| 1 | Genome survey uncovers the secrets of sex and lifestyle in caterpillar fungus显示文摘The caterpillar fungus Ophiocordyceps sinensis (best known as Cordyceps sinensis) mummifies ghost moth larvae exclusively in Tibetan Plateau alpine ecosystems. Touted as 'Himalayan Viagra', the fungus is highly prized due to its medical benefits and dwindling supplies. Attempts to culture the sexual fruiting-body have failed and the huge market demand has led to severe devastation of local ecosystems and to the fungus heading towards extinction. By genome sequencing, we establish that unlike related insect pathogens O. sinensis contains two compatible mating-type genes in its genome and is self-fertile, i.e. homothallic. However, sexual processes are only initiated under native environmental conditions. O. sinensis resembles biotrophic plant pathogens in having a genome shaped by retrotransposon-driven expansions. The resulting changes in gene content suggest that O. sinensis has a biphasic pathogenic mechanism beginning with stealth pathogenesis in early host instars. O. sinensis is the first psychrophilic fungus sequenced and is adapted to extreme cold with putative antifreeze proteins and mechanisms for increasing lipid accumulation and fatty acid unsaturation. | 2 | Fiducial inference in the pivotal family of distributions显示文摘In this paper a family, called the pivotal family, of distributions is considered.A pivotal family is determined by a generalized pivotal model. Analytical results show that a great many parametric families of distributions are pivotal. In a pivotal family of distributions a general method of deriving fiducial distributions of parameters is proposed. In the method a fiducial model plays an important role. A fiducial model is a function of a random variable with a known distribution, called the pivotal random element, when the observation of a statistic is given.The method of this paper includes some other methods of deriving fiducial distributions. Specially the first fiducial distribution given by Fisher can be derived by the method. For the monotone likelihood ratio family of distributions, which is a pivotal family, the fiducial distributions have a frequentist property in the Neyman-Pearson view. Fiducial distributions of regular parametric functions also have the above frequentist property. Some advantages of the fiducial inference are exhibited in four applications of the fiducial distribution. | 5 | Purification and characterization of a novel immunomodulatory protein from the medicinal mushroom Trametes versicolor显示文摘Bioactive proteins represent an important group of functional agents in medicinal mushrooms.Trametes versicolor(L.) Lloyd is a mushroom frequently used in traditional Chinese medicine for its anti-tumor and immunomodulatory activities.A new immunomodulatory protein from T.versicolor,named TVC,was purified by ammonium sulfate precipitation,ion-exchange chromatography and gel filtration chromatography.Sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis of the purified protein revealed a single band with a molecular weight of 15.0 kD.Native polyacrylamide gel analysis revealed a band at 30 kD,indicating that TVC exists in solution as a homodimer.Isoelectric focusing showed that TVC was an acidic protein with an isoelectric point of 4.0.TVC was found to lack carbohydrate modifications(based on periodic acid/Schiff staining) and it does not agglutinate mouse red blood cells,suggesting that TVC is not a lectin-like protein.Biological activity assays dem-onstrated that TVC can enhance the proliferation of splenocytes,while it has no stimulatory effects on CD4+and CD8+T cells. | 7 | Yangtze Three Gorges Reservoir, China: A holistic assessment of organic pollution, mutagenic effects of sediments and genotoxic impacts on fish显示文摘Besides obvious benefits,the Three Gorges Dam's construction resulted in new pollution scenarios with the potentials to threaten the Three Gorges Reservoir(TGR) ecosystem.In order to record organic contamination,to find links to ecotoxicological impacts and to serve as reference for ensuing monitoring,several sites in the TGR area were screened applying the triad approach with additional lines-of-evidence as a holistic assessment method.Sediments and the