| 2 | Influence of methionine/valine-depleted enteral nutrition on nucleic acid and protein metabolism in tumor-bearing rats显示文摘AIM: To investigate the effects of methionine/valine-depleted enteral nutrition (EN) on RNA, DNA and protein metabolism in tumor-bearing (TB) rats.METHODS: Sprague-Dawlley (SD) rats underwent jejunostomy for nutritional support. A suspension of Walker256 carcinosarcoma cells was subcutaneously inoculated.48 TB rats were randomly divided in 4 groups: A, B, C and D. The TB rats had respectively received jejunal feedings supplemented with balanced amino acids, methioninedepleted, balanced amino acids and valine-depleted for 6days before injection of 740 KBq 3H- methionine/valine via jejunum. The 3H incorporation rate of the radioactivity into RNA, DNA and proteins in tumor tissues at 0.5, 1, 2, 4 h postinjection of tracers was assessed with liquid scintillation counter.RESULTS: Incorporation of 3H into proteins in groups B and D was (0.500±0.020) % to (3.670±0.110) % and (0.708±0.019) % to (3.813±0.076) % respectively, lower than in groups A [(0.659±0.055) % to (4.492±0.108) %]and C r(0.805±0.098) % to (4.180±0.018) %]. Incorporation of 3H into RNA, DNA in group B was (0.237±0.075) %and (0.231±0.052) % respectively, lower than in group A (P<0.01). | 3 | Toxicities and therapeutic effect of 5-fluorouracil controlled release implant on tumor-bearing rats显示文摘AIM: To investigate the toxicities, biodistribution and anticancer effect of 5-fluorouracil controlled release implant (5-FUCI) on Walker 256 carcinosarcoma cells in Wistar rats.METHODS: Experiment 1: Wistar rats were randomly divided into three groups (27 rats per group). Blank implant was implanted in left lobe of the liver, and rats were treated with saline solution (in group A) or 5-fluorouracil(subcutaneous injection, group B). 5-FUCI was inserted in left lobe of the liver (group C). The gastrointestinal and hematological toxicities were observed and contents of element F in group C were assayed. Experiment 2: on day 6 after Walker-256carcinosarcoma transplantation in left lobe of the liver, 5-FUCI was implanted in right lobe of the liver (group E) or left lobe (group F), and rats in control group (group D)were inserted blank implant. Tumor inhibition rate and survival time were investigated.RESULTS: 5-FUCI showed no obvious toxic effect, extraction of Evan′s blue from gastrointestinal tissue was normal, the peripheral white blood cells and bone marrow nucleated cells were not reduced, compared with control group (P>0.05).Histological examination revealed that there were no visible changes in small intestinal mucosa, The concentration of 5-fluorouracil in left lobe of the liver was 9.84, 28, 34 times as much as those of right lobe of the liver, heart and kidney respectively after the implantation in group C. They kept a high level of fluorouracil in left lobe of the liver, ranging from (4.414±0.482)% to (7.800±0.804)%, for eight weeks.Survival days were 28.0±2.2, 30.0±3.2 and 38.7±6.7 d in group D, E and F, respectively.CONCLUSION: 5-FUCI shows no obvious toxicities to gastrointestinal tract and myelotoxicity. After implantation,it kept a high level of 5- fluorouracil in surrounding tissues of the implant for eight weeks. | 4 | Polymerase chain reaction of Au nanoparticle-bound primers显示文摘Polymerase chain reaction (PCR) is a useful technique for in vitro amplification of a DNA fragment. In this paper, a PCR procedure using Au nanoparticle (AuNP) -bound primers was systemically studied. The 5′-SH- (CH2)6-modified primers were covalently attached to the AuNP surface via Au-S bonds, and plasmid pBluescript SK was used as a template. The effects of the concentration of AuNP-bound primers, annealing temperature and PCR cy-cles were evaluated, respectively. The results indicate that PCR can proceed successfully under optimized condition, with either forward or reverse primers bound to the AuNP surface or with both the two primers bound to the AuNP surface. | 5 | Effects of freeze–thaw cycles on soil N_2O concentration and flux in the permafrost regions of the Qinghai–Tibetan Plateau显示文摘Nitrous oxide(N_2 O) is one of the most important greenhouse gases in the atmosphere; freeze–thaw cycles(FTCs) might strongly influence the emission of soil N_2 O on the Qinghai–Tibetan Plateau(QTP). However, there is a lack of in situ research on the characteristics of soil N_2 O concentration and flux in response to variations in soil properties caused by FTCs.Here, we report the effect of FTC-induced changes in soil properties on the soil N_2 O concentration and flux in