1Special Section:SARS-CoV-2 Preliminary evidence from a multicenter prospective observational study of the safety and efficacy of chloroquine for the treatment of COVID-19显示文摘Effective therapies are urgently needed for the SARS-CoV-2 pandemic.Chloroquine has been proved to have antiviral effect against coronavirus in vitro.In this study,we aimed to assess the efficacy and safety of chloroquine with different doses in COVID-19.In this multicenter prospective observational study,we enrolled patients older than 18 years old with confirmed SARS-CoV-2 infection excluding critical cases from 12 hospitals in Guangdong and Hubei Provinces.Eligible patients received chloroquinephosphate s00 mg,orally,once(half dose)or twice(full dose)daily.Patients treated with non-chloroquine therapy were included as historical controls.The primary endpoint is the time to undetectable viral RNA.Secondary outcomes include the proportion of patients with undetectable viral RNA by day 10 and 14,hospitalization time,duration of fever,and adverse events.A total of 197 patients completed chloroquine treatment,and 176patients were included as historical controls.The median time to achieve an undetectable viral RNA was shorter in chloroquine than in non-chloroquine(absolute difference in medians-6.0 days;95%CI-6.0 to—4.0).The duration of fever is shorter in chloroquine(geometric mean ratio 0.6;95%CIO.5 to 0.8).No serious adverse events were observed in the chloroquine group.Patients treated with half dose experienced lower rate of adverse events than with full dose.Although randomized trials are needed for further evaluation,this study provides evidence for safety and efficacy of chloroquine in COVID-19 and suggests that chloroquine can be a cost-effective therapy for combating the COVID-19 pandemic.Mingxing Huang Man Li Fei Xiao Pengfei Pang Jiabi Liang Tiantian Tang Shaoxuan Liu Binghui Chen Jingxian Shu Yingying You Yang Li Meiwen Tang Jianhui Zhou Guanmin Jiang Jingfen Xiang Wenxin Hong Songmei He Zhaoqin Wang Jianhua Feng Changqing Lin Yinong Ye Zhilong Wu Yaocai Li Bei Zhong Ruilin Sun Zhongsi Hong Jing Liu Huili Chen Xiaohua Wang Zhonghe Li Duanqing Pei Lin Tian Jinyu Xia Shanping Jiang Nanshan Zhong Hong Shan 2020National Science Review2020,7,9:20
2Remimazolam benzenesulfonate anesthesia effectiveness in cardiac surgery patients under general anesthesia显示文摘BACKGROUND Sedation with propofol injections is associated with a risk of addiction,but remimazolam benzenesulfonate is a comparable anesthetic with a short elimination half-life and independence from cell P450 enzyme metabolism.Compared to remimazolam,remimazolam benzenesulfonate has a faster effect,is more quickly metabolized,produces inactive metabolites and has weak drug interactions.Thus,remimazolam benzenesulfonate has good effectiveness and safety for diagnostic and operational sedation.AIM To investigate the clinical value of remimazolam benzenesulfonate in cardiac surgery patients under general anesthesia.METHODS A total of 80 patients who underwent surgery in the Department of Cardiothoracic Surgery from August 2020 to April 2021 were included in the study.Using a random number table,patients were divided into two anesthesia induction groups of 40 patients each:remimazolam(0.3 mg/kg remimazolam benzenesulfonate)and propofol(1.5 mg/kg propofol).Hemodynamic parameters,inflammatory stress response indices,respiratory function indices,perioperative indices and adverse reactions in the two groups were monitored over time for comparison.RESULTS At pre-anesthesia induction,the remimazolam and propofol groups did not differ regarding heart rate,mean arterial pressure,cardiac index or volume per wave index.After endotracheal intubation and when the sternum was cut off,mean arterial pressure and volume per wave index were significantly higher in the remimazolam