benthic fish species Pelteobagrus vachellii were sampled in 2011 and 2012 to determine organic pollution levels,mutagenic potentials and genotoxic impacts.Two regional hot-spots near the cities of Chongqing and Kaixian were identified and further investigated in 2013.Only polycyclic aromatic hydrocarbons(PAHs) could be detected in sediments in 2011(165–1653 ng/g),emphasizing their roles as key pollutants of the area.Their ubiquity was confirmed at Chongqing(150–433 ng/g) and Kaixian(127–590 ng/g) in2013.Concentrations were comparable to other major Chinese and German rivers.However,the immense sediment influx suggested a deposition of 216–636 kg PAH/day(0.2–0.6 mg PAH/(m2·day)),indicating an ecotoxicological risk.PAH source analysis highlighted primary impacts of combustion sources on the more industrialized upper TGR section,whereas petrogenic sources dominated the mid-low section.Furthermore,sediment extracts from several sites exhibited significant activities of frameshift promutagens in the Ames fluctuation assay.Additionally,significant genotoxic impairments in erythrocytes of P.vachellii were detected(Chongqing/Kaixian),demonstrating the relevance of genotoxicity as animportant mode of action in the TGR's fish.PAHs,their derivatives and non-target compounds are considered as main causative agents. | Tilman Floehr Bj?rn Scholz-Starke Hongxia Xiao Josef Koch Lingling Wu Junli Hou Anja Wolf Axel Bergmann Kerstin Bluhm Xingzhong Yuan Martina Ro?-Nickoll Andreas Sch?ffer Henner Hollert | 2015 | Journal of Environmental Sciences2015,27,12: | 9 |

Because of its high efficiency,it has brought new opportunities for the development of molecular biotechnology. Reversed micelles represent nano-sized aqueous droplets stabilized by surfactant amphiphiles inside the bulk organic solvents. The entrapped enzymes have enhanced activities under those conditions as suited in the lipid bilayers of biological membranes. The fundamentals of enzyme-containing reversed micellar system are described in this paper,with special emphasis on the effects of surfactants varying in concentrations and structures. The latest study progress on the surfactants application in enzyme-containing reversed micelles is reviewed. | 9 | The point defect and electronic structure of K doped LaCo_(0.9)Fe_(0.1)O_(3)perovskite with enhanced microwave absorbing ability显示文摘ABO_(3)perovskites,owning unique properties,have great research prospect in electromagnetic wave absorption field.Normally,doping can significantly regulate the dielectric loss,whereas the magnetic loss can be ignored.In this work,the crystal structure and electromagnetic properties can be regulated systematically by the K,Fe co-doping for LaCoO_(3)perovskites(LKCFO)under the condition of fixed F content.In addition,the obtained samples show the obvious interfacial polarization effect on accounting to the small size effect,which is conducive to the effective microwave absorption.By analyzing the evolution of the positron annihilation lifetime and the first-principles calculation of the oxygen density of states for the series of LKCFO perovskites,it is found that the charge transport characteristics will be controlled by the point defect generated by allelic doping.The point defect content decreases and then increases as the doping level rises.The prepared perovskite exhibits the lowest defect density and the largest dielectric loss capability,which indicates that the lower point defects promote electron migration and thus enhance the dielectric loss;thus,the electromagnetic wave absorption bandwidth up to 6.2 GHz is reached.In contrast,both insufficient and excessive K doping are detrimental to the enhancement of microwave absorption.Especially,the practical application value was investigated using Computer Simulation Technology(CST)simulations.The LKCFO-2 exhibits the smallest RCS value(below-10 dBm^(2))at almost-90°-90°with a thickness of 2 mm,providing an effective method for study excellent microwave absorption and scattering property. | Fan Wang Weihua Gu Jiabin Chen Yue Wu Ming Zhou Shaolong Tang Xingzhong Cao Peng Zhang Guangbin Ji | 2022 | Nano Research2022,15,4: | 7 |
