1人源抗滋养层细胞表面抗原-2基因工程抗体Fab的制备及特性分析显示文摘应用噬菌体抗体库技术制备全人源抗滋养层细胞表面抗原-2(Trop-2)特异性Fab抗体片段.抗体库经细胞筛选和固相抗原筛选,获得特异性的阳性克隆.阳性载体经核酸序列分析后,构建工程菌,经IPTG诱导表达,SDS-PAGE和Western blot分析,呈现28ku和32ku大小的两条蛋白质条带.Fab分子经流式细胞术、细胞免疫荧光检测,结果表明,Fab能够与BxPc3细胞膜蛋白特异性结合,而与NIH3T3细胞不结合.免疫共沉淀与质谱分析结果表明,该Fab分子能够与Trop-2蛋白特异性结合.免疫组化显示,该抗体可结合胰腺癌细胞膜蛋白,在细胞培养液中加入Fab,能够抑制BxPc3细胞的生长.以上研究结果提示,该抗体有望成为胰腺癌临床影像诊断或治疗的候选分子.林红 梁洁 张慧林 唐奇 苏亦平 Brian Cao 朱进 管晓虹 2010生物化学与生物物理进展2010,37,10:5
2Generation and characterization of an anti-GP73 monoclonal antibody for immunoblotting and sandwich ELISA显示文摘Recently,serum Golgi protein 73(GP73) levels have been found to be elevated in patients with hepatocellular carcinoma(HCC),and GP73 has been proposed as a novel marker for HCC.However,GP73 levels in patients remain controversial due to the specificity of the anti-GP73 antibody-based enzyme linked immunosorbent assay(ELISA).Therefore,an anti-GP73 antibody with high specificity was highly demanded.In the present study,by hybridoma screening,we generated an anti-GP73 monoclonal antibody(mAb) designated as 6A2 using recombinant GP73 protein produced by prokaryotic expression.The specificity of 6A2 was evaluated by Western blotting,immunohistochemistry and immunoprecipitation.The results showed that 6A2 recognized GP73 in both native and denatured forms.In addition,we have developed a sandwich ELISA using 6A2 and GP73 polyclonal antibody generated in New Zealand white rabbits according to standard procedures,and measured the serum GP73 level of patients using this assay.Our results showed that serum GP73 levels of HCC patients were significantly higher than those of healthy controls(P = 0.0036).Furthermore,for the first time,GP73 serum level was found to be elevated in patients with breast cancer compared with healthy controls(P = 0.0172).Aixia Zhang Brian Cao 2012The Journal of Biomedical Research2012,26,6:4
3Identification of two aberrant transcripts derived from a hybridoma with amplification of functional immunoglobulin variable genes显示文摘Murine monoclonal antibodies(mAbs)are widely used but have limitations if administered in humans.The use of chimeric or humanized mAbs can reduce immunogenicity.The first step in producing such mAbs is to clone murine variable genes from a hybridoma,but it is possible to amplify both functional and aberrant variable genes,as they coexist in the hybridoma.During the development of a murine–human chimeric antibody,we have cloned from a hybridoma the functional heavy chain variable region(VH)and light chain variable region(VL)genes of a mAb that blocks the binding of anthrax lethal factor to protective antigen.In this study,we report the detection of two aberrant transcripts from a hybridoma produced using myeloma cell line OUR-1,the development of a method to distinguish between the functional and abundant aberrant VL transcripts,and the origins of these aberrant genes.The aberrant VL gene is derived from OUR-1 cells,while the aberrant VH gene might derive from antibody repertoires in B cells or from gene rearrangement in the hybridoma cells.The aberrant VH and VL genes in this study may facilitate discrimination between the functional and aberrant variable genes from hybridoma cells.Guipeng Ding Ximin Chen Jin Zhu Brian Cao 2010Cellular & Molecular Immunology2010,7,5:2