group than in the propofol group(P<0.05).After endotracheal intubation,the oxygenation index and the respiratory index did not differ between the groups.After endotracheal intubation and when the sternum was cut off,the oxygenation index values were significantly higher in the remimazolam group than in the propofol group(P<0.05).Serum interleukin-6 and tumor necrosis factor-αlevels 12 h after surgery were significantly higher than before surgery in both groups(P<0.05).The observation indices were re-examined 2 h after surgery,and the epinephrine,cortisol and blood glucose levels were significantly higher in the remimazolam group than in the propofol group(P<0.05).The recovery and extubation times were significantly lower in the remimazolam group than in the propofol group(P<0.05);there were significantly fewer adverse reactions in the remimazolam group(10.00%)than in the propofol group(30.00%;P<0.05).CONCLUSION Compared with propofol,remimazolam benzenesulfonate benefited cardiac surgery patients under general anesthesia by reducing hemodynamic fluctuations.Remimazolam benzenesulfonate influenced the surgical stress response and respiratory function,thereby reducing anesthesia-related adverse reactions.Fang Tang Jian-Min Yi Hong-Yan Gong Zi-Yun Lu Jie Chen Bei Fang Chen Chen Zhi-Yi Liu 2021World Journal of Clinical Cases2021,9,34:19

4Type 1 CD8^+ T Cells are Superior to Type 2 CD8^+ T Cells in Tumor Immunotherapy due to Their Efficient Cytotoxicity,Prolonged Survival and Type 1 Immune Modulation显示文摘CD8+ cytotoxic T(Tc) cells play a crucial role in host immune responses to cancer,and in this context,adoptive CD8+ Tc cell therapy has been studied in numerous animal tumor models. Its antitumor efficacy is,to a large extent,determined by the ability of Tc cells to survive and infiltrate tumors. In clinical trials,such in vitro-activated T cells often die within hours to days,and this greatly limits their therapeutic efficacy. CD8+ Tc cells fall into two subpopulations based upon their differential cytokine secretion. In this study,we in vitro generated that ovalbumin(OVA) -pulsed dendritic cell(DCOVA) -activated CD8+ type 1 Tc(Tc1) cells secreting IFN-γ,and CD8+ type 2 Tc(Tc2) cells secreting IL-4,IL-5 and IL-10,which were derived from OVA-specific T cell receptor(TCR) transgenic OT I mice. We then systemically investigated the in vitro and in vivo effector function and survival of Tc1 and Tc2 cells,and then assessed their survival kinetics after adoptively transferred into C57BL/6 mice,respectively. We demonstrated that,when compared to CD8+ Tc2,Tc1 cells were significantly more effective in perforin-mediated cytotoxicity to tumor cells,had a significantly higher capacity for in vivo survival after the adoptive T cell transfer,and had a significantly stronger therapeutic effect on eradication of well-established tumors expressing OVA in animal models. In addition,CD8+ Tc1 and Tc2 cells skewed the phenotype of CD4+ T cells toward Th1 and Th2 type,respectively. 6High-performance asymmetric electrodes photodiode based on Sb/WSe2 heterostructure显示文摘Two-dimensional (2D) van der Waals (vdWs) metal-semiconductor heterostructures with atomically sharp interface and matched work functions have recently attracted great attention due to their unique electronic and optoelectronic properties. Here we report the vapor phase epitaxial growth of large-scale vertical Sb/WSe2 metal-semiconductor vdWs heterostructures with uniform stacking orientation. Compared with the growth on S1O2/S1 substrate, the thick ness of Sb nan osheet on WSe2 can be reduced effectively to mono layer. We con struct Sb-WSe2-Au asymmetric electrodes photodiode based on the Sb/WSe2 heterostructures. Electrical transport measurements indicate that the photodiode show obvious rectifying effect. Optoelectronic characterizations show