| 1 | Arabidopsis ANAC092 regulates auxin-mediated root development by binding to the ARF8 and PIN4 promoters显示文摘Auxin is an important plant hormone that is essential for growth and development due to its effects on organogenesis, morphogenesis, tropisms, and apical dominance. The functional diversity of auxin highlights the importance of its biosynthesis, transport, and associated responses. In this study, we show that a NAC transcription factor, ANAC092(also named At NAC2 and ORESARA1),known to positively regulate leaf senescence and contribute to abiotic stress responses, also affects primary root development. Plants overexpressing ANAC092 had altered root meristem lengths and shorter primary roots compared with the wild-type control. Additionally, expression of the pro ANAC092::GUS was strongly induced by indole-3-acetic acid. Quantitative real-time RT-PCR(q RT-PCR) analysis revealed that the YUCCA2, PIN, and ARF expression levels were downregulated in ANAC092-overexpressing plants. Moreover, yeast one-hybrid and chromatin immunoprecipitation assays confirmed that ANAC092 binds to the promoters of AUXIN RESPONSE FACTOR 8(ARF8) and PIN-FORMED 4(PIN4). Furthermore, a dual-luciferase assay indicated that ANAC092 decreases ARF8 and PIN4 promoter activities. We also applied a CRISPR/Cas9 system to mutate ANAC092. The roots of three of the analyzed mutants were longer than normal. | 2 | Elevating H3K27me3 level sensitizes colorectal cancer to oxaliplatin显示文摘Histone methylation is a context-dependent modification that regulates gene expression,and the trimethylation of histone H3 lysine 27(H3K27me3)usually induces gene silencing.Overcoming colorectal cancer(CRC)chemoresistance is currently a huge challenge,but the relationship between H3K27me3 modification and chemoresistance remains largely unclear.Here,we found that H3K27me3 levels positively correlated with the metastasis-free survival of CRC patients and a low H3K27me3 level predicted a poor outcome upon chemotherapeutic drug treatment.Oxaliplatin stimulation significantly induced the expression of H3K27 lysine demethylase 6A/6B(KDM6A/6B),thus decreasing the level of H3K27me3 in CRC cells.Elevation of H3K27me3 level through KDM6A/6B depletion or GSK-J4(a KDM6A/6B inhibitor)treatment significantly enhanced oxaliplatin-induced apoptosis.Conversely,when inhibiting the expression of H3K27me3 by EPZ-6438,an inhibitor of the histone methyltransferase EZH2,the proportion of apoptotic cells remarkably decreased.In addition,the combination of GSK-J4 and oxaliplatin significantly inhibited tumor growth in an oxaliplatin-resistant patient-derived xenograft model.Importantly,we revealed that oxaliplatin treatment dramatically induced NOTCH2 expression,which was caused by downregulation of H3K27me3 level on the NOTCH2 transcription initiation site.Thus,the activated NOTCH signaling promoted the expression of stemness-related genes,which resulted in oxaliplatin resistance.Furthermore,oxaliplatin-induced NOTCH signaling could be interrupted by GSK-J4 treatment.Collectively,our findings suggest that elevating H3K27me3 level can improve drug sensitivity in CRC patients. | Qi Wang Xi Chen Yuhang Jiang Sanhong Liu Hanshao Liu Xiaohua Sun Haohao Zhang Zhi Liu Yu Tao Cuifeng Li Yiming Hu Dandan Liu Deji Ye Yongzhong Liu Mingliang Wang Xiaoren Zhang | 2020 | Journal of Molecular Cell Biology2020,12,2: | 6 |

| 4 | Pyrroloquinoline quinone regulates the redox status in vitro and in vivo of weaned pigs via the Nrf2/HO-1 pathway显示文摘Background:Oxidative stress is a main cause of piglet gut damage and diarrhea.Pyrroloquinoline quinone(PQQ),is a novel redox cofactor with antioxidant properties.However,the effect and mechanism that PQQ supplementation decreases oxidative injury in weaned pigs is not understood.Therefore,the aim of this study is to confirm the effect of PQQ on regulating redox status in weaned pigs and the mechanism for antioxidant function by porcine intestinal epithelial cell line(IPEC-J2)challenged with H_(2)O_(2).Results:Experiment 1,144 Duroc×Landrace×Yorkshire pigs(weaned