| 2 | Reliability analysis of marine risers with narrow and long corrosion defects under combined loads显示文摘A marine riser, one of the most important components of offshore oil/gas transportation, needs to be designed to eliminate the risks caused by complex ocean environments, platform displacement and internal corrosion, etc. In this study, a new analytical-numerical assessment approach is proposed in order to quantitatively investigate the reliability of internally corroded risers under combined loads including axial tension and internal pressure. First, an analytical solution of the limit state function of intact risers under combined loads is obtained, which is further modified by the non-dimensional corrosion depth(d/ t) for the risers with a narrow and long corrosion defect. The relationship between d/t and limited internal pressure is obtained by finite element analysis and nonlinear regression. Through an advanced first-order reliability method(HL-RF) algorithm, reliability analysis is performed to obtain the failure probability, the reliability index and the sensitivity. These results are further verified by Monte-Carlo importance sampling. | 4 | Seam Tracking Technology for Hyperbaric Underwater Welding显示文摘Automatic weld seam tracking technology to be used in hyperbaric underwater damaged pipeline repair welding is much more important, because of poor bevel preparation and severe working condition. A weld seam tracking system based on digital signal processing(DSP) passive light weld image processing technology has been established. A convenient charge coupled device(CCD) camera system was used in the high pressure environment with the help of an aperture and focus altering mechanism to guarantee overall image visibility in the scope of pressure below 0.7 MPa. The system can be used in the hyperbaric environment to pick up the real welding image of both the welding arc and the welding pool. The newly developed DSP technology was adopted to achieve the goal of system real time characteristics. An effective weld groove edge recognition technique including narrow interesting window opening, middle value wave filtering, Sobel operator weld edge detecting and edge searching in a defined narrow area was proposed to remove the guide error and system accuracy was ensured. | 5 | Effects of trace Zr on the microstructure and properties of 2E12 alloy显示文摘Al-4.0Cu-1.4Mg-0.6Mn (2E12) and Al-4.0Cu-1.4Mg-0.6Mn-0.3Zr aluminum billets were manufactured by soft-contact electromagnetic continuous casting (EMC). Subsequent forging and heat treatment were conducted and the effects of Zr on the microstructure and properties of the Al-4.0Cu-1.4Mg-0.6Mn alloy were studied. The results show that the addition of 0.3% Zr can reduce the dendrite and refine grains. During forging and solution treatment, fine and dispersive Al3Zr particles precipitated from the supersaturated α (Al) solid solution in the heating process of the billet can effectively pin dislocations and subgrain boundaries. Because of the addition of Zr, the mechanical properties are improved with the tensile strength, yield strength, elongation, and contraction of the area increasing by 5.4%, 11.3%, 9.7%, and 12.6%, respectively. | 8 | Spatiotemporal Characteristics and Future Scenario Simulation of the Trade-offs and Synergies of Mountain Ecosystem Services: A Case Study of the Dabie Mountains Area, China显示文摘Mountain ecosystems play an essential role in supporting regional sustainable development and improving local ecological environments. However, economic development in mountainous areas has long been lagging, and multiple conflicts related to resource assurance, ecological protection, and economic development have emerged. An accurate grasp of the current status and evolutionary trends of mountain ecosystems is essential to enhance the overall benefits of ecosystem services and maintain regional ecological security. Based on the In VEST(Integrated Valuation of Ecosystem Services and Trade-offs) model, this study analyzed the spatiotemporal evolution patterns and the trade-offs and synergies among ecosystem services(ES) in the Dabie Mountains Area(DMA) of eastern China. The Markov-PLUS(Patch-generating Land Use Simulation) model was used to conduct a multi-scenario simulation of the area's future development. Water yield(WY) and soil conservation(SC) had overall increasing trends during 2000-2020, carbon storage(CS)decreased overall but slowed with time, and habitat quality(HQ) increased and then decreased. The ecological protection scenario is the best scenario for improving ES in the DMA by 2030;compared to 2020, the total WY would decrease by 3.77 × 10^(8) m^(3), SC would increase by 0.65 × 10^(6) t, CS would increase by 1.33 × 10^(6) t, and HQ would increase by 0.06%. The comprehensive development scenario is the second-most effective scenario for ecological improvement, while the natural development scenario did not have a significant effect. However, as the comprehensive development scenario considers both environmental protection and economic development, which are both vital for the sustainable development of the mountainous areas, this scenario is considered the most suitable path for future development. There are trade-offs between WY, CS, and HQ, while there are synergies between SC, CS, and HQ. | 10 | Effects of chronic dexamethasone administration on hyperglycemia and insulin release in goats显示文摘Background: Dexamethasone(Dex), a synthetic glucocorticoid, is among the most commonly used drugs worldwide in animals and humans as an anti-inflammatory and immunosuppressive