2Mesenchymal stem cells derived from human placenta suppress allogeneic umbilical cord blood lymphocyte proliferation显示文摘ABSTRACTHuman placenta-derived mononuclear cells (MNC) were isolated by a Percoll density gradient and cultured in mesen-chymal stem cell (MSC) maintenance medium. The homogenous layer of adherent cells exhibited a typical fibroblast-like morphology, a large expansive potential, and cell cycle characteristics including a subset of quiescent cells. In vitrodifferentiation assays showed the tripotential differentiation capacity of these cells toward adipogenic, osteogenic andchondrogenic lineages. Flow cytometry analyses and immunocytochemistry stain showed that placental MSC was ahomogeneous cell population devoid of hematopoietic cells, which uniformly expressed CD29, CD44, CD73, CD105,CD166, laminin, fibronectin and vimentin while being negative for expression of CD31, CD34, CD45 and α-smoothmuscle actin. Most importantly, immuno-phenotypic analyses demonstrated that these cells expressed class I majorhistocompatibility complex (MHC-I), but they did not express MHC-II molecules. Additionally these cells could sup-press umbilical cord blood (UCB) lymphocytes proliferation induced by cellular or nonspecific mitogenic stimuli. Thisstrongly implies that they may have potential application in allograft transplantation. 3Meta-analysis of defibrase in treatment of acute cerebral infarction显示文摘Background Fibrinogen-depleting agents are promising in the treatment of cerebral ischemic disease. They were studied by many trials, and the outcomes were different because of different regimens and different doses. In this study, we assessed the efficacy and safety of defibrase on acute cerebral infarction in China. Methods A search using Chinese hospital knowledge database (CHKD) and MEDLINE database for randomized controlled trials was carried out. A CHKD (1994 June 2005) search was performed with the keyword 'defibrase', then a second search for the keyword 'acute cerebral infarction'; a MEDLINE search (1950 June 2005) was performed with the following [(cerebral ischemia), OR (acute cerebral infarction), OR (stroke)], AND [defibrase]. Meta-analysis was performed with RevMan software 4.2. Results Included were 14 studies comparing the efficiency and safety of defibrase with other drugs in the treatment of acute cerebral infarction. Patients’ records were pooled (total 646 patients; defibrase, n=328, no defibrase n=318). Neurological deficit score (NDS) before treatment showed weighted mean differences (WMD)=0.95, 95% confidence interval (CI)= (-0.60, 2.50), P=0.23; NDS after treatment showed WMD=- 2.20, 95% CI= (-4.21, -0.18), P=0.03; Barthel index at 3 months showed WMD=4.45, 95% CI= (-0.13, 9.03), P=0.06; the plasma fibrinogen level before treatment showed WMD=0.02, 95% CI= (-0.16, 0.19), P=0.86; plasma fibrinogen level after treatment showed WMD=-1.51, 95% CI= (-1.88, -1.15), P<0.00 001. Conclusions With the given dose and regimen of defibrase in China, defibrase may play a role of anticoagulation. 5Activation of gibberellin 2-oxidase 6 decreases active gibberellin levels and creates a dominant semi-dwarf phenotype in rice (Oryza sativa L.)显示文摘Gibberellin (GA) 2-oxidase plays a key role in the GA catabolic pathway through 2β-hydroxylation.In the present study,we isolated a CaMV 35S-enhancer activation tagged mutant,H032.This mutant exhibited a dominant dwarf and GA-deficient phenotype,with a final stature that was less than half of its wild-type counterpart.The endogenous bioactive GAs are markedly decreased in the H032 mutant,and application of bioactive GAs (GA3 or GA4) can reverse the dwarf phenotype.The integrated T-DNA was detected 12.8 kb upstream of the OsGA2ox6 in the H032 genome by TAIL-PCR.An increased level of OsGA2ox6 mRNA was detected at a high level in the H032 mutant,which might be due to the enhancer role of the CaMV 35S promoter.RNAi and ectopic expression analysis of OsGA2ox6 indicated that the dwarf trait and the decreased levels of bioactive GAs in the H032 mutant were a result of the up-regulation of the OsGA2ox6 gene.BLASTP analysis revealed that OsGA2ox6 belongs to the class III of GA 2-oxidases,which is a novel type of GA2ox that uses C20-GAs (GA12 and/or GA53) as the substrates.Interestingly,we found that a GA biosynthesis inhibitor,paclobutrazol,positively regulated the OsGA2ox6 gene.Unlike the over-expression of OsGA2ox1,which led to a high rate of seed abortion,the H032 mutant retained normal flowering and seed production.These results indicate that OsGA2ox6 mainly affects plant stature,and the dominant dwarf trait of the H032 mutant can be used as an efficient dwarf resource in rice breeding.Jian Huang Ding Tang Yi Shen Baoxiang Qin Lilan Hong Aiqing You Ming Li Xin Wang Hengxiu Yu Minghong Gu Zhukuan Cheng 2010Journal of Genetics and Genomics2010,37,1:36

