1Manipulating mRNA splicing by base editing in plants显示文摘Precursor-mRNAs(pre-mRNA) can be processed into one or more mature m RNA isoforms through constitutive or alternative splicing pathways. Constitutive splicing of pre-mRNA plays critical roles in gene expressional regulation, such as intronmediated enhancement(IME), whereas alternative splicing(AS) dramatically increases the protein diversity and gene functional regulation. However, the unavailability of mutants for individual spliced isoforms in plants has been a major limitation in studying the function of mRNA splicing. Here, we describe an efficient tool for manipulating the splicing of plant genes. Using a Cas9-directed base editor, we converted the 5′ splice sites in four Arabidopsis genes from the activated GT form to the inactive AT form. Silencing the AS of HAB 1.1(encoding a type 2 C phosphatase) validated its function in abscisic acid signaling, while perturbing the AS of RS31 A revealed its functional involvement in plant response to genotoxic treatment for the first time. Lastly,altering the constitutive splicing of Act2 via base editing facilitated the analysis of IME. 2Human placenta-derived mesenchymal stem cells loaded on linear ordered collagen scaffold improves functional recovery after completely transected spinal cord injury in canine显示文摘Traumatic spinal cord injury(SCI) is a major challenge in the clinic. In this study, we sought to examine the synergistic effects of linear ordered collagen scaffold(LOCS) and human placenta-derived mesenchymal stem cells(hPMSCs) when transplanted into completely transected beagle dogs. After 36 weeks observation, we found that LOCS+hPMSCs implants promoted better hindlimb locomotor recovery than was observed in the non-treatment(control) group and LOCS group. Histological analysis showed that the regenerated tissue after treatment was well integrated with the host tissue, and dramatically reduced the volume of cystic and chondroitin sulfate proteoglycans(CSPGs) expression. Furthermore, the LOCS+hPMSCs group also showed more neuron-specific βIII-tubulin(Tuj-1)-and NeuN-positive neurons in the lesion area, as well as axonal regeneration, remyelination and synapse formation in the lesion site. Additionally, dogs in the LOCS+hPMSCs group experienced enhanced sprouting of both ascending(CGRP-positive) sensory fibers and descending(5-HT-and TH-positive) motor fibers at the lesion area. All these data together suggested that the combined treatment had beneficial effects on neuronal regeneration and functional improvement in a canine complete transection model. 3Aberrant activation of latent transforming growth factor-β initiates the onset of temporomandibular joint osteoarthritis显示文摘There is currently no effective medical treatment for temporomandibular joint osteoarthritis(TMJ-OA) due to a limited understanding of its pathogenesis. This study was undertaken to investigate the key role of transforming growth factor-β(TGF-β)signalling in the cartilage and subchondral bone of the TMJ using a temporomandibular joint disorder(TMD) rat model, an ageing mouse model and a Camurati–Engelmann disease(CED) mouse model. In the three animal models, the subchondral bone phenotypes in the mandibular condyles were evaluated by μCT, and changes in TMJ condyles were examined by TRAP staining and immunohistochemical analysis of Osterix and p-Smad2/3. Condyle degradation was confirmed by Safranin O staining, the Mankin and OARSI scoring systems and type X collagen(Col X), p-Smad2/3 a and Osterix immunohistochemical analyses. We found apparent histological phenotypes of TMJ-OA in the TMD, ageing and CED animal models, with abnormal activation of TGF-βsignalling in the condylar cartilage and subchondral bone. Moreover, inhibition of TGF-β receptor I attenuated TMJ-OA progression in the TMD models. 