| 1 | Antioxidant properties of magnesium lithospermate B contribute to the cardioprotection against myocardial ischemia/reperfusion injury in vivo and in vitro显示文摘OBJECTIVE: To determine the cardioprotective effect of magnesium lithospermate B (MLB) on myocardial ischemia/reperfusion (MI/R) injury and to investigate the antioxidant potential in vivo and in vitro. METHODS: MI/R injury was induced by the occlusion of left anterior descending coronary artery for 30 min followed by reperfusion for 3 h in rats. After reperfusion, hearts were harvested to assess infarct size, histopathological damages, the levels of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), reduced glutathione (GSH) and malondialdehyde (MDA). Blood samples were col- lected to determine serum levels of creatine kinase-MB (CK-MB), cardiac troponin (cTnI) and lactate dehydrogenase (LDH). Furthermore, simulatedischemia/reperfusion (SI/R) injury in vitro was established by oxygen and glucose deprivation (OGD) for 2 h followed by 24-hour recovery period in cardiomyocytes. The activity of LDH in the cultured supernatant and the levels of intracellular reactive oxygen species (ROS), SOD and MDA in cardiomyocytes were also measured. Finally, cardiomyocytes apoptosis was determined with flow cytometry. RESULTS: MLB significantly limited infarct size, ameliorated histopathological damages and prevented leakage of CK-MB, cTnI and LDH. Additionally, SOD, CAT, GPx and GSH activities were notably increased by MLB, along with the MDA content decreased as compared with the model group in rats. In vitro study, MLB also decreased LDH activity in the cultured supernatant, increased SOD activity in cardiomyocytes, reduced intracellular ROS and MDA levels, and significantly suppressed cardiomyocytes apoptosis. CONCLUSION: MLB possessed remarkably cardioprotective effects on MI/R injury in vivo and in vitro. | 2 | Late Mesozoic Ore-forming Events in the Ningwu Ore District, Middle-Lower Yangtze River Polymetallic Ore Belt, East China: Evidence from Zircon U-Pb Geochronology and Hf Isotopic Compositions of the Granodioritic Stocks显示文摘Late Mesozoic volcanic-subvolcanic rocks and related iron deposits, known as porphyry iron deposits in China, are widespread in the Ningwu ore district (Cretaceous basin) of the middle-lower Yangtze River polymetallic ore belt, East China. Two types of Late Mesozoic magmatic rocks are exposed: one is dioritic rocks closely related to iron mineralization as the hosted rock, and the other one is granodioritic (-granitic) rocks that cut the ore bodies. To understand the age of the iron mineralization and the ore-forming event, detailed zircon U-Pb dating and Hf isotope measurement were performed on granodioritic stocks in the Washan, Gaocun-Nanshan, Dongshan and Heshangqiao iron deposits in the basin. Four emplacement and crystallization (typically for zircons) ages of granodioritic rocks were measured as 126.1±0.5 Ma, 126.8±0.5 Ma, 127.3±0.5 Ma and 126.3±0.4 Ma, respectively in these four deposits, with the LA-MC-ICP-MS zircon U-Pb method. Based on the above results combined with previous dating, it is inferred that the iron deposits in the Ningwu Cretaceous basin occurred in a very short period of 131-127 Ma. In situ zircon Hf compositions of εHf(t) of the granodiorite are mainly from -3 to -8 and their corresponding 176Hf/177Hf ratio are from 0.28245 to 0.28265, indicating similar characteristics of dioritic rocks in the basin. We infer that granodioritic rocks occurring in the Ningwu ore district have an original relationship with dioritic rocks. | 3 | Effects of Shuanghuanglian oral liquids on patients with COVID-19:a randomized,open-label,parallel-controlled,multicenter clinical trial显示文摘We conducted a randomized,open-label,parallel-controlled,multicenter trial on the use of Shuanghuanglian(SHL),a traditional Chinese patent medicine,in treating cases of COVID-19.A total of 176 patients received SHL by three doses(56 in low dose,61 in middle dose,and 59 in high dose)in addition to standard care.The control group was composed of 59 patients who received standard therapy alone.Treatment with SHL was not associated with a difference from standard care in the time to disease recovery.Patients with 14-day SHL treatment had significantly higher rate in negative conversion of SARS-CoV-2 in nucleic acid swab tests than the patients from the control group(93.4%vs.73.9%,P=0.006).Analysis of chest computed tomography images showed that treatment with high-dose SHL significantly promoted absorption of inflammatory focus of pneumonia,which was evaluated by density reduction of inflammatory focus from baseline,at day 7(mean difference(95%CI),−46.39(−86.83 to−5.94)HU;P=0.025)and day 14(mean difference(95%CI),−74.21(−133.35 to−15.08)HU;P=0.014).No serious adverse events occurred in the SHL groups.This study illustrated that SHL in combination with standard care was safe and partially effective for the treatment of COVID-19. | Li Ni Zheng Wen Xiaowen Hu Wei Tang Haisheng Wang Ling Zhou Lujin Wu Hong Wang Chang Xu Xizhen Xu Zhichao Xiao Zongzhe Li Chene Li Yujian Liu Jialin Duan Chen Chen Dan Li Runhua Zhang Jinliang Li Yongxiang Yi Wei Huang Yanyan Chen Jianping Zhao Jianping Zuo Jianping Weng Hualiang Jiang Dao Wen Wang | 2021 | Frontiers of Medicine2021,15,5: | 7 |
