1Staged angioplasty versus regular carotid artery stenting in patients with carotid artery stenosis at high risk of hyperperfusion:a randomised clinical trial显示文摘Background and purpose Hyperperfusion(HP)is a devastating complication associated with carotid artery stenting(CAS)or endarterectomy.The efficacy and safety of staged angioplasty(SAP)in patients with CAS at high risk of HP remains unclear.We sought to determine whether SAP is superior to regular CAS in patients with high risk of HP.Methods A randomised,multicentre open-label clinical trial with blinded outcome assessment(STEP)was conducted.Patients with severe carotid stenosis at high risk of HP were randomly assigned(1:1)to the SAP or regular CAS group.The primary endpoint was hyperperfusion syndrome(HPS)and intracerebral haemorrhage(ICH)within 30 days after the procedure.Results From November 2014 to January 2017,a total of 64 patients were enrolled in 11 centres.33 patients were allocated to the SAP group and 31 to the regular CAS group.At 30 days,the rate of primary endpoint was 0.0%(0/33)in the SAP group and 9.7%(3/31)in the regular CAS group(absolute risk reduction(ARR),9.7%;95%CI−20.1% to 0.7%;p=0.11).As one of the secondary endpoints,the incidence of HP phenomenon(HPP)was lower in the SAP group than the regular CAS group(0.0%vs 22.6%,ARR,−22.6%;95% CI−36.8%to−10.2%;p=0.04).Conclusion The rate of HPS and ICH was not significantly lower in SAP group;the extended secondary endpoint of HPP,however,significantly reduced,which suggested that SAP may be a safe and effective carotid revascularisation procedure to prevent HP.Dapeng Mo Baixue Jia Huaizhang Shi Yaxuan Sun Qingan Liu Chengzhe Fan Jianping Deng Jinglin Yuan Wei Wu Changchun Jiang Guilian Zhang Hanjun Du Ning Ma Feng Gao Xuan Sun Ligang Song Lian Liu Guangge Peng Yongjun Wang Yilong Wang Zhongrong Miao the STEP Study Group 2021Stroke & Vascular Neurology2021,6,1:3

3Effects of febrile seizures in mesial temporal lobe epilepsy with hippocampal sclerosis on gene expression using bioinformatical analysis显示文摘Background:To investigate the effect of long-term febrile convulsions on gene expression in mesial temporal lobe epilepsy with hippocampal sclerosis(MTLE-HS)and explore the molecular mechanism of MTLE-HS.Methods:Microarray data of MTLE-HS were obtained from the Gene Expression Omnibus database.Differentially expressed genes(DEGs)between MTLE-HS with and without febrile seizure history were screened by the GEO2R software.Pathway enrichment and gene ontology of the DEGs were analyzed using the DAVID online database and FunRich software.Protein–protein interaction(PPI)networks among DEGs were constructed using the STRING database and analyzed by Cytoscape.Results:A total of 515 DEGs were identified in MTLE-HS samples with a febrile seizure history compared to MTLEHS samples without febrile seizure,including 25 down-regulated and 490 up-regulated genes.These DEGs were expressed mostly in plasma membrane and synaptic vesicles.The major molecular functions of those genes were voltage-gated ion channel activity,extracellular ligand-gated ion channel activity and calcium ion binding.The DEGs were mainly involved in biological pathways of cell communication signal transduction and transport.Five genes(SNAP25,SLC32A1,SYN1,GRIN1,and GRIA1)were significantly expressed in the MTLE-HS with prolonged febrile seizures.Conclusion:The pathogenesis of MTLE-HS involves multiple genes,and prolonged febrile seizures could cause differential expression of genes.Thus,investigations of those genes may provide a new perspective into the mechanism of MTLE-HS.Yinchao Li Chengzhe Wang Peiling Wang Xi Li Liemin Zhou 2020Acta Epileptologica2020,2,1:1
4Technetium-99m-labeled annexin V imaging for detecting prosthetic joint infection in a rabbit model显示文摘Accurate and timely diagnosis of prosthetic joint infection is essential to initiate early treatment and achieve a favorable outcome.In this study,we used a rabbit model to assess the feasibility of technetium-99m-labeled annexin V for detecting prosthetic joint infection.Right knee arthroplasty was performed on 24 New Zealand rabbits.After surgery,methicillin-susceptible Staphylococcus aureus was intra-articularly injected to create a model of prosthetic joint infection(the infected group,n = 12).Rabbits in the control group were injected with sterile saline(n=12).Seven and 21 days after surgery,technetium-99m-labeled annexin V imaging was performed in 6 rabbits of each group.Images were acquired 1 and 4 hours after injection of technetium-99 mlabeled annexin V(150 MBq).The operated-to-normal-knee activity ratios were calculated for quantitative analysis.Seven days after surgery,increased technetium-99m-labeled annexin V uptake was observed in all cases.However,at 21 days a notable decrease was found in the control group,but not in the infected group.The operated-to-normal-knee activity ratios of the infected group were 1.84 ± 0.29 in the early phase and 2.19 ±0.34 in the delay phase,both of which were significantly higher than those of the control group(P=0.03 and P=0.02).The receiver operator characteristic curve analysis showed that the operated-to-normal-knee activity ratios of the delay phase at 21 days was the best indicator,with an accuracy of 80%.In conclusion,technetium-99m-labeled annexin V imaging could effectively distinguish an infected prosthetic joint from an uninfected prosthetic joint in a rabbit model.Cheng Tang Feng Wang Yanjie Hou Shanshan Lu Wei Tian Yan Xu Chengzhe Jin Liming Wang 2015The Journal of Biomedical Research2015,29,3:1

