| 2 | LncRNA PU.1 AS regulates arsenic-induced lipid metabolism through EZH2/Sirt6/SREBP-1c pathway显示文摘Arsenic (As) is an omnipresent metalloid toxicant,which has elicited serious environmental pollution and health risky problems.Previous studies have uncovered that the As exposure could also cause markedly reduction of serum triglycerides in mice.However,the regulation mechanisms are still largely unknown.The present study is aimed to elucidate the molecular mechanisms of lncRNAs in As-induced lipid metabolic disequilibrium.We demonstrated that lncRNA PU.1 AS was significantly induced in the liver of As-feed mice companied with lower serum triglycerides contents;further in vitro experiment confirmed that PU.1 AS regulated liver cells lipid accumulation by nile red fluorescence staining.Intensive mechanistic investigations illustrated that PU.1 AS could interact with EZH2 protein to regulate its downstream target gene expression,and Asinduced PU.1 AS attenuated EZH2-supppressed Sirt6 expression,thereafter leading to a decreased SREBP-1c protein expression,as well as the diminished synthesis of triglycerides in hepatocytes.In conclusion,this study provided a new lncRNA-related regulatory signaling pathway participating in As-induced abnormal lipid metabolism. | Zheng Dong Changying Li Chunyang Yin Ming Xu Sijin Liu Ming Gao | 2019 | Journal of Environmental Sciences2019,31,11: | 8 |

| 6 | Lithospheric composition and structure beneath the northern margin of the Qinling orogenic belt——On deep-seated xenoliths in Minggang region of Henan Province显示文摘Swarms of mafic-intermediate volcaniclastic bodies occur in the Minggang region of Henan Province, a tectonic boundary between the North Qinling and the North China Block, and emplaced at (178.31±3.77) Ma. These volcanic rocks are subalkaline basaltic andesites and contain abundance of lower crust and mantle xenoliths. Thus this area is an ideal place to reveal the lithospheric composition and structure beneath the northern margin of the Qinling orogenic belt. Geochemical data indicate that these mafic granulites, eclogites and metagabbros have trace elemental and Pb isotopic characteristics very similar to those rocks from the South Qinling Block, representing the lower part of lower crust of the South Qinling which subducted beneath the North China Block. Talcic peridotites represent the overlying mantle wedge materials of the North China Block, which underwent the metasomatism of the acidic melt/fluid released from the underlying lower crust of the South Qinling Block. Deep tectonic model proposed in this paper is that after the Late Paleozoic South Qinling lithosphere subducted northward and decoupled, the upper part of the lithosphere emplaced under the North Qinling and the lower part continuously subducted northward under the North China Block. | 7 | Disuccinimidyl suberate cross-linked hemoglobin as a novel red blood cell substitute显示文摘Disuccinimidyl suberate (DSS) intramolecularly cross-linked hemoglobin (Hb) was developed as a novel red blood cell substitute. A multi-angle laser light scattering detector coupled with size exclusion HPLC was applied to determine the molecular weight of the modified Hb. SDS-PAGE was also used as a complement. It was proved that 83.8% of the product was intramolecularly cross-linked Hb with weight-average molecular weights (Mw) of 67.5 kD, 12% was dimeric Hb with Mw of 146.6 kD, and 4.2% was trimeric Hb with Mw of 306.4 kD. The tetramer structure of the cross-linked Hb was stable as shown in size-exclusion chromatography using a mobile phase containing 1 mol/L MgCl2. Analysis by LC-MS demonstrated that the reaction of DSS with Hb mainly took place between the twoα subunits within a Hb molecule, resulting in stabilization of the tetramer structure. However, the cross-linking was not site-specific. The P50 of the cross-linked Hb decreased from 21.8 mmHg to 14.3 mmHg, and the Hill coefficient decreased from 2.22 to 1.41. Result of isoelectric focusing showed that the pI of DSS cross-linked Hb was in the range of 4.6-5.2, similar to that of serum albumin. The safety of DSS cross-linked Hb was favored by animal tests on rats and guinea pigs. Exchange transfusion experiment with DSS cross-linked Hb using rats as a model indicated no pressor effect or other significant side effects. | 9 | Safety and immunogenicity