2The role of m6A modification in the biological functions and diseases显示文摘N6-methyladenosine(m6A)is the most prevalent,abundant and conserved internal cotranscriptional modification in eukaryotic RNAs,especially within higher eukaryotic cells.m6A modification is modified by the m6A methyltransferases,or writers,such as METTL3/14/16,RBM15/15B,ZC3H3,VIRMA,CBLL1,WTAP,and KIAA1429,and,removed by the demethylases,or erasers,including FTO and ALKBH5.It is recognized by m6A-binding proteins YTHDF1/2/3,YTHDC1/2 IGF2BP1/2/3 and HNRNPA2B1,also known as'readers'.Recent studies have shown that m6A RNA modification plays essential role in both physiological and pathological conditions,especially in the initiation and progression of different types of human cancers.In this review,we discuss how m6A RNA methylation influences both the physiological and pathological progressions of hematopoietic,central nervous and reproductive systems.We will mainly focus on recent progress in identifying the biological functions and the underlying molecular mechanisms of m6A RNA methylation,its regulators and downstream target genes,during cancer progression in above systems.We propose that m6A RNA methylation process offer potential targets for cancer therapy in the future.Xiulin Jiang Baiyang Liu Zhi Nie Lincan Duan Qiuxia Xiong Zhixian Jin Cuiping Yang Yongbin Chen 2021Signal Transduction and Targeted Therapy2021,6,3:16
3Source rupture process of Lushan M_S7.0 earthquake, Sichuan,China and its tectonic implications显示文摘The source rupture process of the MS7.0 Lushan earthquake was here evaluated using 40 long-period P waveforms with even azimuth coverage of stations.Results reveal that the rupture process of the Lushan MS7.0 event to be simpler than that of the Wenchuan earthquake and also showed significant differences between the two rupture processes.The whole rupture process lasted 36 s and most of the moment was released within the first 13 s.The total released moment is 1.9×1019N m with MW=6.8.Rupture propagated upwards and bilaterally to both sides from the initial point,resulting in a large slip region of 40 km×30 km,with the maximum slip of 1.8 m,located above the initial point.No surface displacement was estimated around the epicenter,but displacement was observed about 20 km NE and SW directions of the epicenter.Both showed slips of less than 40 cm.The rupture suddenly stopped at 20 km NE of the initial point.This was consistent with the aftershock activity.This phenomenon indicates the existence of significant variation of the medium or tectonic structure,which may prevent the propagation of the rupture and aftershock activity.The earthquake risk of the left segment of Qianshan fault is worthy of attention.ZHAO CuiPing ZHOU LianQing CHEN ZhangLi 2013Chinese Science Bulletin2013,58,28:13
4Electrical stimulation of acupoint combinations against deep venous thrombosis in elderly bedridden patients after major surgery显示文摘OBJECTIVE:To compare the effects of electrical stimulation of different acupoint combinations among postoperative bedridden elderly patients on hemorheology and deep venous blood flow velocity and investigate the role of electrical stimulation against deep vein thrombosis(DVT).METHODS:From November 2010 to October 2011,a total of 160 elderly bedridden patients after major surgery were divided into the conventional care group,invigorating and promoting Qi group,blood-activating and damp-eliminating group,and acupoint-combination stimulation group.Whole blood viscosity,plasma viscosity,D-dimer levels,lower limb skin temperature,lower limb circumference,and flow velocities of the external iliac vein,femoral vein,popliteal vein,and deep calf veins in all patients were documented and compared among the four groups.RESULTS:Whole blood viscosity,plasma viscosity,D-dimer levels,and lower limb circumference were significantly reduced in the blood-activating and damp-eliminating group compared with the conventional care group(P<0.05) and were almost equal to those in the acupoint-combination stimulation group(P>0.05).Lower limb venous flow velocities were accelerated in the invigorating and promoting Qi group compared with the other groups,excluding the acupoint-combination stimulation group(P<0.05).CONCLUSION:Hemorheological indices in postoperative bedridden elderly patients were improved after combined electrical stimulation at Yinlingquan(SP 9) and Sanyinjiao(SP 6).Combined electrical stimulation at Zusanli(ST 36) and Taichong(LR 3),on the other hand,accelerated lower limb venous flow.Lili Hou Cuiping Chen Lei Xu PeihaoYin Wen Peng 2013Journal of Traditional Chinese Medicine2013,33,2:13