the permafrost region of the higher reaches of the Shule River Basin on the northeastern margin of the QTP. We measured chemical properties of the topsoil, activities of soil microorganisms, and air temperature(AT), as well as soil N_2 O concentration and flux, over an annual cycle from July 31, 2011, to July 30, 2012. The results showed that soil N_2 O concentration was significantly affected by soil temperature(ST), soil moisture(SM), soil salinity(SS), soil polyphenol oxidase(SPO), soil alkaline phosphatase(SAP), and soil culturable actinomycetes(SCA), ranked as SM>SS>ST>SPO>SAP>SCA, whereas ST significantly increased soil N_2 O flux, compared with SS. | 6 | TAA family contributes to auxin production during de novo regeneration of adventitious roots from Arabidopsis leaf explants显示文摘Many differentiated plant organs have the ability to regenerate into a new plant after detachment via de novo organogenesis. During de novo root organogenesis from Arabidopsis thaliana leaf explants, wounding first induces endogenous auxin production in mesophyll cells. Auxin is then polar transported to, and accumulates in, regenerationcompetent cells near the wound to trigger the cell-fate transition. The TRYPTOPHAN AMINOTRANSFERASE OF ARABIDOPSIS(TAA) family proteins and the YUCCA(YUC) family proteins catalyze two successive biochemical steps in auxin biogenesis, and YUCs have been shown to be involved in auxin production in mesophyll cells during de novo root organogenesis. In thisstudy, we show that the TAA family is also required for adventitious rooting. Inhibition of TAA blocked adventitious root formation from leaf explants. Intriguingly,whereas YUC1 and YUC4 have been shown to be highly induced by wounding, TAA genes retained consistent expression levels before and after leaf detachment.Therefore, we suggest that TAAs and YUCs are both required for auxin biogenesis in leaf explants, but they play different roles in regeneration. | 10 | Evaluation of the relationships and uncertainties of airborne and ground-based sea ice surface temperature measurements against remotely sensed temperature records显示文摘Sea ice surface temperature(IST)is an important indicator of environmental changes in the Arctic Ocean.In this study,the relative performance of four mainstream IST records,i.e.airborne IST,infrared radiometer measured IST(IR IST),longwave radiation derived IST(LWR IST),and snow and ice mass balance array buoy derived IST(Buoy IST),were evaluated against the MODIS IST product.Bias,standard deviation(STD),and root mean square error(RMSE)were used to evaluate the data quality.Results revealed that airborne IST had the best accuracy,which was 0.21 K colder than MODIS IST,with STD of 1.46 K and RMSE of 1.47 K.Ground-based ISTs were biased with each other but all warmer than the MODIS IST.The IR IST had the best overall accuracy(bias=0.55 K;STD=1.52 K;RMSE=1.61 K),while the LWR IST was the noisiest measurement with the largest outlier data percent.Besides,co-located IR and LWR ISTs were more consistent than any type of evaluated IST against MODIS IST(correlation coefficient=0.99).Airborne and IR ISTs are thus the premier choice for monitoring the rapidly changing Arctic sea ice,together with satellite observations. | Pei Fan Xi Zhao Meng Qu Zhongnan Yan Ying Chen Zeyu Liang Xiaoping Pang | 2022 | International Journal of Digital Earth2022,15,1: | 0 |
| 11 | Retrieval of snow depth on Antarctic sea ice from the FY-3D MWRI data显示文摘The snow depth on sea ice is an extremely critical part of the cryosphere.Monitoring and understanding changes of snow depth on Antarctic sea ice is beneficial for research on sea ice and global climate change.The Microwave Radiation Imager(MWRI)sensor aboard the Chinese FengYun-3D(FY-3D)satellite has great potential for obtaining information of the spatial and temporal distribution of snow depth on the sea ice.By comparing in-situ snow depth measurements during the 35th Chinese Antarctic Research Expedition(CHINARE-35),we took advantage of the combination of multiple gradient ratio(GR(36V,10V)and GR(36V,18V))derived from the measured brightness temperature of FY-3D MWRI to estimate the snow depth.This method could simultaneously introduce the advantages of high and low GR in the snow depth retrieval model and perform well in both deep and shallow snow layers.Based on this,we constructed a novel model to retrieve the FY-3D MWRI snow depth on Antarctic sea ice.The new model validated by the ship-based observational snow depth data from CHINARE-35 and the snow depth measured by snow buoys from the Alfred Wegener