| 1 | Epigenetic modification in histone deacetylase deletion strain of Calcarisporium arbuscula leads to diverse diterpenoids显示文摘Epigenetic modifications have been proved to be a powerful way to activate silent gene clusters and lead to diverse secondary metabolites in fungi. Previously, inactivation of a histone H3 deacetylase in Calcarisporium arbuscula had led to pleiotropic activation and overexpression of more than 75% of the biosynthetic genes and isolation of ten compounds. Further investigation of the crude extract of C. arbuscula Δhda A strain resulted in the isolation of twelve new diterpenoids including three cassanes(1-3), one cleistanthane(4), six pimaranes(5-10), and two isopimaranes(11 and 12) along with two know cleistanthane analogues. Their structures were elucidated by extensive NMR spectroscopic data analysis. In this paper, we consider objective Bayesian analysis for the ICCin the context of normal linear regression model. We first derive two objective priors for theunknown parameters and show that both result in proper posterior distributions. Within aBayesian decision-theoretic framework, we then propose an objective Bayesian solution to theproblems of hypothesis testing and point estimation of the ICC based on a combined use of theintrinsic discrepancy loss function and objective priors. The proposed solution has an appealinginvariance property under one-to-one reparametrisation of the quantity of interest. | 17 | Application of a jigsaw puzzle flap based on free-style perforator to repair large scalp defects after tumor resection:A case series显示文摘Background:Reconstruction of large scalp defects after tumor resection is common and challenging.Free skin grafting or free flap transfer is always performed.However,these techniques can result in poor functional and cosmetic outcomes.Methods:In the Second Affiliated Hospital of Soochow University and the Second Affiliated Hospital of Chengdu Medical College,35 patients underwent jigsaw puzzle flap procedures based on a free-style perforator to repair large scalp defects after tumor resection from May 2013 to November 2022.The key to this procedure was to divide a large defect into several smaller parts that correspond to adjacent free-style perforators around the scalp defect.The free-style perforator flaps were designed in different ways,such as propeller flaps,rotation flaps,and V-Y advancement flaps,which were pedicled with free-style perforators.All of the small flaps were then mobilized to the defect and sutured to each other to create a new,large jigsaw puzzle-like flap to cover the large scalp defect.Finally,all the donor sites were closed directly.Results:Overall,35 patients were treated.The average defect size was 72 cm^(2)(range,25-91 cm^(2)).All flaps had a satisfactory appearance after follow-up periods of 6 months-10 years.Local tumor recurrence occurred in one case.Conclusion:The application of a jigsaw puzzle flap based on a free-style perforator is a suitable option for the reconstruction of large scalp defects after resection,yielding satisfactory functional and cosmetic results. | Daojiang Yu Jing Wang Lei Chen Lu An Yahui Feng Sheng Jiang Shuyu Zhang Xiaoming Chen Guozhong Lv | 2023 | Chinese Journal Of Plastic and Reconstructive Surgery2023，5，3： | 0 |
| 18 | RXRα agonist bexarotene attenuates radiation-induced skin injury by relieving oxidative stress显示文摘Objective: To investigate the protective effect of bexarotene in radiation-induced skin injury and elucidate underlying mechanism.Methods: Irradiated cellular and animal models were established using an X-ray linear accelerator. Cell viabilityand apoptosis were evaluated by CCK8 and flow cytometry. In vivo protective effect of bexarotene was measuredin irradiated SD rats. The antioxidant capacity of bexarotene was validated by DCF-DA method. The signalingpathways involved in bexarotene-mediated skin repair were enriched by RNA sequencing.Results: Bexarotene could significantly restore the proliferation and inhibit the apoptosis of WS1 cells with radiation damage (P < 0.05). Moreover, bexarotene could effectively shorten the process of skin damage andpromote skin repair in a rat model of radiation-induced skin injury (P < 0.05). Mechanistically, bexaroteneeffectively reduced the expression of RXRα (P < 0.05), thus leading to an early decrease in reactive oxygen species(ROS) after radiation exposure.
