1Ecological provinces of spring phytoplankton in the Yellow Sea:species composition显示文摘Phytoplanktonic ecological provinces of the Yellow Sea(31.20°–39.23°N, 121.00°–125.16°E) is derived in terms of species composition and hydrological factors(temperature and salinity). 173 samples were collected from 40 stations from April 28 to May 18, 2014, and a total of 185 phytoplanktonic algal species belonging to 81 genera of 7phyla were identified by Uterm?hl method. Phytoplankton abundance in surface waters is concentrated in the west coast of Korean Peninsula and Korea Bay, and communities in those areas are mainly composed of diatoms and cyanobacteria with dominant species of Cylindrotheca closterium, Synechocystis pevalekii, Chroomonas acuta,Paralia sulcata, Thalassiosira pacifica and Karenia mikimotoi, etc. The first ten dominant species of the investigation area are analyzed by multidimensional scaling(MDS) and cluster analysis, then the Yellow Sea is divided into five provinces from Province I(P-I) to Province V(P-V). P-I includes the coastal areas near southern Liaodong Peninsula, with phytoplankton abundance of 35 420×10~3–36 163×10~3 cells/L and an average of 35 791×10~3 cells/L, and 99.84% of biomass is contributed by cyanobacteria. P-II is from Shandong Peninsula to Subei coastal area. Phytoplankton abundance is in a range of 2×10~3–48×10~3 cells/L with an average of 24×10~3cells/L, and 63.69% of biomass is contributed by diatoms. P-III represents the Changjiang(Yangtze River) Diluted Water. Phytoplankton abundance is 10×10~3–37×10~3 cells/L with an average of 24×10~3 cells/L, and 73.14% of biomass is contributed by diatoms. P-IV represents the area affected by the Yellow Sea Warm Current.Phytoplankton abundance ranges from 6×10~3 to 82×10~3 cells/L with an average of 28×10~3 cells/L, and 64.17% of biomass is contributed by diatoms. P-V represents the cold water mass of northern Yellow Sea. Phytoplankton abundance is in a range of 41×10~3–8 912×10~3 cells/L with an average of 1 763×10~3 cells/L, and 89.96% of biomass is contributed by diatoms. Overall, structures of phytoplankton community in spring are quite heterogeneous in different provinces. 2Enhancement of innate immune system in monocot rice by transferring the dicotyledonous elongation factor Tu receptor EFR显示文摘The elongation factor Tu(EF-Tu) receptor(EFR) in cruciferous plants specifically recognizes the N-terminal acetylated elf18 region of bacterial EF-Tu and thereby activates plant immunity.It has been demonstrated that Arabidopsis EFR confers broad-spectrum bacterial resistance in the EFR transgenic solanaceous plants.Here,the transgenic rice plants(Oryza sativa L.ssp.japonica cv.Zhonghua 17) and cell cultures with constitutive expression of AtEFR were developed to investigate whether AtEFR senses EF-Tu and thus enhances bacterial resistance in the monocot plants.We demonstrated that the Xanthomonas oryzae-derived elf18 peptide induced oxidative burst and mitogen-activated protein kinase activation in the AtEFR transgenic rice cells and plants,respectively.Pathogenesis-related genes,such as OsPBZ1,were upregulated dramatically in transgenic rice plant and cell lines in response to elf18 stimulation.Importantly,pretreatment with elf18 triggered strong resistance to X.oryzae pv.oryzae in the transgenic plants,which was largely dependent on the AtEFR expression level.These plants also exhibited enhanced resistance to rice bacterial brown stripe,but not to rice fungal blast.Collectively,the results indicate that the rice plants with heterologous expression of AtEFR recognize bacterial EF-Tu and exhibit enhanced broad-spectrum bacterial disease resistance and that pattern recognition receptor-mediated immunity may be manipulated across the two plant classes,dicots and monocots.Fen Lu Huiqin Wang Shanzhi Wang Wendi Jiang Changlin Shan Bin Li Jun Yang Shiyong Zhang Wenxian Sun 2015Journal of Integrative Plant Biology2015,57,7:6
3Obtaining digital elevation data in different terrain and physiognomy regions with spaceborne InSAR and its application analysis显示文摘Synthetic Aperture Radar Interferometry (for short, InSAR) is a new kind of earth observation technology, which has obtained great development in recent ten years and has a great development potential and successful future. In this note, three typical regions with different physiognomies and terrains have been selected as study regions to extract their Digital Elevation Model (OEMs). Compared with the existing 1 : 250000 DEM and by analyzing their results, we have obtained its accuracy and applicable scopes. The results show that in the region (plains, mountains or highlands) with dry surface and sparse vegetation, because of the better correlatability of images, the DEM obtained by InSAR is evidently better than the existing 1 : 250000 DEM and the accuracy can reach 4-6m; in the thick-vegetation-covering region, correlatability between images descends and the accuracy of InSAR DEM can only reach about 30 m worse than its existing 1:250000 DEM; in the middle covering field, the accuracy of InSAR DEMSHAN Xinjian SONG Xiaoyu LIU Jiahang WANG Changlin 2002Chinese Science Bulletin2002,47,10:3