of a modified COVID-19 mRNA vaccine,SYS6006,as a fourth-dose booster following three doses of inactivated vaccines in healthy adults:an open-labeled Phase 1 trial显示文摘The continuous emergence of the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)variants led to a rapid decline in protection efficacy and neutralizing titers even after three doses of COVID-19 vaccines.Here,we report an open-labeled Phase I clinical trial of a modified mRNA vaccine(SYS6006)as a fourth-dose booster in healthy adults.Eighteen eligible participants,who had completed three doses of inactivated COVID-19 vaccines,received a fourth boosting dose of SYS6006-20μg.Eighteen convalescent COVID-19 patients were enrolled for the collection of serum samples as a comparator of immunogenicity.The primary endpoint of this trial was titers of anti-receptor binding domain of spike glycoprotein(RBD)antibodies of the Omicron strain(BA.2 and BA.4/5)in serum;titers of neutralizing antibodies against pseudovirus of the Omicron strain(BA.2 and BA.4/5).The secondary endpoint was the incidence of adverse events within 30 days after the boosting.The exploratory endpoint was the cellular immune responses(interferon gamma,IFN-γ).This trial was registered with the Chinese Clinical Trial Registry website.No serious adverse events were reported within 30 days after vaccination.No Grade 3 fever or serious adverse event was reported in the SYS6006 group.Notably,SYS6006 elicited higher titers and longer increases in anti-RBD antibodies and neutralizing antibodies(>90 days)compared with the convalescent group(P<0.0001)against Omicron strain(BA.2 and BA.4/5).Besides,higher positive spots of T-cell-secreting IFN-γwere observed in the SYS6006 group than those in the convalescent group(P<0.05).These data demonstrated that SYS6006 was well tolerated and highly immunogenic,generating a stronger and more durable immune response against different variants of SARS-CoV-2. | Yuzhou Gui Ye Cao Jiajin He Chunyang Zhao Wei Zheng Ling Qian Jie Cheng Chengyin Yu Chen Yu Kun Lou Gangyi Liu Jingying Jia | 2023 | Life Metabolism2023,2,3: | 1 |

| 11 | Characterization of SHARPIN knockout Syrian hamsters developed using CRISPR/Cas9 system显示文摘Background : SHARPIN (SHANK- associated RH domain interactor) is a component ofthe linear ubiquitination complex that regulates the NF- κB signaling pathway. To betterunderstand the function of SHARPIN, we sought to establish a novel geneticallyengineered Syrian hamster with SHARPIN disruption using the CRISPR/Cas9 system.Methods : A single- guide ribonucleic acid targeting exon 1 of SHARPIN gene was designedand constructed. The zygotes generated by cytoplasmic injection of the Cas9/gRNA ribonucleoprotein were transferred into pseudopregnant hamsters. Neonatalmutants were identified by genotyping. SHARPIN protein expression was detectedusing Western blotting assay. Splenic, mesenteric lymph nodes (MLNs), and thymicweights were measured, and organ coefficients were calculated. Histopathologicalexamination of the spleen, liver, lung, small intestine, and esophagus was performedindependently by a pathologist. The expression of lymphocytic markers and cytokineswas evaluated using reverse transcriptase- quantitative polymerase chain reaction.Results : All the offspring harbored germline- transmitted SHARPIN mutations.Compared with wild- type hamsters, SHARPIN protein was undetectable in SHARPIN −/−hamsters. Spleen enlargement and splenic coefficient elevation were spotted inSHARPIN −/− hamsters, with the descent of MLNs and thymuses. Further, eosinophilinfiltration and structural alteration in spleens, livers, lungs, small intestines, and esophagiwere obvious after the deletion of SHARPIN. Notably, the expression of CD94 and CD22 was downregulated in the spleens of knockout (KO) animals. Nonetheless,the expression of CCR3, CCL11, Il4 , and Il13 was upregulated in the esophagi. Theexpression of NF- κB and phosphorylation of NF- κB and IκB protein significantly diminishedin SHARPIN −/− animals.Conclusions : A novel SHARPIN KO hamster was successfully established using theCRISPR/Cas9 system. | 13 | Biphasic mineralized collagen-based composite scaffold for cranial bone regeneration in developing sheep显示文摘Appropriate mechanical support and excellent osteogenic capability are two essential prerequisites of customized implants for