5Development and optimization of an antibody array method for potential cancer biomarker detection显示文摘Biomarkers play an important role in the detection at an early stage of pancreatic cancer. The aim of the present study was to optimize the conditions of antibody arrays for detecting Hippocalcin-like 1 (HPCAL1), phosphatidylethanolamine binding protein 1 (PEBP1), lectin galactoside-binding soluble 7 (LGALS7), and serpin peptidase inhibitor clade E member 2 (SERPINE2) as biomarkers for pancreatic cancer detection in a single assay and to investigate antibodies' specificity and cross-reactivity. Capture antibodies against HPCAL1, PEBP1, LGALS7 and SERPINE2 were printed on nitrocellulose coated glass slides. HPCAL1, PEBP1, LGALS7 and SERPINE2 proteins with different concentrations were incubated with the capture antibodies at different temperatures for different time periods. Biotinylated detection antibodies recognizing a different epitope on the captured proteins and a secondary detection molecule (Streptavidin-PE) were used to detect fluorescent signals. The arrays showed the strongest signals when the concentration of the capture antibodies was at 500 μg/mL in PBST0.05 (PBS with 0.05% Tween-20), and the slides were incubated overnight at 4°C. The lowest protein concentration for detection was 2 ng/mL. Each antibody demonstrated high specificity to the corresponding antigen in detecting a mixture of 4 proteins without significant cross-reactivity. The fluorescence and biomarker concentration displayed a linear correlation. 6Methane oxidation by green oxidant to methanol over zeolite-based catalysts显示文摘To reduce greenhouse gas emission from oil and gas production,it is essential to better convert methane to useful chemicals(rather) than to flare it.Conversion of methane to liquid oxygenates(mainly methanol) has attracted extensive attention and countless efforts have been made;however,running this reaction in a green,efficient,and practical way has remained elusive.The novel catalyst and oxidants play a critical role in activating methane and converting it to oxygenates(methanol).In this review,the work of commonly used oxidants for methane partial oxidation have been summarized,in which,earth abundant oxidants,O;and H;O are promising.Moreover,H;or CO can activate O;to produce H;O;that catalyzes methane partial oxidation more efficiently and selectively than O;or H;O.Therefore,the work of using reducing agent,such as CO and H;have been reviewed,focusing on rational catalyst design that features multifunction(H;O;production and CH;activation).The novel catalyst design has advanced this reaction towards practicality with green oxidants and H;using zeolites-based catalyst.Environmentally friendly zeolite preparation methods and novel two-dimensional(2 D) zeolites that can reduce waste,improve synthesis and catalytical performance substantially are also reviewed in this work to provide insights for a more comprehensive approach to meet the environment protection needs.Sufeng Cao Ke Zhang Brian Hanna Essam Al-Sayed 2022Chinese Chemical Letters2022,33,4:1

9Translational research case studies:Development of antibodies for cancer diagnosis and therapy显示文摘Antibodies are primary tools in several areas of biomedical sciences,including basic research,diagnostics,and molecular therapeutics.Antibodies are widely used in diagnostic applications for clinical medicine.Analysis of cells and tissues in pathology laboratories includes the use of antibodies on tissue sections.Further,antibodies are making rapid inroads into medical therapeutics,driven by technological evolution from chimeric and humanized to fully human antibodies.The therapeutic antibody market has the potential to reach $30 billion by 2010.Our lab has developed a monoclonal antibody,named Met4 that was raised against the extracellular domain of Met specifically with the goal of measuring Met in FFPE tissues.The Met receptor kinase is expressed on the cell surface of a significant number and variety of human primary solid tumors and in their metastases.The characterization of the Met4 antibody suggests it should possess adequate performance for quantification of Met expression in clinical specimens.We have also generated a fully human Fab fragment against EGFR;conjugated it to taxol as an immuno-chemotherapy agent;and investigated its in vitro anti-tumor efficacy on EGFR positive A431 epidermoid carcinoma cells.The Fab-Taxol conjugate inhibited A431 cell proliferation at low concentrations and in a dose-responsive manner;furthermore,almost 100% of cells underwent apoptosis after treatment with Fab-Taxol.Our findings suggest that this Fab-Taxol conjugate could be a potential immuno-chemotherapeutic drug for clinical treatment of EGFR over-expressing tumors.Brian Cao 2007Journal of Nanjing Medical University2007,21,6:1

12A mechanistic consideration of oxygen enhancement ratio, oxygen transport and their relevancies for normal tissue sparing under FLASH irradiation显示文摘Our study investigated the role of oxygen in mediating the FLASH efect.This efect,which was frst reported in vitro in the 1950s and in vivo in the 1970s,recently gained prominence with a number of publications showing diferential sparing between normal tissues and tumors.Oxygen depletion(and subsequent induction of transient hypoxia)is the oldest and most prominent hypothesis to explain this efect.To better understand how the oxygen depletion hypothesis and oxygen enhancement ratio(OER)are relevant for interpreting FLASH benefts,an analytical model was proposed to estimate the sparing factor.The model incorporated factors such as OER,oxygen partial pressure(pO_(2)),loco-regional oxygen difusion/metabolism,total dose and dose rate.The sparing factor,was used to quantify the sparing of normal tissue(initially physoxic).The radiosensitivity parameters of two cell types(V79 Chinese hamster cells and T1 human kidney cells)were selected.Furthermore,the transient behavior of OER during fnite time intervals was modeled,for both without and with the presence of oxygen transport using a difusion model.For tissues with an oxygen consumption rate of 20mmHg/s and a distance of 60μm away from blood vessels,the sparing factor demonstrates an increase from 1.03/1.06(V79/T1)at 2.5Gy/s up to 1.28/1.72(V79/T1)at 100Gy/s(total dose:10Gy).For normal tissues of initial pO_(2) between 1.5 and 8mmHg,the beneft from pushing the dose rate above 100Gy/s is found to be marginal.Preliminary animal experiments have been conducted for validation.Overall,our study predicts that the dose rate associated with maximum normal tissue protection is between 50Gy/s and 100Gy/s.Other than the postulation of the hypoxic stem cell niches in normal tissues,we believe that a framework based upon the oxygen depletion hypothesis and OER is not able to efciently interpret diferential responses between normal and tumor tissue under FLASH irradiation.Mengyu Jia Xu Cao Brian W.Pogue Hao Peng 2022Holistic Integrative Oncology2022,1,1:0