6Influence of nano-mechanical evolution of Ti_(3)AlC_(2) ceramic on the arc erosion resistance of Ag-based composite electrical contact material显示文摘Al-containing MAX phase ceramic has demonstrated great potential in the field of high-performance low-voltage electrical contact material.Elucidating the anti-arc erosion mechanism of the MAX phase is crucial for further improving performance,but it is not well-understood.In this study,Ag/Ti_(3)AlC_(2) electrical contact material was synthesized by powder metallurgy and examined by nanoindentation techniques such as constant loading rate indentation,creep testing,and continuous stiffness measurements.Our results indicated a gradual degradation in the nano-mechanical properties of the Ti_(3)AlC_(2) reinforcing phase with increasing arc erosion times,although the rate of this degradation appeared to decelerate over arc erosion times.Specifically,continuous stiffness measurements highlighted the uneven mechanical properties within Ti_(3)AlC_(2),attributing this heterogeneity to the phase’s decomposition.During the early(1-100 times)and intermediate(100-1000 times)stages of arc erosion,the decline in the nano-mechanical properties of Ti_(3)AlC_(2) was primarily ascribed to the decomposition of Ti_(3)AlC_(2) and limited surface oxidation.During the later stage of arc erosion(1000-6200 times),the inner region of Ti_(3)AlC_(2) also sustained arc damage,but a thick oxide layer formed on its surface,enhancing the mechanical properties and overall arc erosion resistance of the Ag/Ti_(3)AlC_(2).Xuelian Wu Chengzhe Wu Xinpeng Wei Wanjie Sun Chengjian Ma Yundeng Zhang Gege Li Liming Chen Dandan Wang Peigen Zhang Zhengming Sun Jianxiang Ding 2024Journal of Advanced Ceramics2024,13,2:0
7PRRT2 gene and protein in human:characteristics,evolution and function显示文摘Background:This study was designed to characterize human PRRT2 gene and protein,in order to provide theoretical reference for research on regulation of PRRT2 expression and its involvement in the pathogenesis of paroxysmal kinesigenic dyskinesia and other related diseases.Method:Biological softwares Protparam,Protscale,MHMM,SignalP 5.0,NetPhos 3.1,Swiss-Model,Promoter 2.0,AliBaba2.1 and EMBOSS were used to analyze the sequence characteristics,transcription factors of human PRRT2 and their binding sites in the promoter region of the gene,as well as the physicochemical properties,signal peptides,hydrophobicity property,transmembrane regions,protein structure,interacting proteins and functions of PRRT2 protein.Results:(1)Evolutionary analysis of PRRT2 protein showed that the human PRRT2 had closest genetic distance from Pongo abelii.(2)The human PRRT2 protein was an unstable hydrophilic protein located on the plasma membrane.(3)The forms of random coil(67.65%)and alpha helix(23.24%)constituted the main secondary structure elements of PRRT2 protein.There were also multiple potential phosphorylation sites in the protein.(4)The results of ontology analysis showed that the cellular component of PRRT2 protein was located in the plasma membrane;the molecular function of PRRT2 included syntaxin-1 binding and SH3 domain binding;the PRRT2 protein is involved in biological processes of negative regulation of soluble NSF attachment protein receptor(SNAR E)complex assembly and calcium-dependent activation of synaptic vesicle fusion.(5)String database analysis revealed 10 proteins with close interactions with the human PRRT2 protein.(6)There were at least two promoter regions in the PRRT2 gene within 2000 bp upstream the 5'flank,a 304-bp CpG island in the promoter region and four GC boxes in the 5'regulatory region of PRRT2 gene and we found 13 transcription factors that could bind the promoter region of the PRRT2 gene.Conclusion:These results provide important information for further studies on the role of PRRT2 gene and identify their functions.Yinchao Li Shuda Chen Chengzhe Wang Peiling Wang Xi Li Liemin Zhou 2021Acta Epileptologica2021,3,1:0
8Enhanced structural damage behavior of liquid-filled tank by reactive material projectile impact显示文摘A series of ballistic experiments were performed to investigate the damage behavior of high velocity reactive material projectiles(RMPs) impacting liquid-filled tanks,and the corresponding hydrodynamic ram(HRAM) was studied in detail.PTFE/Al/W RMPs with steel-like and aluminum-like densities were prepared by a pressing/sintering process.The projectiles impacted a liquid-filled steel tank with front aluminum panel at approximately 1250 m/s.The corresponding cavity evolution characteristics and HRAM pressure were recorded by high-speed camera and pressure acquisition system,and further compared to those of steel and aluminum projectiles.Significantly different from the conical cavity formed by the inert metal projectile,the cavity formed by the RMP appeared as an ellipsoid with a conical front.The RMPs were demonstrated to enhance the radial growth velocity of cavity,the global HRAM pressure amplitude and the front panel damage,indicating the enhanced HRAM and structural damage behavior.Furthermore,combining the impact-induced fragmentation and deflagration characteristics,the cavity evolution of RMPs under the combined effect of kinetic energy impact and chemical energy release was analyzed.The mechanism of enhanced HRAM pressure induced by the RMPs was further revealed based on the theoretical model of the initial impact wave and the impulse analysis.Finally,the linear correlation between the deformation-thickness ratio and the non-dimensional impulse for the front panel was obtained and analyzed.It was determined that the enhanced near-field impulse induced by the RMPs was the dominant reason for the enhanced structural damage behavior.Jianwen Xie Yuanfeng Zheng Zhenyang Liu Chengzhe Liu Aoxin Liu Pengwan Chen Haifu Wang 2024Defence Technology(防务技术)2024,31,1:0
