| 1 | Genomic insights into the fast growth of paulownias and the formation of Paulownia witches' broom显示文摘Paulownias are among the fastest growing trees in the world,but they often suffer tremendous loss of wood production due to infection by Paulownia witches'broom(PaWB)phytoplasmas.In this study,we have sequenced and assembled a high-quality nuclear genome of Paulownia fortunei,a commonly cultivated paulownia species.The assembled genome of P.fortunei is 511.6 Mb in size,with 93.2%of its sequences anchored to 20 pseudo-chromosomes,and it contains 31985 protein-coding genes.Phylogenomic analyses show that the family Paulowniaceae is sister to a clade composed of Phrymaceae and Orobanchaceae.Higher photosynthetic efficiency is achieved by integrating C3 photosynthesis and the crassulacean acid metabolism pathway,which may contribute to the extremely fast growth habit of paulownia trees.Comparative transcriptome analyses reveal modules related to cambial growth and development,photosynthesis,and defense responses.Additional genome sequencing of PaWB phytoplasma,combined with functional analyses,indicates that the effector PaWB-SAP54 interacts directly with Paulownia PfSPLa,which in turn causes the degradation of PfSPLa by the ubiquitin-mediated pathway and leads to the formation of witches'broom.Taken together,these results provide significant insights into the biology of paulownias and the regulatory mechanism for the formation of PaWB. | Yabing Cao Guiling Sun Xiaoqiao Zhai Pingluo Xu Liming Ma Minjie Deng Zhenli Zhao Haibo Yang Yanpeng Dong Zhonghai Shang Yujie Lv Lijun Yan Haifang Liu Xibing Cao Bingbing Li Zhe Wang Xiaogai Zhao Haiyan Yu Fan Wang Wen Ma Jinling Huang Guoqiang Fan | 2021 | Molecular Plant2021,14,10: | 7 |

| 3 | High-risk phenotypes of genetic disease in a Neonatal Intensive Care Unit population显示文摘To the Editor: Genetic diseases contribute to 35% of deaths during the first year of life and are a significant cause of intensive care.[1] A previous study based on the China Neonatal Genomes Project investigated the genetic causes of early infant deaths and found that >25% of deceased neonates with genetic diagnoses can be cured if diagnosed in time.[2] Therefore, it is crucial to target and diagnose neonates with genetic diseases as early as possible. According to our experience, the typical phenotypes, such as special facial features or multiple congenital anomalies (MCAs), indicate a high risk of genetic disease and lead physicians to perform genetic testing in neonates as early as possible. However, in practice, infants without typical phenotypes typically undergo a long and costly diagnostic process before genetic diagnoses are confirmed. Moreover, a recent survey by the American College of Medical Genetics and Genomics (ACMG) and other national professional organizations indicated that there are insufficient numbers of qualified geneticists to fulfil genetic service needs.[3] The ACMG published the general clinical features for genetic testing indications. For example, patients with phenotypes or family history data that strongly implicate a genetic cause may undergo genetic testing.[1] However, the study indicated that many genetic conditions arise de novo or are inherited with no family history.[1] A previous study attempted to apply the non-phenotype-driven panel approach in neonates admitted to the neonate intensive care unit (NICU).[4] However, at present, the diagnostic yield is only 3.45% (1/29).[4] In addition, the economic and ethical issues associated with genomic screening remain challenging. | 15 | PICOTEES:a privacy-preserving online service of phenotype exploration for genetic-diagnostic variants from Chinese children cohorts显示文摘The growth in biomedical data resources has raised potential privacy concerns and risks of genetic information leakage. For instance, exome sequencing aids clinical decisions by comparing data through web services, but it requires significant trust between users and providers. To alleviate privacy concerns, the most commonly used strategy is to anonymize sensitive data. Unfortunately, studies have shown that anonymization is insufficient to protect against reidentification