Institute(AWI)suggest that the model proposed in this study performs better than traditional models,with root mean square deviations(RMSDs)of 8.59 cm and 7.71 cm,respectively.A comparison with the snow depth measured from Operation IceBridge(OIB)project indicates that FY-3D MWRI snow depth was more accurate than the released snow depth product from the U.S.National Snow and Ice Data Center(NSIDC)and the National Tibetan Plateau Data Center(NTPDC).The spatial distribution of the snow depth from FY-3D MWRI agrees basically with that from ICESat-2;this demonstrates its reliability for estimating Antarctic snow depth,and thus has great potential for understanding snow depth variations on Antarctic sea ice in the context of global climate change. | Zhongnan Yan Xiaoping Pang Qing Ji Yizhuo Chen Chongxin Luo Pei Fan Zeyu Liang | 2023 | Acta Oceanologica Sinica2023,42,12: | 0 |

| 13 | Multiple Dirac points including potential spin-orbit Dirac points in nonsymmorphic HfGe0.92Te显示文摘The search for new materials with Dirac points has been a fascinating subject of condensed matter physics.Here we first report the growth and band structure of HfGe_(0.92)Te single crystals featuring three different types of Dirac points.HfGe_(0.92)Te crystallizes in a nonsymmorphic tetragonal space group P4/nmm(No.129),having a square Ge-atom plane with vacancies of about 8%.Using angle-resolved photoemission spectroscopy(ARPES),the Dirac nodal line composed of conventional Dirac points vulnerable to spin-orbit coupling(SOC)is observed,accompanied by robust Dirac points protected by the nonsymmorphic symmetry against SOC and vacancies.In particular,spin-orbit Dirac points(SDPs)originating from the surface formed under significant SOC could exist based on ARPES and calculations.Quasi-two-dimensional(quasi-2D)characteristics are confirmed by angular-resolved magnetoresistance.HfGe_(0.92)Te bulk crystals can be easily exfoliated to flakes with a thickness of approximately 5 nm for the quasi-2D nature.Thus,HfGe_(0.92)Te provides a good platform to explore exotic topological phases or topological properties with three different types of Dirac points,which is a potential candidate to achieve novel 2D SDPs. | Long Chen Liqin Zhou Ying Zhou Chen Liu Zhongnan Guo Ke Liao Shunye Gao Wenhui Fan Jinfeng Xu Yuxuan Guo Jia'ou Wang Tian Qian Hongming Weng Gang Wang | 2023 | Science China(Physics,Mechanics & Astronomy)2023,66,1: | 0 |
| 14 | Retrieval of Antarctic sea ice freeboard and thickness from HY-2B satellite altimeter data显示文摘Antarctic sea ice is an important part of the Earth’s atmospheric system,and satellite remote sensing is an important technology for observing Antarctic sea ice.Whether Chinese Haiyang-2B(HY-2B)satellite altimeter data could be used to estimate sea ice freeboard and provide alternative Antarctic sea ice thickness information with a high precision and long time series,as other radar altimetry satellites can,needs further investigation.This paper proposed an algorithm to discriminate leads and then retrieve sea ice freeboard and thickness from HY-2B radar altimeter data.We first collected the Moderate-resolution Imaging Spectroradiometer ice surface temperature(IST)product from the National Aeronautics and Space Administration to extract leads from the Antarctic waters and verified their accuracy through Sentinel-1 Synthetic Aperture Radar images.Second,a surface classification decision tree was generated for HY-2B satellite altimeter measurements of the Antarctic waters to extract leads and calculate local sea surface heights.We then estimated the Antarctic sea ice freeboard and thickness based on local sea surface heights and the static equilibrium equation.Finally,the retrieved HY-2B Antarctic sea ice thickness was compared with the CryoSat-2 sea ice thickness and the Antarctic Sea Ice Processes and Climate(ASPeCt)ship-based observed sea ice thickness.The results indicate that our classification decision tree constructed for HY-2B satellite altimeter measurements was reasonable,and the root mean square error of the obtained sea ice thickness compared to the ship measurements was 0.62 m.The proposed sea ice thickness algorithm for the HY-2B radar satellite fills a gap in this application domain for the HY-series satellites and can be a complement to existing Antarctic sea ice thickness products;this algorithm could provide long-time-series and large-scale sea ice thickness data that contribute to research on global climate change. | Yizhuo Chen Xiaoping Pang Qing Ji Zhongnan Yan Zeyu Liang Chenlei Zhang | 2024 | Acta Oceanologica Sinica2024,43,3: | 0 |
