A heavy mineral viewpoint on sediment provenance and environment in the Qiongdongnan Basin显示文摘Based on heavy mineral data in core samples from eleven drillings, supplemented by paleontological, element geochemical and seismic data, the evolution of sediment provenance and environment in the Qiongdongnan Basin(QDNB) was analysed. The results show that the basement in the QDNB was predominantly composed of terrigenous sediments. Since the Oligocene the QDNB has gradually undergone transgressions and evolution processes in sedimentary environment from terrestrial-marine transitional to littoral-neritic, neritic, and bathyal roughly. The water depth showed a gradually increasing trend and was generally greater in the southern region than that in the northern region in the same time. With changes in sedimentary environment, provenances of the strata(from the Yacheng Formation to the Yinggehai Formation) showed principal characteristics of multisources, evolving from autochthonous source, short source to distant source step by step. During the Early Oligocene, the sediments were mainly proximal basaltic pyroclastic source and adjacent terrigenous clastic source, afterwards were becoming distant terrigenous clastic sources, including Hainan Island on the north,Yongle Uplift on the south, Shenhu Uplift on the northeast, the Red River System on the northwest and Indochina Peninsula on the southwest, or even a wider region. The Hainan Island provenance began to develop during the Early Oligocene and has become a main provenance in the QDNB since the Middle Miocene. The provenances from Yongle Uplift and Shenhu Uplift most developed from the Late Oligocene to the Early Miocene and gradually subsided during the Middle Miocene. During the Late Miocene, as a main source of sediments filled in the central canyon, the Red River System provenance added to the QDNB massively, whose impact terminated at the end of the Pliocene. The western Yinggehai Basin(YGHB) provenance derived from Indochina Peninsula had developed from the Pliocene on to the Pleistocene. In addition, the material contribution of marine authigenous source to the basin(especially to the southern region) could not be ignored.LIU Xiaofeng ZHANG Daojun ZHAI Shikui LIU Xinyu CHEN Hongyan LUO Wei LI Na XIU Chun 2015Acta Oceanologica Sinica2015,34,4:10
Two gap-free reference genomes and a global view of the centromere architecture in rice显示文摘Rice(Oryza sativa),a major staple throughout the world and a model system for plant genomics and breeding, was the first crop genome sequenced almost two decades ago. However, reference genomes for all higher organisms to date contain gaps and missing sequences. Here,we report the assembly and analysis of gap-free reference genome sequences for two elite O.sativa xian/indica rice varieties,Zhenshan 97 and Minghui 63,which are being used as a model system for studying heterosis and yield.Gap-free reference genomes provide the opportunity for a global view of the structure and function of centromeres. We show that all rice centromeric regions share conserved centromere-specific satellite motifs with different copy numbers and structures. In addition, the similarity of CentO repeats in the same chromosome is higher than across chromosomes, supporting a model of local expansion and homogenization. Both genomes have over 395 non-TE genes located in centromere regions,of which∼41%are actively transcribed. Two large structural variants at the end of chromosome 11 affect the copy number of resistance genes between the two genomes. The availability of the two gap-free genomes lays a solid foundation for further understanding genome structure and function in plants and breeding climate-resilient varieties.Jia-Ming Song Wen-Zhao Xie Shuo Wang Yi-Xiong Guo Dal-Hoe Koo Dave Kudrna Chenbo Gong Yicheng Huang Jia-Wu Feng Wenhui Zhang Yong Zhou Andrea Zuccolo Evan Long Seunghee Lee Jayson Talag Run Zhou Xi-Tong Zhu Daojun Yuan Joshua Udall Weibo Xie Rod AWing Qifa Zhang Jesse Poland Jianwei Zhang Ling-Ling Chen 2021Molecular Plant2021,14,10:9