6Cross-cultural adaption and psychometric properties of the Chinese version of the Diabetes Behavior Rating Scale: a pilot study显示文摘Self-care behavior plays a major role in diabetic management. However, in China, a satisfactory instrument has not yet been developed to evaluate the compliance of self-care behavior for young patients with type 1 diabetes mellitus(T1 DM). The Diabetes Behavior Rating Scale(DBRS) has a potential to be the first mature instrument. The purpose of this study is to cross-culturally adapt the DBRS, and preliminarily evaluate its psychometric properties. The instrument translation included translation, back translation and culture adaptation. Psychometric properties were assessed in a sample of 116 young patients with T1 DM adapting insulin injection therapy. The Chinese version of the DBRS was divided to four subscales. Cronbach's α for the total scale was 0.92. The mean inter-item and item-total correlations were 0.35 and 0.54 respectively. Test-retest reliability showed good temporal stability(r=0.81, P=0.001). Negative correlations were found between DBRS scores with the Diabetes Distress Scale scores(r=.0.32, P=0.003) and hemoglobin A1 c(HbA1 c) levels(r=.0.36, P=0.002). Higher DBRS scores correlated with better glycemic control. The Chinese insulin injection therapy version of the DBRS is well translated and culturally adapted. 7Expression and intercellular trafficking of the VP22 protein of CVI988/Rispens vaccine strain of Marek’s disease virus显示文摘The viral protein 22 (VP22) in the tegument of Marek’s disease virus serotype 1 (MDV-1) plays an im-portant role in cell-to-cell spread and viral propagation. Antiserum against the carboxyl terminus of VP22 was prepared by immunizing mice with recombinant VP22 expressed in E. coli, and used to in-vestigate its expression in chicken embryo fibroblast (CEF) cells infected with different MDV-1 strains. At an infection dose of PFU=50, intercellular trafficking of the VP22 into the nuclei of the surrounding receipt cells was detected as early as 3 hours post infection. By 6 hours after infection (before viral plague formation), the protein was detected in the whole nuclei of the recipient cells with no difference among MDV-1 strains CVI988/Rispens, GA and RB1B. Intra-nuclear accumulation of the VP22 protein was further increased when the viral plagues started to form. 8Rupture process of the Wenchuan M8.0 earthquake of Sichuan, China: the segmentation feature显示文摘Moment tensor solution, rupture process and rupture characteristics of the great Wenchuan M8.0 earthquake are studied by using 39 long-period P and SH waveforms with evenly azimuth coverage of stations. Our results reveal that the Wenchuan M8.0 event consisted of 5 sub-events of Mw≥7.3 occurring succesively in time and space. Rupture started with a Mw7.3 introductory strike-slip faulting in the first 12 s, then within 12?40 s, two sub-events with Mw7.6 and Mw7.4 occurred within 80 km northeast from the initial point with the dominant rupture type of thrust moving. From 40 to 62 s, a Mw7.5 and Mw7.4 right-lateral strike-slip type of sub-events occurred on the two sides of Beichuan, 120 km away northeast from the initial point. The whole rupture process lasted 105 s and unilaterally propagated from the initial point on the WS section of the Yingxiu-Beichuan fault to the NE direction, resulting in a 230-km-long surface rupture zone and the average surface dislocation is up to 4 m. Two asperities are identified and the whole rupture process is formed by WS and NE parts. In the WS part named