1Ferroptosis:molecular mechanisms and health implications显示文摘Cell death can be executed through different subroutines.Since the description of ferroptosis as an iron-dependent form of nonapoptotic cell death in 2012,there has been mounting interest in the process and function of ferroptosis.Ferroptosis can occur through two major pathways,the extrinsic or transporter-dependent pathway and the intrinsic or enzyme-regulated pathway.Ferroptosis is caused by a redox imbalance between the production of oxidants and antioxidants,which is driven by the abnormal expression and activity of multiple redox-active enzymes that produce or detoxify free radicals and lipid oxidation products.Accordingly,ferroptosis is precisely regulated at multiple levels,including epigenetic,transcriptional,posttranscriptional and posttranslational layers.The transcription factor NFE2L2 plays a central role in upregulating anti-ferroptotic defense,whereas selective autophagy may promote ferroptotic death.Here,we review current knowledge on the integrated molecular machinery of ferroptosis and describe how dysregulated ferroptosis is involved in cancer,neurodegeneration,tissue injury,inflammation,and infection.Daolin Tang Xin Chen Rui Kang Guido Kroemer 2021Cell Research2021,31,2:182

3Cuproptosis:a copper-triggered modality of mitochondrial cell death显示文摘Various heavy metals can induce regulated cell death through different subroutines.A recent study published in Science found that intracellular copper accumulation triggers the aggregation of mitochondrial lipoylated proteins and the destabilization of Fe–S cluster proteins,leading to a unique type of cell death termed cuproptosis.Beyond classical apoptosis,several forms of regulated cell death(RCD)have been identified.These RCD subroutines differ in the initiating stimuli,intermediate activation events,and end effectors.1 Heavy metal ions are essential micronutrients,but either insufficient or excessive abundance of metals can trigger cell death.For example,ferroptosis has been defined as an iron-dependent form of oxidative cell death caused by unrestricted lipid peroxidation.2 Surprisingly,a recent study by Tsvetkov and colleagues showed that intracellular copper(Cu)induces a novel form of RCD that is different from oxidative stress-related cell death(e.g.,apoptosis,ferroptosis,and necroptosis)and has been termed“cuproptosis”.3 In contrast,mitochondrial stress,especially the aggregation of lipoylated mitochondrial enzymes and the loss of Fe–S cluster proteins,ignites cuproptosis(Fig.1).Daolin Tang Xin Chen Guido Kroemer 2022Cell Research2022,32,5:19
4Current management of spontaneous intracerebral haemorrhage显示文摘Intracerebral haemorrhage(ICH)is the most devastating and disabling type of stroke.Uncontrolled hypertension(HTN)is the most common cause of spontaneous ICH.Recent advances in neuroimaging,organised stroke care,dedicated Neuro-ICUs,medical and surgical management have improved the management of ICH.Early airway protection,control of malignant HTN,urgent reversal of coagulopathy and surgical intervention may increase the chance of survival for patients with severe ICH.Intensive lowering of systolic blood pressure to<140 mm Hg is proven safe by two recent randomised trials.Transfusion of platelets in patients on antiplatelet therapy is not indicated unless the patient is scheduled for surgical evacuation of haematoma.In patients with small haematoma without significant mass effect,there is no indication for routine use of mannitol or hypertonic saline(HTS).However,for patients with large ICH(volume>30 cbic centmetre)or symptomatic perihaematoma oedema,it may be beneficial to keep serum sodium level at 140–150 mEq/L for 7–10 days to minimise oedema expansion and mass effect.Mannitol and HTS can be used emergently for worsening cerebral oedema,elevated intracranial pressure(ICP)or pending herniation.HTS should be administered via central line as continuous infusion(3%)or bolus(23.4%).Ventriculostomy is indicated for patients with severe intraventricular haemorrhage,hydrocephalus or elevated ICP.Patients with large cerebellar or temporal ICH may benefit from emergent haematoma evacuation.It is important to start intermittent pneumatic compression devices at the time of admission and subcutaneous unfractionated heparin in stable patients within 48 hours of admission for prophylaxis of venous thromboembolism.There is no benefit for seizure prophylaxis or aggressive management of fever or hyperglycaemia.Early aggressive comprehensive care may improve survival and functional recovery.Cyrus K Dastur Wengui Yu 2017Stroke & Vascular Neurology2017,2,1:18
