1Investigation of Myocardial Stunning after Cardiopulmonary Resuscitation in Pigs显示文摘Objective To investigate cardiac function and myocardial perfusion during 48 h after cardiopulmonary resuscitation (CPR), further to test myocardial stunning and seek indicators for long‐term survival after CPR. Methods After 4 min of untreated ventricular fibrillation, fifteen anesthetized pigs were studied at baseline and 2 h, 4 h, 24 h, and 48 h after restoration of spontaneous circulation (ROSC). Hemodynamic data, echocardiography and gated‐single photon emission computed tomography myocardial perfusion images were carried out. Results Mean arterial pressure (MAP), coronary perfusion pressure (CPP) and cardiac troponin I (CTNI) showed significant differences between eventual survival animals and non‐survival animals at 4 h after ROSC (109.2±10.7 mmHg vs. 94.8±12.3 mmHg, P=0.048; 100.8±6.9 mmHg vs. 84.4±12.6 mmHg, P=0.011; 1.60±0.13 ug/L vs. 1.75±0.10 ug/L, P=0.046). Mitral valve early‐to‐late diastolic peak velocity ratio, mitral valve deceleration time recovered 24 h; ejection faction and the summed rest score recovered 48 h after ROSC. Conclusion Cardiac systolic and early active relaxation dysfunctions were reversible within survival animals; cardiac stunning might be potentially adaptive and protective after CPR. 2Enhancing production of ergosterol in Pichia pastoris GS115 by over-expression of 3-hydroxy-3-methylglutaryl CoA reductase from Glycyrrhiza uralensis显示文摘The rate-limiting enzyme in the mevalonic acid(MVA)pathway which can lead to triterpenoid saponin glycyrrhizic acid(GA)is 3-hydroxy-3-methylglutaryl-CoA reductase(HMGR).In order to reveal the effect of copy number variation in the HMGR gene on the MVA pathway,the HMGR gene from Glycyrrhiza uralensis Fisch.(GuHMGR)was cloned and over-expressed in Pichia pastoris GS115.Six recombinant P.pastoris strains containing different copy numbers of the GuHMGR gene were obtained and the content of ergosterol was analyzed by HPLC.The results showed that all the recombinant P.pastoris strains contained more ergosterol than the negative control and the strains with 8 and 44 copies contained significantly more ergosterol than the other strains.However,as the copy number increased,the content of ergosterol showed an increasing–decreasing–increasing pattern.This study provides a rationale for increasing the content of GA through over-expressing the GuHMGR gene in cultivars of G.uralensis.Ying Liu Xiaoqing Zhu Wendong Li Hao Wen Ya Gao Yong Liu Chunsheng Liu 2014Acta Pharmaceutica Sinica B2014,4,2:7

5Deformation and failure characteristics and fracture evolution of cryptocrystalline basalt显示文摘Cryptocrystalline basalt is one of the two major types of rocks exposed in the super large-scale underground powerhouse in Baihetan hydropower station in China.The rock of this type shows various sitespecific mechanical responses(e.g.fragmentation,fracturing,and relaxation)during excavation.Using conventional triaxial testing facility MTS 815.03,we obtained the stressestrain curves,macroscopic failure characteristics,and strength characteristics of cryptocrystalline basalt.On this basis,evolution of crack initiation and propagation was explored using the finite-discrete element method(FDEM)to understand the failure mechanism of cryptocrystalline basalt.The test results showed that:(1)under different confining stresses,almost all the pre-peak stressestrain curves of cryptocrystalline basalt were linear and the post-peak stresses decreased rapidly;(2)the cryptocrystalline basalt showed a failure mode in a form of fragmentation under low and medium confining stresses while fragmentation-shear coupling failure dominated at high confining stresses;and(3)the initial strength ratio(sci/sf,where sci and sf are the crack initiation strength and peak strength,respectively)ranged from 0.45 to 0.55 and the damage strength ratio(scd/sf,where scd is the crack damage strength)exceeded 0.9.The stressestrain curve characteristics and failure modes of cryptocrystalline basalt could be reflected numerically.For this,FDEM simulation was employed to reveal the characteristics of cryptocrystalline basalt,including high scd/sf values and rapid failure after scd,with respect to the microscopic characteristics of mineral structures.The results showed that the fragmentation characteristics of cryptocrystalline basalt were closely related to the development of tensile cracks in rock samples prior to failure.Moreover,the decrease in degree of fragmentation with increasing confining stress was also correlated with the dominant effect of confining stress on the tensile cracks.Zhenjiang Liu Chuanqing Zhang Chunsheng Zhang Yang Gao Hui Zhou Zhaorong Chang 2019Journal of Rock Mechanics and Geotechnical Engineering2019,11,5:4
