1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29
2Cotton-derived oxygen/sulfur co-doped hard carbon as advanced anode material for potassium-ion batteries显示文摘Hard carbon is regarded as promising anode materials for potassium-ion batteries(KIBs)owing to their low price and easy availability.However,the limited rate capability still needs to be improved.Herein,we demonstrate the fabrication of oxygen/sulfur co-doped hard carbon through a facile hydrolyzationsulfuration process of skimmed cotton.The simultaneous dopants significantly improve potassium ion diffusion rate.When served as the anode for KIBs,this hydrolyzed hard carbon delivered a high reversible capacity(409 mAh/g at 0.1 A/g),superior rate capability(135 mAh/g at 2 A/g)and excellent cyclability(about 120 mAh/g overt 500 cycles at 2 A/g).This work provides a facile strategy to prepare low-cost doped-hard carbon with superior potassium storage property.Baolin Xu Shihan Qi Fang Li Xiaoxin Peng Jinfeng Cai Jiaojiao Liang Jianmin Ma 2020Chinese Chemical Letters2020,31,1:12
3The Epitope Study on the SARS-CoV Nucleocapsid Protein显示文摘The nucleocapsid protein (N protein) has been found to be an antigenic protein in a number of coronaviruses. Whether the N protein in severe acute respiratory syndrome-associated coronavirus (SARS-CoV) is antigenic remains to be elucidated. Using Western blot and Enzyme-linked Immunosorbent Assay (ELISA),the recombinant N proteins and the synthesized peptides derived from the N protein were screened in sera from SARS patients. All patient sera in this study displayed strong positive immunoreactivities against the recombinant N proteins,whereas normal sera gave negative immunoresponses to these proteins, indicating that the N protein of SARS-CoV is an antigenic protein. Furthermore, the epitope sites in the N protein were determined by competition experiments, in which the recombinant proteins or the synthesized peptides competed against the SARS-CoV proteins to bind to the antibodies raised in SARS sera. One epitope site located at the C-terminus was confirmed as the most antigenic region in this protein. A detailed screening of peptide with ELISA demonstrated that the amino sequence from Codons 371 to 407 was the epitope site at the C-terminus of the N protein. 4Genetic diversity and DNA fingerprinting in jute(Corchorus spp.) based on SSR markers显示文摘Genetic diversity analysis and DNA finger printing are very useful in breeding programs,seed conservation and management. Jute(Corchorus spp.) is the second most important natural fiber crop after cotton. DNA fingerprinting studies in jute using SSR markers are limited. In this study, 58 jute accessions, including two control varieties(Huangma 179 and Kuanyechangguo) from the official variety registry in China were evaluated with 28 pairs of SSR primers. A total of 184 polymorphic loci were identified. Each primer detected 3 to 15 polymorphic loci, with an average of 6.6. The 58 jute accessions were DNA-fingerprinted with 67 SSR markers from the 28 primer pairs. These markers differentiated the 58 jute accessions from one another, with Co SSR305-120 and Co SSR174-195 differentiating Huangma 179 and Kuanyechangguo, respectively. NTSYS-pc2.10 software was used to analyze the genetic diversity in the 58 jute accessions. Their genetic similarity coefficients ranged from 0.520 to 0.910 with an average of 0.749, indicating relatively great genetic diversity among them. The 58 jute accessions were divided into four groups with the coefficient 0.710 used as a value for classification, consistent with their species and pedigrees. 5Safety and efficacy of stent-assisted coiling for acutely ruptured wide-necked intracranial aneurysms:comparison of LVIS stents with laser-cut stents显示文摘Background:To compare the safety and efficacy of LVIS stent-assisted coiling with those of laser-cut stent-assisted coiling for the treatment of acutely ruptured wide-necked intracranial aneurysms.Methods:Patients with acutely ruptured wide-necked intracranial aneurysms treated with LVIS stent-assisted coiling(LVIS stent group)and laser-cut stent-assisted coiling(laser-cut stent group)were retrospectively reviewed from January 2014 to December 2017.Propensity score matching was used to adjust for potential differences in age,sex,aneurysm location,aneurysm size,neck width,Hunt-Hess grade,and modified Fisher grade.Perioperative procedure-related complications and clinical and angiographic follow-up outcomes were compared.Univariate and multivariate analyses were performed to determine the associations between procedure-related complications and potential risk factors.Results:A