| 4 | Proteolytic processing of SDF-1α by matrix metalloproteinase-2 impairs CXCR4 signaling and reduces neural progenitor cell migration显示文摘Neural stem cells and neural progenitor cells(NPCs)exist throughout life and are mobilized to replace neurons,astrocytes and oligodendrocytes after injury.Str-omal cell-derived factor 1(SDF-1,now named CXCL12)and its receptor CXCR4,an α-chemokine receptor,are critical for NPC migration into damaged areas of the brain.Our previous studies demonstrated that immune activated and/or HIV-1-infected human monocyte-derived-macrophages(MDMs)induced a substantial increase of SDF-1 production by human astrocytes.However,matrix metalloproteinase(MMP)-2,a protein up-regulated in HIV-1-infected macrophages,is able to cleave four amino acids from the N-terminus of SDF-1,resulting in a truncated SDF-1(5-67).In this study,we investigate the diverse signaling and function induced by SDF-1α and SDF-1(5-67)in human cortical NPCs.SDF-1(5-67)was generated by incubating human recombinant SDF-1α with MMP-2 followed by protein determination via mass spectrometry,Western blotting and ELISA.SDF-1α induced time-dependent phosphorylation of extracellular signal-regulated kinases(ERK)1/2,Akt-1,and diminished cyclic adenosine monophosphate(cAMP).In contrast,SDF-1(5-67)failed to induce these signaling.SDF-1α activation of CXCR4 induced migration of NPCs,an effect that is dependent on ERK1/2 and Akt-1 pathways;whereas SDF-1(5-67)failed to induce NPC migration.This observation provides evidence that MMP-2 may affect NPC migration through post-translational processing of SDF-1α. | Hui Peng Yumei Wu Zhiyuan Duan Pawel Ciborowski Jialin C.Zheng | 2012 | Protein & Cell2012,3,11: | 3 |

| 7 | Characterization of 3D DNA Assemblies Using Cryogenic Electron Microscopy显示文摘DNA nanotechnology utilizes DNA double strands as building units for self-assembly of DNA nanostructures.The specific base-pairing interaction between DNA molecules is the basis of these assemblies.After decades of development,this technology has been able to construct complex and programmable structures.With the increase in delicate nature and complexity of the synthesized nanostructures,a characterization technology that can observe these structures in three dimensions has become necessary,and developing such a technology is considerably challenging.DNA assemblies have been studied using different characterization methods including atomic force microscopy(AFM),scanning electron microscopy(SEM),and transmission electron microscopy(TEM).However,the three-dimensional(3D)DNA assemblies always collapse locally due to the dehydration during the drying process.Cryogenic electron microscopy(cryo-EM)can overcome the challenge by maintaining three-dimensional morphologies of the cryogenic samples and reconstruct the 3D models from cryogenic samples accordingly by collecting thousands of two-dimensional(2D)projection images,which can restore their original morphologies in solution.Here,we have reviewed several typical cases of 3D DNA-assemblies and highlighted the applications of cryo-EM in characterization of these assemblies.By comparing with some other characterization methods,we have shown how cryo-EM promoted the development of structural characterization in the field of DNA nanotechnology. | WANG Mingyang DUAN Jialin DAI Lizhi XIN Xiaodong WANG Fangfang LI Zheng TIAN Ye | 2020 | Chemical Research in Chinese Universities2020,36,2: | 0 |

| 9 | Role of Intracellular Distribution of Feline and Bovine SAMHD1 Proteins in Lentiviral Restriction显示文摘Human SAMHD1(h SAM)restricts lentiviruses at the reverse transcription step through its d NTP triphosphohydrolase(d NTPase)activity.Besides humans,several mammalian species such as cats and cows that carry their own lentiviruses also express SAMHD1.However,the intracellular distribution of feline and bovine SAMHD1(f SAM and b SAM)and its significance in their lentiviral restriction function is not known.Here,we demonstrated that f SAM and b SAM were both predominantly localized to the nucleus and nuclear localization signal(11KRPR14)-deleted f SAM and b SAM relocalized to the cytoplasm.Both cytoplasmic f SAM and b SAM retained the antiviral function against different lentiviruses and cytoplasmic f SAM could restrict Vpx-encoding SIV and HIV-2 more efficiently than its wild-type(WT)protein as cytoplasmic h SAM.Further investigation revealed that cytoplasmic f SAM was resistant to Vpx-induced degradation like cytoplasmic h SAM,while cytoplasmic b SAM was not,but they all demonstrated the same in vitro d NTPase activity and all could interact with Vpx as their WT proteins,indicating that cytoplasmic h SAM and f SAM can suppress more SIV and HIV-2 by being less sensitive to Vpx-mediated degradation.Our results suggested that f SAM-and b SAM-mediated lentiviral restriction does not require their nuclear localization and that f SAM shares more common features with h SAM.These findings may provide insights for the establishment of alternative animal models to study SAMHD1 in vivo. | Chu Wang Lina Meng Jialin Wang Kaikai Zhang Sizhu Duan Pengyu Ren Yingzhe Wei Xinyu Fu Bin Yu Jiaxin Wu Xianghui Yu | 2021 | Virologica Sinica2021,36,5: | 0 |