6Macrophage membrane-coated nanocarriers Co-Modified by RVG29 and TPP improve brain neuronal mitochondria-targeting and therapeutic efficacy in Alzheimer's disease mice显示文摘Neuronal mitochondrial dysfunction caused by excessive reactive oxygen species(ROS)is an early event of sporadic Alzheimer's disease(AD),and considered to be a key pathologic factor in the progression of AD.The targeted delivery of the antioxidants to mitochondria of injured neurons in brain is a promising therapeutic strategy for AD.A safe and effective drug delivery system(DDS)which is able to cross the blood-brain barrier(BBB)and target neuronal mitochondria is necessary.Recently,bioactive materials-based DDS has been widely investigated for the treatment of AD.Herein,we developed macrophage(MA)membrane-coated solid lipid nanoparticles(SLNs)by attaching rabies virus glycoprotein(RVG29)and triphenylphosphine cation(TPP)molecules to the surface of MA membrane(RVG/TPP-MASLNs)for functional antioxidant delivery to neuronal mitochondria.According to the results,MA membranes camouflaged the SLNs from being eliminated by RESrich organs by inheriting the immunological characteristics of macrophages.The unique properties of the DDS after decoration with RVG29 on the surface was demonstrated by the ability to cross the BBB and the selective targeting to neurons.After entering the neurons in CNS,TPP further lead the DDS to mitochondria driven by electric charge.The Genistein(GS)-encapsulated DDS(RVG/TPP-MASLNs-GS)exhibited the most favorable effects on reliveing AD symptoms in vitro and in vivo by the synergies gained from the combination of MA membranes,RVG29 and TPP.These results demonstrated a promising therapeutic candidate for delaying the progression of AD via neuronal mitochondria-targeted delivery by the designed biomimetic nanosystems.Yang Han Chunhong Gao Hao Wang Jiejie Sun Meng Liang Ye Feng Qianqian Liu Shiyao Fu Lin Cui Chunsheng Gao Yi Li Yang Yang Baoshan Sun 2021Bioactive Materials2021,6,2:4

8Carrier-free highly drug-loaded biomimetic nanosuspensions encapsulated by cancer cell membrane based on homology and active targeting for the treatment of glioma显示文摘Nanosuspensions,as a new drug delivery system for insoluble drugs,are only composed of a drug and a small amount of stabilizer,which is dispersed in an aqueous solution with high drug-loading,small particle size,high dispersion,and large specific surface area.It can significantly improve the dissolution,bioavailability,and efficacy of insoluble drugs.In this study,paclitaxel nanosuspensions((PTX)NS)were prepared by an ultrasonic precipitation method,with the characteristics of simple preparation and easy repetition.With the help of a homologous targeting mechanism,a kind of glioma C6 cancer cell membrane(CCM)-coated(PTX)NS was developed and modified with DWSW peptide to obtain DWSW-CCM-(PTX)NS with the functions of BBB penetration and tumor targeting.The results showed that the cancer cell membrane could effectively camouflage the nanosuspensions so that it was not cleared by the immune system and could cross the blood-brain-barrier(BBB)and selectively target tumor tissues.Cell uptake experiments and in vivo imaging confirmed that the uptake of DWSW-CCM-(PTX)NS by tumor cells and the distribution in intracranial gliomas increased.Cytotoxicity test and in vivo anti-glioma studies showed that DWSW-CCM-(PTX)NS could significantly inhibit the growth of glioma cells and significantly prolong the survival time of glioma-bearing mice.Finally,the cancer cell membrane coating endowed the nanosuspensions with the biological properties of homologous adhesion and immune escape.This study provides an integrated solution for improving the targeting of nanosuspensions and demonstrates the encouraging potential of biomimetic nanosuspensions applicable to tumor therapy.Yueyue Fan Yuexin Cui Wenyan Hao Mengyu Chen Qianqian Liu Yuli Wang Meiyan Yang Zhiping Li Wei Gong Shiyong Song Yang Yang Chunsheng Gao 2021Bioactive Materials2021,6,12:3