8Radiofrequency ablation or microwave ablation combined with transcatheter arterial chemoembolization in treatment of hepatocellular carcinoma by comparing with radiofrequency ablation alone显示文摘Objective:To compare radiofrequency ablation(RFA) or microwave ablation(MWA) and transcatheter arterial chemoembolization(TACE) with RFA or MWA monotherapy in hepatocellular carcinoma(HCC).Methods:A prospective,randomized,controlled trial was conducted on 94 patients with HCC ≤7 cm at a single tertiary referral center from June 2008 to June 2010 at the Department of Hepatobiliary Surgery,the Second Affiliated Hospital of Southeast University.The patients were randomly assigned into the TACERFA or TACE-MWA(combined treatment group) and the RFA-alone or MWA-alone groups(control group).The primary end point was overall survival.The secondary end point was recurrence-free survival,and the tertiary end point was adverse effects.Results:Until the time of censor,17 patients in the TACE-RFA or TACE-MWA group had died.The median follow-up time of the patients who were still alive for the TACE-RFA or TACE-MWA group was 47.5±11.3 months(range,29 to 62 months).The 1-,3- and 5-year overall survival for the TACE-RFA or TACE-MWA group was 93.6%,68.1% and 61.7%,respectively.Twenty-five patients in the RFA or MWA group had died.The median follow-up time of the patients who were still alive for the RFA or MWA group was 47.0±12.9 months(range,28 to 62 months).The 1-,3- and 5-year overall survival for the RFA or MWA group was 85.1%,59.6% and 44.7%,respectively.The patients in the TACE-RFA or TACE-MWA group had better overall survival than the RFA or MWA group [hazard ratio(HR),0.526;95% confidence interval(95% CI),0.334-0.823;P=0.002],and showed better recurrence-free survival than the RFA or MWA group(HR,0.582;95% CI,0.368-0.895;P=0.008).Conclusions:RFA or MWA combined with TACE in the treatment of HCC ≤7 cm was superior to RFA or MWA alone in improving survival by reducing arterial and portal blood flow due to TACE with iodized oil before RFA.Yongxiang Yi Yufeng Zhang Qiang Wei Liang Zhao Jianbo Han Yan Song Ying Ding Guilan Lu Junmao Liu Huaiying Ding Feng Dai Xiaojun Tang 2014Chinese Journal of Cancer Research2014,26,1:29

10Behaviors and kinetics of toluene adsorption-desorption on activated carbons with varying pore structure显示文摘This work was undertaken to investigate the behaviors and kinetics of toluene adsorption and desorption on activated carbons with varying pore structure. Five kinds of activated carbon from different raw materials were selected. Adsorption isotherms and breakthrough curves for toluene were measured. Langmuir and Freundlich equations were fitted to the equilibrium data, and the Freundlich equation was more suitable for simulating toluene adsorption. The process consisted of monolayer, multilayer and partial active site adsorption types. The effect of the pore structure of the activated carbons on toluene adsorption capacity was investigated. The quasi-first-order model was more suitable for describing the process than the quasi-second-order model. The adsorption data was also modeled by the internal particle diffusion model and it was found that the adsorption process could be divided into three stages. In the external surface adsorption process, the rate depended on the specific surface area. During the particle diffusion stage, pore structure and volume were the main factors affecting adsorption rate. In the final equilibrium stage, the rate was determined by the ratio of meso-and macro-pores to total pore volume. The rate over the whole adsorption process was dominated by the toluene concentration. The desorption behavior of toluene on activated carbons was investigated,and the process was divided into heat and mass transfer parts corresponding to emission and diffusion mechanisms, respectively. 13Research on the slope spectrum of the Loess Plateau显示文摘A new concept dealing with digital analysis of loess terrain,slope spectrum,is presented and discussed in this paper,by introducing its characteristic,represen-tation and extracting method from DEMs. Using 48 geomorphological units in dif-ferent parts of the loess as test areas and 5 m-resolution DEMs as original test data,the quantitative depiction and spatial distribution of slope spectrum in China's Loess Plateau have been studied on the basis of a series of carefully-designed experiments. In addition,initial experiment indicates a strong relationship between the slope spectrum and the loess landform types,displaying a potential importance of the slope spectrum in geomorphological studies. Based on the slope spectrums derived from the 25 m-resolution DEM data in whole loess terrain in northern part of Shaanxi,13 slope spectrum indices were extracted and integrated into a compre-hensive layer with image integration method. Based on that,a series of unsuper-vised classifications was applied in order to make a landform classification in northern Shaanxi Loess Plateau. Experimental results show that the slope spec-trum analysis is an effective method in revealing the macro landform features. A continuous change of slope spectrum from south to north in northern Shaanxi Loess Plateau shows an obvious spatial distribution of different loess landforms. 