| 6 | Pore Size Distribution and Its Quantitative Relationship with Strength of Corundum Based Castables显示文摘A series of corundum based castables with 0,2%,4%,6%,and 8% α-Al2O3 micropowders were prepared using tabular alumina aggregates (6-3,3-1 and ≤1 mm) and fines (≤0.088 and ≤0.045 mm),calcium aluminate cement,and α-Al2O3 micropowders (d50=1.754 μm) as starting materials. Cold mechanical strength and pore size distribution of the castables specimens after heat treatment at 110,1 100 and 1 500 ℃ were tested,respectively. The quantitative relationship between strength and apparent porosity,and that between strength and median pore diameter were verified by Atzeni equation. The correlation between interval of pore size and mechanical strength of specimens was also studied by means of gray relational theory. The results show that:(1) the pore size distribution of castables is strongly influenced by both micropowders filling and matrix sintering; the addition of micropowders decreases median pore diameter while the sintering process increases it; (2) when adding a constant correction term,Atzeni equation can substantially describe the quantitative relationship between median pore diameter and strength of castables specimens after heat treatment at the same temperature; the significant differences of the gray relational degree between the interval of pore size and castables strength are characterized; it is also found that for the same interval of pore size,the gray relational degree isaffected by the heat treatment temperature; the pore size interval <0.5 μm has the highest gray relational degree with the strength at 110-1 500 ℃. | FANG Binxiang ZHU Boquan LI Xiangcheng WAN Hongbo | 2010 | China's Refractories2010,19,4: | 2 |

| 14 | Enzyme-linked immunosorbent assay of changes in serum levels of growth hormone(cGH)in common carps(Cyprinus carpio)显示文摘The aim of the present study was to purify the common native carp growth hormone (ncGH), produce monoclonal antibodies (mAbs) to common native carp growth hormone (ncGH), and further enhance the sensitivity of enzyme-linked immunosorbent assays (ELISA) for ncGH. Additionally, we investigated changes in serum ncGH levels in carps raised in different environmental conditions. The recombinant grass carp (Ctenopharyngodon idella) growth hormone was purified and used as antigen to immunize the rabbit. The natural ncGH was isolated from the pituitaries of common carp. SDS-PAGE and Western blot utilizing the polyclonal anti-rgcGH antibody confirmed the purification of ncGH from pituitaries. Purified ncGH was then used as an immunogen in the B lymphocyte hybridoma technique. A total of 14 hybridoma cell lines (FMU-cGH 1-14) were established that were able to stably secrete mAbs against ncGH. Among them, eight clones (FMU-cGH1-6, 12 and 13) were successfully used for Western blot while nine clones (FMU-cGH 1-7, 9 and 10) were used in fluorescent staining and immunohistochemistry. Epitope mapping by competitive ELISA demonstrated that these mAbs recognized five different epitopes. A sensitive sandwich ELISA for detection of ncGH was developed using FMU-cGH12 as the coating mAb and FMU-cGH6 as the enzyme labeled mAb. This detection system was found to be highly stable and sensitive, with detection levels of 70 pg/mL. | 17 | Thrombopoietin induced proliferation and differentiation of fetal liver CD34^+ cells with phenotype change from hemopoiesis to neurogenesis显示文摘BACKGROUND: Previous studies have reported a neurotrophin-like motif in the N-terminal receptor binding region of the thrombopoietin (TPO) molecule, and have described localization of TPO and TPO receptor in the brain. Therefore, it is believed that TPO may be involved in regulation of neurogenesis. OBJECTIVE: To validate the effect of TPO on trans-differentiation, or differentiation from hematopoietic stem cells (HSCs) to neural stem cells (NSCs). DESIGN, TIME AND SETTING: Comparative studies were performed from March 2004 to April 2007 at the Department of Experimental Medicine, Northern Hospital, and the Department of Immunology, Fourth Military Medical University of Chinese PLA. MATERIALS: Human fetal liver (FL) was obtained from fetuses after water-balloon abortion. Gestational age ranged from 16 to 20 weeks. The study was approved by the Institutional Review Board and Ethics Committee of the Northern Hospital. TPO was kindly provided by Genentech Inc (USA). Iscove's Modified Dulbecco's Medium (IMDM) and neurobasalTM medium were purchased from Invitrogen (USA). MACS CD34 multisort kit was purchased from Miltenyi Biotec (Germany). METHODS: CD34+ cells were isolated from human FL mononuclear cells using MACS CD34 multisort kit and cultured at 1 × 105/mL in IMDM, containing TPO for 60 days with weekly changes of half of the medium. After culturing for 30 and 60 days, the TPO-induced cells were resuspended in neurobasalTM medium containing 10% fetal brain extracts and plated in an 8-well BIOCOAT poly-D-Lysine Culture Slide and cul- tured for another 7 days. MAIN OUTCOME MEASURES: Cell number, viability, phenotype and expression of hemopoiesis-related and neurogenesis-related proteins were examined by trypan blue exclusion with hemocytometer, immunoblot, immunocytochemistry and flow cytometry. RESULTS: After 60 days of induction with TPO, the cell number increased by 4.6-fold compared to the ini- tial culture. Although the proportion of the cells expressing the hemopoietic stem cell associated antigen (CD34) decreased steadily, both proportions of the cultured FL-derived CD34+ cells expressing CD41a and CD61 remained unchanged, which still accounted for 10%. Noticeably, the proportions of the cells express- ing nestin and epidermal growth factor receptor increased significantly (both > 50%), whereas the expression of more mature neural or glial proteins [microtubule-associated protein-2 (MAP2), glial fibrillary acidic pro- tein (GFAP), oligodendrocyte marker O4 (O4)] markers on the cultured fetal liver derived-CD34+ cells were at lower levels. After another 7 days incubation in neurobasal? medium, these TPO-induced cells formed neurospheres, which were labeled with nestin, and differentiated into cells with morphological characteristics of neurons, astrocytes and oligodendrocytes, which were labeled with MAP-2, GFAP, and O4, respectively.