attacks. Recently, privacy-preserving technologies have been applied to preserve application utility while protecting the privacy of biomedical data. We present the PICOTEES framework, a privacy-preserving online service of phenotype exploration for genetic-diagnostic variants (https://birthdefectlab.cn:3000/). PICOTEES enables privacy-preserving queries of the phenotype spectrum for a single variant by utilizing trusted execution environment technology, which can protect the privacy of the user's query information, backend models, and data, as well as the final results. We demonstrate the utility and performance of PICOTEES by exploring a bioinformatics dataset. The dataset is from a cohort containing 20,909 genetic testing patients with 3,152,508 variants from the Children's Hospital of Fudan University in China, dominated by the Chinese Han population (>99.9%). | 19 | High area-capacity Mg batteries enabled by sulfur/copper integrated cathode design显示文摘Rechargeable Mg batteries potentially display lower cost and competitive energy density compared with their Li-ion counterparts.However,the practical implementation of high area-capacity cathodes still remains a formidably challenging task.This work presents the sulfur/copper integrated cathodes fabricated by the conventional blade-coating process and slurry-dipping method.The sulfur/copper foil integrated cathodes deliver a high area-capacity of 2.6 mAh cm^(-2)after 40 cycles,while the sulfur/copperfoam integrated cathode exhibits an ultrahigh area-capacity of 35.4 mAh cm^(-2),corresponding to 743.1 Wh L^(-1)at the electrode level(1.5 times higher than the LiCoO_(2)-graphite system).The in-situ formed copper sulfide intermediates with sufficient cation defects can act as functional intermediates to regulate the sulfur electrochemistry during the first discharge process.The subsequent cycles are operated by the reversible displacement reaction between Mg-ions and copper sulfide active substances.In particular,the copper ions prefer to extrude along the[001]direction in copper sulfides lattice and simultaneously the rock-salt MgS crystals are generated.Besides,the nonuniform surface topography of the cycled Mgmetal anode,caused by the spatial inhomogeneity in current distribution,is demonstrated to lead to the battery performance degradation for high area-capacity Mg batteries. | Zhenfang Zhou Aobing Du Weijie Kong Zhuang Chen Zhonghua Zhang Bingbing Chen Yitao He Shanmu Dong Zhenjiang Li Guicun Li Guanglei Cui | 2022 | Journal of Energy Chemistry2022,31,9: | 0 |
| 20 | Effect of CO_(2) dilution on laminar burning velocities,combustion characteristics and NO_(x) emissions of CH_(4)/air mixtures显示文摘The laminar combustion characteristics of CH_(4)/air premixed flames with CO_(2) addition are systemically studied.Experimental measurements and numerical simulations of the laminar burning velocity(LBV)are performed in CH_(4)/CO_(2)/Air flames with various CO_(2) doping ratio under equivalence ratios of 1.0–1.4.GRI 3.0 mech and Aramco mech are employed for predicting LBV,adiabatic flame temperature(AFT),important intermediate radicals(CH_(3),H,OH,O)and NO_(x) emissions(NO,NO_(2),N2O),as well as the sensitivity analysis is also conducted.The detail analysis of experiment and simulation reveals that as the CO_(2) addition increases from 0%to 40%,the LBVs and AFTs decrease monotonously.Under the same CO_(2) doping ratio,the LBVs and AFTs increase first and then decrease with the increase of equivalence ratio,and the maximum of LBV is reached at equivalence ratio of 1.05.The mole fraction tendency of important intermediates and NO_(x) with equivalence ratio and CO_(2) doping ratio are similar to the LBVs and AFTs.Reaction H+O_(2)⇔O+OH is found to be responsible for the promotion of the generation of important intermediates and NO_(x) under the equivalence ratios and CO_(2) addition through sensitivity analysis.The sensitivity coefficients of elementary reactions that the increasing of CO_(2) doping ratio promotes or inhibits formation of intermediate radicals and NO_(x) decreases. | Wenlong Dong Longkai Xiang Jian Gao Bingbing Qiu Huaqiang Chu | 2023 | International Journal of Coal Science & Technology2023,10,5: | 0 |
