1Special Section:SARS-CoV-2 Preliminary evidence from a multicenter prospective observational study of the safety and efficacy of chloroquine for the treatment of COVID-19显示文摘Effective therapies are urgently needed for the SARS-CoV-2 pandemic.Chloroquine has been proved to have antiviral effect against coronavirus in vitro.In this study,we aimed to assess the efficacy and safety of chloroquine with different doses in COVID-19.In this multicenter prospective observational study,we enrolled patients older than 18 years old with confirmed SARS-CoV-2 infection excluding critical cases from 12 hospitals in Guangdong and Hubei Provinces.Eligible patients received chloroquinephosphate s00 mg,orally,once(half dose)or twice(full dose)daily.Patients treated with non-chloroquine therapy were included as historical controls.The primary endpoint is the time to undetectable viral RNA.Secondary outcomes include the proportion of patients with undetectable viral RNA by day 10 and 14,hospitalization time,duration of fever,and adverse events.A total of 197 patients completed chloroquine treatment,and 176patients were included as historical controls.The median time to achieve an undetectable viral RNA was shorter in chloroquine than in non-chloroquine(absolute difference in medians-6.0 days;95%CI-6.0 to—4.0).The duration of fever is shorter in chloroquine(geometric mean ratio 0.6;95%CIO.5 to 0.8).No serious adverse events were observed in the chloroquine group.Patients treated with half dose experienced lower rate of adverse events than with full dose.Although randomized trials are needed for further evaluation,this study provides evidence for safety and efficacy of chloroquine in COVID-19 and suggests that chloroquine can be a cost-effective therapy for combating the COVID-19 pandemic.Mingxing Huang Man Li Fei Xiao Pengfei Pang Jiabi Liang Tiantian Tang Shaoxuan Liu Binghui Chen Jingxian Shu Yingying You Yang Li Meiwen Tang Jianhui Zhou Guanmin Jiang Jingfen Xiang Wenxin Hong Songmei He Zhaoqin Wang Jianhua Feng Changqing Lin Yinong Ye Zhilong Wu Yaocai Li Bei Zhong Ruilin Sun Zhongsi Hong Jing Liu Huili Chen Xiaohua Wang Zhonghe Li Duanqing Pei Lin Tian Jinyu Xia Shanping Jiang Nanshan Zhong Hong Shan 2020National Science Review2020,7,9:20

4Prognostic value of lymphovascular invasion in Bismuth-Corlette type Ⅳ hilar cholangiocarcinoma显示文摘AIM To assess the prognostic value of lymphovascular invasion(LVI)in Bismuth-Corlette typeⅣhilar cholangiocarcinoma(HC)patients. METHODS A retrospective analysis was performed on 142consecutively recruited typeⅣHC patients undergoing radical resection with at least 5 years of followup.Survival analysis was performed by the KaplanMeier method,and the association between the clinicopathologic variables and survival was evaluated by log-rank test.Multivariate analysis was adopted to identify the independent prognostic factors for overall survival(OS)and disease-free survival(DFS).Multiple logistic regression analysis was performed to determine the association between LVI and potential variables. RESULTS LVI was confirmed histopathologically in 29(20.4%)patients.Multivariate analysis showed that positive resection margin(HR=6.255,95%CI:3.485-11.229,P<0.001),N1 stage(HR=2.902,95%CI:1.132-7.439,P=0.027),tumor size>30 mm(HR=1.942,95%CI:1.176-3.209,P=0.010)and LVI positivity(HR=2.799,95%CI:1.588-4.935,P<0.001)were adverse prognostic factors for DFS.The independent risk factors for OS were positive resection margin(HR=6.776,95%CI:3.988-11.479,P<0.001),N1 stage(HR=2.827,95%CI:1.243-6.429,P=0.013),tumor size>30 mm(HR=1.739,95%CI:1.101-2.745,P=0.018)and LVI positivity(HR=2.908,95%CI:1.712-4.938,P<0.001).LVI was associated with N1 stage and tumor size>30 mm.Multiple logistic regression analysis indicated that N1 stage(HR=3.312,95%CI:1.338-8.198,P=0.026)and tumor size>30 mm(HR=3.258,95%CI:1.288-8.236,P=0.013)were associated with LVI. 5Geochronology and Geochemistry of the Granites from the Longtoushan Hydrothermal Gold Deposit in the Dayaoshan Area,Guangxi:Implication for Petrogenesis and Mineralization显示文摘The gold mineralization in the Longtoushan hydrothermal gold deposit is concentrated within the contact zone of the granitic complex. Whole rock geochemistry and in-situ U-Pb and Hf isotopic data were used to constrain the genesis and age of the granites and related Cu-Au mineralization in