5Thrombus aspiration in acute myocardial infarction:Rationale and indication显示文摘Reperfusion of myocardial tissue is the main goal of primary percutaneous coronary intervention(PPCI) with stent implantation in the treatment of acute ST-segment elevation myocardial infarction(STEMI). Although PPCI has contributed to a dramatic reduction in cardiovascular mortality over three decades, normal myocardial perfusion is not restored in approximately one-third of these patients. Several mechanisms may contribute to myocardial reperfusion failure, in particular distal embolization of the thrombus and plaque fragments. In fact, this is a possible complication during PPCI, resulting in microvascular obstruction and no-reflow phenomenon. The presence of a visible thrombus at the time of PPCI in patients with STEMI is associated with poor procedural and clinical outcomes. Aspiration thrombectomy during PPCI has been proposed to prevent embolization in order to improve these outcomes. In fact, the most recent guidelines suggest the routine use of manual aspiration thrombectomy during PPCI(class Ⅱa) to reduce the risk of distal embolization. Even though numerous international studies have been reported, there are conflicting results on the clinical impact of aspiration thrombectomy during PPCI. In particular, data on long-term clinical outcomes are still inconsistent. 7De novo exploration and self-guided learning of potentialenergy surfaces显示文摘Interatomic potential models based on machine learning(ML)are rapidly developing as tools for material simulations.However,because of their flexibility,they require large fitting databases that are normally created with substantial manual selection and tuning of reference configurations.Here,we show that ML potentials can be built in a largely automated fashion,exploring and fitting potential-energy surfaces from the beginning(de novo)within one and the same protocol.The key enabling step is the use of a configuration-averaged kernel metric that allows one to select the few most relevant and diverse structures at each step.The resulting potentials are accurate and robust for the wide range of configurations that occur during structure searching,despite only requiring a relatively small number of single-point DFT calculations on small unit cells.We apply the method to materials with diverse chemical nature and coordination environments,marking an important step toward the more routine application of ML potentials in physics,chemistry,and materials science.Noam Bernstein Gábor Csányi Volker L.Deringer 2019npj Computational Materials2019,,1:10

9Clopidogrel loading dose versus maintenance dose to treat patients with acute ischaemic stroke in China(CLASS-China):results from a prospective double-blind randomised clinical trial显示文摘Aim The role of clopidogrel in treating patients with acute ischaemic stroke is unclear.We have conducted the clinical trial in order to evaluate the efficacy and safety of clopidogrel with a loading dose in treating patients with non-cardiogenic acute ischaemic stroke.Method Clopidogrel loading dose versus maintenance dose to treat patients with acute ischaemic stroke in China(CLASS-China)was a prospective,randomised,double-blind and placebo-controlled clinical trial in China.Patients with acute ischaemic stroke of non-cardiogenic origin within 48 hours of onset were enrolled and those received thrombolysis were excluded.Enrolled patients were divided into two treatment groups:loading dose and routine dose.The primary outcome was the incidence of stroke recurrence or progression within 7 days.Primary safety outcome was measured by life-threatening haemorrhage.An intent-to-treat analysis was used for the statistical analysis.results From March 2008 to March 2010,a total of 303 patients from 16 centres were recruited into this study;six were excluded because of lack of basic information.Since the enrolment was slow and the study drug expired in March 2010,this clinical trial was stopped earlier than planned.No significant baseline and demographic differences were seen between the two groups.There was no difference