| 1 | In vitro antibacterial activity of thymol and carvacrol and their effects on broiler chickens challenged with Clostridium perfringens显示文摘Background:In the post-antibiotic era,essential oils(EO) are promising alternatives to growth-promoting antibiotics.The aim of the present study was to investigate the antibacterial activities of an EO product and its components thymol and carvacrol in vitro,and the efficacy of EO to control Clostridium perfringens challenge in broiler chickens.Results:The in vitro minimum inhibitory concentration assay showed strong antibacterial activity of the EO product,thymol,and carvacrol against pathogenic Escherichia coli,C.perfringens,and Salmonella strains,and weak activity towards beneficial Lactobacillus strains.Besides,an additive effect was observed between thymol and carvacrol.The in vivo study was carried out with 448 male broiler chicks following a 4 × 2 factorial arrangement to test the effects of EO supplementation(0,60,120,or 240 mg/kg EO in wheat-based diet),pathogen challenge(with or without oral gavage of C perfringens from day 14 to day 20) and their interactions.Each treatment consisted of eight replicate pens(seven birds/pen).The challenge led to macroscopic gut lesions,and resulted in a significant increase in ileal populations of C.perfringens and Escherichia subgroup(P< 0.05) on day 21.Dietary EO supplementation did not influence C perfringens numbers,but linearly alleviated intestinal lesions on day 21 and 28(P=0.010 and 0.036,respectively),and decreased Escherichia populations in ileum with increased EO dosages(P=0.027 and 0.071 for day 21 and 28,respectively).For caecum,EO quadratically influenced Lactobacillus populations on day 21(p=0.002),and linearly decreased the numbers of total bacteria and Escherichia on day 28(P=0.026 and 0.060,respectively).Mean thymol and carvacrol concentrations in the small intestine were 0.21 and 0.20 μg/g in intestinal digesta(wet weight),respectively,for birds fed 60 mg/kg EO,and 0.80 and 0.71 μg/g,respectively,for birds fed 240 mg/kg EO.Conclusions:These results indicated that dietary EO supplementation could affect intestinal microbiota and alleviate intestinal lesions in broilers,which may contribute in controlling C.perfringens infection in broiler chickens. | Encun Du Liping Gan Zhui Li Weiwei Wang Dan Liu Yuming Guo | 2016 | Journal of Animal Science and Biotechnology2016,7,1: | 14 |

| 3 | Toxicity of Graphene Quantum Dots in Zebrafish Embryo显示文摘Objective To evaluate the bio-safety of graphene quantum dots(GQDs), we studied its effects on the embryonic development of zebrafish. Methods In vivo, biodistribution and the developmental toxicity of GQDs were investigated in embryonic zebrafish at exposure concentrations ranging from 12.5-200 μg/m L for 4-96 h post-fertilization(hpf). The mortality, hatch rate, malformation, heart rate, GQDs uptake, spontaneous movement, and larval behavior were examined. Results The fluorescence of GQDs was mainly localized in the intestines and heart. As the exposure concentration increased, the hatch and heart rate decreased, accompanied by an increase in mortality. Exposure to a high level of GQDs(200 μg/mL) resulted in various embryonic malformations including pericardial edema, vitelline cyst, bent spine, and bent tail. The spontaneous movement significantly decreased after exposure to GQDs at concentrations of 50, 100, and 200 μg/mL. The larval behavior testing(visible light test) showed that the total swimming distance and speed decreased dose-dependently. Embryos exposed to 12.5 μg/mL showed hyperactivity while exposure to higher concentrations(25, 50, 100, and 200 μg/mL) caused remarkable hypoactivity in the light-dark test. Conclusion Low concentrations of GQDs were relatively non-toxic. | 9 | Seismic behavior of thin-walled circular and stiffened square steel tubed-reinforced-concrete columns显示文摘Steel tubed-reinforced-concrete(TRC) columns have been gradually used in the construction of high-rise buildings recently because of their high axial load-carrying capacities and excellent seismic behavior. Existing studies about their seismic behavior were focused on columns with relatively thick tubes, i.e., diameter-to-thickness/width-to-thickness(D/t) ratios were below 100,while little is known about thin-walled TRC columns, especially for square TRC columns. Considering the infilled concrete of square TRC columns is non-uniformly and non-effectively confined, accordingly, stiffened square TRC columns are usually adopted in practice. Thus, two thin-walled circular TRC columns(D/t=120) and two stiffened square ones with diagonal stiffeners in plastic hinge regions(D/t=106) were tested under a constant axial compression combined with cyclic lateral loading.Both the circular and stiffened square TRC columns had the same cross sectional area, tube thickness, reinforcing bar ratio and column height. Flexural failure occurred for all the four specimens. Test results showed the strengths of the stiffened square TRC columns were a little higher in comparison to their circular counterparts; the ductility and energy dissipation capacities were excellent for both the stiffened and circular TRC columns, indicating very good confinement was gained from the yielded steel tubes of the plastic hinge regions at the peak loads. And shear stresses(35–90 MPa) in the sheared plates showed their moderate contribution of carrying lateral loads. | 11 | Cellular Tolerance, Accumulation and Distribution of Cadmium in Leaves of Hyperaccumulator Picris divaricata显示文摘Knowledge of cellular metal homeostasis will provide a better understanding of the mechanisms involved in metal tolerance and hyperaccumulation in metal-hyperaccumulating plants. Energy dispersive X-ray spectrometry (EDS) was used to determine the localization of cadmium (Cd) in leaves of the Zn/Cd hyperaccumulator Picris divaricata which had a shoot Cd concentration of 565 mg kg 1 after 2 weeks of growth in solution culture supplying 10 μmol L-1 CdCl2 . The results indicated that Cd was distributed mainly in the trichomes, upper and lower epidermis and bundle sheath cells, with a relatively low level of Cd in mesophyll cells. Mesophyll protoplasts isolated from leaves remained viable after 24 h exposure to CdCl2 at a concentration up to 1 mmol L-1 , indicating their high tolerance to Cd. The intracellular Cd was visualized by staining with Leadmium Green dye, a cellular permeable Cd fluorescence probe. The results showed that the majority of protoplasts (> 82%) did not accumulate Cd, with only a minority (< 18%) showing Cd accumulation. In the Cd-accumulating protoplasts, Cd accumulation was depressed by the addition of Fe2+ , Mn2+ and the metabolic inhibitor carbonyl cyanide m-chlorophenylhydrazone (CCCP), but not by Ca 2+ or Zn2+ . Furthermore, the entire process of Cd uptake from external solution into the cytoplasm and subsequent sequestration into vacuoles was successfully recorded by confocal images.