Evaluation of efficacy and safety of gandouling plus sodium dimercaptosulphonate in treatment of patients with neurological Wilson's disease from China显示文摘OBJECTIVE: To evaluate the efficacy and safety of gandouling plus sodium dimercaptosulphonate(DMPS) on neurological Wilson's disease(WD) in patients.METHODS: We retrospectively evaluated the clinical records of 125 WD patients with neurological syndromes who were treated with gandouling plus sodium DMPS or DMPS used alone. All patients had a history of neurological deterioration during their diseases courses. The clinical efficacies, adverse reactions, and results of the various hematological and biochemical investigations were recorded for statistical analysis.RESULTS: 92.30%(60 patients) of the WD patients treated with the combined therapy experienced an improved or stable neurological condition paral-leled by a significantly improved GAS score. Meanwhile, the WBC and PLT counts stabilized, liver function and renal function were improved or remained stable. The combined therapy also obviously promoted the 24-h urinary copper excretion. In particular, only 30.76% of the WD patients experienced mild adverse reactions, including neurological deterioration in 5 patients(7.69%), hepatic worsening in 1 subject(1.89%), which was less frequently than those in the control group treated with DMPS only.CONCLUSION: Our findings indicate that the safety and efficacy of gandou-ling plus DMPS is superior to those of DMPS used alone in the WD patients with neurological symptoms.Zhang Jing Xie Daojun Guo Yanbing Li Yajun Li Liangyong Han Hui Zhang Juan Chen Huaizhen Wang Meixia 2018Journal of Traditional Chinese Medicine2018,38,5:7
Abrogation of Hn RNP L enhances anti-PD-1 therapy efficacy via diminishing PD-L1 and promoting CD8^(+) T cell-mediated ferroptosis in castration-resistant prostate cancer显示文摘Owing to incurable castration-resistant prostate cancer(CRPC)ultimately developing after treating with androgen deprivation therapy(ADT),it is vital to devise new therapeutic strategies to treat CRPC.Treatments that target programmed cell death protein 1(PD-1)and programmed death ligand-1(PD-L1)have been approved for human cancers with clinical benefit. However, many patients, especially prostate cancer, fail to respond to anti-PD-1/PD-L1 treatment,so it is an urgent need to seek a support strategy for improving the traditional PD-1/PD-L1 targeting immunotherapy. In the present study, analyzing the data from our prostate cancer tissue microarray,we found that PD-L1 expression was positively correlated with the expression of heterogeneous nuclear ribonucleoprotein L(Hn RNP L).Hence,we further investigated the potential role of Hn RNP L on the PD-L1 expression, the sensitivity of cancer cells to T-cell killing and the synergistic effect with anti-PD-1 therapy in CRPC.Indeed,Hn RNP L knockdown effectively decreased PD-L1 expression and recovered the sensitivity of cancer cells to T-cell killing in vitro and in vivo,on the contrary,Hn RNP L overexpression led to the opposite effect in CRPC cells. In addition, consistent with the previous study,we revealed that ferroptosis played a critical role in T-cell-induced cancer cell death, and Hn RNP L promoted the cancer immune escape partly through targeting YY1/PD-L1 axis and inhibiting ferroptosis in CRPC cells.Furthermore,Hn RNP L knockdown enhanced antitumor immunity by recruiting infiltrating CD8^(+)T cells and synergized with anti-PD-1 therapy in CRPC tumors. This study provided biological evidence that Hn RNP L knockdown might be a novel therapeutic agent in PD-L1/PD-1 blockade strategy that enhanced anti-tumor immune response in CRPC.Xumin Zhou Libin Zou Hangyu Liao Junqi Luo Taowei Yang Jun Wu Wenbin Chen Kaihui Wu Shengren Cen Daojun Lv Fangpeng Shu Yu Yang Chun Li Bingkun Li Xiangming Mao 2022Acta Pharmaceutica Sinica B2022,12,2:4