4Biochemical properties of K_(11,48)-branched ubiquitin chains显示文摘As one of the most widely existing post-translational modification models, ubiquitination regulates diverse cellular activities. In eukaryotes, K_(11,48)-branched ubiquitin chains play key roles in cell cycle and protein quality control. However, the structural and biochemical properties of K_(11,48)-branched ubiquitin chains have not been well examined. Here we employed the synthetic linkage-and length-defined K_(11,48)-branched ubiquitin chains to examine their binding and hydrolysis properties in vitro. Quantitatively affinity determination of ubiquitin chains to the proteasome ubiquitin receptor S5 a indicated that the S5 a exhibited preference binding to K_(11,48)-branched chains over K_(11)-linked chains, but not K_(48)-conjugated chains. 6Recent advances in chemical protein synthesis:method developments and biological applications显示文摘The central dogma of modern biology underscores the pivotal roles proteins play in diverse biological processes,the study of which necessitates advanced methods to produce proteins with precision and versatility.Chemical protein synthesis,a powerful approach utilizing chemical reactions for the de novo construction of structurally accurate proteins,has emerged as a transformative tool for studying proteins and generating protein derivatives/mimics inaccessible by natural biological machinery,including post-translationally modified proteins,proteins comprised of unnatural amino acids,as well as mirror-image proteins.This review summarizes recent strides in synthetic method developments for chemical protein synthesis,including innovative techniques in solid-phase peptide synthesis,the challenges presented by difficult sequences in either synthesis or folding and the exploration of novel ligation reactions using both chemical and enzymatic methods.Furthermore,the review also delves into newly developed protocols for site-selective protein modifications and the generation of stapled or macrocyclized peptides/miniproteins,highlighting the power of chemical methods to make structurally diverse proteins.Recent applications of synthetic proteins in investigating post-translational modifications(phosphorylation,lipidation,glycosylation,ubiquitination,etc.),mirror-image biological processes and drug development are further discussed.Together,these topics provide a comprehensive overview of the current landscape of chemical protein synthesis.Suwei Dong Ji-Shen Zheng Yiming Li Huan Wang Gong Chen Yongxiang Chen Gemin Fang Jun Guo Chunmao He Honggang Hu Xuechen Li Yanmei Li Zigang Li Man Pan Shan Tang Changlin Tian Ping Wang Bian Wu Chuanliu Wu Junfeng Zhao Lei Liu 2024Science China Chemistry2024,67,4:0

8Identification of coexistence of biological and non-biological aerosol particles with DAPI (4′,6-diamidino-2-phenylindole) stain显示文摘The fluorescent dye 4′,6-diamidino-2-phenylindole (DAPI) has been widely used to stain microorganisms in various environment media. We applied DAPI fluorescence enumeration to airborne microorganisms and found that non-biological particles, including organic compounds, minerals, and soot, were also visible upon exposure to UV excitation under fluorescence microscope. Using laboratory-prepared biological particles as the control, we investigated the feasibility of identifying both biological and non-biological particles in the same sample with DAPI staining. We prepared biological (bacterial, fungi, and plant detritus) and non-biological (biochar, soot, mineral, metal, fly ash, salt) particles in the laboratory and enumerated the particles and their mixture with DAPI. We found that mineral particles were transparent, and biochar, soot, metals and fly ash particles were black under a filter set at excitation 350/50 nm and emission 460/50 nm bandpass (DAPI-BP), while biological particles were blue, as expected. Particles of the water-soluble salts NaCl and (NH_(4))_(2)SO_(4) were yellow under a filter set at excitation 340–380 nm and emission 425 nm long pass (DAPI-LP). Case studies with samples of dustfall, atmospheric aerosols and surface soils could allow for the quantification of the relative number of different types of particles and particles with organic matter or salt coating as well.
