3N-AcetyI-L-cysteine and pyrrolidine dithiocarbamate inhibited nuclear factor-κB activation in alveolar macrophages by different mechanisms显示文摘Aim:To study the effects of N-acetyl-L-cysteine(NAC)and pyrrolidine dithio-carbamate(PDTC)on the phosphorylation of IκB kinase(IKK)β,IKKα,and IλBαin alveolar macrophages(AM),and to explore the pharmacological mechanisms ofNAC and PDTC as inhibitors of NF-κB activation.Methods:AM were collectedfrom bronchoalveolar lavage fluid from the patients with chronic obstructive pul-monary disease.The AM were incubated for 1.5 h with NAC and PDTC,and thenstimulated for 90 min by either tumor necrosis factor(TNF)-α or interleukin(IL)-1.Western blotting was used to detect the protein phosphorylation levels of IKKβ,IKKα,and IκBα.NF-κB activity was analyzed by using an electrophoretic mobil-ity shift assay.Results:NAC inhibited the phosphorylation of IKKβ,IKKα,andIκBα induced by TNF-α,but had no effect on the phosphorylation of IKKβ,IKKαand IκBα induced by IL-1.PDTC did not inhibit the phosphorylation of IκBαinduced by TNF-α or IL-1.Similarly,NAC inhibited the activation of NF-κBinduced by TNF-α,but had no effect on the activation of NF-κB induced by IL-1.PDTC significantly inhibited the activation of NF-κB induced by TNF-α and IL-1.The electrophoretic mobility shift assay also showed that PDTC and NAC do notdirectly inhibit NF-κB DNA binding activity in vitro.Conclusion:PDTC preventsthe degradation of IκBα via the ubiquitylation-proteasome proteolytic pathway.NAC can inhibit the processes upstream of IKK activation induced by TNF-α,which results in the decline of NF-κB activity.Ya-qing LI Zhen-xiang ZHANGI Yong-jian XU Wang NI Shi-xin CHEN Zhao YANG Dan MA 2006Acta Pharmacologica Sinica2006,27,3:29

5Comparative analysis of the complete chloroplast genome among Prunus mume,P.armeniaca,and P.salicina显示文摘Prunus mume Sieb.et Zucc.,P.armeniaca L.,and P.salicina L.are economically important fruit trees in temperate regions.These species are taxonomically perplexing because of shared interspecific morphological traits and variation,which are mainly attributed to hybridization.The chloroplast is cytoplasmically inherited and often used for evolutionary studies.We sequenced the complete chloroplast genomes of P.mume,P.armeniaca,and P.salicina using Illumina sequencing followed by de novo assembly.The three chloroplast genomes exhibit a typical quadripartite structure with conserved genome arrangement,structure,and moderate divergence.The lengths of the genomes are 157,815,157,797,and 157,916 bp,respectively.The length of the large single-copy region(LSC)region is 86,113,86,283,and 86,122 bp,and the length of the SSC region is 18,916,18,734,and 19,028 bp;the IR region is 26,393,26,390,and 26,383 bp,respectively.Each of the three chloroplast genomes encodes 133 genes,including 94 protein-coding,31 tRNA,and eight rRNA genes.Differential gene analysis for the three species revealed that trnY-ATA is a unique gene in P.armeniaca;in contrast,the gene trnI-TAT is only present in P.mume and P.salicina,though the position of the gene in these chloroplast genomes differs.Further comparative analysis of the complete chloroplast genome sequences revealed that the ORF genes and the sequences of linked regions rps16 and atpA,atpH and atpI,trnc-GCA and psbD,ycf3 and atpB,and rpL32 and ndhD are significantly different and may be used as molecular markers in taxonomic studies.Phylogenetic evolution analysis of the three species suggests that P.mume has a closer genetic relationship to P.armeniaca than to P.salicina.Song Xue Ting Shi Wenjie Luo Xiaopeng Ni Shahid Iqbal Zhaojun Ni Xiao Huang Dan Yao Zhijun Shen Zhihong Gao 2019Horticulture Research2019,6,1:10