| 10 | What are the common anthracnose pathogens of tropical fruits?显示文摘t Species of Colletotrichum are associated with anthracnose of a wide range of host plants including cultivated and wild tropical fruits.The genetic and ecological diversity of species associated with wild fruits are poorly explored,as compared to those associated with pre and postharvest diseases of cultivated fruits.In the present study,isolates of Colletotrichum were obtained from commercially available cultivated fruits,wild fruits(from native trees in natural habitats)and a few herbaceous hosts collected in northern Thailand.These isolates were initially characterized based on analysis of complete sequences of nuclear ribosomal internal transcribed spacer(ITS),into the genetically defined species complexes of Colletotrichum gloeosporioides,C.acutatum,C.boninense and C.truncatum.The isolates were primarily identified in the C.gloeosporioides species complex,based on a strongly supported clade within the ITS gene tree and were further characterized using multi-gene phylogenetic analyses and morphology.Phylogenetic analyses of ITS,partial sequences of actin(ACT),calmodulin(CAL),glyceraldehyde-3-phosphate dehydrogenase(GAPDH),glutamine synthetase(GS)andβtubulin(TUB2)genetic markers were performed individually and in combination.Colletotrichum gloeosporioides sensu stricto was identified from lime(Citrus aurantifolia)and rose apple(Syzygium samarangense).Colletotrichum fructicola was isolated from dragon fruit(Hylocerous undatus)and jujube(Ziziphus sp.).Colletotrichum endophytica was found only from an unknown wild fruit.We observed a considerable genetic and host diversity of species occurring on tropical fruits within the clade previously known as Colletotrichum siamense sensu lato.The clade consists of isolates identified as pre and postharvest pathogens on a wide range of fruits,including coffee(Coffea arabica),custard apple(Annona reticulata),Cerbera sp.,figs(Ficus racemosa)mango(Mangifera indica),neem(Azadirachta indica)and papaya(Carica papaya)and was the dominant group of species among most wild fruits studied.With the exception of one isolate from banana,which grouped in the C.siamense clade,all the other isolates were identified as Colletotrichum musae.A new species,Colletotrichum syzygicola,associated with Syzygium samarangense in Thailand,is introduced with descriptions and illustrations.This study highlights the need to re-assess the evolutionary relationships of Colletotrichum species occurring on cultivated and wild fruits with emphasis on their ecology and cryptic diversification including sampling at regional and global scales. | Dhanushka Udayanga Dimuthu S.Manamgoda Xingzhong Liu Ekachai Chukeatirote Kevin D.Hyde | 2013 | Fungal Diversity2013,,4: | 6 |

| 13 | Effect of a modified Banxia Xiexin decoction plus chemotherapy on stage Ⅲ colon cancer显示文摘OBJECTIVE: To investigate the effect of a modified Banxia Xiexin decoction(MBXD) plus chemotherapy on postoperative adverse reaction in patients with stage Ⅲ colon cancer in patients whose symptoms were identified as cold-heat complicated pattern in terms of the theory of Traditional Chinese Medicine(TCM).METHODS: A prospective non-randomized control study of patients with stage Ⅲ colon cancer in Beijing Shijitan Hospital and Guang'anmen Hospital between January 2012 and December 2013. A total of 80 patients were divided into experimental group(MBXD + chemotherapy) and control group(chemotherapy). The adverse reactions, life quality and disease-free survival(DFS) were compared between the two groups.RESULTS: The cold-heat complex pattern was released in 94.3% of patients in experimental group whereas 62.2% in control group(P < 0.05). Life quality was improved in 24 vs 14 cases, stabilized in 9 vs14 cases whereas decreased in 2 vs 15 cases in experimental group and control group, respectively(P < 0.05). | 14 | Construction of human liver cancer vascular endothelium cDNA expression library and screening of the endothelium-associated antigen genes显示文摘AIM: To gain tumor endothelium associated antigen genes from human liver cancer vascular endothelial cells (HLCVECs)cDNA expression library, so as to find some new possible targets for the diagnosis and therapy of liver tumor.METHODS: HLCVECs were isolated and purified from a fresh hepatocellular carcinoma tissue sample, and were