2Neuroprotective Autophagic Flux Induced by Hyperbaric Oxygen Preconditioning is Mediated by Cystatin C显示文摘We have previously reported that Cystatin C(CysC) is a pivotal mediator in the neuroprotection induced by hyperbaric oxygen(HBO) preconditioning; however,the underlying mechanism and how CysC changes after stroke are not clear. In the present study, we demonstrated that CysC expression was elevated as early as 3 h after reperfusion, and this was further enhanced by HBO preconditioning. Concurrently, LC3-II and Beclin-1, two positive-markers for autophagy induction, exhibited increases similar to CysC, while knockdown of CysC blocked these elevations. As a marker of autophagy inhibition, p62 was downregulated by HBO preconditioning and this was blocked by CysC knockdown. Besides, the beneficial effects of preserving lysosomal membrane integrity and enhancing autolysosome formation induced by HBO preconditioning were abolished in CysC-/-rats.Furthermore, we demonstrated that exogenous CysC reduced the neurological deficits and infarct volume after brain ischemic injury, while 3-methyladenine partially reversed this neuroprotection. 3Toripalimab plus chemotherapy as second-line treatment in previously EGFR-TKI treated patients with EGFR-mutant-advanced NSCLC:a multicenter phase-II trial显示文摘This multicenter phase-II trial aimed to investigate the efficacy,safety,and predictive biomarkers of toripalimab plus chemotherapy as second-line treatment in patients with EGFR-mutant-advanced NSCLC.Patients who failed from first-line EGFR-TKIs and did not harbor T790M mutation were enrolled.Toripalimab plus carboplatin and pemetrexed were administrated every three weeks for up to six cycles,followed by the maintenance of toripalimab and pemetrexed.The primary endpoint was objective-response rate(ORR).Integrated biomarker analysis of PD-L1 expression,tumor mutational burden(TMB),CD8+tumor-infiltrating lymphocyte(TIL)density,whole-exome,and transcriptome sequencing on tumor biopsies were also conducted.Forty patients were enrolled with an overall ORR of 50.0%and disease-control rate(DCR)of 87.5%.The median progression free survival(PFS)and overall survival were 7.0 and 23.5 months,respectively.The most common treatment-related adverse effects were leukopenia,neutropenia,anemia,ALT/AST elevation,and nausea.Biomarker analysis showed that none of PD-L1 expression,TMB level,and CD8+TIL density could serve as a predictive biomarker.Integrated analysis of whole-exome and transcriptome sequencing data revealed that patients with DSPP mutation had a decreased M2 macrophage infiltration and associated with longer PFS than those of wild type.Toripalimab plus chemotherapy showed a promising anti-tumor activity with acceptable safety profiles as the second-line setting in patients with EGFR-mutant NSCLC.DSPP mutation might serve as a potential biomarker for this combination.A phase-III trial to compare toripalimab versus placebo in combination with chemotherapy in this setting is ongoing(NCT03924050).Tao Jiang Pingyang Wang Jie Zhang Yanqiu Zhao Jianying Zhou Yun Fan Yongqian Shu Xiaoqing Liu Helong Zhang Jianxing He Guanghui Gao Xiaoqian Mu Zhang Bao Yanjun Xu Renhua Guo Hong Wang Lin Deng Ningqiang Ma Yalei Zhang Hui Feng Sheng Yao Jiarui Wu Luonan Chen Caicun Zhou Shengxiang Ren 2021Signal Transduction and Targeted Therapy2021,6,11:8
4Biotransformation of aesculin by human gut bacteria and identification of its metabolites in rat urine显示文摘AIM:To observe the biotransformation process of a Chinese compound,aesculin,by human gut bacteria,and to identify its metabolites in rat urine.METHODS:Representative human gut bacteria were collected from 20 healthy volunteers,and then utilized in vitro to biotransform aesculin under anaerobic conditions.At 0,2,4,8,12,16,24,48 and 72 h post- incubation,10 mL of culture medium was collected.Metabolites of aesculin were extracted 3×from rat urine with methanol and analyzed by HPLC.For in vivo metabolite analysis,aesculetin(100 mg/kg)was administered to rats via stomach gavage,rat urine was collected from 6 to 48 h post-administration,and metabolite analysis was performed by LC/ESI-MS and MS/MS in the positive and negative modes.RESULTS:Human gut bacteria could completely convert aesculin into aesculetin in vitro.The biotransformation process occurred from 8 to 24 h post-incubation,with its highest activity was seen from 8 to 12 h.The in vitro process was much slower than the in vivo process.In contrast to the in vitro model,six aesculetin metabolites were identified in rat urine,including 6-hydroxy-7-gluco- coumarin(M1),6-hydroxy-7-sulf-coumarin(M2),6,7-di- gluco-coumarin(M3),6-glc-7-gluco-coumarin(M4),6-O-methyl-7-gluco-coumarin(M5)and 6-O-methyl-7- sulf-coumarin(M6).Of which,M2 and M6 were novel metabolites.CONCLUSION:Aesculin can be transferred into aesculetin by human gut bacteria and is further modified by the host in vivo.The diverse metabolites of aesculin may explain its pleiotropic pharmaceutical effects.Wei-Jun Ding Yun Deng Hao Feng Wei-Wei Liu Rong Hu Xiang Li Zhe-Ming Gu Xiao-Ping Dong 2009World Journal of Gastroenterology2009,15,12:5