prominent photoresponse with a high photoresposivity of 364 mA/W, a fast response time of less than 8 ms, a large open-circuit voltage of 0.27 V and a maximum electrical power output of 0.11 nW. 9Site-directed mutagenesis identified the key active site residues of alcohol acyltransferase PpAAT1 responsible for aroma biosynthesis in peach fruits显示文摘The aroma of peach fruit is predominantly determined by the accumulation of γ-decalactone and ester compounds.A previous study showed that the biosynthesis of these aroma compounds in peach fruit is catalyzed by PpAAT1,an alcohol acyltransferase.In this work,we investigated the key active site residues responsible for γ-decalactone and ester biosynthesis.A total of 14 candidate amino acid residues possibly involved in internal esterification and 9 candidate amino acid residues possibly involved in esterification of PpAAT1 were assessed via site-directed mutagenesis.Analyses of the in vitro enzyme activities of PpAAT1 and its site-directed mutant proteins(PpAAT1-SMs)with different amino acid residue mutations as well as the contents of γ-decalactone in transgenic tobacco leaves and peach fruits transiently expressing PpAAT1 and PpAAT1-SMs revealed that site-directed mutation of H165 in the conserved HxxxD motif led to lost enzymatic activity of PpAAT1 in both internal esterification and its reactions,whereas mutation of the key amino acid residue D376 led to the total loss ofγ-decalactone biosynthesis activity of PpAAT1.Mutations of 9 and 7 other amino acid residues also dramatically affected the enzymatic activity of PpAAT1 in the internal esterification and esterification reactions,respectively.Our findings provide a biochemical foundation for the mechanical biosynthesis ofγ-decalactone and ester compounds catalyzed by PpAAT1 in peach fruits,which could be used to guide the molecular breeding of new peach species with more favorable aromas for consumers.Zhi-Zhong Song Bin Peng Zi-Xia Gu Mei-Ling Tang Bei Li Mei-Xia Liang Li-Min Wang Xiao-Tong Guo Jian-Ping Wang Yu-Fen Sha Hong-Xia Zhang 2021Horticulture Research2021,8,1:3
10Direct van der Waals epitaxial growth of 1D/2D Sb2Se3/WS2 mixed-dimensional p-n heterojunctions显示文摘The mixed-dimensional integration of two-dimensional (2D) materials with non-2D materials can give rise to prominent advances in performance or function.To date,the mixed-dimensional one-dimensional (1D)/2D heterostructures have been fabricated using various physical assembly approaches.However,direct epitaxial growth method which has notable advantages in preparing large-scale products and obtaining perfect interfaces is rarely investigated.Herein,we demonstrate for the first time the direct synthesis of the 1D/2D mixed-dimensional heterostructures by sequential vapor-phase growth of Sb2Se3 nanowires on WS2 monolayers.X-ray diffraction (XRD) pattern and Raman spectrum confirm the composition of the Sb2Se3/NS2 heterostructures.Transmission electron microscope (TEM) measurement demonstrates high quality of the heterostructures.Electrical transport characterization reveals that Sb2Se3 nanowire exhibits p-type characteristic and that WS2 monolayer exhibits n-type behavior,and that the p-n diode from 1D/2D mixed-dimensional Sb2Se3/WS2 heterostructure possesses obvious current rectification behavior.Optoelectronic measurements of the heterostructures show apparent photovoltaic response with an open-circuit voltage of 0.19 V,photoresponsivity of 1.51 A/W (Vds =5 V) and fast response time of less than 8 ms.The van der Waals epitaxial growth mode of Sb2Se3 nanowires on WS2 monolayers is verified by stripping the Sb2Se3 nanowire from the heterostructures using tape.Together,the direct van der Waals epitaxy opens a facile pathway to 1D/2D mixed-dimensional heterostructures for functional electronic and optoelectronic devices.Guangzhuang Sun Bo Li Jia Li Zhengwei Zhang Huifang Ma Peng Chen Bei Zhao Ruixia Wu Weiqi Dang Xiangdong Yang Xuwan Tang Chen Dai Ziwei Huang Yuan Liu Xidong Duan Xiangfeng Duan 2019Nano Research2019,12,5:2