5The analysis of water vapor distribution over Taklimakan Desert显示文摘Up to now, all analysis of the distribution of water vapor over the Taklimakan desert area only depends on limited ground measurements and radio soundings setting mostly on the outer margin area. This paper establishes an approach to retrieve the water vapor over the desert at high temporal and spatial resolutions by the use of FY2C geostationary satellite split-window channels in cooperation with ground-based GPS water vapor measurement. Results show that the water vapor distribution over the Taklimakan desert is affected highly by topography and surface properties. The outer margin area has generally more water vapor than the inner area. Over the outer margin area, the western part has more water vapor than the eastern part, and the northern part has more than the southern part. The driest area lies to the south of Tazhong, east of Hotan River, and extended to the south boundary of the desert. Similar to elsewhere, water vapor over the desert area shows diurnal, monthly, seasonal and annual variations even at the driest inner area of the desert. 8Zircon U-Pb Age,Geochemical,and Sr-Nd-Pb-Hf Isotopic Constraints on the Time Frame and Origin of Early Cretaceous Mafic Dykes in the Wuling Mountain Gravity Lineament,South China显示文摘In view of the importance of mafic dyke swarms and their contribution to current scientific problems relating to South China,herein,we present the findings of studies on twenty–five representative mafic dykes cropping out in Hunan Province and Guangxi Zhuang Autonomous Region,within the southern Wuling Mountain gravity lineament,China.These results include new zircon LA-ICP-MS U-Pb age,whole rock geochemical,Sr-Nd-Pb isotopic,and zircon Hf isotopic data for these dykes.The dykes formed between 131.5±1.2 and 121.6±1.1 Ma,and have typical doleritic textures.They fall into the alkaline and shoshonitic series,are enriched in light rare earth elements(LREE),some large ion lithophile elements(LILE;e.g.,Rb,Ba,and Sr),Th,U,and Pb,and are depleted in Nb,Ta,Hf,and Ti.Moreover,the dolerites have high initial 87 Sr/86 Sr ratios(0.7055–0.7057),negativeεNd(t)and zirconεHf(t)values(-14.8 to-11.9,-30.4 to-14.9),and relatively constant initial Pb isotopic ratios(that are EM1-like,16.77–16.94,15.43–15.47,and 36.84–36.92 for 206 Pb/204 Pb,207 Pb/204 Pb,and 208 Pb/204 Pb,respectively).These results indicate that the dykes were likely derived from magma generated through low-degree partial melting(1.0%–10%)of an EM1-like garnet–lherzolite mantle source.The parental magmas fractionated olivine,clinopyroxene,plagioclase,and Ti-bearing phases with negligible crustal contamination,during ascent and dyke emplacement.Several possible models have been proposed to explain the origin of Mesozoic magmatism along the Wuling Mountain gravity lineament.Herein we propose a reasonable model for the origin of these mafic dykes,involving the collision between the paleo-Pacific Plate and South China,which led to subsequent lithospheric extension and asthenosphere upwelling,resulting in partial melting the underlying mantle lithosphere in the Early Cretaceous,to form the parental magmas to the WMGL mafic dykes,as studied.CHEN Xiaoqing LIU Shen FENG Caixia XU Jingyan ZHAO Huibo FENG Guangying Ian M.COULSON SUN Jinggui FAN Yan 2021Acta Geologica Sinica(English Edition)2021,95,2:1
9Tricarbocyclic core formation of tyrosine-decahydrofluorenes implies a three-enzyme cascade with XenF-mediated sigmatropic rearrangement as a prerequisite显示文摘Tyrosine-decahydrofluorene derivatives feature a fused[6.5.6]tricarbocyclic core and a 13-membered para-cyclophane ether.Herein,we identified new xenoacremones A,B,and C(1-3)from the fungal strain Xenoacremonium sinensis ML-31 and elucidated their biosynthetic pathway using gene deletion in the native strain and heterologous expression in Aspergillus nidulans.The hybrid polyketide synthaseenonribosomal peptide synthetase(PKS-NRPS)XenE together with enoyl reductase XenG were confirmed to be responsible for the formation of the tyrosine-nonaketide skeleton.This skeleton was subsequently dehydrated by XenA to afford a pyrrolidinone moiety.XenF catalyzed a novel sigmatropic rearrangement to yield a key cyclohexane intermediate as a prerequisite for the formation of the multi-ring system.Subsequent oxidation catalyzed by XenD supplied the substrate for XenC to link the para-cyclophane ether,which underwent subsequent spontaneous Diels-Alder reaction to give the end products.Thus,the results indicated that three novel enzymes XenF,XenD,and XenC coordinate to assemble the[6.5.6]tricarbocyclic ring and para-cyclophane ether during biosynthesis of complex tyrosine-decahydrofluorene derivatives.Zhiguo Liu Wei Li Peng Zhang Jie Fan Fangbo Zhang Caixia Wang Shuming Li Yi Sun Shilin Chen Wenbing Yin 2021Acta Pharmaceutica Sinica B2021,11,11:1