6Comparative efficacy and safety of Chinese herbal injections combined with transcatheter hepatic arterial chemoembolization in treatment of liver cancer: a bayesian network Meta-analysis显示文摘OBJECTIVE: To address the optimal Chinese herbal injections(CHIs) against liver cancer, the present network Meta-analysis is designed to investigate the comparative efficacy and safety of different CHIs.METHODS: Several electronic databases were searched up to June 1 st, 2017. The quality assessment was conducted and network Meta-analysis was performed to compare the efficacy and safety of different CHIs plus transcatheter hepatic arterial chemoembolization(TACE). Primary outcomes were 1-year and 2-year survival rate, the secondary outcomes includes the clinical effective rate, performance status and the adverse reactions(ADRs). Data analysis was applied Stata 13.0 and Win BUGS 1.4 software.RESULTS: A total of 105 randomized controlled trials(RCTs) were identified for inclusion in this analysis, with data for 7683 patients and 13 CHIs. The results suggested that Javanica oil emulsion,Huachansu injection plus TACE were more favorable for 1-year and 2-year survival rate than other CHIs. 8Improving fatigue performance of Ti-6Al-4V alloy via ultrasonic surface rolling process显示文摘The effect of a gradient nanostructured(GNS) surface layer obtained by ultrasonic surface rolling process(USRP) on the fatigue behavior of Ti-6Al-4V alloy has been studied in this paper. Microstructure, surface topography, surface roughness and residual stress measurements were performed to characterize the surface under different conditions. Rotating bending fatigue tests were carried out to evaluate the fatigue behavior of different treatments. The results present a remarkable fatigue performance enhancement for the Ti-6Al-4V alloy with a GNS surface layer obtained by application of USRP with respect to the untreated condition, notwithstanding its considerable surface roughness due to severe ultrasonic impacts and extrusions. Mechanical surface polishing treatment further enhances the beneficial effects of USRP on the fatigue performance. The significantly improved fatigue performance can mainly be ascribed to the compressive residual stress. 9Effects of Shuanghuanglian oral liquids on patients with COVID-19:a randomized,open-label,parallel-controlled,multicenter clinical trial显示文摘We conducted a randomized,open-label,parallel-controlled,multicenter trial on the use of Shuanghuanglian(SHL),a traditional Chinese patent medicine,in treating cases of COVID-19.A total of 176 patients received SHL by three doses(56 in low dose,61 in middle dose,and 59 in high dose)in addition to standard care.The control group was composed of 59 patients who received standard therapy alone.Treatment with SHL was not associated with a difference from standard care in the time to disease recovery.Patients with 14-day SHL treatment had significantly higher rate in negative conversion of SARS-CoV-2 in nucleic acid swab tests than the patients from the control group(93.4%vs.73.9%,P=0.006).Analysis of chest computed tomography images showed that treatment with high-dose SHL significantly promoted absorption of inflammatory focus of pneumonia,which was evaluated by density reduction of inflammatory focus from baseline,at day 7(mean difference(95%CI),−46.39(−86.83 to−5.94)HU;P=0.025)and day 14(mean difference(95%CI),−74.21(−133.35 to−15.08)HU;P=0.014).No serious adverse events occurred in the SHL groups.This study illustrated that SHL in combination with standard care was safe and partially effective for the treatment of COVID-19.Li Ni Zheng Wen Xiaowen Hu Wei Tang Haisheng Wang Ling Zhou Lujin Wu Hong Wang Chang Xu Xizhen Xu Zhichao Xiao Zongzhe Li Chene Li Yujian Liu Jialin Duan Chen Chen Dan Li Runhua Zhang Jinliang Li Yongxiang Yi Wei Huang Yanyan Chen Jianping Zhao Jianping Zuo Jianping Weng Hualiang Jiang Dao Wen Wang 2021Frontiers of Medicine2021,15,5:7
