cofilin. Via the analysis of Gene Ontology and pathways, these proteins in IPECJ2 cells were related with some physiological functions, such as protein metabolism, glycolysis, cell proliferation, apoptosis and phagocytosis. | 10 | Recent advances in laser self-injection locking to high-Q microresonators显示文摘The stabilization and manipulation of laser frequency by means of an external cavity are nearly ubiquitously used in fundamental research and laser applications. While most of the laser light transmits through the cavity, in the presence of some back-scattered light from the cavity to the laser, the self-injection locking effect can take place, which locks the laser emission frequency to the cavity mode of similar frequency. The self-injection locking leads to dramatic reduction of laser linewidth and noise. Using this approach, a common semiconductor laser locked to an ultrahigh-Q microresonator can obtain sub-Hertz linewidth, on par with state-of-the-art fiber lasers. Therefore it paves the way to manufacture high-performance semiconductor lasers with reduced footprint and cost. Moreover, with high laser power, the optical nonlinearity of the microresonator drastically changes the laser dynamics, offering routes for simultaneous pulse and frequency comb generation in the same microresonator. Particularly, integrated photonics technology, enabling components fabricated via semiconductor CMOS process, has brought increasing and extending interest to laser manufacturing using this method. | 11 | A high-quality sponge gourd(Luffa cylindrica)genome显示文摘Sponge gourd(Luffa cylindrica)is an important cultivated vegetable and medicinal plant in the family Cucurbitaceae.In this study,a draft genome sequence of the sponge gourd inbred line P93075 was analyzed.Using Illumina,PacBio,and 10×Genomics sequencing techniques as well as new assembly techniques such as FALCON and chromatin interaction mapping(Hi-C),a chromosome-scale genome of approximately 656.19 Mb,with an N50 scaffold length of 48.76 Mb,was generated.From this assembly,25,508 protein-coding gene loci were identified,and 63.81%of the whole-genome consisted of transposable elements,which are major contributors to the expansion of the sponge gourd genome.According to a phylogenetic analysis of conserved genes,the sponge gourd lineage diverged from the bitter gourd lineage approximately 41.6 million years ago.Additionally,many genes that respond to biotic and abiotic stresses were found to be lineage specific or expanded in the sponge gourd genome,as demonstrated by the presence of 462 NBS-LRR genes,a much greater number than are found in the genomes of other cucurbit species;these results are consistent with the high stress resistance of sponge gourd.Collectively,our study provides insights into genome evolution and serves as a valuable reference for the genetic improvement of sponge gourd. | Haibin Wu Gangjun Zhao Hao Gong Junxing Li Caixia Luo Xiaoli He Shaobo Luo Xiaoming Zheng Xiaoxi Liu Jinju Guo Junqiu Chen Jianning Luo | 2020 | Horticulture Research2020,7,1: | 2 |
| 12 | Understanding enzyme catalysis by means of supramolecular artificial enzymes显示文摘Enzymes are biomacromolecules responsible for the abundant chemical biotransformations that sustain life. Recently, biochemists have discovered that multiple conformations and numerous parallel paths are involved during the processes catalyzed by enzymes. It is plausible that the entire macromolecular scaffold is involved in catalysis via cooperative motions that result in incredible catalytic efficiency. Moreover, some enzymes can very strongly bind the transition state with an association constant of up to 1024 M-1, suggesting that covalent bond formation is a possible process during the conversion of the transition state in enzyme catalysis, in addition to the concatenation of noncovalent interactions. Supramolecular chemistry provides fundamental knowledge about the relationships between the dynamic structures and functions of organized molecules. By tak-ing advantage of supramolecular concepts, numerous supramolecular enzyme mimics with complex and hierarchical structures have been designed and investigated. Through the study of supramolecular enzyme models, a great deal of information to aid our understanding of the mechanism of catalysis by natural enzymes has been acquired. By analyzing the co-seismic responses,we found that firstly there was essentially a linear correlation between response time and epicentral distance. Secondly,there was some correlation between maximum response amplitude and earthquake magnitude as well as between the duration and earthquake magnitude. Thirdly,the response amplitudes and decay rates were different for different types of instruments. Due to less data-sampling frequency,the deformation instruments,could not display the first motion of P and S waves,but responded mainly to far-field surface waves. Before the