1Selective blockade of NF-kappa B by novel mutated I kappa B alpha suppresses CD3/CD28-induced activation of memory CD4(+) T cells in asthma显示文摘Background:Nuclear factor kappa B(NF-kappa B) overactivation plays a crucial role in T-helper 2 (Th2)-biased allergic airway inflammation by increased activation and decreased apoptosis of CD4(+) T cells. We have shown that targeted NF-kappa B suppression in dendritic cells by adenoviral gene transfer of a novel mutated inhibitor of NF-kappa B(I kappa B alpha) (AdI kappa B alpha M) contributes to T-cell tolerance, but the immunosuppressive action of AdI kappa B alpha M on memory(CD45RO(+)) CD4(+) T cells remains enigmatic. Methods:CD45RO(+) T cells from Dermatophagoides farinaei-sensitized asthmatic patients, untransfected or transfected with AdI kappa B alpha M or AdLacZ(beta-galactosidase) for 24 h, were stimulated with anti-CD3(1.0 mu g/ml) plus anti-CD28(0.5 mu g/ml) monoclonal antibody for an additional 24 h. I kappa B alpha M transgene expression and NF-kappa B activation were detected by polymerase chain reaction(PCR), reverse transcription-PCR(RT-PCR), Western blot analysis, and electrophoretic mobility shift assay. Phenotype and apoptosis were measured by flow cytometry, annexin V binding, and terminal deoxyribonucleotidyl transferase-mediated dUTP nick end labeling analyses. Cytokine production and cell proliferation were determined using enzyme-linked immunosorbent assay andH-3 thymidine incorporation. Results:A unique 801-bp I kappa B alpha M cDNA and a dose-dependent increase in I kappa B alpha M transgene expression were observed in AdI kappa B alpha M-transfected CD45RO(+) T cells. Significantly, AdI kappa B alpha M inhibited CD3/CD28-mediated NF-kappa B activation in CD45RO(+) T cells, leading to evident apoptosis, reduction of eotaxin, RANTES, Th1 interferon (IFN)-gamma and interleukin (IL)-2, and Th2 (IL-4, IL-5, and IL-13 despite a slight decrease in IL-10) cytokines and secondary proliferative response. AdI kappa B alpha M also upregulated cytotoxic T lymphocyte-associated antigen 4(CTLA-4) and downregulated CD69 besides no change in CD28. Conclusion:I kappa B alpha M might be beneficial to augment memory CD4(+) T-cell tolerance through modulating B7-CD28/CTLA-4 co-stimulatory pathways and NF-kappa B-dependent cytokine profiles in allergic inflammatory diseases including asthma.Zhou,L. 2Features of near-inertial motions observed on the northern South China Sea shelf during the passage of two typhoons显示文摘Features of near-inertial motions on the shelf(60 m deep) of the northern South China Sea were observed under the passage of two typhoons during the summer of 2009. There are two peaks in spectra at both sub-inertial and super-inertial frequencies. The super-inertial energy maximizes near the surface, while the sub-inertial energy maximizes at a deeper layer of 15 m. The sub-inertial shift of frequency is induced by the negative background vorticity. The super-inertial shift is probably attributed to the near-inertial wave propagating from higher latitudes. The near-inertial currents exhibit a two-layer pattern being separated at mid-depth(25–30 m), with the phase in the upper layer being nearly opposite to that in the lower layer. The vertical propagation of phase implies that the near-inertial energy is not dominantly downward. The upward flux of the near-inertial energy is more evident at the surface layer(<17 m). There exist two boundaries at 17 and 40 m, where the near-inertial energy is reflected upward and downward. The near-inertial motion is intermittent and can reach a peak of as much as 30 cm/s. The passage of Typhoon Nangka generates an intensive near-inertial event, but Typhoon Linfa does not. This difference is attributed to the relative mooring locations, which is on the right hand side of Nangka's path(leading to a wind pattern rotating clockwise with time) and is on the left hand side of Linfa's path(leading to a wind pattern rotating anti-clockwise with time).CHEN Shengli HU Jianyu POLTON Jeff A. 2015Acta Oceanologica Sinica2015,34,1:6

4Laser Welding,Microwelding,Nanowelding and Nanoprocessing(Invited Paper)显示文摘Laser has been widely applied for advanced material processing,such as welding of advanced high strength steels(AHSS) for automotive applications and Pt,Ti,and Nitinol alloys for medical applications.Recently,ultrafast pulse laser,especially femtosecond laser,is identified as powerful tools for joining and surface engineering of nanomaterials.This paper reviews our research on these fields at the Centre of Advanced Materials Joining,University of Waterloo.Representative conclusions are listed for laser material processing at three continuing scales.1) For AHSS welding,a thin Al layer can dramatically enhanced the weldability of galvannealed AHSS.The movement of the welding line and softening in the vicinity has significantly influence on global mechanical properties in dual phase(DP) and transformation induced plasticity(TRIP) steels.The increase of the strength ratio decreases the formability and results in the non-uniform strain distribution.2) For medical micro-welding,oxygen is found to be a key role for welding strength and microstructure.The formation of intermetallics reduces the relative ductility and increases the susceptibility of cracking.Different laser processing parameters can significantly influence weld mechanical performance.3) For femtosecond laser nanofabrication,the joining of Au nanoparticles is successfully achieved by controlling laser energy.Surface nanostructure of Ag induced by femtosecond laser irradiation can work effective probes for surface enhanced Raman spectroscopy.A. 5Donor denervation and elimination of Kupffer cells affect expression of P-selectin and ICAM-1 in liver graft显示文摘BACKGROUND:The non-function and dysfunction of primary liver graft likely involves dependence on Kupffer cells and hepatic innervation. The present experiment was designed to study the expression of P-selectin and intercellular adhesion molecule-1(ICAM-1) mRNA in liver graft and to elucidate the role of Kupffer cells and the sympathetic nerve of the liver in down-regulating this expression. METHODS:Donor rats were given hexamethonium,a sympathetic ganglionic blocking agent,and/or gadolinium chloride,an inhibitor of Kupffer cells. Then the changes of graft P-selectin and ICAM-1 mRNA expression were measured after liver transplantation. RESULTS:The expressions of P-selectin and ICAM-1 mRNA were increased after liver transplantation,and down-regulated by liver denervation and elimination of Kupffer cells. CONCLUSIONS:Live donor denervation and elimination of Kupffer cells down-regulated the expressions of P-selectin and ICAM-1 mRNA in grafts. 