14Protein convertase subtilisin/Kexin type 9 inhibits hepatocellular carcinoma growth by interacting with GSTP1 and suppressing the JNK signaling pathway显示文摘Objective:Protein convertase subtilisin/Kexin type 9(PCSK9)has been found to be closely associated with the occurrence and development of numerous tumors.However,the precise role of PCSK9 and its relationship to the development of hepatocellular carcinoma(HCC)remain largely unknown.This study aimed to clarify these issues.Methods:The expression levels of PCSK9 in HCC tissues and HCC cell lines were determined by the quantitative reverse transcription polymerase chain reaction,Western blot,and immunohistochemical analyses,and the effects of PCSK9 expression on HCC cell biological traits were investigated by overexpressing and downregulating PCSK9 expression in vivo and in vitro.Additionally,the mechanism by which PCSK9 mediated dissociation of glutathione S-transferase Pi 1(GSTP1)dimers and phosphorylation of the Jun N-terminal kinase(JNK)pathway components were investigated.Results:PCSK9 expression levels were significantly lower in HCC tissues than in adjacent non-tumor samples.In vivo and in vitro experiments suggested that PCSK9 inhibited HCC cell proliferation and metastasis.Further analysis showed that PCSK9 interacted with GSTP1 and promoted GSTP1 dimer dissociation and JNK signaling pathway inactivation in HCC cells.Moreover,the relationships between PCSK9 protein expressions and clinical outcomes were investigated.The PCSK9-lo group displayed a significantly shorter overall survival(OS;median OS:64.2 months vs.83.2 months;log-rank statistic:4.237;P=0.04)and recurrencefree survival(RFS;median RFS:26.5 months vs.46.6 months;log-rank statistic:10.498;P=0.001)time than the PCSK9-hi group.Conclusions:PCSK9 inhibited HCC cell proliferation,cell cycle progression,and apoptosis by interacting with GSTP1 and suppressing JNK signaling,suggesting that PCSK9 might act as a tumor suppressor and be a therapeutic target in HCC patients.Mingyan He Jing Hu Tingting Fang Wenqing Tang Bei Lv Biwei Yang Jinglin Xia 2022Cancer Biology & Medicine2022,19,1:1

16EPSTI1 promotes monocyte adhesion to endothelial cells in vitro via upregulating VCAM-1 and ICAM-1 expression显示文摘Atherosclerosis is a chronic inflammatory disease of arterial wall, and circulating monocyte adhesion to endothelial cells is a crucial step in the pathogenesis of atherosclerosis. Epithelial-stromal interaction 1 (EPSTI1) is a novel gene, which is dramatically induced by epithelial-stromal interaction in human breast cancer. EPSTI1 expression is not only restricted to the breast but also in other normal tissues. In this study we investigated the role of EPSTI1 in monocyte-endothelial cell adhesion and its expression pattern in atherosclerotic plaques. We showed that EPSTI1 was dramatically upregulated in human and mouse atherosclerotic plaques when compared with normal arteries. In addition, the expression of EPSTI1 in endothelial cells of human and mouse atherosclerotic plaques is significantly higher than that of the normal arteries. Furthermore, we demonstrated that EPSTI1 promoted human monocytic THP-1 cell adhesion to human umbilical vein endothelial cells (HUVECs) via upregulating VCAM-1 and ICAM-1 expression in HUVECs. Treatment with LPS (100, 500, 1000 ng/mL) induced EPSTI1 expression in HUVECs at both mRNA and protein levels in a dose- and time-dependent manner. Knockdown of EPSTI1 significantly inhibited LPS-induced monocyte-endothelial cell adhesion via downregulation of VCAM-1 and ICAM-1. Moreover, we revealed that LPS induced EPSTI1 expression through p65 nuclear translocation.