total of 142 patients who underwent LVIS stent-assisted coiling and 93 patients who underwent laser-cut stent-assisted coiling were enrolled after 1:2 propensity score matching.The angiographic follow-up outcomes showed that the LVIS stent group had a slightly higher complete occlusion rate and lower recurrence rate than the laser-cut stent group(92.7%vs 80.6%;3.7%vs 9.7%,P=0.078).The clinical outcomes at discharge and follow-up between the two groups demonstrated no significant differences(P=0.495 and P=0.875,respectively).The rates of intraprocedural thrombosis,postprocedural thrombosis,postoperative early rebleeding,and procedure-related death were 0.7%(1/142),1.4%(2/142),2.8%(4/142),and 2.1%(3/142)in the LVIS stent group,respectively,and 4.3%(4/93),2.2%(2/93),1.1%(1/93),and 3.2%(3/93)in the laser-cut stent group,respectively(P=0.082,0.649,0.651,and 0.683).Nevertheless,the rates of overall procedure-related complications and intraprocedural rupture in the LVIS stent group were significantly lower than those in the laser-cut stent group(5.6%vs 14.0%,P=0.028;0.7%vs 6.5%,P=0.016).Multivariate analysis showed that laser-cut stent-assisted coiling was an independent predictor for overall procedurerelated complications(OR=2.727,P=0.037);a history of diabetes(OR=7.275,P=0.027)and other cerebrovascular diseases(OR=8.083,P=0.022)were independent predictors for ischemic complications,whereas none of the factors were predictors for hemorrhagic complications.Conclusions:Compared with laser-cut stent-assisted coiling,LVIS stent-assisted coiling for the treatment of acutely ruptured wide-necked intracranial aneurysms could reduce the rates of overall procedure-related complications and intraprocedural rupture.Gaici Xue Qiao Zuo Xiaoxi Zhang Haishuang Tang Rui Zhao Qiang Li Yibin Fang Pengfei Yang Bo Hong Yi Xu Qinghai Huang Jianmin Liu 2021Chinese Neurosurgical Journal2021,7,2:8
6De novo assembly of chloroplast genomes of Corchorus capsularis and C.olitorius yields species-specific InDel markers显示文摘Jute(Corchorus spp.)is a member of the Malvaceae family,which comprises more than 100 species.The systematic positions of jute species have remained unsettled.Chloroplasts are maternally inherited and their genomes are widely used for plant phylogenetic studies.In the present study,the chloroplast genomes of Corchorus capsularis and C.olitorius were assembled,with sizes of respectively 161,088 and 161,766 bp.Both genomes contained 112 unique genes(78 protein-coding,four rRNA,and 30 tRNA genes).Four regionswith high variation between the two species were located in single-copy rather than inverted-repeat regions.A total of 66 simple sequence repeats(SSRs)were identified in the C.capsularis chloroplast genome and 56 in that of C.olitorius.Comparison of the two chloroplast genome sequences permitted the evaluation of nucleotide variation including 2417 single-nucleotide polymorphisms sites and 294 insertion or deletion sites,of which one marker(cpInDel 205)could discriminate the two jute species.Comparison of the C.capsularis and C.olitorius chloroplast genomeswith those of other species in theMalvaceae revealed breakpoints in the accD locus,which is involved in fatty acid synthesis,in C.capsularis and C.olitorius.This finding suggests that genes from the chloroplast genomemight have been transferred to the nuclear genome in some Corchorus species.This hypothesis was supported by synteny analysis of the accD region among the nuclear,chloroplast,and mitochondrial genomes.To our knowledge,this is the first report of the assembled chloroplast genome sequences of C.capsularis and C.olitorius.C.capsularis and C.olitorius are closely related to Gossypium species and there are abundant microstructure variations between these two genera.These results will expand our understanding of the systematics of species in the Malvaceae.Shusheng Fang Liemei Zhang Jianmin Qi Liwu Zhang 2021The Crop Journal2021,9,1:5
7Value of Ecosystem Services in Conventional and Organic Rice Paddies:A Case Study in Wannian,Jiangxi,China显示文摘In the current study,the ecosystem services(ES) of conventional and organic rice paddies in Wannian,Jiangxi Province,China are investigated.First,the ES at the field level under organic and conventional paddies were investigated.Total economic value of ES in organic rice paddies was 30093.08 yuan RMB/ha per year and that of conventional rice paddies was 22 793.31 yuan RMB/ha per year.The total indirect value of ES was 14 813.7 yuan RMB/ha per year in organic rice paddies and 12 424.56 yuan RMB/ha per year in conventional ones.There were significant differences between organic and