Dujiangyang-Wenchuan, where the initial point is located, the rupture process showed reverse faulting with the maximum slip of 8.2 m. Around Mianzhu, rupture changed to right-lateral strike slip faulting and formed a Beichuan-Qingchuan large slip area. The rupture area on this part is about 10 km in depth, shallower than on the WS part. The maximum slip is 6.53 m. Consequently, there formed 2 segments with the surface dislocation larger than 6 m. One is the Dujiangyan-Wenchuan segment with the maximum surface displacement of 6.44 m, the other is the Beichuan-Qingchuan segment with the maximum surface displacement of 6.53 m. 12Optimized prime editing efficiently generates glyphosate-resistant rice plants carrying homozygous TAP-IVS mutation in EPSPS显示文摘Dear Editor,Prime editing is a novel and universal CRISPR-Cas-derived precise genome-editing technology and has great potentials for applications in basic plant research and crop molecular breeding(Anzalone et al.,2019).Although low efficiency has restrained the original prime editors(PEs)from being used as a routine tool for precise genome editing in plants,an iterative update of the PEs is removing this obstacle(Lin et al.,2021;Xu et al.,2022).Recently,the Liu group reported three optimization strategies for improving prime editing efficiency(Chen et al.,2021;Nelson et al.,2022).The first strategy is based on engineered prime editing guide RNAs(epegRNAs),which were generated by incorporating structured RNA motifs to the 3′terminus of pegRNAs.This strategy enhances pegRNA stability and prevents degradation of the 3′extension(Nelson et al.,2022).The second strategy is based on the optimized PE2 protein(PEmax),which harbors a SpCas9 variant with increased nuclease activity,an additional nuclear localization signal(NLS)sequences,and a new linker between nCas9 and reverse transcriptase(Chen et al.,2021).The third strategy is based on inhibition of DNA mismatch repair(MMR)in cells(Chen et al.,2021).In this work,we tested the optimized PEs generated with these three strategies in rice,demonstrating that the optimized PEs greatly improved prime editing efficiency in rice.We named the two optimized PEs ePE3max and ePE5max:the former is comprised of the PEmax protein,an epegRNA with evopreQ1 appended to its 3′end,and a nicking sgRNA;the latter is comprised of the ePE3max system and a dominant negative OsMLH1 variant for inhibiting MMR.Using the two optimized PEs,we efficiently generated homozygous and heterozygous T173I,A174V,and P177S(TAP-IVS)mutation in EPSPS in rice,which lays a solid foundation for rice non-transgenic glyphosate-resistance breeding.Yuanyuan Jiang Yiping Chai Dexin Qiao Junya Wang Cuiping Xin Wei Sun Zhenghong Cao Yu Zhang Yun Zhou Xue-Chen Wang Qi-Jun Chen 2022Molecular Plant2022,15,11:3
13Object-Based Classification Method for PolSAR Images with Improved Scattering Powers and Contextual Features显示文摘This paper proposes a new object-based classification method for Polarimetric synthetic aperture radar(Pol SAR) images, which considers scattering powers from an improved model-based polarimetric decomposition approach, as well as the spatial and textural features. With the decomposition, the scattering ambiguities between oriented buildings and vegetation are reduced. Furthermore,various contextual features are extracted from the object and incorporated into the K-nearest neighbors(k-NN)based classification. To reduce the feature redundancy, a new Supervised locally linear embedding(S-LLE) dimensionality reduction method is introduced to map the high dimensional polarimetric signatures into the most compact low-dimensional structure for classification. 14Biocompatible, small-sized and well-dispersed gold nanoparticles regulated by silk fibroin fiber from Bombyx mori cocoons显示文摘Biocompatible, small-sized but well-dispersed gold nanoparticles (Au NPs) remain a major challenge for their synthesis. Here a convenient solution impregnation technique is developed to prepare such Au NPs under the regulation of degummed silk fibroin fibers (SFFs) extracted from Bombyx mori cocoons. SFFs play multiple roles in the formation of Au NPs such as reactive substrate to capture AuCl4^- ions by the chelation of -C = 0, reducing agent for Au(0) by the reduction of -OH , and modifiers to render biocompatible Au NPs by some functional groups and biomolecules. The as-prepared Au NPs with a size of 7-10 nm are embedded in the solid SFF substrate, and can disperse well in the liquid system by the disintegration of SFFs into silk fibroin (SF) in a certain CaCl2 solution. 