14Wx^lv、the Ancestral Allele of Rice Waxy Gene显示文摘In rice grains,the Waxy (Wx) gene is responsible for the synthesis of amylose,the most important determinant for eating and cooking quality.The effects of several Wx alleles on amylose content and the taste of cooked rice have been elucidated.However,the relationship between artificial selection and the evolution of various Wx alleles as well as their distribution remain unclear.Here we report the identification of an ancestral allele,Wx^lv,which dramatically affects the mouthfeel of rice grains by modulating the size of amylose molecules.We demonstrated that WF originated directly from wild rice,and the three major Wx alleles in cultivated rice (Wx^b,Wx^a,and Wx^in) differentiated after the substitution of one base pair at the functional sites.These data indicate that the Wx^lv allele played an important role in artificial selection and domestication.The findings also shed light on the evolution of various Wx alleles,which have greatly contributed to improving the eating and cooking quality of rice.Changquan Zhang Jihui Zhu Shengjie Chen Xiaolei Fan Qianfeng Li Yan Lu Min Wang Hengxiu Yu Chuandeng Yi Shuzhu Tang Minghong Gu Qiaoquan Liu 2019Molecular Plant2019,12,8:22

16p16 gene methylation in colorectal cancers associated with Duke's staging显示文摘AIM To explore the association of methylation of the CpG island in the promotor of the p16 tumor suppressor gene with the clinicopathological characteristics of the colorectal cancers.``METHODS Methylation-specific PCR (MSP) was used to detect p16 methylation of 62 sporadic colorectal cancer specimens.``RESULTS pi6 methylation was detected in 42% of the tumors. 18Abdominal paracentesis drainage ameliorates severe acute pancreatitis in rats by regulating the polarization of peritoneal macrophages显示文摘AIM To investigate the role of peritoneal macrophage(PM) polarization in the therapeutic effect of abdominal paracentesis drainage(APD) on severe acute pancreatitis(SAP).METHODS SAP was induced by 5% Na-taurocholate retrograde injection in Sprague-Dawley rats. APD was performed by inserting a drainage tube with a vacuum ball into the lower right abdomen of the rats immediately after the induction of SAP. To verify the effect of APD on macrophages, PMs were isolated and cultured in an environment, with the peritoneal inflammatory environment simulated by the addition of peritoneal lavage in complete RPMI 1640 medium. Hematoxylin and eosin staining was performed. The levels of pancreatitis biomarkers amylase and lipase as well as the levels of inflammatory mediators in the blood and peritoneal lavage were determined. The polarization phenotypes of the PMs were identified by detecting the marker expression of M1/M2 macrophages via flow cytometry, qPCR and immunohistochemical staining. The protein expression in macrophages that had infiltrated the pancreas was determined by Western blot.RESULTS APD treatment significantly reduced the histopathological scores and levels of amylase, lipase, tumor necrosis factor-α and interleukin(IL)-1β, indicating that APD ameliorates the severity of SAP. Importantly, we found that APD treatment polarized PMs towards the M2 phenotype, as evidenced by the reduced number of M1 macrophages and the reduced levels of proinflammatory mediators, such as IL-1β and L-selectin, as well as the increased number of M2 macrophages and increased levels of anti-inflammatory mediators, such as IL-4 and IL-10. Furthermore, in an in vitro study wherein peritoneal lavage from the APD group was added to the cultured PMs to simulate the peritoneal inflammatory environment, PMs also exhibited a dominant M2 phenotype, resulting in a significantly lower level of inflammation. 20The poisoning and regeneration effect of alkali metals deposed over commercial V_2O_5-WO_3/TiO_2 catalysts on SCR of NO by NH_3显示文摘In this study,commercial V2O5-WO3/TiO2catalysts were deactivated by loading with alkali metals(K and Na).These catalysts were then regenerated by washing with either deionized water or 0.5 mol/L H2SO4(through the ultrasonic-assisted method).The samples used in this research were characterized by NH3-temperature programmed desorption(TPD),and X-ray photoelectron spectroscopy(XPS).Results showed that Na2O and K2O doping can poison the V2O5-WO3/TiO2catalyst and that the poisoning effect of Na2O was stronger than that of K2O.However,the Na2O-loaded sample was easier to regenerate than the K2O-loaded sample.The surfaces of catalysts can be sulfated by washing with dilute sulfuric acid because strong acid sites adhere to the catalyst surface.SO42-could also promote catalyst activity.As indicated by the NH3-TPD findings,the deposition of Na2O and K2O could also reduce the amount of desorbed ammonia and destabilize the acid sites,especially strong chemisorption sites.XPS results revealed that catalysts were deactivated by the decrease in the concentration of chemisorbed oxygen[the Oa/(Oα+Oβ)ratio].In the Na2O-doped catalyst,much chemisorbed oxygen was lost(from 28.8%to10.6%).However,the decrease in the Oa/(Oα+Oβ)ratio was less significant in the K2O-doped catalyst(from28.8%to 23.5%).Nonetheless,the binding energies of O1s broadened with respect to both high and low energy.In particular,the binding energy of chemisorbed oxygen increased from 531.5 to 531.8 eV.Fengyu Gao Xiaolong Tang Honghong Yi Shunzheng Zhao Tongtong Zhang Dong Li Ding Ma 2014Chinese Science Bulletin2014,59,31:19