the Longtoushan Deposit. The granites mainly consist of the granite porphyry, rhyolite porphyry, porphyritic granite and quartz porphyry. LA-ICP-MS U-Pb dating of zircons from the granite porphyry, rhyolite porphyry and quartz porphyry indicates that they intruded from ca. 94 to 97 Ma. These intrusions exhibit similar trace element characteristics, i.e., right-dipping REE patterns, depletion of Ba, Sr, P and Ti, and enrichment of Th, U, Nd, Zr and Hf. The εHf(t) values of zircons from the granite porphyry, rhyolite porphyry and quartz porphyry range from-26.81 to-8.19,-8.12 to-5.33, and-8.99 to-5.83, respectively, suggesting that they were mainly derived from the partial melting of the Proterozoic crust. The Cu-Au mineralization is mainly related to the rhyolite porphyry and porphyritic granite, respectively. 6Layers of interstitial fluid flow along a“slit-shaped”vascular adventitia显示文摘Interstitial fluid(ISF)flow through vascular adventitia has been discovered recently.However,its kinetic pattern was unclear.We used histological and topographical identification to observe ISF flow along venous vessels in rabbits.By magnetic resonance imaging(MRI)in live subjects,the inherent pathways of ISF flow from the ankle dermis through the legs,abdomen,and thorax were enhanced by paramagnetic contrast.By fluorescence stereomicroscopy and layer-by-layer dissection after the rabbits were sacrificed,the perivascular and adventitial connective tissues(PACTs)along the saphenous veins and inferior vena cava were found to be stained by sodium fluorescein from the ankle dermis,which coincided with the findings by MRI.The direction of ISF transport in a venous PACT pathway was the same as that of venous blood flow.By confocal microscopy and histological analysis,the stained PACT pathways were verified to be the fibrous connective tissues,consisting of longitudinally assembled fibers.Real-time observations by fluorescence stereomicroscopy revealed at least two types of spaces for ISF flow:one along adventitial fibers and another one between the vascular adventitia and its covering fascia.Using nanoparticles and surfactants,a PACT pathway was found to be accessible by a nanoparticle of<100 nm and contained two parts:a transport channel and an absorptive part.The calculated velocity of continuous ISF flow along fibers of the PACT pathway was 3.6-15.6 mm/s.These data revealed that a PACT pathway was a'slit-shaped'porous biomaterial,comprising a longitudinal transport channel and an absorptive part for imbibition.The use of surfactants suggested that interfacial tension might play an essential role in layers of continuous ISF flow along vascular vessels.A hypothetical'gel pump'is proposed based on interfacial tension and interactions to regulate ISF flow.These experimental findings may inspire future studies to explore the physiological and pathophysiological functions of vascular ISF or interfacial fluid flow among interstitial connective tissues throughout the body.Hongyi LI You LYU Xiaoliang CHEN Bei LI Qi HUA Fusui JI Yajun YIN Hua LI 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,8:3

8Time and Location Aware Points of Interest Recommendation inLocation-Based Social Networks显示文摘The wide spread of location-based social networks brings about a huge volume of user check-in data, whichfacilitates the recommendation of points of interest (POIs). Recent advances on distributed representation shed light onlearning low dimensional dense vectors to alleviate the data sparsity problem. Current studies on representation learningfor POI recommendation embed both users and POIs in a common latent space, and users' preference is inferred basedon the distance/similarity between a user and a POI. Such an approach is not in accordance with the semantics of usersand POIs as they are inherently different objects. In this paper, we present a novel translation-based, time and locationaware (TransTL) representation, which models the spatial and temporal information as a relationship connecting users andPOIs. Our model generalizes the recent advances in knowledge graph embedding. The basic idea is that the embedding ofa 〈time, location〉 pair corresponds to a translation from embeddings of users to POIs. Since the POI embedding shouldbe close to the user embedding plus the relationship vector, the recommendation can be performed by selecting the top-kPOIs similar to the translated POI, which are all of the same type of objects. We conduct extensive experiments on tworeal-world data.sets. The results demonstrate that our TransTL model achieves the state-of-the-art performance. 