agent. GC has profound effects on plasma glucose level and other metabolic conditions. However, the effect of prolonged use of Dex on glucose metabolism in ruminants is still unclear.Results: Ten goats were randomly assigned to two groups: the control goats were injected with saline, and the Dex-treated goats were intramuscularly injected daily for 21 d with 0.2 mg/kg Dex. The results showed that plasma glucose and insulin concentrations were significantly increased after Dex administration(P < 0.05). Additionally, the content of hepatic glycogen was also markedly increased in Dex-treated goats(P < 0.01), while the content of glycogen in dorsal longissimus was unchanged by Dex(P > 0.05). The expression of several key genes, involved in blood glucose regulation, was detected by real-time PCR in the small intestine, skeletal muscle and liver. The expression of glucose transporter type 2(GLUT2), sodium-glucose transporter 1(SGLT1) and sodium-potassium ATPase(Na-K/ATPase) in the small intestine were generally increased by Dex, and GLUT2 m RNA expression was significantly up-regulated(P < 0.05). In liver, the expression of genes involved in gluconeogenesis including glucose-6-phosphatase catalytic subunit(G6 PC), cytosolic form of phosphoenolpyruvate carboxykinase(PCK1) and pyruvate carboxylase(PC), were significantly down-regulated by Dex. However, the protein expression levels of PCK1 & PCK2 were significantly increased by Dex, suggesting a post-transcriptional regulation. | 11 | Negative effects of long-term feeding of high-grain diets to lactating goats on milk fat production and composition by regulating gene expression and DNA methylation in the mammary gland显示文摘Background: It is well known that feeding a high concentrate(HC) diet to lactating ruminants likely induces subacute ruminal acidosis(SARA) and leads to a decrease in milk fat production. However, the effects of feeding a HC diet for long periods on milk fatty acids composition and the mechanism behind the decline of milk fat still remains poorly understood. The aim of this study was to investigate the impact of feeding a HC diet to lactating dairy goats on milk fat yield and fatty acids composition with an emphasis on the mechanisms underlying the milk fat depression. Seventeen mid-lactating dairy goats were randomly allocated to three groups. The control treatment was fed a low-concentrate diet(35% concentrate, n = 5, LC) and there were two high-concentrate treatments(65% concentrate, HC), one fed a high concentrate diet for a long period(19 wks, n = 7, HL); one fed a high concentrate diet for a short period of time(4 wk, n = 5, HS). Milk fat production and fatty acids profiles were measured. In order to investigate the mechanisms underlying the changes in milk fat production and composition,the gene expression involved in lipid metabolism and DNA methylation in the mammary gland were also analyzed.Results: Milk production was increased by feeding the HC diet in the HS and HL groups compared with the LC diet(P < 0.01), while the percentage of milk fat was lower in the HL(P < 0.05) but not in the HS group. The total amount of saturated fatty acids(SFA) in the milk was not changed by feeding the HC diet, whereas the levels of unsaturated fatty acids(UFA) and monounsaturated fatty acids(MUFA) were markedly decreased in the HL group compared with the LC group(P < 0.05). Among these fatty acids, the concentrations of C15:0(P < 0.01), C17:0(P < 0.01), C17:1(P < 0.01), C18:1 n-9 c(P < 0.05), C18:3 n-3 r(P < 0.01) and C20:0(P < 0.01) were markedly lower in the HL group, and the concentrations of C20:0(P < 0.05) and C18:3 n-3 r(P < 0.01) were lower in the HS group compared with the LC group. However, the concentrations of C18:2 n-6 c(P < 0.05) and C20:4 n-6(P < 0.05) in the milk fat were higher in the HS group. Real-time PCR results showed that the m RNA expression of the genes involved in milk fat production in the mammary gland was generally decreased in the HL and HS groups compared with the LC group. Among these genes, ACSL1, ACSS1 & 2, ACACA, FAS, SCD, FADS2, and SREBP1 were downregulated in the mammary gland of the HL group(P < 0.05), and the expressions of ACSS2, ACACA, and FADS2 m RNA were markedly decreased in the HS goats compared with the LC group(P < 0.05). In contrast to the gene expression, the level of DNA methylation in the promoter regions of the ACACA and SCD genes was increased in the HL group compared with the LC group(P < 0.05). The levels of ACSL1 protein expression and FAS enzyme activity were also decreased in the mammary gland of the HL compared with the LC group(P < 0.05).Conclusions: Long-term feeding of a HC diet to lactating goats induced milk fat depression and FAs profile shift with lower MUFAs but higher SFAs. | 17 | Effects of Spiral Magnetic Field on Structures Transformation and Macrosegregation of Sn-Pb Alloy显示文摘The effect of spiral magnetic field (SMF) on the solidification process of Sn-20 wt.%Pb hypoeutectic alloy was studied in this work. The temperature distribution and the macrosegregation together with the eutectic lamellar spacing during solidification were measured under different magnetic fields. The results indicate that the compositions in the top and bottom portions of the sample are nearly the same and the segregation phenomenon is basically eliminated by SMF at an exciting current of 125 A. The morphology of the primary phase transforms from coarse dendrite to rosette or ellipsoidal equiaxed crystal and the spacing of the eutectic lamellar increases from 1.6 to 3.9 μm when the exciting current increases from 0 to 125 A.