A Deletable and Modifiable Blockchain Scheme Based on Record Verification Trees and the Multisignature Mechanism显示文摘As one of the most valuable technologies, blockchains have received extensive attention from researchers and industry circles and are widely applied in various scenarios. However, data on a blockchain cannot be deleted. As a result,it is impossible to clean invalid and sensitive data and correct erroneous data. This,to a certain extent, hinders the application of blockchains in supply chains and Internet of Things. To address this problem, this study presents a deletable and modifiable blockchain scheme(DMBlockChain)based on record verification trees(RVTrees)and the multisignature scheme.(1)In this scheme,an RVTree structure is designed and added to the block structure. The RVTree can not only ensure that a record is true and valid but, owing to its unique binary structure, also verify whether modification and deletion requests are valid.(2)In DMBlockChain, the multisignature mechanism is also introduced. This mechanism requires the stakeholders’signatures for each modification or deletion request and thus ensures that a record will not be modified arbitrarily.A user’s request is deemed valid only if it is dually verified by the RVTree and the multisignature mechanism. The analysis finds that DMBlockChain can provide a secure and valid means for modifying and deleting records in a block while ensuring the integrity of the block and that DMBlockChain can effectively save space in some scenarios that require frequent records modification.Daojun Han Jinyu Chen Lei Zhang Yatian Shen Yihua Gao Xueheng Wang 2021Computer Modeling in Engineering & Sciences2021,,7:3
Mode for reducing wind curtailment based on battery transportation显示文摘Renewable energy sources, such as wind and solar, face several obstacles, including curtailment, where the generated energy exceeds local demand and production must be reduced because of limited transmission capacity.Simultaneously, consumer demand for large-capacity batteries is expanding given the recent rapid development of electric vehicles(EVs) and plug-in hybrid EVs. A batterycharging mode, in which discharged batteries are transported from battery exchange stations in high-load areas to wind farms, is proposed to alleviate curtailment. In this study, batteries store otherwise curtailed energy and smooth the wind power output simultaneously. The structure of the battery-charging device is discussed, and the concept of a battery-charging container is proposed. The control principle of the battery-charging management unit is presented with a simulation model constructed in the PSCAD/EMTDC environment. A case study is simulated, and the feasibility of the mode is analyzed considering the levelized cost and energy losses. Simulation results show that this mode is a feasible solution to alleviating wind curtailment and providing fresh impetus for developing EV battery exchanges.Zhanlong ZHANG Daojun MEI Han JIANG Guohua LIU Hongpeng HE Yue CHEN 2018Journal of Modern Power Systems and Clean Energy2018,6,6:2

Effect of manganese and/or ceria loading on V_2O_5-MoO_3/TiO_2 NH_3 selective catalytic reduction catalyst显示文摘The effect of manganese and/or ceria loading of V2 O5-MoO3/TiO2 catalysts was investigated for selective catalytic reduction(SCR) of NOx by NH3.The manganese and/or ceria loaded V2 O5-MoO3/TiO2 catalysts we re prepared by the wetness impregnation method. The physicochemical characteristics of the catalysts were thoroughly characterized. The catalytic performance of 1.5 wt% V2 O5-3 wt% MoO3/TiO2(V1.5 Mo3/Ti) is greatly enhanced by addition of 2.5 wt% MnOx and 3.0 wt% CeO2(V1.5 Mo3 Mn2.5 Ce3/Ti) below450℃.Compared with the V1.5 Mo3/Ti catalyst with NOx conversion of 75% at 275 ℃,V1.5 Mo3 Mn2.5 Ce3/Ti exhibits higher NOx conversion of 84% with good resistance to SO2 and H2 O at a gas hourly space velocity value of 150000 h-1.The active manganese, cerium, molybdenum, and vanadium oxide species are highly dispersed on the catalyst surface and some synergistic effects exist among these species.Addition of MnOx significantly enhances the redox ability of the cerium, vanadium, and molybdenum species.Addition of Ce increases the acidity of the catalyst.More active oxygen species, including surface chemisorbed oxygen, form with addition of Mn and/or Ce.Because of the synergistic effects, appropriate proportions of manganese in different valence states exist in the catalysts. In summary, the good redox ability and the strong acidity contribute to the high NH3-SCR activity and N2 selectivity of the V1.5 Mo3 Mn2.5 Ce3/Ti catalyst in a wide temperature range. And the V1.5 Mo3 Mn2.5 Ce3/Ti catalyst shows good resistance to H2 O and SO2 in long-time catalytic testing, which can be ascribed to the highly sulfated species adsorbed on the catalyst.Daojun Zhang Ziran Ma Baodong Wang Tao Zhu Duan Weng Xiaodong Wu Jingyun Chen Hongyan Wang Ge Li Jiali Zhou 2020Journal of Rare Earths2020,38,7:1