11Ultra-homogeneous Cu_(2-x)Se nanoparticles as near-infraredⅡplasmonic phototheranstics for photothermal/chemodynamic synergistic therapy of liver cancer显示文摘Laser mediated photothermal/chemodynamic co-therapy is widely applied for anti-cancer treatment due to its good therapeutic effect.However,the laser wavelength for effectively targeting deep tumor tissues can decide whether or not it is a key premise of irradiation treatment.In this study,a super-homogenous and highly dispersive spherical nanoparticle Cu_(2-x)Se was prepared,which displayed a strong surface plasmon resonance(SPR)peak in the near-infrared region(NIR)-Ⅱand high Fenton catalytic efficiency.The results of in vitro experiments showed that Cu_(2-x)Se nanoparticles exhibited high photothermal conversion efficiency under the excitation of NIR-Ⅱlaser irradiation,which significantly improved its ability to produce OH.In a tumor-bearing mouse model,the combination of Cu_(2-x)Se and photothermal/chemodynamic co-therapy guided by the real-time photoacoustic imaging in the NIR-Ⅱcould effectively inhibit tumor growth and exhibit good in vivo/in vitro biocompatibility.In summary,this Cu_(2-x)Se nanoparticle-based NIR-Ⅱlaser-mediated photothermal/chemodynamic co-therapy strategy is expected to provide a new option for the diagnosis and treatment of liver cancer.Bei Li Jing Li Yi Fu Xiwen Ye Maodi Xie Lei Feng Xiaoya Niu Zhen You 2023Particuology2023,,5:0

14Constraints on black hole spins with a general relativistic accretion disk corona model显示文摘The peaks in the spectra of the accretion disks surrounding massive black holes in quasars are in the far-UV or soft X-ray band, which are usually not observed. However, in the disk corona model, soft photons from the disk are Comptonized to high energy in the hot corona, and the hard X-ray spectra(luminosity and spectral shape) contain information on the incident spectra from the disk. The values of black hole spin parameter a*are inferred from the spectral fitting, which are spread over a large range, ^-0.94 to 0.998. We find that the inclination angles and mass accretion rates are well determined by the spectral fitting, but the results are sensitive to the accuracy of black hole mass estimates. No tight constraints on the black hole spins are achieved, if the uncertainties in black hole mass measurements are a factor of four,which are typical for the single-epoch reverberation mapping method. Recently, the accuracy of black hole mass measurement has been significantly improved to 0.2- 0.4 dex with the velocity resolved reverberation mapping method. The black hole spin can be well constrained if the mass measurement accuracy is<50%. In the accretion disk corona scenario, a fraction of power dissipated in the disk is transported into the corona, and therefore the accretion disk is thinner than a bare disk for the same mass accretion rate,because the radiation pressure in the disk is reduced. 15Correlation between excitation index and Eddington ratio in radio galaxies显示文摘We use a sample of 111 radio galaxies with redshift z < 0.3 to investigate their nuclear properties.The black hole masses of the sources in this sample are estimated with the velocity dispersion/luminosity of the galaxies, or the width of the broad-lines. We find that the excitation index, the relative intensity of low and high excitation lines, is correlated with the Eddington ratio for this sample. The size of the narrow-line region(NLR) was found to vary with ionizing luminosity as RNLR∝L_(ion)^(0.25)(Liu et al. 2013). Using this empirical relation, we find that the correlation between the excitation index and the Eddington ratio can be reproduced by photoionization models. We adopt two sets of spectral energy distributions(SEDs), with or without a big blue bump in ultraviolet as the ionizing continuum, and infer that the modeled correlation between the excitation index and the Eddington ratio is insensitive to the applied SED. This means that the difference between high excitation galaxies and low excitation galaxies is not caused by the different accretion modes in these sources.