5Grade classification of neuroepithelial tumors using high-resolution magic-angle spinning proton nuclear magnetic resonance spectroscopy and pattern recognition显示文摘Clinical data have shown that survival rates vary considerably among brain tumor patients,according to the type and grade of the tumor.Metabolite profiles of intact tumor tissues measured with high-resolution magic-angle spinning proton nuclear magnetic resonance spectroscopy (HRMAS 1H NMRS) can provide important information on tumor biology and metabolism.These metabolic fingerprints can then be used for tumor classification and grading,with great potential value for tumor diagnosis.We studied the metabolic characteristics of 30 neuroepithelial tumor biopsies,including two astrocytomas (grade I),12 astrocytomas (grade II),eight anaplastic astrocytomas (grade III),three glioblastomas (grade IV) and five medulloblastomas (grade IV) from 30 patients using HRMAS 1H NMRS.The results were correlated with pathological features using multivariate data analysis,including principal component analysis (PCA).There were significant differences in the levels of N-acetyl-aspartate (NAA),creatine,myo-inositol,glycine and lactate between tumors of different grades (P<0.05).There were also significant differences in the ratios of NAA/creatine,lactate/creatine,myo-inositol/creatine,glycine/creatine,scyllo-inositol/creatine and alanine/creatine (P<0.05).A soft independent modeling of class analogy model produced a predictive accuracy of 87% for high-grade (grade III-IV) brain tumors with a sensitivity of 87% and a specificity of 93%.HRMAS 1H NMR spectroscopy in conjunction with pattern recognition thus provides a potentially useful tool for the rapid and accurate classification of human brain tumor grades.CHEN WenXue LOU HaiYan ZHANG HongPing NIE Xiu LAN WenXian YANG YongXia XIANG Yun QI JianPin LEI Hao TANG HuiRu CHEN FenEr DENG Feng 2011Science China(Life Sciences)2011,54,7:5

7Hypoxia induces T-cell apoptosis by inhibiting chemokine C receptor 7 expression:the role of adenosine receptor A2显示文摘Hypoxia is a major characteristic of the tumor microenvironment,and its effects on immune cells are proposed to be important factors for the process of tumor immune escape.It has been reported that hypoxia affects the function of dendritic cells and the antitumor function of T cells.Here we discuss the effects of hypoxia on T-cell survival.Our results showed that hypoxia induced apoptosis of T cells.Adenosine and adenosine receptors(AR)are important to the hypoxia-related signaling pathway.Using AR agonists and antagonists,we demonstrated that hypoxia-induced apoptosis of T cells was mediated by A^(2a)and A^(2b)receptors.Furthermore,we are the first,to our knowledge,to report that hypoxia significantly inhibited the expression of chemokine C receptor 7(CCR7)of T cells via the A^(2)R signal pathway,perhaps representing a mechanism of hypoxia-induced apoptosis of T cells.Collectively,our research demonstrated that hypoxia induces T-cell apoptosis by the A^(2)R signaling pathway partly by suppressing CCR7.Blocking the A^(2)R signaling pathway and/or activation of CCR7 can increase the anti-apoptosis function of T cells and may become a new strategy to improve antitumor potential.Jintang Sun Yan Zhang Meixiang Yang Yun Zhang Qi Xie Zewu Li Zhaogang Dong Yongmei Yang Biping Deng Alei Feng Weixu Hu Haiting Mao Xun Qu 2010Cellular & Molecular Immunology2010,7,1:3

9Evaluation of the New Automatic Mycob.T Stainer andScanner for Detecting Acid-fast Bacilli in China显示文摘Objective To validate the performance of Mycob. T Stainer and Scanner(MTSS) for detecting acid-fast bacilli(AFB). Methods A total of 3,816 sputum samples from 1,515 tuberculosis(TB) suspects were tested at the Anhui Provincial Chest Hospital and the Linyi People's Hospital from April-August, 2016. Each specimen was placed on two smear slides. One slide was stained by the ziehl-neelsen(ZN) method to be read by conventional microscopy(CM). The other slide was stained and scanned by MTSS. All specimens were decontaminated with 4% NaO H, and then inoculated into solid culture. The performance of MTSS was assessed. Results MTSS produced higher average positivity rate(27.96%) as compared with the CM(26.83%). The overall sensitivity and specificity of MTSS were 78.9% and 93.9%, respectively. The sensitivity and specificity of CM was 77.4% and 95.0%, respectively. Conclusion MTSS exhibited a favorable performance in the detection of AFB. 11Unsupervised content-preserving transformation for optical microscopy显示文摘The development of deep learning and open access to a substantial collection of imaging data together provide a potential solution for computational image transformation,which is gradually changing the landscape of optical imaging and biomedical research.However,current implementations of deep learning usually operate in a supervised manner,and their reliance on laborious and error-prone data annotation procedures remains a barrier to more general applicability.Here,we propose an unsupervised image