at 28 d)were allocated to four groups:received a basal diet(control)and diets supplemented with 0.15%,0.30%and 0.45%PQQ,respectively.On d 28,growth performance,diarrhea incidence and redox factors were measured.Experiment 2,IPEC-J2 were treated with or without PQQ in the presence or absence of H_(2)O_(2)for indicated time points.Experiment 3,IPEC-J2 were transfected with or without Nrf2 siRNA,then treated according to Experiment 2.The cell viability,redox factors,protein of tight junctions and Nrf2 pathway were determined.In vivo,PQQ supplementation demonstrated dose-related improvements in average daily gain,and gain to feed ratio(Linear P<0.05).During d 0–28,compared to controls,0.45%PQQ supplementation for pigs decreased diarrhea incidence and MDA content in liver and jejunum,and increased concentration of SOD in liver;0.3%PQQ supplementation decreased ileal and liver MDA concentration;and 0.15%PQQ supplementation decreased ileal MDA concentration(P<0.05).In vitro,compared to cells cultured with H_(2)O_(2),pre-treatment with PQQ increased cell viability,tight junction proteins expression including ZO-1,ZO-2,Occludin and Claudin-1;and decreased ROS concentration and level of Caspase-3(P<0.05);as well as upregulated the ratio of Bcl-2 to Bax and protein expression of nuclear Nrf2,HO-1.Notably,Nrf2 knockdown by transfection with Nrf2 siRNA largely abrogated the positive effects of PQQ pretreatment on H_(2)O_(2)-induced intracellular changes.Conclusions:PQQ administration attenuated oxidative stress in weaned pigs which is associated with activation of Nrf2/HO-1 pathway. | Caiyun Huang Zijuan Fan Dandan Han Lee J.Johnston Xi Ma Fenglai Wang | 2021 | Journal of Animal Science and Biotechnology2021,12,4: | 4 |

| 7 | Scientific objectives of the Hot Universe Baryon Surveyor(HUBS) mission显示文摘The Hot Universe Baryon Surveyor(HUBS) is a proposed space-based X-ray telescope for detecting X-ray emissions from the hot gas content in our universe. With its unprecedented spatially-resolved high-resolution spectroscopy and large field of view,the HUBS mission will be uniquely qualified to measure the physical and chemical properties of the hot gas in the interstellar medium, the circumgalactic medium, the intergalactic medium, and the intracluster medium. These measurements will be valuable for two key scientific goals of HUBS, namely to unravel the AGN and stellar feedback physics that governs the formation and evolution of galaxies, and to probe the baryon budget and multi-phase states from galactic to cosmological scales. In addition to these two goals, the HUBS mission will also help us solve some problems in the fields of galaxy clusters, AGNs, difuse X-ray backgrounds, supernova remnants, and compact objects. | 8 | Magnetic Nanomotor-Based Maneuverable SERS Probe显示文摘Surface-enhanced Raman spectroscopy(SERS)is a powerful sensing technique capable of capturing ultrasensitive fingerprint signal of analytes with extremely low concentration.However,conventional SERS probes are passive nanoparticles which are usually massively applied for biochemical sensing,lacking controllability and adaptability for precise and targeted sensing at a small scale.Herein,we report a“rod-like”magnetic nanomotor-based SERS probe(MNM-SP)that integrates a mobile and controllable platform of micro-/nanomotors with a SERS sensing technique.The“rod-like”structure is prepared by coating a thin layer of silica onto the self-assembled magnetic nanoparticles.Afterwards,SERS hotspots of silver nanoparticles(AgNPs)are decorated as detecting nanoprobes.The MNM-SPs can be navigated on-demand to avoid obstacles and target sensing sites by the guidance of an external gradient magnetic field.Through applying a rotating magnetic field,the MNM-SPs can actively rotate to efficiently stir and mix surrounding fluid and thus contact with analytes quickly for SERS sensing.Innovatively,we demonstrate the self-cleaning capability of the MNM-SPs which can be used to overcome the contamination problem of traditional single-use SERS probes.Furthermore,the MNM-SPs could