9Defect-density control of platinum-based nanoframes with high-index facets for enhanced electrochemical properties显示文摘Structure-engineered platinum-based nanoframes(NFs)at the atomic level can effectively improve the catalytic performance for fuel cells and other heterogeneous catalytic fields.We report herein,a microwave-assisted wet-chemical method for the preparation of platinum-copper-cobalt NFs with tunable defect density and architecture,which exhibit enhanced activity and durability towards the electro-oxidation reactions of methanol(MOR)and formic acid(FAOR).By altering the reduction/capping agents and thus the nucleation/growth kinetics,trimetallic platinum-copper-cobalt hexapod NFs with different density high-index facets are achieved.Especially,the rough hexapod nanoframes(rh-NFs)exhibit excellent specific activities towards MOR and FAOR,7.25 and 5.20 times higher than those of benchmark Pt/C,respectively,along with prolonged durability.The excellent activities of the rh-NFs are assigned to a synergistic effect,including high density of defects and high-index facets,suitable d-band center,and open-framework structure.This synergistic working mechanism opens up a new way for enhancing their electrocatalytic performances by increasing defect density and high-index facets in open-framework platinum-based NFs.Shaohan Yang Shuna Li Lianghao Song Yipin Lv Zhongyao Duan Chunsheng Li Raphael Francesco Praeg Daowei Gao Guozhu Chen 2019Nano Research2019,12,11:3
10A Novel Wavenumber Domain SAR Imaging Algorithm Based on the Fractional Fourier Transform显示文摘The conventional Wavenumber domain algorithm(WDA) employs Fast Fourier transform(FFT) to implement matched filter. It is characterized by its high precision SAR imaging. Compared with FFT, Fractional Fourier transform(Fr FT) can provide lower and narrower sidelobes when it is applied to compress chirp signals at the optimal transform. This paper proposed a novel SAR imaging algorithm, termed Fr WDA which replaces FFT in WDA with Fr FT. The Fr WDA was derived and its implementation was presented. How to choose the optimal transformation order was given subsequently. The Fr WDA preserves high precision focusing, and has better performance on sidelobe suppression. The Fr WDA is more suitable for the high precision SAR imaging, especially in target detection and recognition. 19Cell relay-delivery improves targeting and therapeutic efficacy in tumors显示文摘Cell-mediated drug delivery system(CDDS)has shown great potential for cancer therapy.However,a single cell-mediated drug delivery mechanism has not generally been successful,particularly for systemic administration.To augment the antitumor therapy efficacy,herein,we propose a strategy of cell relay-delivery for the use of artificially damaging/aging erythrocytes to hitchhike on circulating monocytes/macrophages for intratumoral accumulation of anticancer drugs.This biomimetic relay-delivery strategy was derived from the manner in which circulating monocytes/macrophages in body specifically engulf damaged/senescent erythrocytes and actively transmigrate into the tumor bulk.The strategy elegantly combines the natural functions of both cells,which therefore provides a new perspective to challenge current obstacles in drug delivery.According to the strategy,we developed biotinylated erythrocyte-poly(lactic-co-glycolic acid)(PLGA)nanoparticle hybrid DDSs(bE-NPs)using avidin-biotin coupling.In such a system,biotinylated erythrocytes can mimic the natural property of damaged/senescent erythrocytes,while PLGA NPs are capable of encapsulating anticancer drugs and promoting sustained drug release.Anticancer drugs can effectively target tumor sites by two steps.First,by using biotinylated erythrocytes as the carrier,the drug-loaded PLGA NPs could be specifically phagocytized by monocytes/macrophages.Second,by taking advantage of the tumor-tropic property of monocytes/macrophages,the drug-loaded PLGA NPs could be efficiently transported into the tumor bulk.After encapsulating vincristine(VIN)as the model drug,bE-NPs exhibited the most favorable antitumor effects in vitro and in vivo by the cell relay-delivery effect.These results demonstrate that the cell relay-delivery provides a potential method for improving tumor treatment efficacy.Ye Feng Qianqian Liu Yi Li Yang Han Meng Liang Hao Wang Qing Yao Yuli Wang Meiyan Yang Zhiping Li Wei Gong Yang Yang Chunsheng Gao 2021Bioactive Materials2021,6,6:0
20Distinct superconducting behaviors of pressurized WB2 and ReB2 with different local B layers显示文摘The recent discovery of superconductivity up to 32 K in the pressurized MoBreignites the interest in exploring high-Tc superconductors in transition-metal diborides. Inspired by that work, we turn our attention to the 5 d transition-metal diborides.Here we systematically investigate the responses of both structural and physical properties of WBand ReBto external pressure,which possess different types of boron layers. Similar to MoB, the pressure-induced superconductivity was also observed in WBabove 60 GPa with a maximum Tcof 15 K at 100 GPa, while no superconductivity was detected in ReBin this pressure range. Interestingly, the structures at ambient pressure for both WBand ReBpersist to high pressure without structural phase transitions. Theoretical calculations suggest that the ratio of flat boron layers in this class of transition-metal diborides may be crucial for the appearance of high Tc.
