| 1 | Controlling N-doping type in carbon to boost single-atom site Cu catalyzed transfer hydrogenation of quinoline显示文摘Single-atom site(SA)catalysts on N-doped carbon(CN)materials exhibit prominent performance for their active sites being M-Nx.Due to the commonly random doping behaviors of N species in these CN,it is a tough issue to finely regulate their doping types and clarify their effect on the catalytic property of such catalysts.Herein,we report that the N-doping type in CN can be dominated as pyrrolic-N and pyridinic-N respectively through compounding with different metal oxides.It is found that the proportion of distinct doped N species in CN depends on the acidity and basicity of compounded metal oxide host.Owing to the coordination by pyrrolic-N,the SA Cu catalyst displays an enhanced activity(two-fold)for transfer hydrogenation of quinoline to access the valuable molecule tetrahydroquinoline with a good selectivity(99%)under mild conditions.The higher electron density of SA Cu species induced by the predominate pyrrolic-N coordination benefits the hydrogen transfer process and reduces the energy barrier of the hydrogenation pathway,which accounts for the improved catalytic effeciency. | Jian Zhang Caiyan Zheng Maolin Zhang Yajun Qiu Qi Xu Weng-Chon Cheong Wenxing Chen Lirong Zheng Lin Gu Zhengpeng Hu Dingsheng Wang Yadong Li | 2020 | Nano Research2020,13,11: | 29 |
| 2 | Improving Plant Genome Editing with High-Fidelity xCas9 and Non-canonical PAM-Targeting Cas9-NG显示文摘Two recently engineered SpCas9 variants, namely xCas9 and Cas9-NG, show promising potential in improving targeting specificity and broadening the targeting range. In this study, we evaluated these Cas9 variants in the model and crop plant, rice. We first tested xCas9-3.7, the most effective xCas9 variant in mammalian cells, for targeted mutagenesis at 16 possible NGN PAM (protospacer adjacent motif) combinations in duplicates. xCas9 exhibited nearly equivalent editing efficiency to wild-type Cas9 (Cas9-WT) at most canonical NGG PAM sites tested, whereas it showed limited activity at non-canonical NGH (H = A, C, T) PAM sites. High editing efficiency of xCas9 at NGG PAMs was further demonstrated with C to T base editing by both rAPOBECI and PmCDAI cytidine deaminases. With mismatched sgRNAs, we found that xCas9 had improved targeting specificity over the Cas9-WT. Furthermore, we tested two Cas9-NG variants, Cas9-NGv1 and Cas9-NG, for targeting NGN PAMs. Both Cas9-NG variants showed higher editing efficiency at most non-canonical NG PAM sites tested, and enabled much more efficient editing than xCas9 at AT-rich PAM sites such as GAT, GAA, and CAA. Nevertheless, we found that Cas9-NG variants showed significant reduced activity at the canonical NGG PAM sites. In stable transgenic rice lines, we demonstrated that Cas9-NG had much higher editing efficiency than Cas9-NGv1 and xCas9 at NG PAM sites. To expand the base-editing scope, we developed an efficient C to T base-editing system by making fusion of Cas9-NG nickase (D10A version), PmCDAI, and UGI. | 4 | Local and long-range transport influences on PM_(2.5) at a cities-cluster in northern China,during summer 2008显示文摘Hourly PM2.5 concentrations were observed simultaneously at a cities-cluster comprising 10 cities/towns in Hebei province in China from July 1 to 31,2008. Among the 10 cities/towns, Baoding showed the highest average concentration level(161.57(jig/m3) and Yanjiao exhibited the lowest(99.35(jig/m3). These observed data were also studied using the joint potential source contribution function with 24-h and72-h backward trajectories, to identify more clearly the local and countrywide-scale long-range transport sources. For the local sources, three important influential areas were found, whereas five important influential areas were defined for long-range transport sources. Spatial characteristics of PM2.5 were determined by multivariate statistical analyses. Soil dust, coal combustion, and vehicle emissions might be the potential contributors in these areas. The results of a hierarchical cluster analysis for back trajectory endpoints and PM2.5 concentrations datasets show that the spatial characteristics of PM2.5 in the cities-cluster were influenced not only by local sources, but also by long-range transport sources. Different cities in the cities-cluster obtained different weighted contributions