precisely approach the targeted single cell and then enter into the cell by endocytosis.It is worth mentioning that by the effective mixing of intracellular biocomponents,much more informative Raman signals with improved signal-to-noise ratio can be captured after active rotation.Therefore,the demonstrated magnetically activated MNM-SPs that are endowed with SERS sensing capability pave way to the future development of smart sensing probes with maneuverability for biochemical analysis at the micro-/nanoscale. | Yong Wang Yuhuan Liu Yang Li Dandan Xu Xi Pan Yuduo Chen Dekai Zhou Bo Wang Huanhuan Feng Xing Ma | 2020 | Research2020,,1: | 1 |

| 10 | Prediction and observation of defect-induced room-temperature ferromagnetism in halide perovskites显示文摘The possibility to induce a macroscopic magnetic moment in lead halide perovskites(LHPs),combined with their excellent optoelectronic properties,is of fundamental interest and has promising spintronic applications.However,these possibilities remain an open question in both theory and experiment.Here,theoretical and experimental studies are performed to explore ferromagnetic states in LHPs originated from lattice defects.First-principle calculations reveal that shallow-level Br vacancies in defective CsPbBr3 can produce spin-splitting states and the coupling between them leads to a ferromagnetic ground state.Experimentally,ferromagnetism at 300 K is observed in room-temperature synthesized CsPbBr3 nanocrystals,but is not observed in hot-injection prepared CsPbBr3 quantum dots and in CsPbBr3 single crystals,highlighting the significance played by vacancy defects.Furthermore,the ferromagnetism in the CsPbBr3 nanocrystals can be enhanced fourfold with Ni2+ion dopants,due to enhancement of the exchange coupling between magnetic polarons.Room-temperature ferromagnetism is also observed in other LHPs,which suggests that vacancy-induced ferromagnetism may be a universal feature of solution-processed LHPs,which is useful for future spintronic devices. | Zhiguo Sun Bo Cai Xi Chen Wenxian Wei Xiaoming Li Dandan Yang Cuifang Meng Ye Wu Haibo Zeng | 2020 | Journal of Semiconductors2020,41,12: | 0 |
| 11 | Bcl-3 promotes Wnt signaling by maintaining the acetylation ofβ-catenin at lysine 49 in colorectal cancer显示文摘Wnt/β-catenin signaling plays a critical role in colorectal cancer(CRC)tumorigenesis and the homeostasis of colorectal cancer stem cells(CSCs),but its molecular mechanism remains unclear.B-cell lymphoma 3(Bcl-3),a member of the IκB family,is overexpressed in CRC and promotes tumorigenicity.Here,we report a novel function of Bcl-3 in maintaining colorectal CSC homeostasis by activating Wnt/β-catenin signaling.Silencing Bcl-3 suppresses the self-renewal capacity of colorectal CSCs and sensitizes CRC cells to chemotherapeutic drugs through a decrease in Wnt/β-catenin signaling.Moreover,our data show that Bcl-3 is a crucial component of Wnt/β-catenin signaling and is essential forβ-catenin transcriptional activity in CRC cells.Interestingly,Wnt3a increases the level and nuclear translocation of Bcl-3,which binds directly toβ-catenin and enhances the acetylation ofβ-catenin at lysine 49(Ac-K49-β-catenin)and transcriptional activity.Bcl-3 depletion decreases the Ac-K49-β-catenin level by increasing the level of histone deacetylase 1 to remove acetyl groups fromβ-catenin,thus interrupting Wnt/β-catenin activity.In CRC clinical specimens,Bcl-3 expression negatively correlates with the overall survival of CRC patients.A significantly positive correlation was found between the expression of Bcl-3 and Ac-K49-β-catenin.Collectively,our data reveal that Bcl-3 plays a crucial role in CRC chemoresistance and colorectal CSC maintenance via its modulation of the Ac-K49-β-catenin,which serves as a promising therapeutic target for CRC. | Xi Chen Chen Wang Yuhang Jiang Qi Wang Yu Tao Haohao Zhang Yongxu Zhao Yiming Hu Cuifeng Li Deji Ye Dandan Liu Wenxia Jiang Eugene Y.Chin Sheng Chen Yongzhong Liu Mingliang Wang Sanhong Liu Xiaoren Zhang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 0 |