10Three-dimensional theoretical analysis of seepage field in front of shield tunnel face显示文摘To evaluate hydraulic head distribution in front of a shield tunnel in a saturated soil layer,theoretical analysis and numerical simulations are carried out in this study.Based on the partial differential equilibrium equation of seepage flow,a three-dimensional(3D)theoretical analytical model of the shield tunnel face and the seepage field in front of it is established using the eigenfunction and the Fourier series expansion methods,and the hydraulic head calculation formula is derived.Combined with engineering cases,the theoretical analysis results and the 3D numerical simulation results are compared and analyzed.The effect of the water pressure of the tunnel face on the hydraulic head distribution is also analyzed.The results of the proposed analytical solution are in agreement with those of the numerical simulation solutions;moreover,the proposed analytical solution requires less time to calculate the seepage hydraulic head than the numerical simulation.The ratio of the initial water table to the diameter(D)of tunnel face has a more significant impact on the hydraulic head distribution at a position 0.5D above the tunnel vault.When the water pressure on the tunnel face is not considered,the values of the hydraulic head are significantly underestimated.Qiguang Di Pengfei Li Mingju Zhang Caixia Guo Fan Wang Jie Wu 2022Underground Space2022,7,4:1
11Changes in the Urban Surface Thermal Environment of a Chinese Coastal City Revealed by Downscaling MODIS LST with Random Forest Algorithm显示文摘Analysis of the formation and evolution of urban surface thermal environment is crucial for urban planning and improving the environment of a settlement.Qingdao was selected in this study as a typical coastal city undergoing rapid urbanization,and the spatiotemporal dynamic characteristics of its urban surface thermal environment from2010 to 2019 were evaluated.The random forest(RF)algorithm was adopted to obtain its land surface temperature(LST)map with 30-m resolution by downscaling the Moderate Resolution Imaging Spectroradiometer(MODIS)LST product;the remote sensing indices emphasizing different land cover types,LST calculated by the radiative transfer equation,and elevation were used as input variables in the algorithm.The heat island intensity(HII),urban heat island(UHI)volume,and UHI grade were used to analyze the spatiotemporal dynamic characteristics of the urban surface thermal environment in Qingdao.The results show an increasing trend in average HII between 1.1 and 2.52°C in the study area over the past 10 years.The northern city appeared to have the highest UHI volume,while change of the UHI volume in Huangdao District of southwestern Qingdao was the most significant.The areas with high HII have gradually expanded during the last 10 years,and the areas with a 10-yr persistently high HII are distributed mainly in old urban areas with high building density and a dense population.Different factors may influence UHI,such as artificial heat sources,surface heat sources,and hybrid heat sources.Finally,adjusting the urban structure,increasing the vegetated area,and changing building colors are proposed to mitigate UHI in the areas with continuously high HII.Nuo XU Fan DENG Bingqi LIU Caixia LI Hancong FU Huan YANG Jiahua ZHANG 2021Journal of Meteorological Research2021,35,5:1