from local or long-range transport sources. | 5 | Mass-transport Deposits of the Upper Cretaceous Qingshankou Formation, Songliao Terrestrial Basin, Northeast China: Depositional Characteristics, Recognition Criteria and External Geometry显示文摘Extensive mass transport deposits(MTDs), which form a significant component of the stratigraphic record in ancient and modern deep water systems, mostly distributed in the continental margin of ocean basins worldwide. To date, no large-scale MTDs have been found in Chinese terrestrial basins. Based on integrated analysis of the seismic, drilling, core and logging data, this article report MTDs have been found in Songliao terrestrial basin(SLTB), Northeast China. During the depositional period of the Upper Cretaceous Qingshankou Formation, slope break of lacustrine basins was an ideal discharge place for gravity depositions. Regional sedimentary study of MTDs reveals that Cretaceous-age MTDs in SLTB were deposited in slope-break environments by slide, slump and debris flow, and MTDs were mixed with little turbidite simultaneously. Besides the massive aspect, key features of MTDs are the common presence of deformational structures or softsedimentary deformation, floating shale clasts, boudins, microfaults, distortion beddings, rotated sand debris. Detailed mapping of two three-dimensional(3-D) seismic surveys acquired in Yingtai(YT) and Haituozi(HTZ) areas reveals that MTDs are characterized by chaotic, mounded, and transparent seismic reflection. Growth fault, slope gully and geomorphology of the slope break greatly influenced the sedimentary process and external geometry of MTDs, and as a consequence, MTDs in SLTB exhibit classic fan-like geometry and some special(non-fan-like) external geometry. Furthermore, fanlike MTDs can be divided into three subcategories, including isolated fan, mother-son fan and stacked fan. Non-fan-like MTDs can be divided into two subcategories, strip-like MTDs and faulted-pit MTDs. A new sedimentary model has been built for MTDs in SLTB. | 6 | Downregulation of LINC01296 suppresses non-small-cell lung cancer via targeting miR-143-3p/ATG2B显示文摘The 5-year survival rate of lung cancer is one of the lowest among various malignant tumors.Long noncoding RNAs(lncRNAs),noncoding RNAs longer than 200 nucleotides,can function either as tumor suppressors or as oncogenes.The aim of this study is to investigate the function of lncRNA LINC01296 and its molecular mechanism in non-small-cell lung cancer(NSCLC).According to the Gene Expression Omnibus database,10 differentially expressed lncRNAs in NSCLC cells and patient tissues are upregulated.LINC01296 is the one with the most significant overexpression.Knockdown of LINC01296 inhibits the growth and migration,arrests the cell cycle,and promotes the apoptosis of NSCLC cells.Knocking down LINC01296 in vivo suppresses tumor growth and metastasis.LINC01296 also acts as the sponge of miR-143-3p.Lowering the expression of LINC01296 leads to decreased expression of autophagy-related 2B,ATG2B,a target gene of miR-143-3p.Moreover,downregulation of LINC01296 promotes paclitaxel sensitivity in NSCLC.These results demonstrated that the LINC01296/miR-143-3p/ATG2B axis is crucial in promoting the development of NSCLC and paclitaxel resistance.Our study may provide new ideas for the further research of clinical chemotherapy of NSCLC in the near future. | Yanli Li Hui Zhang Jing Guo Wanqiu Li Xianyi Wang Caiyan Zhang Qiangling Sun Zhongliang Ma | 2021 | Acta Biochimica et Biophysica Sinica2021,53,12: | 8 |

Tibet can be classified as three parts through zoning the danger degree, the mountain canyon high danger zone of east and southeast Tibet, the plateau mountain lake basin and valley middle danger zone of south Tibet , and the Plateau Mountain lake basin low danger zone of south Tibet. This paper takes the debris flow, collapse, landslide as the key points to analyze the distribution characteristics of geological hazards, and analyze the factors which influence the distribution of geological hazards, such as terrain landform, formation lithology, geologic structure pattern, precipitation, earthquake, human activity and so on. finally, as a conclusion: in whole Tibet, the geological hazards are more in southeast than in northwest, more in mountainous area which in the edge of plateau and river valley than in the interior of plateau