Molecular Characterization, Tissue Distribution and Localization of Larimichthys crocea Kif3a and Kif3b and Expression Analysis of Their Genes During Spermiogenesis显示文摘KIF3A and KIF3B are two N-terminal motor proteins belonging to the kinesin-II superfamily that play essential roles in spermiogenesis. To understand the roles played by KIF3 A/3B during spermatogenesis of large yellow croaker Larimichthys crocea,we studied the testis characteristics at different developmental stages of L.crocea, and determined the spatiotemporal expression patterns of kif3a and kif3b during spermiogenesis.Quantitative real-time PCR(qR T-PCR)showed that the overall trends of kif3 a/3 b m RNA abundance during testis development are similar. From stage Ⅱ to stage V,kif3a/3b m RNA abundances first increased and then fell after reaching a peak at stage IV.Interestingly, the m RNA abundances of both genes at stage V were higher than those at stages Ⅱ and Ⅲ.In addition,it is worth of noting that kif3 b m RNA abundance was higher than that of kif3a at all stages.Fluorescence in situ hybridization results revealed that kif3a/3b m RNA abundance dynamics were consistent with the migration of mitochondria, the deformation of nucleus, and the formation of tail. The m RNA hybridization signals of both genes first appeared either around the nuclear periphery or on the side of the nuclei, then appeared at one side of nuclei, and finally were mainly on the tail during spermiogenesis. Our findings contributed to better understanding the molecular mechanisms of spermiogenesis in fish;and suggested that KIF3A and KIF3B may participate in the intracellular transport of mitochondria, nuclear deformation, and the formation of tail during the spermiogenesis in L.crocea.MU Danli DU Chen FU Suyan WANG Jingqian HOU Congcong TANG Daojun ZHU Junquan 2019Journal of Ocean University of China2019,18,6:0
Characterization, expression dynamics, and potential function of OPA1 for regulation of mitochondrial morphology during spermiogenesis in Phascolosoma esculenta显示文摘Mitochondria undergo morphological changes during spermatogenesis in some animals. The mechanism and role of mitochondrial morphology regulation, however, remain somewhat unclear. In this study,we analyzed the molecular characteristics, expression dynamics and subcellular localization of optic atrophy protein 1(OPA1),a mitochondrial fusion and cristae maintenance-related protein,to reveal the possible regulatory mechanisms underlying mitochondrial morphology in Phascolosoma esculenta spermiogenesis. The full-length cDNA of the P.esculenta opa1 gene(Pe-opa1)is 3743 bp in length and encodes 975 amino acids. The Pe-OPA1 protein is highly conservative and includes a transmembrane domain,a GTPase domain, two helical bundle domains, and a lipid-interacting stalk.Gene and protein expression was higher in the coelomic fluid(a site of spermatid development)of male P.esculenta and increased first and then decreased from March to December. Moreover, their expression during the breeding stage was significantly higher than during the non-breeding stage, suggesting that Pe-OPA1 is involved in P.esculenta reproduction. The Pe-OPA1 protein was more abundant in components consisting of many spermatids than in components without, indicating that Pe-OPA1 mainly plays a role in the spermatid in coelomic fluid.Moreover,Pe-OPA1 was mainly detected in the spermatid mitochondria.Immunofluorescence experiments showed that the Pe-OPA1 are constitutively expressed and co-localized with mitochondria during spermiogenesis, suggesting its involvement in P.esculenta spermiogenesis. These results provide evidence for Pe-OPA1's involvement in the regulation of mitochondrial morphology during spermiogenesis.Xinming GAO Binbin FENG Chen DU Congcong HOU Shan JIN Daojun TANG Junquan ZHU 2024Journal of Oceanology and Limnology2024,42,1:0
