1Genetic Analysis of Two Weak Dormancy Mutants Derived from Strong Seed Dormancy Wild Type Rice N22 (Oryza sativa)显示文摘Two weak dormancy mutants,designated Q4359 and Q4646,were obtained from the rice cultivar N22 after treatment with 400 Gy 60Co gamma-radiation.Compared to the N22 cultivar,the dormancy of the mutant seeds was more readily broken when exposed to a period of room temperature storage.The mutants also showed a reduced level of sensitivity to abscisic acid compared to the N22 cultivar,although Q4359 was more insensitive than Q4646.A genetic analysis indicated that in both mutants,the reduced dormancy trait was caused by a single recessive allele of a nuclear gene,but that the mutated locus was different in each case.The results of quantitative trait locus(QTL)mapping,based on the F2 population from Q4359 x Nanjing35,suggested that Q4359 lacks the QTL qSdn-1 and carries a novel allele at QTL qSdn-9, while a similar analysis of the Q4646 x Nanjing35 F2 population suggested that Q4646 lacks QTL qSdn-5, both qSdn-1 and qSdn-5 are major effect seed dormancy QTL in N22.Therefore,these two mutants were helpful to understand the mechanism of seed dormancy in N22.Bingyue Lu KunXie Chunyan Yang Long Zhang Tao Wu Xi Liu Ling Jiang Jianmin Wan 2011Journal of Integrative Plant Biology2011,53,5:7
2Combination of acupuncture and traditional Chinese medicine for functional constipation:A meta-analysis显示文摘To systematically assess the effectiveness and safety of using acupuncture with traditional Chinese medicine for functional constipation so as to provide the evidence-based data for clinical practice.Wanfang databases,CBM,VIP,CNKI,PubMed,Cochrane Library and EMbase were searched for trials based on randomization and control for use of acupuncture in combination with CHM for functional constipation published from the inception of the database to Dec.2019.We used the risk of bias of Cochrane Collaboration for literature quality assessment,extracted trial data in duplicate,and then assessed the risk of bias.The data were discussed by adopting Review Manager 5.3 program.A total of 22 RCTs with 2574 cases were retrieved.Meta-analysis showed that acupuncture with CHM could improve the clinical effectiveness(OR6.10,95%CI 4.72–7.87,P<0.00001);heterogeneity was not identified(Chi^(2)=32.84,P=0.43,I^(2)=2%),and the difference was statistically significant.Second-level outcomes(e.g.,defecation interval time(P<0.00001),Bristol Stool Scale(P=0.002),defecation difficulty(P=0.01)and accompanying symptoms(P<0.00001))in the treatment groups were significantly different after the acupuncture with CHM compared with control groups,whereas defecation symptoms were not significantly different(P=0.06).The use of acupuncture with CHM can significantly improve the clinical efficacy of functional constipation,ameliorate Bristol Stool Scale,defecation difficulty,defecation symptoms,and accompanying symptoms,as well as shorten the defecation interval time,with better safety.Dongxue Lu Feng Liu Jing Yan Haixia Liu Zhiguang Sun Bingyu Chen Shilin Zhu Fei Ge 2022Medicine in Novel Technology and Devices2022,,4:1

4Neo-intline: integrated pipeline enables neoantigen design through the in-silico presentation of T-cell epitope显示文摘Neoantigen vaccines are one of the most effective immunotherapies for personalized tumour treatment.The current immunogen design of neoantigen vaccines is usually based on whole-genome sequencing(WGS)and bioinformatics prediction that focuses on the prediction of binding affinity between peptide and MHC molecules,ignoring other peptide-presenting related steps.This may result in a gap between high prediction accuracy and relatively low clinical effectiveness.In this study,we designed an integrated in-silico pipeline,Neo-intline,which started from the SNPs and indels of the tumour samples to simulate the presentation process of peptides in-vivo through an integrated calculation model.Validation on the benchmark dataset of TESLA and clinically validated neoantigens illustrated that neo-intline could outperform current state-of-the-art tools on both sample level and melanoma level.Furthermore,by taking the mouse melanoma model as an example,we verified the effectiveness of 20 neoantigens,including 10 MHC-I and 10 MHC-II peptides.The in-vitro and in-vivo experiments showed that both peptides predicted by Neo-intline could recruit corresponding CD4^(+)T cells and CD8^(+)T cells to induce a T-cell-mediated cellular immune response.Moreover,although the therapeutic effect of neoantigen vaccines alone is not sufficient,combinations with other specific therapies,such as broad-spectrum immune-enhanced adjuvants of granulocyte-macrophage colony-stimulating factor(GM-CSF)and polyinosinic-polycytidylic acid(poly(I:C)),or immune checkpoint inhibitors,such as PD-1/PD-L1 antibodies,can illustrate significant anticancer effects on melanoma.Neo-intline can be used as a benchmark process for the design and screening of immunogenic targets for neoantigen vaccines.Bingyu Li Ping Jing Genhui Zheng Chenyu Pi Lu Zhang Zuojing Yin Lijun Xu Jingxuan Qiu Hua Gu Tianyi Qiu Jianmin Fang 2023Signal Transduction and Targeted Therapy2023,8,11:0
5Decrypting the Influence of Axial Coordination on the Electronic Microenvironment of Co-N_(5)Site for Enhanced Electrocatalytic Reaction显示文摘Metal porphyrins are star molecules that possess welldefined coordination metal centers for versatile catalytic reactions.However,most previous work has focused on the correlations between in-plane symmetric configuration of metal-N_(4)sites and their catalytic performance.Addressing the catalytic contribution of additional axial coordination to such symmetric configuration remains a challenge.Theoretical calculations revealed that axially anchoring an extra pyridine on the tetra-coordinated cobalt porphyrin(Co-N4)to construct penta-coordinated cobalt porphyrin(Co-N_(5))renders cobalt a higher electron density,thereby favoring the rate-determining O_(2)adsorption/activation and reducing the oxygen electroreduction barrier.Therefore,a well-defined Co-N_(5)site is rationally introduced into the azo-linked polymer framework for a fundamental structure-catalytic performance correlation study.As-prepared Co-N_(5)catalyst exhibits a 26 mV positive shift in half-wave potential compared with the pyridine-free Co-N_(4)counterpart,discloses a markedly higher power density(141.4 mW cm^(−2)),and possesses better long-term durability(over 160 h cycles)in a Zn-air battery.Moreover,such a Co-N_(5)catalyst also showcases potential applications for CO_(2)reduction with high CO_(2)-to-CO conversion faradic efficiency and better selectivity than the Co-N_(4)counterpart because coordination of the fifth pyridine evokes electronic localization that suppresses a competitive side reaction.This work proves the positive electrocatalytic contribution of axial penta-coordination on well-defined metalporphyrin-based catalysts and offers atomic understanding of the structure-performance correlation on single atom catalysts for future catalyst design.Bingyu Huang Senhe Huang Chenbao Lu Longbin Li Judan Chen Ting Hu Dirk Lützenkirchen-Hecht Kai Yuan Xiaodong Zhuang Yiwang Chen 2023CCS Chemistry2023,5,8:0