in primary outcome between the loading dosage group 16.1%(24/149)and control group 14.9%(22/148),respectively(p=0.782).The mortality and disability rate within 90 days in loading dose group(19.6%)was slightly lower than that in controlled group(23.4%),p=0.444.Loading dose group had two(1.3%)cases of fatal haemorrhage and control group had four(2.7%)within 90 days,p=0.674.No significant difference was detected in other adverse events between the groups.conclusion In our study stopped early due to slow enrolment,loading dose of clopidogrel does not reduce the risk of recurrent stroke.Future trials with sufficient number of patients enrolled are needed to re-examine this hypothesis.Ying Zhao Wanyong Yang Zefeng Tan Wenmin Wang Weimin Xiao Jinsheng Zeng Anding Xu 2017Stroke & Vascular Neurology2017,2,3:7
10Would integrated Western and traditional Chinese medicine have more benefits for stroke rehabilitation? A systematic review and meta-analysis显示文摘Background Stroke is a major cause of death or long-term disability worldwide.Many patients with stroke receive integrative therapy consisting of Western medicine(WM)and routine rehabilitation in conjunction with Chinese medicine(CM),such as acupuncture and Chinese herbal medicine.However,there is no available evidence on the effectiveness of the combined use of WM and CM interventions in stroke rehabilitation.Aims The purpose of this meta-analysis is to evaluate the results of all individual studies to assess the combined use of CM and WM in stroke rehabilitation compared with WM only.Methods The Preferred Reporting Items for Systematic Reviews and Meta-Analysis guidelines were followed.MEDLINE,EMBASE,Cochrane and China National Knowledge Infrastructure(CNKI)were searched.The included outcomes were dependency,motor function,depression and swallowing function.Subgroup analysis was performed,and publication bias was assessed using funnel plots.Summary of review 58 studies and 6339 patients were included in the meta-analysis.Subgroup analysis revealed that combined therapy comprising both acupuncture and WM had a superior effect on improving dependency and swallowing function compared with standard WM therapy alone.Potential superiority of combined therapy comprising CM and WM in improving depression compared with standard WM therapy was also found.Conclusions Our results indicate that the combined use of CM and WM could be more efficacious in stroke rehabilitation compared with the use of WM therapy alone.However,most studies were short in duration(2 to 4 weeks)and prone to different types of biases,which prevents making any conclusion regarding the long-term effects and raises concerns regarding true efficacy in context of high likelihood of Hawthorn bias.So,more randomised controlled trials with more rigorous design and longer duration of treatment and follow-up need to be conducted to compare WM alone versus WM and CM combined.Linda LD Zhong Ya Zheng Alexander Y Lau Norman Wong Liang Yao Xingyao Wu Tengteng Shao Zhenxing Lu Huijuan Li Chun Sum Yuen Jianwen Guo Suzanne Lo Janita Chau Kam Wa Chan Bacon Fung Leung Ng Zhaoxiang Bian Edwin Chau-leung Yu 2022Stroke & Vascular Neurology2022,7,1:7
11Effectiveness of Catch-Up Vaccinations after COVID-19 Containment--China,2020显示文摘Introduction:In the early stages of the coronavirus disease 2019(COVID-19)epidemic in China,the highest level of public health emergency response(Level 1 Response)was launched in all province-level administrative divisions(PLADs)across the country.Provision of vaccination services was selectively suspended due to the physical distancing and gathering restrictions required of Level 1 Response.The purpose of this paper is to evaluate the impact of COVID-19 on vaccination services and the effectiveness of selective vaccination service suspension and catch-up vaccinations in China.Methods:Vaccine doses administered,routine vaccination coverage levels,catch-up vaccine doses administered,and coverage levels after the catch-up campaign were determined from individual vaccination records in sampled clinics,standard routine immunization administrative coverage reports,and catch-up vaccination activity reports submitted by PLADs between October 2019 and October 2020.Results:National Immunization Program(NIP)and non-NIP vaccine doses administered in sampled clinics decreased by 80% and 90%,respectively,compared with