Prohibitin Protein Expression During Spermatogenesis in the Large Yellow Croaker, Larimichthys crocea显示文摘Mitochondria are important for animals’fertility, and their morphologies and functions during spermatogenesis are un-der investigation. However, the molecular mechanism that regulates the mitochondrial dynamic during spermatogenesis is still un-known.In this study, the cytological features of spermatogenesis were investigated in Larimichthys crocea. In addition, the structure and function of prohibitin(PHB),which is associated with mitochondrial structure and dynamic, was also investigated. The full-length cDNA and protein(Lc-PHB)from the L.crocea phb gene(Lc-phb)contained 1625 base pairs and 271 amino acids,respec-tively.Lc-PHB had a conserved primary structure that resulted in a transmembrane,SPFH(the analogous region of proteins stomat-ins,prohibitins,flotillins and HflK/C),and coiled-coil domains. It was detected at high levels in the muscle, liver, and heart, and at intermediate levels in the testis, gill, and brain.Lc-phb mRNA expression was detected in spermatogenic cells by fluorescence in situ hybridization. An immunofluorescence assay revealed that PHB protein was localized in the mitochondria during spermatogenesis.Specifically,PHB expression was detected in the perinuclear cytoplasm of spermatogonia, spermatocytes, and spermatids in the early developmental stage, and mainly localized on one side of the nuclei in the cytoplasm of spermatids in a middle developmental stage, and finally on the sperm midpiece.Western blotting showed that PHB was located in the extracted mitochondria protein fraction but not in the cytoplasm protein fraction of testes.Conclusively, these results indicated that PHB was expressed in the mitochondria dur-ing spermatogenesis. In addition, the study explained the mitochondrial dynamic during fish spermatogenesis and proposed a possi-ble relationship among PHB,spermatogenesis, and male fertility.LIN Chenwen GAO Xinming NI Jie ZHANG Shengshuo LIU Cheng LUO Shengyu DU Chen ZHENG Xuebin HOU Congcong TANG Daojun ZHANG Chundan ZHU Junquan 2022Journal of Ocean University of China2022,21,5:0
Physiological and histological responses of Phascolosoma esculenta (Sipuncula: Phascolosomatidea) to acute heat stress显示文摘Temperature is an important factor affecting the growth, development, and survival of organisms. The effects of temperature stress on aquatic organisms have received increasing attention,as these organisms are mostly poikilotherms and their body temperature are directly corresponding changes with ambient temperature, resulting in they are easily exposed in temperature stress. However, little is known about the effects of high temperature on Sipuncula. In this study,we investigated the effects of acute heat stress on malondialdehyde(MDA)concentration, the activities of antioxidant(superoxide dismutase and glutathione peroxidase)and immunity-related(acidic and alkaline phosphatase)enzymes, heat shock protein 70(hsp70)and hsp90 gene expression, and the histological structure of the sipunculid Phascolosoma esculenta.Within the coelom fl uid,the MDA concentration and all detected antioxidant enzyme activities increased during high temperature stress;signifi cant increases were also observed here and in the intestine in the hsp70 and hsp90 mRNA expression levels. These results indicated that acute heat stress caused oxidative stress;antioxidants and heat shock proteins probably act to protect P.esculenta against oxidative damage, constituting part of its physiological mechanism for adaptation to high temperatures. In addition, the increased activity of the acidic and alkaline phosphatases indicated eff ects on its nonspecifi c immune system.Furthermore, damaged tissue structures were observed in the body wall, retractor muscle, intestine, and nephridium after 96 h of 40-℃ stress. The damaged cells of these tissues showed obvious condensed chromatin around the nuclear membrane. This histological damage suggests that heat stress could affect movement, food absorption, digestion, and excretion in P.esculenta.These results elucidate the effects of temperature stress on P.esculenta and its physiological response mechanisms and provide practical indicators for assessing heat stress status and determining suitable culture temperatures for P.esculenta.Xinming GAO Haiyan YANG Daojun TANG Chen DU Shan JIN Congcong HOU Chundan ZHANG Junquan ZHU Jianping WANG 2022Journal of Oceanology and Limnology2022,40,2:0