12Geochronology and Genetic Model for Early Cretaceous Volcanic Rocks from the Southern Qiangtang Terrane,Northern Tibet,China: Constraints from U-Pb Zircon Dating, Whole-Rock Geochemical and Sr-Nd Isotopic Data显示文摘Post-collisional volcanic rocks of Mesozoic age occur in the regions adjacent to Gerze, part of the southern Qiangtang Terrane of northern Tibet, China. Geochronological, geochemical, and wholerock Sr-Nd isotopic analyses were performed on the volcanic rocks to better characterize their emplacement age and models for their origin. Laser ablation-inductively coupled plasma-mass spectrometry(LA-ICP-MS) U-Pb zircon analyses yielded consistent ages ranging from 123.1±0.94 Ma to 124.5±0.89 Ma for six volcanic rocks from the study area. The intermediate volcanic rocks belong to the alkaline and sub-alkaline magma series in terms of K2 O+Na2 O contents(5.9%–9.0%), and to the shoshonitic and calc-alkaline series on the basis of their high K2 O contents(1.4%–3.3%). The Gerze volcanic rocks are characterized by the enrichment of light rare earth elements [(La/Yb)N=34.9–49.5] and large–ion lithophile elements(e.g., Rb, Ba, Th, U, K, Pb, and Sr), slightly negative Eu anomalies(Eu/Eu*=0.19–0.24), and negative anomalies in high field strength elements(e.g., Nb, Ta, Hf and Ti), relative to primitive mantle. The samples show slightly elevated(87 Sr/86 Sr)i values that range from 0.7049 to 0.7057, and low εNd(t) values from-0.89 to-2.89. These results suggest that the volcanic rocks studied derived from a compositionally heterogeneous mantle source and that their parent magmas were basaltic. The more mafic, parental magmas to the Gerze volcanic rocks likely underwent fractional crystallization of clinopyroxene, hornblende, biotite, and potassium feldspar, during ascent, with little to no crustal contamination, prior to their eruption/emplacement. 13Identifying Heteroatomic and Defective Sites in Carbon with Dual-Ion Adsorption Capability for High Energy and Power Zinc Ion Capacitor显示文摘Aqueous zinc-based batteries(AZB s)attract tremendous attention due to the abundant and rechargeable zinc anode.Nonetheless,the requirement of high energy and power densities raises great challenge for the cathode development.Herein we construct an aqueous zinc ion capacitor possessing an unrivaled combination of high energy and power characteristics by employing a unique dual-ion adsorption mechanism in the cathode side.Through a templating/activating co-assisted carbonization procedure,a routine protein-rich biomass transforms into defect-rich carbon with immense surface area of 3657.5 m^(2) g^(-1) and electrochemically active heteroatom content of 8.0 at%.Comprehensive characterization and DFT calculations reveal that the obtained carbon cathode exhibits capacitive charge adsorptions toward both the cations and anions,which regularly occur at the specific sites of heteroatom moieties and lattice defects upon different depths of discharge/charge.The dual-ion adsorption mechanism endows the assembled cells with maximum capacity of 257 mAh g^(-1) and retention of72 mAh g^(-1) at ultrahigh current density of 100 A g^(-1)(400 C),corresponding to the outstanding energy and power of 168 Wh kg^(-1)and 61,700 W kg^(-1).Furthermore,practical battery configurations of solid-state pouch and cable-type cells display excellent reliability in electrochemistry as flexible and knittable power sources.Wenjie Fan Jia Ding Jingnan Ding Yulong Zheng Wanqing Song Jiangfeng Lin Caixia Xiao Cheng Zhong Huanlei Wang Wenbin Hu 2021Nano-Micro Letters2021,13,4:1

16Geochemical constraints on the origin of Early Cretaceous alkaline intrusions and its tectonic implication,Sulu Orogenic Belt,Eastern North China Craton显示文摘Post-orogenic alkaline intrusions from the Sulu Orogenic Belt of eastern North China Craton consist of A-type granites.In this study,we report U-Pb zircon ages,geochemical data,Sr-Nd-Pb,and zircon Hf isotopic data for these rocks.The LA-ICP-MS U-Pb zircon analyses yield consistent ages ranging from 127.1±2.4 to 119.5±4.8 Ma for four samples.The alkaline rocks are characterized by high total alkalis(K2 O+Na2 O=8.32-10.11 wt%),light rare-earth element enrichment,and heavy rare-earth element depletion,with a wide range(La/Yb)N values(20-48),moderate negative Eu anomalies(Eu/Eu*=0.50-0.74),enrichment in large-ion lithophile elements(LILEs,i.e.,Rb,Th,U and Pb),and depletion in Ba,Sr and high field strength elements(HFSEs,i.e.,Nb,Ta,and Ti),high(87Sr/86Sr)i ranging from 0.708 to 0.7089,low sNd(t)values from-19.4 to-16.8,(206Pb/204Pb)i=16.751-16.935,(207Pb/204Pb)i=15.381-15.535,(208-Pb/204Pb)i=37.472-37.838,negativeεHf(t)values between-21.3 and-25.7 for the magmatic zircons,and larger TDM2 model ages from 2.5 to 2.8 Ga.These results suggest that the rocks were derived from a common enriched lithospheric mantle