doses administered before the COVID-19 pandemic.Coverage with the birth dose of hepatitis B vaccine(HepB1)and the Bacillus Calmette-Guérin vaccine(BCG)—two vaccinations recommended to continue throughout the epidemic due to their timecritical nature—were maintained at over 85%,while coverage of other NIP vaccines decreased to below 60%by February 2020.Catch-up vaccination coverage of the 29 PLADs,excluding Xinjiang and Xizang(Tibet),began in April 2020 and exceeded 90%;Hubei catch-up coverage was 95% by October 2020.Conclusions and Implications for Public Health Practice:The COVID-19 pandemic had a great impact on China’s vaccination services.During the epidemic and time of local transmission of the coronavirus,HepB1,BCG,rabies vaccine for postexposure prophylaxis,and tetanus antitoxin(TAT)for wound prophylaxis were maintained at high levels.Of the 69 million vaccine doses postponed during the time of local transmission of the coronavirus,94% were administered in the catch-up campaign.China’s pandemic immunization practices may provide evidence for other countries’immunization programs.Jing Wu Wenzhou Yu Lei Cao Lingsheng Cao Lance Rodewald Jiakai Ye Yifan Song Li Li Xiaoxue Liu Ning Wen Fuzhen Wang Lixin Hao Yixing Li Hui Zheng Keli Li Chao Ma Dan Wu Yanmin Liu Guomin Zhang Zhijie An Huaqing Wang Zundong Yin 2020China CDC weekly2020,2,50:6

14Saliva as a non-invasive diagnostic tool for inflammation and insulin-resistance显示文摘Saliva has been progressively studied as a non-invasive and relatively stress-free diagnostic alternative to blood. Currently, saliva testing is used for clinical assessment of hormonal perturbations, detection of HIV antibodies, DNA analysis, alcohol screening, and drug testing. Recently, there has been increasing interest in evaluating the diagnostic potential of saliva in obesity, inflammation, and insulin-resistance. Current literature has demonstrated elevated levels of inflammatory biomarkers including C-reactive protein, tumor necrosis factor-α, interleukin-6, and interferon-γ in saliva of obese/overweight children and adults. Salivary antioxidant status has also been studied as a measure of oxidative stress in individuals with type 2 diabetes. Further, several studies have demonstrated correlations of salivary markers of stress and insulin resistance including cortisol, insulin, adiponectin, and resistin with serum concentrations. These findings suggest the potential diagnostic value of saliva in health screening and risk stratification studies, particularly in the pediatric population, with implications for inflammatory, metabolic and cardiovascular conditions. However, additionalstudies are required to standardize saliva collection and storage procedures, validate analytical techniques for biomarker detection, and establish reference ranges for routine clinical use. 20Regulated cell death(RCD)in cancer:key pathways and targeted therapies显示文摘Regulated cell death(RCD),also well-known as programmed cell death(PCD),refers to the form of cell death that can be regulated by a variety of biomacromolecules,which is distinctive from accidental cell death(ACD).Accumulating evidence has revealed that RCD subroutines are the key features of tumorigenesis,which may ultimately lead to the establishment of different potential therapeutic strategies.Hitherto,targeting the subroutines of RCD with pharmacological small-molecule compounds has been emerging as a promising therapeutic avenue,which has rapidly progressed in many types of human cancers.Thus,in this review,we focus on summarizing not only the key apoptotic and autophagy-dependent cell death signaling pathways,but the crucial pathways of other RCD subroutines,including necroptosis,pyroptosis,ferroptosis,parthanatos,entosis,NETosis and lysosomedependent cell death(LCD)in cancer.Moreover,we further discuss the current situation of several small-molecule compounds targeting the different RCD subroutines to improve cancer treatment,such as single-target,dual or multiple-target small-molecule compounds,drug combinations,and some new emerging therapeutic strategies that would together shed new light on future directions to attack cancer cell vulnerabilities with small-molecule drugs targeting RCD for therapeutic purposes.Fu Peng Minru Liao Rui Qin Shiou Zhu Cheng Peng Leilei Fu Yi Chen Bo Han 2022Signal Transduction and Targeted Therapy2022,7,9:4