10Mechanism of Chaihu Shugan Powder(柴胡疏肝散) for Treating Depression Based on Network Pharmacology显示文摘Objective:To analyze the effective components of Chinese medicine(CM)contained in Chaihu Shugan Powder(柴胡疏肝散,CSP)in the treatment of depressive disorders and to predict its anti-depressant mechanism by network pharmacology.Methods:Absorption,distribution,metabolism,excretion,and toxicity calculation method was used to screen the active components of CSP.Traditional Chinese Medicine System Pharmacological Database Analysis Platform and text mining tool(GoPuMed database)were used to predict and screen the active ingredients of CSP and anti-depressive targets.Through Genetic Association Database,Therapeutic Target Database,and PharmGkb database targets for depression were obtained.Cytoscape3.2.1 software was used to establish a network map of the active ingredients-targets of CSP,and to analyze gene function and metabolic pathways through Database for Annotation,Visualization and Integrated Discovery and the Omicshare database.Results:The 121 active ingredients and 15 depression-related targets which were screened from the database can exert antidepressant effects by improving the neural plasticity,growth,transfer condition and gene expression of neuronal cell,and the raise of the expression of gap junction protein.The 15 targets passed 14 metabolic pathways,mainly involved in the regulation of neurotransmitters(5-hydroxytryptamine,dopamine and epinephrine),inflammatory mediator regulation of TRP channels,calcium signaling pathway,cyclic adenosine monophosphate signaling pathway and neuroactive ligand-receptor interaction and other signal channels to exert anti-depressant effects.Conclusion:This article reveals the possible mechanism of CSP in the treatment of depression through network pharmacology research,and lays a foundation for further target studies.LIU Yuan-yue HU Dan FAN Qi-qi ZHANG Xiao-hao ZHU Yi-cheng NI Miao-yan WANG Yan-ming ZHANG Lan-kun SHENG Lei 2020Chinese Journal of Integrative Medicine2020,26,12:6
11Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). 12KLF4 initiates sustained YAP activation to promote renal fibrosis in mice after ischemia-reperfusion kidney injury显示文摘Acute renal injury(AKI)causes a long-term risk for progressing into chronic kidney disease(CKD)and interstitial fibrosis.Yes-associated protein(YAP),a key transcriptional cofactor in Hippo signaling pathway,shuttles between the cytoplasm and nucleus,which is required for the renal tubular epithelial cells repair in the acute phase of AKI.In this study we investigated the role of YAP during ischemia-reperfusion(IR)-induced AKI to CKD.Mice were subjected to left kidney IR followed by removal of the right kidney on the day before tissue harvests.Mouse shRNA expression adenovirus(Ad-shYAP or Ad-shKLF4)and mouse KLF4 expression adenovirus(Ad-KLF4)were delivered to mice by intrarenal injection on D7 after IR.We showed that the expression and nucleus distribution of YAP were persistently increased until the end of experiment(D21 after IR).The sustained activation of YAP in post-acute phase of AKI was accompanied by renal dysfunction and interstitial fibrosis.Knockdown of YAP significantly attenuated IR-induced renal dysfunction and decreased the expression of fibrogenic factors TGF-βand CTGF in the kidney.We showed that the expression of the transcription factor KLF4,lined on the upstream of YAP,was also persistently increased.Knockdown on KLF4 attenuated YAP increase and nuclear translocation as well as renal functional deterioration and interstitial fibrosis in IR mice,whereas KLF4 overexpression caused opposite effects.KLF4 increased the expression of ITCH,and ITCH facilitated YAP nuclear translocation via degrading LATS1.Furthermore,we demonstrated in primary cultured renal tubular cells that KLF4 bound to the promoter region of YAP and positively regulates YAP expression.In biopsy sample from CKD patients,we also observed increased expression and nuclear distribution of YAP.In conclusion,the activation of YAP in the post-acute phase of AKI is implicated in renal functional deterioration and fibrosis although it exhibits beneficial effect in acute phase.Reprogramming factor KLF4 is responsible for the persistent activation of YAP.Blocking the activation of KLF4-YAP pathway might be a way to prevent the transition of AKI into CKD.Dan Xu Pan-pan Chen Pei-qing Zheng Fan Yin Qian Cheng Zhuan-li Zhou Hong-yan Xie Jing-yao Li Jia-yun Ni Yan-zhe Wang Si-jia Chen Li Zhou Xiao-xia Wang Jun Liu Wei Zhang Li-min Lu 2021Acta Pharmacologica Sinica2021,42,3:6