7FERROELECTRIC CHARACTERISTICS OF SrBi_2Ta_2O_9 THIN FILMS ANNEALED IN FORMING GAS显示文摘The degradation behavior of SrBi2 Ta2O9 (SBT) thin films annealed in forming gas (5%H2 +95%N2) was investigated. The ferroelectric remnant polarization P. of SBT had a severe decrease in the containing H2 atmosphere at 400℃. SEM results showed that Bi could be reduced from SBT thin films for the effect of H2. The reduction role increased with a rise of annealing temperature. SBT thin films lost their ferroelectricity when the annealing temperature was above 450℃. It implied that the degradation of P. value or the disappearance of ferroelectricity in SBT thin films had the relationship with the reductive amounts of Bi in SBT. H2 played an important role in degradation of ferroelectric SBT thin films. This role could be attributed to its strong reduction activity during forming gas annealing.D.S. Wang, T. Yu and A. 8Development and evaluation of a safe and effective sugar-free herbal lollipop that kills cavity-causing bacteria显示文摘Dental caries (tooth decay) is caused by a specific group of cariogenic bacteria, like Streptococcus mutans, which convert dietary sugars into acids that dissolve the mineral in tooth structure. Killing cariogenic bacteria is an effective way to control or prevent tooth decay. In a previous study, we discovered a novel compound (Glycyrrhizol A), from the extraction of licorice roots, with strong antimicrobial activity against cariogenic bacteria. In the current study, we developed a method to produce these specific herbal extracts in large quantities, and then used these extracts to develop a sugar-free lollipop that effectively kills cariogenic bacteria like Streptococcus mutans. Further studies showed that these sugar-free lollipops are safe and their antimicrobial activity is stable. Two pilot human studies indicate that a brief application of these lollipops (twice a day for ten days) led to a marked reduction of cariogenic bacteria in oral cavity among most human subjects tested. 10Progress in TILLING as a tool for functional genomics and improvement of crops显示文摘Food security is a global concern and substantial yield increases in crops are required to feed the growing world population. Mutagenesis is an important tool in crop improvement and is free of the regulatory restrictions imposed on genetically modified organisms. Targeting Induced Local Lesions in Genomes(TILLING), which combines traditional chemical mutagenesis with high‐throughput genome‐wide screening for point mutations in desired genes, offers a powerful way to create novel mutant alleles for both functional genomics and improvement of crops. TILLING is generally applicable to genomes whether small or large, diploid or evenallohexaploid, and shows great potential to address the major challenge of linking sequence information to the function of genes and to modulate key traits for plant breeding. TILLING has been successfully applied in many crop species and recent progress in TILLING is summarized below, especially on the developments in mutation detection technology, application of TILLING in gene functional studies and crop breeding. The potential of TILLING/EcoTILLING for functional genetics and crop improvement is also discussed. 14Nonvolatile waveguide transmission tuning with electrically-driven ultra-small GST phase-change material显示文摘Low-power reconfigurable optical circuits are highly demanded to satisfy a variety of different applications. Conventional electro-optic and thermo-optic refractive index tuning methods in silicon photonics are not suitable for reconfiguration of optical circuits due to their high static power consumption and volatility. We propose and demonstrate a nonvolatile tuning method by utilizing the reversible phase change property of GST integrated on top of the silicon waveguide. The phase change is enabled by applying electrical pulses to the lm-sized GST active region in a sandwich structure. 19Metal Exposure and Risk of Diabetes and Prediabetes among Chinese Occupational Workers显示文摘Objective To study the association between metal exposure and risk of diabetes and prediabetes among Chinese workers exposed to metals.Methods We used data obtained from the baseline survey of the Jinchang Cohort Study of workers in Jinchang Industry,the largest nickel production company in China.A total of 42,122 workers ≥20 years of age were included in the study.A standardized,structured questionnaire was used to collect epidemiological information.Physical examinations and laboratory tests were conducted to evaluate the health status of the participants and to measure various biomarkers including blood sugar,lipids,and urinary metal concentrations.Logistic regression was used to study the association between occupational groups categorized according to the measured metal levels(office workers,low-level;mining/production workers,mid-level;and smelting/refining workers,high-level)and risk of diabetes and prediabetes.Results The overall prevalence of diabetes and prediabetes was 7.5% and 16.8%,respectively.The adjusted odds ratios for diabetes among mining/production workers and smelting/refining workers compared to office workers were 1.5(95% CI:1.3,1.7)and 3.8(95% CI:3.4,4.3),respectively.No association was observed between these occupational groups and prediabetes in this study.Conclusion Occupations associated with higher levels of metal exposure were associated with an increased risk of diabetes in this cohort.More studies are needed to confirm this observed association.YANG Ai Min CHENG Ning PU Hong Quan LIU Si Min LI Juan Sheng BASSIG Bryan A.