cultured and proliferated in vitro. A cDNA expression library was constructed with the mRNA extracted from HLCVECs.Anti-sera were prepared from immunized BALB/c mice through subcutaneous injection with high dose of fixed HLCVECs, and were then tested for their specificity against HLCVECs and angiogenic effects in vitro, such as inhibiting proliferation and inducing apoptosis of tumor endothelial cells, using immunocytochemistry, immunofluorescence,cell cycle analysis and M-I-l- assays, etc. The identified xenogeneic sera from immunized mice were employed to screen the library of HLCVECs by modified serological analyses of recombinant cDNA expression libraries (SEREX).The positive clones were sequenced and analyzed by bioinformatics.RESULTS: The primary cDNA library consisted of 2×10^6 recombinants. Thirty-six positive clones were obtained from 6× 10^5 independent dones by immunoscreening. Bio-informatics analysis of cDNA sequences indicated that 36 positive clones represented 18 different genes. Among them, 3 were new genes previously unreported, 2 of which were hypothetical genes. The other 15 were already known ones. Series analysis of gene expression (SAGE) database showed that ERP70,GRP58, GAPDH, SSB, $100A6, BMP-6, DVS27, HSP70 and NAC alpha in these genes were associated with endothelium and angiogenesis, but their effects on HLCVECs were still unclear. GAPDH, $100A6, BMP-6 and hsp70 were identified by SEREX in other tumor cDNA expression libraries.CONCLUSION: By screening of HLCVECs cDNA expression library using sera from immunized mice with HLCVECs,the functional genes associated with tumor endothelium or angiogenesis were identified. The modified SEREX,xenogeneic functional serum screening, was demonstrated to be effective for isolation and identification of antigen genes of tumor endothelium, and also for other tumor cell antigen genes. | 15 | Study on ultrasonic spray technology for the coating of vascular stent显示文摘The ultrasonic spray technology is studied by the method of theoretical derivation,CFD simulation,spray particle diameter detection and analysis,and experimental analysis. And the ultrasonic spray process for the coating of vascular stent is also optimized. Firstly,the ultrasonic atomization physical model is established and the equation of atomization particle diameter is derived. Secondly,the ultrasonic atomization process is simulated by the CFD method,and shows three atomization patterns:incomplete atomization pattern,critical atomization pattern and jet atomization pattern. The critical amplitude and power equation for ultrasonic atomization is derived. Thirdly,experiment is conducted to study the influence of parameters including power,gas pressure,and surface tension. The results show that the spray is stable though few particles are likely to collide each other during spray moving,and the droplet diameter is about 10-m. The Rosin-Rammler distribution equation for ultrasonic spray is created,and the uniform index number is between 7.11 and 11.48. The uniformity of spray particle diameter,the efficiency of adjustment and the energy consumption are better than traditional spray technology. | 19 | Electrochemical Preparation and Photoelectric Properties of Cu_2O-loaded TiO_2 Nanotube Arrays显示文摘TiO2 nanotube(TNT) arrays were fabricated by anodic oxidation of titanium foil in a fluoridebased solution, on which Cu2O particles were loaded via galvanostatic pulse electrodeposition in cupric acetate solutions in the absence of any other additives. The structure and optical properties of Cu2O-loaded TiO2 nanotube arrays(Cu2O-TNTs) were analyzed by scanning electron microscopy(SEM), X-ray diffraction(XRD) and UV-Vis absorption, and the photoelectrochemical performance was measured using an electrochemical work station with a three-electrode configuration. The results show that the Cu2O particles distribute uniformly on the highly ordered anatase TiO2 nanotube arrays. The morphologies of Cu2O crystals change from branched, truncated octahedrons to dispersive single octahedrons with increasing deposition current densities. The Cu2O TNTs exhibited remarkable visible light responses with obvious visible light absorption and greatly enhanced visible light photoelectrochemical performance.