source that was metasomatized by foundered lower crustal eclogitic materials before magma generation.Furthermore,the geochemical and isotopic feature implies that the primary magma of these rocks originated through partial melting of ancient lithospheric mantle that was variably hybridized by melts derived from lower crust eclogite.These rocks in this study may have been generated by subsequent fractionation of potassium feldspar,plagioclase,ilmenite,and/or rutile.However,negligible crustal contamination occurred during the diagenesis process.Shen Liu Caixia Feng Yan Fan Kairui Tai Tianjing Gao Ian M.Coulson 2020Acta Geochimica2020,39,5:1
17Sphingolipid synthesis inhibitor fumonisin B1 causes verticillium wilt in cotton显示文摘Verticillium wilt caused by Verticillium dahliae is a major disease of cotton. Acidic protein–lipopolysaccharide complexes are thought to be the toxins responsible for its symptoms. Here,we determined that the sphingolipid biosynthesis inhibitor fumonisin B1(FB1) acts as a toxin and phenocopies the symptoms induced by V. dahliae. Knocking out genes required for FB1 biosynthesis reduced V. dahliae pathogenicity. Moreover, we showed that overexpression of a FB1 and V. dahliae both downregulated gene, Gh IQD10, enhanced verticillium wilt resistance by promoting the expression of brassinosteroid and anti-pathogen genes. 18One-Pot Preparation of Highly Dispersed Second Near-Infrared J-Aggregate Nanoparticles Based on FD-1080 Cyanine Dye for Bioimaging and Biosensing显示文摘Bioimaging and biosensing in the second nearinfrared(NIR-II)window have attracted great attention due to their unprecedented high temporal–spatial resolution,sensitivity,and penetration depth.Although some organic fluorescence dyes have been developed in this window,it is still a great challenge to synthesize hydrophilic organic contrast agents with both absorbance and emission wavelengths beyond 1300 nm.J-aggregation is a facile pathway to achieve the wavelength red-shift of organic dyes to the NIR-II window and simultaneously improve their hydrophilicity.Here,we report FD-1080 J-aggregates(FD-J)with absorbance of 1360 nm and emission of 1370 nm through heating H-aggregated FD-1080 cyanine dyes in an aqueous solution.With FD-J administration,real-time imaging of mice brain and hindlimb vasculatures can be performed beyond 1500 nm.Meanwhile,arterial and venous vessels can be clearly distinguished through dynamic imaging after injection of FD-J.In addition,reactive oxygen species-responsive NIR-II ratiometric fluorescence sensors were available based on FD-J and lanthanide nanoparticles to enable the detection of inflammation in living mice.Caixia Sun Mengyao Zhao Xinyan Zhu Peng Pei Fan Zhang 2022CCS Chemistry2022,4,2:0

20Qu-2,a robust poplar suspension cell line for molecular biology显示文摘Populus spp.have long been used as model woody plant species for molecular biology research.However,tissues of poplar are often recalcitrant to experimental procedures for molecular studies.We generated a hormone autotrophic poplar suspension cell line from a hybrid of Populus alba×P.berolinensis‘Yinzhong’,named Qu-2.Qu-2 cells are suitable as a model biological system for studying woody plants.Qu-2 cells have many advantages over suspension cell lines derived so far from any other woody plants.Qu-2 cells are very easy to cultivate and can grow on several common plant culture media without the addition of any plant hormone.They show exceptionally high growth rates,reaching an approximately 150-fold increase in biomass after one week of culturing.Another important unique characteristic of Qu-2 cells is that they can be cryopreserved and readily reactivated.Qu-2 cells are suitable for molecular manipulations such as protoplast production,transient transformation,and RNA-seq analysis.Therefore,Qu-2 cells have the great potential to be an excellent model cell line in tree molecular biological research,ranging from physiology to gene function.The Qu-2 cells will be made available to the plant community for research.Caixia Liu Kailong Li Meng Wang Erqin Fan Chuanping Yang Junhui Wang Pengyue Fu Xiaolan Ge Heike W.Sederoff Ronald RSederoff Vincent LChiang Sui Wang Guanzheng Qu 2021Journal of Forestry Research2021,32,2:0
