| 2 | Suppression of murine breast cancer metastasis by selective inhibition of CXCR4 by synthetic polypeptide derived from viral macrophage inflammatory protein Ⅱ显示文摘Stromal cell-derived factor-1 and its receptor CXC chemokine receptor-4(CXCR4)have been implicated in breast cancer metastasis.A significant association between HER2 and CXCR4 expression has been observed in human breast tumor tissues,and overexpression of CXCR4 is essential for HER2-mediated tumor metastasis.Moreover,CXCR4 expression is low in normal breast tissues and high in malignant tumors,suggesting that a blockade of CXCR4 may limit tumor metastasis.The present study investigated the action of a synthetic antagonist 21-mer peptide derived from viral macrophage inflammatory protein Ⅱ against CXCR4(NT21MP)in inhibiting metastasis in vitro and in vivo.The results showed that chemotaxis of SKBR3 cells toward SDF-1αwas reduced by NT21MP in a dose-dependent manner(P<0.05).NT21MP inhibited tumor growth at 500μg/kg and in combination with Herceptin,the anti-HER2 antibody.The in vivo metastatic assay showed that NT21MP significantly inhibited pulmonary metastasis,and the number of metastatic tumor nodes on the surface of the lung was greatly decreased.Compared with the saline-treated control group,PCNA expression was dose-dependently decreased by NT21MP,the percentage of apoptotic cells was increased,and CXCR4 mRNA and protein expression were downregulated.In conclusion,NT21MP inhibits cellular proliferation,promotes apoptosis by downregulating CXCR4 expression,and suppresses the progression of primary and metastatic tumors.CXCR4 may be a useful therapeutic target for breast cancer,and NT21MP may serve as a potential target drug for the treatment of breast cancer metastasis. | YANG QingLin DING YongXing CHEN ChangJie TANG Jie ZHANG Ju YANG ZhiFeng | 2010 | Chinese Science Bulletin2010,55,20: | 6 |
| 3 | The Significance of CXCR4 Expression for the Prediction of Lymph Node Metastasis in Breast Cancer Patients显示文摘OBJECTIVE The chemokine receptor(CXCR4)CXC chemokine receptor 4)plays an important role in cancer metastasis.We therefore studied differential expression of the CXCR4,as well as that of the biomarker HER2,so as to evaluate whether these biomarkers can be used to predict axillary lymph node metastasis in breast cancer patients. METHODS Immunohistochemistry was used to evaluate the CXCR4 and HER2 expressions and to examine the paraffin sections of the breast cancers at various stages.Positive lymph node expression was found in 80 of the cases,and in 7 there was negative expression. RESULTS Compared to the cases with negative lymph nodes, there was a high expression of CXCR4(26.3% vs.14.3%,P=0.013), and an over-expression of HER2(28.8% vs.14.3%,P=0.011). Moreover,there was a direct correlation between the CXCR4 and HER2 expressions and the tumor staging(P=0.000)and lymph node metastasis(P=0.032).When the two biomarkers,i.e.CXCR4 and HER2,were concurrently labeled,a high expression of one of the biomarkers could be seen in the cases with positive lymph nodes(51.3% vs.28.6%,P<0.003). It is mainly composed of basic and intermediate-basic volcanic lava,pyroclasts and terrestrial sedimentary rocks. Based on the regularity of volcanic activity,the Yixian Formation was divided by the present authors into four members in ascending order:the first member is of basal conglomerate,basic and intermediate-basic volcanic rocks; the second member is of lake phrase sedimentary rocks,or in another word,precious fossil-rich sedimentary beds; the third is of basic volcanic rocks; and the fourth is of upper conglomerate. Field mapping and comprehensive study also indicate that there are abundant vertebrate fossils (mainly of Psittacosaurus) in the first member of the Yixian Formation,and the Jehol Biota (including Sinosauropterxy,Confuciusorns sanctus,Archaefructus,etc.) is yielded in the second member of Yixian Formation. | 18 | Novel Evolution Process of Zn-Induced Nanoclusters on Si(111)-(737) Surface显示文摘A tiny number of Zn atoms were deposited on Si(111)-(797) surface to study the evolution process of Zninduced nanoclusters. After the deposition, three types(type I, II, and III) of Zn-induced nanoclusters were observed to occupy preferably in the faulted half-unit cells. These Zn-induced nanoclusters are found to be related to one, two, and three displaced Si edge adatoms, and simultaneously cause the depression of one, two, and three closest Si edge adatoms in the neighboring unfaulted half-unit cells at negative voltages, respectively. First-principles adsorption energy calculations show that the observed type I, II, and III nanoclusters can reasonably be assigned as the Zn3Si1, Zn5Si2, and Zn7Si3 clusters,respectively. And Zn3Si1, Zn5Si2, and Zn7Si3 clusters are, respectively, the most stable structures in cases of one, two, and three displaced Si edge adatoms. Based on the above energy-preferred models, the simulated bias-dependent STM images are all well consistent with the experimental observations. Therefore, the most stable Zn7Si3 nanoclusters adsorbed on the Si(111)-(797) surface should grow up on the base of Zn3Si1 and Zn5Si2clusters.