and lake basin. | 11 | Numerical Simulations of an Advection Fog Event over Shanghai Pudong International Airport with the WRF Model显示文摘A series of numerical simulations is conducted to understand the formation, evolution, and dissipation of an advection fog event over Shanghai Pudong International Airport(ZSPD) with the Weather Research and Forecasting(WRF) model. Using the current operational settings at the Meteorological Center of East China Air Traffic Management Bureau, the WRF model successfully predicts the fog event at ZSPD. Additional numerical experiments are performed to examine the physical processes associated with the fog event. The results indicate that prediction of this particular fog event is sensitive to microphysical schemes for the time of fog dissipation but not for the time of fog onset. The simulated timing of the arrival and dissipation of the fog, as well as the cloud distribution, is substantially sensitive to the planetary boundary layer and radiation(both longwave and shortwave) processes. Moreover, varying forecast lead times also produces different simulation results for the fog event regarding its onset and duration, suggesting a trade-off between more accurate initial conditions and a proper forecast lead time that allows model physical processes to spin up adequately during the fog simulation. | 13 | miR-7/TGF-β2 axis sustains acidic tumor microenvironment-induced lung cancer metastasis显示文摘Acidosis,regardless of hypoxia involvement,is recognized as a chronic and harsh tumor microenvironment(TME)that educates malignant cells to thrive and metastasize.Although overwhelming evidence supports an acidic environment as a driver or ubiquitous hallmark of cancer progression,the unrevealed core mechanisms underlying the direct effect of acidification on tumorigenesis have hindered the discovery of novel therapeutic targets and clinical therapy.Here,chemical-induced and transgenic mouse models for colon,liver and lung cancer were established,respectively.miR-7 and TGF-β2 expressions were examined in clinical tissues(n=184).RNA-seq,miRNA-seq,proteomics,biosynthesis analyses and functional studies were performed to validate the mechanisms involved in the acidic TME-induced lung cancer metastasis.Our data show that lung cancer is sensitive to the increased acidification of TME,and acidic TME-induced lung cancer metastasis via inhibition of miR-7-5 p.TGF-β2 is a direct target of miR-7-5 p.The reduced expression of miR-7-5 p subsequently increases the expression of TGF-β2 which enhances the metastatic potential of the lung cancer.Indeed,overexpression of miR-7-5 p reduces the acidic p H-enhanced lung cancer metastasis.Furthermore,the human lung tumor samples also show a reduced miR-7-5 p expression but an elevated level of activated TGF-β2;the expressions of both miR-7-5 p and TGF-β2 are correlated with patients’survival.We are the first to identify the role of the miR-7/TGF-β2 axis in acidic p H-enhanced lung cancer metastasis.Our study not only delineates how acidification directly affects tumorigenesis,but also suggests miR-7 is a novel reliable biomarker for acidic TME and a novel therapeutic target for non-small cell lung cancer(NSCLC)treatment.Our study opens an avenue to explore the p H-sensitive subcellular components as novel therapeutic targets for cancer treatment. | Tao Su Suchao Huang Yanmin Zhang Yajuan Guo Shuwei Zhang Jiaji Guan Mingjing Meng Linxin Liu Caiyan Wang Dihua Yu Hiu-Yee Kwan Zhiying Huang Qiuju Huang Elaine Lai-Han Leung Ming Hu Ying Wang Zhongqiu Liu Linlin Lu | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 2 |

[Method] A total of 244 fecal samples(102 from Huizhou, 83 from Jiangmen, 35 from Chaozhou and 24 from Guangzhou, respectively) in four pigeon breeding farms between June 2012 and March2013 were collected and examined microscopically. [Results] Eimeria oocysts were seen in 223(91.4%) fecal samples, with three species, namely E. labbeana, E.columbae and E. kapotei. E. labbeana was the most common species in Guangdong provinces with an overall prevalence of 91.4%, while a slightly lower incidence of E. columbae and E. kapotei species was detected with11.1% and 6.6%, representatively. The prevalence in different months varied ranging from 83.3% to 91.7%, with the highest prevalence in summer. Nestling groups showed obviously high infection than adult pigeons.