16Thermodynamic database of the phase diagrams in the Mg-Al-Zn-Y-Ce system显示文摘The Mg-Al-Zn-Y-Ce system is one of the key systems for designing high-strength Mg alloys. The purpose of the present article is to develop a thermodynamic database for the Mg-Al-Zn-Y-Ce multicomponent system to design Mg alloys using the calculation of phase diagrams (CALPHAD) method, where the Gibbs energies of solution phases such as liquid, fcc, bcc, and hcp phases were described by the subregular solution model, whereas those of all the compounds were de- scribed by the sublattice model. The thermodynamic parameters describing Gibbs energies of the different phases in this database were evaluated by fitting the experimental data for phase equilibria and thermodynamic properties. On the basis of this database, a lot of information concerning stable and metastable phase equilibria of isothermal and vertical sections, mo- lar fractions of constituent phases, the liquidus projection, etc., can be predicted. 17Tidal Asymmetry Changes in a Shallow Mud Estuary by a Restoration Project显示文摘The tidal asymmetry-induced siltation below tidal barriers is a worldwide problem that restricts regional socio-economic and environmental development. The hydrodynamic processes of the small mud estuary also feature a high uncertainty after estuary restoration measures. In this study, a hydrodynamic model based on the MIKE21 is used to quantify the responses of tidal asymmetry to a two-phase restoration project in Shuanglong Estuary, Bohai Bay, China. According to the numerical modeling results, the tidal flat removal in the upper estuary(first-phase restoration) induces the flood asymmetry switching to the ebb asymmetry in unrestored reach but enhances flood asymmetry in widening restored reach. Although the tidal asymmetry reverts to flood-dominated pattern after full restoration over the estuary, the imbalance between flood and ebb velocities is relieved. A possible net sediment transport pattern based on a comparison of dominant asymmetric current and actual sediment transport period shows net sediments in the upper estuary and inlet transport seaward and landward, respectively, in the first-phase restoration, whereas landward net sediment transport occurs in the whole estuary under the second-phase restoration scenario. Given these results, we assume that a switch from the flood-dominated estuary to ebb-dominated estuary can be caused by redesigning the cross-sectional profile. The quantitative comparison of Lagrangian residual currents also implies that a channel–shoal structure rather than a flat bathymetry can promote the mass transport. 18The Role of Ti in the Preparation of SiC/7075 Aluminum Matrix Composite显示文摘SiC/7075 aluminum matrix composites were prepared by a liquid stirring method.The role of Ti facilitating the preparation of SiC/7075 aluminum matrix were studied by means of direct-reading spectrometer,scanning electron microscope,energy dispersive spectrometer,X-ray diffraction and the sessile drop method.The results show that the SiC content in the SiC/7075 composite increases with an increase of Ti addition.The addition of Ti can significantly improve the wettability of SiC/Al system,there is a critical value of above 0.5%of Ti content in improving the wettability of the Al/SiC system at 1173K.The temperature of the'non wetting-wetting'transition for the(Al-2Ti)/SiC system is about 1123K,the contact angle decreases to 88°at 200 seconds and reaches a stable contact angle of 28°at 2100 seconds.ZHANG Wenda CHEN Chengcheng XU Hong CHE Luchang GUO Cuiping DU Zhenmin 2020Journal of Wuhan University of Technology(Materials Science)2020,35,2:2