regenerating large-sized cranial bone defect.Although porous bone scaffolds have been widely proven to promote bone regeneration,their weak mechanical properties limit the clinical applications in cranioplasty.Herein,we applied two previously developed mineralized collagen-based bone scaffolds(MC),porous MC(pMC)and compact MC(cMC)to construct a biphasic MC composite bone scaffold(bMC)to repair the large-sized cranial bone defect in developing sheep.A supporting frame composed of cMC phase in the shape of tic–tac–toe structure was fabricated first and then embedded in pMC phase.The two phases had good interfacial bond,attributing to the formation of an interfacial zone.The in vivo performance of the bMC scaffold was evaluated by using a cranial bone defect model in 1-month-old sheep.The computed tomography imaging,X-ray scanning and histological evaluation showed that the pMC phase in the bMC scaffold,similar to the pMC scaffold,was gradually replaced by the regenerative bone tissues with comprehensively increased bone mineral density and complete connection of bone bridge in the whole region.The cMC frame promoted new bone formation beneath the frame without obvious degradation,thus providing appropriate mechanical protection and ensuring the structural integrity of the implant.In general,the sheep with bMC implantation exhibited the best status of survival,growth and the repair effect.The biphasic structural design may be a prospective strategy for developing new generation of cranioplasty materials to regenerate cranial bone defect in clinic. | Jingchuan Zheng Zhijun Zhao Yongdong Yang Shuo Wang Yonggang Zhao Yang Xiong Shuhui Yang Zhiye Qiu Tianxi Song Chunyang Zhang Xiumei Wang | 2022 | Regenerative Biomaterials2022,9,1: | 0 |
| 14 | Boosting the performance of deep-ultraviolet photodetector arrays based on phase-transformed heteroepitaxial β-Ga_(2)O_(3) films for solar-blind imaging显示文摘Solar-blind imaging has attracted considerable interest in both military and civilian applications,spurring the development of high-performance deep-ultraviolet photodetector arrays(PDAs) with wide-bandgap semiconductor materials.Herein,we present a novel method to enhance the performance of solar-blind PDAs(SBPDs) using β-Ga_(2)O_(3) films obtained by the phase transition of heterogeneous epitaxial sub-stable ε-Ga_(2)O_(3),achieved through high-temperature rapid annealing.Metal-semiconductor-metaltype SBPDs based on phase-transformed β-Ga_(2)O_(3) films exhibited superior performance,including an ultrahigh responsivity of 459.38 A/W,detectivity of 10^(14)–10^(15) Jones,external quantum efficiency of 10^(4)%–10^(5)%,rejection ratio(R_(254)/R_(365)) of 10^(5)–10^(6),photo-to-dark current ratio of 10^(4)–10^(6),fast response speed of 1.01 s/0.06 s,and favorable stability.Notably,the ultrahigh responsivity of β-Ga_(2)O_(3)-film-based devices was approximately 222-fold higher than that of ε-Ga_(2)O_(3) film-based devices.The assembled 4×5 β-Ga_(2)O_(3) film-based PDAs exhibited favorable uniformity,repeatability,and high spatial resolution for solarblind imaging.Our study offers a promising approach for the development of high-performance β-Ga_(2)O_(3)-based PDAs for solarblind ultraviolet imaging with potential applications in both military and civilian fields. | ZHENG QiQi CHEN LingRui LI XuDong DING Ke PANG Di LI HongLin XIONG YuanQiang RUAN HaiBo FANG Liang LI WanJun YE LiJuan ZHANG Hong KONG ChunYang | 2023 | Science China(Technological Sciences)2023,66,9: | 0 |

| 19 | Recognition for avian influenza virus proteins based on support vector machine and linear discriminant analysis显示文摘Total 200 properties related to structural characteristics were employed to represent structures of 400 HA coded proteins of influenza virus as training samples. Some recognition models for HA proteins of avian influenza virus (AIV) were developed using support vector machine (SVM) and linear discriminant analysis (LDA). The results obtained from LDA are as follows: the identification accuracy (Ria) for training samples is 99.8% and Ria by leave one out cross validation is 99.5%. Both Ria of 99.8% for training samples and Ria of 99.3% by leave one out cross validation are obtained using SVM model, respectively. External 200 HA proteins of influenza virus were used to validate the external predictive power of the resulting model.