conventional rice paddies for the economic values.Then,this economic information was used to extrapolate and to calculate the total and indirect value of ES from rice paddies in Wangnian.The total and indirect economic values of ES from conventional rice paddies in Wannian were 6791 million and 3702 million yuan RMB per year respectively,and the total and indirect economic values of ES from organic rice paddies in Wannian were 1311 million and 646 million yuan RMB per year.If half the area had being converted to organic farming in Wannian,the total and indirect economic values of ES from conventional rice paddies were 3397 million and 1851 million yuan RMB per year,and the total and indirect economic values of ES from organic rice paddies were 5794 million and 2852 million yuan RMB per year.Finally,the total economic value of ES in rice paddies in Wannian was demonstrated through geographic information system techniques.Zhang Dan Min Qingwen He Lu Cheng Shengkui Fang Jianmin 2010Chinese Journal of Population,Resources and Environment2010,8,2:5

It can also be used for the macro-study of repeat sequences. Since HMW DNA during preparation is inclined to be sheared physically and digested by internal nucleases, it is very difficult to prepare the HMW DNA. 10First experimental constraints on WIMP couplings in the effective field theory framework from CDEX显示文摘We present weakly interacting massive particles(WIMPs) search results performed using two approaches of effective field theory from the China Dark Matter Experiment(CDEX), based on the data from both CDEX-1B and CDEX-10 stages. In the nonrelativistic effective field theory approach, both time-integrated and annual modulation analyses were used to set new limits for the coupling of WIMP-nucleon effective operators at 90% confidence level(C.L.) and improve over the current bounds in the low mχregion. In the chiral effective field theory approach, data from CDEX-10 were used to set an upper limit on WIMP-pion coupling at 90% C.L. 12Structurally tunable characteristics of ionic liquids for optimizing lithium plating/stripping via electrolyte engineering显示文摘Electrolyte chemistry offers the opportunity to regulate the solid electrolyte interphase(SEI) and Li^(+)solvation,which is considered to be crucial to the growth of lithium crystals for safe lithium metal batteries(LMBs).Structurally tunable characteristics of ionic liquids(ILs) from anion type,cationic substituent chain length and cationic substituents,will contribute this field.Here,we explore the influence mechanism of imidazole-based ILs as electrolyte additives on Li+solvation and the formation of SEI.ILs can participate into the formation of efficient SEI,together with cathode electrolyte interphase(CEI).Moreover,ILs can also regulate the sheath structure of Li^(+)solvation,to fasten the kinetics of Li.Furthermore,the imidazole-based cations with long alkyl chain can form an electrostatic shield around newly formed Li nucleus,and suppress further Li plating at this site.Under the optimized condition,the 1-octyl-3-methylimidazolium bis(trifluoromethylsulfonyl)imide([OMIm]TFSI) additive shows the best ability to enhance the electrochemical performance,endowing the Li||Li symmetric cell with a stable life(over800 h) at 0.5 mA cm^(-2) and the Li||LiNi_(0.6)Mn_(0.2)Co_(0.2)O_(2)(NMC622) full cell with a high capacity of 141.7 mAh g^(-1) after 200 cycles at 0.5 C.Shihan Qi Jiandong Liu Jian He Huaping Wang Mingguang Wu Daxiong Wu Junda Huang Fang Li Xin Li Yurong Ren Jianmin Ma 2021Journal of Energy Chemistry2021,30,12:2

14Pseudo-concentrated electrolytes for lithium metal batteries显示文摘Lithium metal batteries suffer from short lifespans and low Coulombic efficiency (CE) due to the high reactivity of Li and the poor stability of the solid electrolyte interphase (SEI). Herein, we propose the concept of a pseudo-concentrated electrolyte (PCE) induced by an electron-deficient additive (4-pyridylboronic acid;4-PBA) to form a robust, LiF-rich SEI, thus addressing the above issues. Molecular dynamics simulations confirm that 4-PBA can increase the coordination number of PF6^(-) anions in the Li+ solvation sheath to achieve pseudo-concentrated LiPF6 in the electrolyte. Moreover, the 4-PBA can scavenge harmful PF5 decomposed from LiPF6 to stabilize the LiF-rich SEI. The resulting robust LiF-rich SEI promotes Li growth along the SEI/Li interface and represses the growth of Li dendrites. Thus, excellent performance is achieved, with a high CE of 97.1% for a Li||Cu cell at 1.0 ​mA ​cm^(−2), and over 950 cycles at 0.5 ​mA ​cm^(−2) for Li||Li symmetric cells with 1.0 ​wt% 4-PBA electrolyte.