transformation to facilitate the utilization of deep learning for optical microscopy,even in some cases in which supervised models cannot be applied.Through the introduction of a saliency constraint,the unsupervised model,named Unsupervised content-preserving Transformation for Optical Microscopy(UTOM);can learn the mapping between two image domains without requiring paired training data while avoiding distortions of the image content.UTOM shows promising performance in a wide range of biomedical image transformation tasks,including in silico histological staining,fluorescence image restoration,and virtual fluorescence labeling.Quantitative evaluations reveal that UTOM achieves stable and high-fidelity image transformations across different imaging conditions and modalities.We anticipate that our framework will encourage a paradigm shift in training neural networks and enable more applications of artificial intelligence in biomedical imaging.Xinyang Li Guoxun Zhang Hui Qiao Feng Bao Yue Deng Jiamin Wu Yangfan He Jingping Yun Xing Lin Hao Xie Haoqian Wang Qionghai Dai 2021Light(Science & Applications)2021,10,3:1

18The propensity for tumorigenesis in human induced pluripotent stem cells is related with genomic instability显示文摘The discovery of induced pluripotent stem cells (iPSCs) is a promising advancement in the field of regenerative medicine. Previous studies have indicated that the teratoma-forming propensity of iPSCs is variable; however, the relationship between tumorigenic potential and genomic instability in human iPSCs (HiPSCs) remains to be fully elucidated. Here, we evaluated the malignant potential of HiPSCs by using both colony formation assays and tumorigenicity tests. We demonstrated that HiPSCs formed tumorigenic colonies when grown in cancer cell culture medium and produced malignancies in immunodeficient mice. Furthermore, we analyzed genomic instability in HiPSCs using whole-genome copy number variation analysis and determined that the extent of genomic instability was related with both the cells′ propensity to form colonies and their potential for tumorigenesis. These findings indicate a risk for potential malignancy of HiPSCs derived from genomic instability and suggest that quality control tests, including comprehensive tumorigenicity assays and genomic integrity validation, should be rigorously executed before the clinical application of HiPSCs. 19Recycling Carbon Resources from Waste PET to Reduce Carbon Dioxide Emission:Carbonization Technology Review and Perspective显示文摘Greenhouse gas emissions from waste plastics have caused global warming all over the world,which has been a central threat to the ecological environment for humans,flora and fauna.Among waste plastics,waste polyethylene terephthalate(PET)is attractive due to its excellent stability and degradation-resistant.Therefore,merging China’s carbon peak and carbon neutrality goals would be beneficial.In this review,we summarize the current state-of-the-art of carbon emission decrease from a multi-scale perspective technologically.We suggest that the carbon peak for waste PET can be achieved by employing the closed-loop supply chain,including recycling,biomass utilization,carbon capture and utilization.Waste PET can be a valuable and renewable resource in the whole life cycle.Undoubtedly,all kinds of PET plastics can be ultimately converted into CO_(2),which can also be feedstock for various kinds of chemical products,including ethyl alcohol,formic acid,soda ash,PU,starch and so on.As a result,the closed-loop supply chain can help the PET plastics industry drastically reduce its carbon footprint.Xing Zhou Qi Wang Sai Feng Jingrui Deng Keming Zhu Yun Xing Xiaolian Meng Xiaojun Wang Lu Li 2023Journal of Renewable Materials2023,11,5:0
20Molecular phylogenomic analysis reveals a single origin of monkeypox virus transmission in Guangzhou,China in June 2023显示文摘The monkeypox(mpox)virus has caused worldwide transmission since its initial report in England in early May 2022.Available data from the World Health Organization(WHO)show that Europe and the Americas experienced a huge wave of mpox virus infection.Now the number of infected cases is on the rise in Asia.Several sporadic infections have been reported in China.In this study,we obtained high‐quality whole viral genomic sequences using a mpox virus‐specific amplicon‐based sequencing strategy.Our analysis of the phylogenomic characteristics indicated that all eight mpox virus sequences from Guangzhou belonged to the clade IIb lineage B.1.3 cluster.However,we could not locate the exact origins where the virus was imported,based on all the available mpox virus sequences from the Global Initiative on Sharing Avian Influenza Data(GISAID)database(https://gisaid.org/),except for their closest sequence similarity to that was reported from Japan.Novel amino acid mutations were found among the eight cases,suggesting that a local transmission may have occurred in Guangzhou,China.Mengling Jiang Xizi Deng Qinghong Fan Fei Gu Huiqin Yang Jian Wang Xiaoping Tang Fengyu Hu Yun Lan Feng Li 2023Biosafety and Health2023,5,5:0