earthquake,the NS earthtide component recorded by the cave stainmeter at Yichang was distorted for nearly eight hours. | 18 | Foundry manufacturing of tight-confinement,dispersion-engineered,ultralow-loss silicon nitride photonic integrated circuits显示文摘The foundry development of integrated photonics has revolutionized today’s optical interconnect and datacenters.Over the last decade,we have witnessed the rising of silicon nitride(Si_(3)N_(4)) integrated photonics,which is currently transferring from laboratory research to foundry manufacturing.The development and transition are triggered by the ultimate need for low optical loss offered by Si_(3)N_(4),which is beyond the reach of silicon and III-V semiconductors.Combined with modest Kerr nonlinearity,tight optical confinement,and dispersion engineering,Si_(3)N_(4) has today become the leading platform for linear and Kerr nonlinear photonics,and it has enabled chip-scale lasers featuring ultralow noise on par with table-top fiber lasers.However,so far all the reported fabrication processes of tight-confinement,dispersion-engineered Si_(3)N_(4) photonic integrated circuits(PICs)with optical loss down to few dB/m have only been developed on 4-inch(100 mm diameter)or smaller wafers.Yet,to transfer these processes to established CMOS foundries that typically operate 6-inch or even larger wafers,challenges remain.In this work,we demonstrate the first foundry-standard fabrication process of Si_(3)N_(4) PICs with only 2.6 dB/m loss,thickness above 800 nm,and near 100%fabrication yield on 6-inch(150 mm diameter)wafers.Such thick and ultralow-loss Si_(3)N_(4) PIC enables low-threshold generation of soliton frequency combs.Merging with advanced heterogeneous integration,active ultralow-loss Si_(3)N_(4) integrated photonics could pave an avenue to addressing future demands in our increasingly information-driven society. | ZHICHAO YE HAIYAN JIA ZHANGJUN HUANG CHEN SHEN JINBAO LONG BAOQI SHI YI-HAN LUO LAN GAO WEI SUN HAIRUN GUO JIJUN HE JUNQIU LIU | 2023 | Photonics Research2023,11,4: | 1 |
| 19 | Yeast hydrolysate attenuates lipopolysaccharide-induced inflammatory responses and intestinal barrier damage in weaned piglets显示文摘Background Intestinal inflammation is the main risk factor causing intestinal barrier dysfunction and lipopolysaccharide(LPS)can trigger inflammatory responses in various eukaryotic species.Yeast hydrolysate(YH)possesses multibiological effects and is received remarkable attention as a functional ingredient for improving growth performance and promoting health in animals.However,there is still inconclusive on the protective effects of dietary YH supplementation on intestinal barrier of piglets.This study was conducted to investigate the attenuate effects of YH supplementation on inflammatory responses and intestinal barrier injury in piglets challenged with LPS.Methods Twenty-four piglets(with an average body weight of 7.42±0.34 kg)weaned at 21 days of age were randomly assigned to one of two dietary treatments(12 replications with one pig per pen):a basal diet or a basal diet containing YH(5 g/kg).On the 22nd d,6 piglets in each treatment were intraperitoneally injected with LPS at 150μg/kg BW,and the others were injected with the same amount of sterile normal saline.Four hours later,blood samples of each piglet were collected and then piglets were euthanized.Results Dietary YH supplementation increased average daily feed intake and average daily gain(P<0.01),decreased the ratio of feed intake to gain of piglets(P sponse,evidenced by the increase o=0.048).Lipopolysaccharide(LPS)injection induced systemic inflammatory ref serum concentrations of haptoglobin(HP),adrenocorticotropic hormone(ACTH),cortisol,and interleukin-1β(IL-1β).Furthermore,LPS challenge resulted in inflammatory intestinal damage,by up-regulation of the protein or mRNA abundances of tumor necrosis factor-α(TNF-α),IL-1β,toll-like receptors 4(TLR4)and phosphor-nuclear factor-κB-p65(p-NFκB-p65)(P<0.01),and down-regulation of the jejunal villus height,the protein and mRNA abundances of zonula occludens-1(ZO-1)and occludin(OCC;P<0.05)in jejunal mucosa.Dietary YH supplementation decreased the impaired effects of ACTH,cortisol,HP,IL-1βand diamine oxidase in serum(P<0.05).Moreover,YH supplementation also up-regulated the jejunal villus height,protein and mRNA abundances of ZO-1 and OCC(P<0.05),down-regulated the mRNA expressions of TNF-αand IL-1βand the protein abundances of TNF-α,IL-1β,TLR4 and p-NFκB-p65 in jejunal mucosa in LPS-challenged pigs(P<0.01).Conclusion Yeast hydrolysate could attenuate inflammatory response and intestinal barrier injury in weaned piglets challenged with LPS,which was associated with the inhibition of TLR4/NF-κB signaling pathway activation. | Runqi Fu Chan Liang Daiwen Chen Gang Tian Ping Zheng Jun He Jie Yu Xiangbing Mao Yuheng Luo Junqiu Luo Bing Yu | 2023 | Journal of Animal Science and Biotechnology2023,14,4: | 0 |