14Aggregation-induced emission luminogen-assisted stimulated emission depletion nanoscopy for super-resolution mitochondrial visualization in live cells显示文摘Aggregation-induced emission luminogens (AIEgens) are fluorescent agents thatare ideal for bioimaging and have been widely used for organeUe targeting, cellularmapping, and tracing. Owing to their promising characteristics, AIEgen-basednanoparticles have recently been used for the stimulated emission depletion(STED) super-resolution imaging of fixed cells. In the present study, and for thefirst time, we used an AIEgen for dynamic STED nanoscopic imaging of a specificorganelle in live cancer cells. TPA-T-CyP is a synthetic red&NIR-emittingluminogen with AIE features that can spontaneously and specifically aggregateon mitochondria without the need for encapsulation or surface modification.The STED efficiency of aggregated TPA-T-CyP can reach more than 80%, andsuper-resolution imaging of TPA-T-CyP-stained mitochondria in live HeLa cellsis possible, with a lateral spatial resolution of 74 nm. 15Mining and Integrating Reliable Decision Rules for Imbalanced Cancer Gene Expression Data Sets显示文摘There have been many skewed cancer gene expression datasets in the post-genomic era. Extraction of differential expression genes or construction of decision rules using these skewed datasets by traditional algorithms will seriously underestimate the performance of the minority class, leading to inaccurate diagnosis in clinical trails. This paper presents a skewed gene selection algorithm that introduces a weighted metric into the gene selection procedure. The extracted genes are paired as decision rules to distinguish both classes, with these decision rules then integrated into an ensemble learning framework by majority voting to recognize test examples; thus avoiding tedious data normalization and classifier construction. 18Portal hypertension exacerbates intrahepatic portosystemic venous shunt and further induces refractory hepatic encephalopathy: A case report显示文摘BACKGROUND Intrahepatic portosystemic venous shunt(IPSVS)is a rare hepatic disease with different clinical manifestations.Most IPSVS patients with mild shunts are asymptomatic,while the patients with severe shunts present complications such as hepatic encephalopathy.For patients with portal hypertension accompanied by intrahepatic shunt,portal hypertension may lead to hemodynamic changes that may result in exacerbated portal shunt and increased shunt flow.CASE SUMMARY A 57-year-old man,with the medical history of chronic hepatitis B and liver cirrhosis,was admitted to our hospital with abnormal behavior for 10 mo.He had received the esophageal varices ligation and entecavir therapy 1 year ago.Comparing with former examination results,the degree of esophageal varices was significantly reduced,while the right branch of the portal vein was significantly expanded and tortuous.Meanwhile,abdominal ultrasound presented the right posterior branch of portal vein connected with the retrohepatic inferior vena cava.The imaging findings indicated the diagnosis of IPSVS and hepatic encephalopathy.Instead of radiologic interventions or surgical therapies,this patient had only accepted symptomatic treatment.No recurrence of hepatic encephalopathy was observed during 1-year follow-up.CONCLUSION Hemodynamic changes may exacerbate intrahepatic portosystemic shunt.The intervention or surgery should be carefully applied to patients with severe portal hypertension due to the risk of hemorrhage.Ying-Hao Chang Xiao-Lei Zhou Dan Jing Zhen Ni Shan-Hong Tang 2021World Journal of Clinical Cases2021,9,2:3
