1Palaeozoic tectonic evolution of the Tianshan belt,NW China显示文摘The Chinese Tianshan belt is a major part of the southern Central Asian Orogenic Belt,extending westward to Kyrgyzstan and Kazakhstan.Its Paleozoic tectonic evolution,crucial for understanding the amalgamation of Central Asia,comprises two stages of subduction-collision.The first collisional stage built the Eo-Tianshan Mountains,before a Visean unconformity,in which all structures are verging north.It implied a southward subduction of the Central Tianshan Ocean beneath the Tarim active margin,that induced the Ordovician-Early Devonian Central Tianshan arc,to the south of which the South Tianshan back-arc basin opened.During the Late Devonian,the closure of this ocean led to a collision between Central Tianshan arc and the Kazakhstan-Yili-North Tianshan Block,and subsequently closure of the South Tianhan back-arc basin,producing two suture zones,namely the Central Tianshan and South Tianshan suture zones where ophiolitic mélanges and HP metamorphic rocks were emplaced northward.The second stage included the Late Devonian-Carboniferous southward subduction of North Tianshan Ocean beneath the Eo-Tianshan active margin,underlined by the Yili-North Tianshan arc,leading to the collision between the Kazakhstan-Yili-NTS plate and an inferred Junggar Block at Late Carboniferous-Early Permian time.The North Tianshan Suture Zone underlines likely the last oceanic closure of Central Asia Orogenic Belt;all the oceanic domains were consumed before the Middle Permian.The amalgamated units were affected by a Permian major wrenching,dextral in the Tianshan.The correlation with the Kazakh and Kyrgyz Tianshan is clarified.The Kyrgyz South Tianshan is equivalent to the whole part of Chinese Tianshan(CTS and STS) located to the south of Narat Fault and Main Tianshan Shear Zone;the so-called Middle Tianshan thins out toward the east.The South Tianshan Suture of Kyrgyzstan correlates with the Central Tianshan Suture of Chinese Tianshan.The evolution of this southern domain remains similar from east(Gangou area) to west until the Talas-Ferghana Fault,which reflects the convergence history between the Kazakhstan and Tarim blocks.Jacques CHARVET SHU LiangShu Sebastien LAURENT-CHARVET WANG Bo Michel FAURE Dominique CLUZEL CHEN Yan Koen De JONG 2011Science China Earth Sciences2011,54,2:57
2Increase of TNFα-stimulated Osteoarthritic Chondrocytes Apoptosis and Decrease of Matrix Metalloproteinases 9 by NF-κB Inhibition显示文摘Objective To investigate the in vitro effect of caffeic acid phenethyl ester(CAPE),a NF-κB inhibitor,on the apoptosis of osteoarthritic(OA)chondrocytes and on the regulation of the gelatinases matrix metalloproteinase 2(MMP-2)and matrix metalloproteinase 9(MMP-9).Methods Annexin V-FITC/propidium iodide(PI)labeling and western blotting were used to observe and determine the apoptosis in TNFα-stimulated primary cultured osteoarthritic chondrocytes.Also,gelatin zymography was applied to examine MMP-2 and MMP-9 activities in supernatants.Results It was confirmed by both flow cytometry and western blotting that chondrocytes from OA patients have an apoptotic background.Use of CAPE in combination with 10 ng/mL of TNFα for 24 h facilitated the apoptosis.MMP-9 in the supernatant could be autoactivated(from proMMP-9 to active MMP-9),and the physiologic calcium concentration(2.5 mmol/L)could delay the autoactivation of MMP-9.The activities of MMP-2 and MMP-9 in the fresh supernatant increased significantly in response to stimulation by 10 ng/mL of TNFα for 24 h.The stimulatory effect of TNFα just on proMMP-9 was counteracted significantly by CAPE.Conclusion NF-κB could prevent chondrocytes apoptosis though its activation was attributed to the increase of proMMP-9 activity induced by TNFα(a pro-apoptotic factor).Therefore,therapeutic NF-κB inhibitor was a 'double-edged swords' to the apoptosis of chondrocytes and the secretion of MMP-9.WANG Yan LI De Ling ZHANG Xin Bo DUAN Yuan Hui WU Zhi Hong HAO Dong Sheng CHEN Bao Sheng QIU Gui Xing 2013Biomedical and Environmental Sciences2013,26,4:13
3The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 4SIZ1-Mediated SUMOylation of TPR1 Suppresses Plant Immunity in Arabidopsis显示文摘Plant immune responses are tightly regulated to ensure their appropriate deployment. Overexpression of TOPLESS-RELATED 1 (TPR1), a SUPPRESSOR OF npr1-1, CONSTITUTIVE 1 (SNC1)-interacting protein, results in autoimmunity that reduces plant growth and development. However, how TPR1 activity is regulated remains unknown. Loss of function of SIZ1, a (SUMO) E3 ligase, induces an autoimmune response, partially due to elevated SNC1 levels. Here we show that SNC1 expression is upregulated in Arabidopsis thaliana siz1-2 due to positive-feedback regulation by salicylic acid. SIZ1 physically interacts with TPR1 and facilitates its SUMO modification. The K282 and K721 residues in TPR1 serve as critical SUMO attachment sites. Simultaneous introduction of K282R and K721R substitutions in TPR1 blocked its SUMOylation, enhaneed its transcriptional co-repressor activity, and increased its association with HISTONE DEACETYLASE 19 (HDA19), suggesting that SUMOylation of TPR1 represses its transcriptional co-repressor activity and inhibits its interaction with HDA19. In agreement with this finding, the simultaneous introduction of K282R and K721R substitutions enhanced TPR1 mediated immunity, and the tpr1 mutation partially suppressed autoimmunity in siz1-2. 6A pair of light signaling factors FHY3 and FAR1 regulates plant immunity by modulating chlorophyll biosynthesis显示文摘Light and chloroplast function is known to affect the plant immune response; however, the underlying mechanism remains elusive. We previously demonstrated that two light signaling factors, FAR-RED ELONGATED HYPOCOTYL 3(FHY3)and FAR-RED IMPAIRED RESPONSE 1(FAR1), regulate chlorophyll biosynthesis and seedling growth via controlling HEMB1 expression in Arabidopsis thaliana. In this study, we reveal that FHY3 and FAR1 are involved in modulating plant immunity. We showed that the fhy3 far1 double null mutant displayed high levels of reactive oxygen species and salicylic acid(SA) and increased resistance to Pseudomonas syringae pathogen infection. Microarray analysis revealed that a large proportion of pathogen-related genes, particularly genes encoding nucleotide-binding and leucine-rich repeat domain resistant proteins, are highly induced in fhy3 far1. Genetic studies indicated that the defects of fhy3 far1 can be largely rescued by reducing SA signaling or blocking SA accumulation, and by overexpression of HEMB1, which encodes a 5-aminolevulinic acid dehydratase in the chlorophyll biosynthetic pathway.Furthermore, we found that transgenic plants with reduced expression of HEMB1 exhibit a phenotype similar to fhy3 far1.Taken together, this study demonstrates an important role of FHY3 and FAR1 in regulating plant immunity, through integrating chlorophyll biosynthesis and the SA signaling pathway.Wanqing Wang Weijiang Tang Tingting Ma De Niu Jing Bo Jin Haiyang Wang Rongcheng Lin 2016Journal of Integrative Plant Biology2016,58,1:8
7Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 9Inclusion of purified dietary fiber during gestation improved the reproductive performance of sows显示文摘Background: This study aimed to investigate the impacts of guar gum and cellulose as the source of dietary fiber during gestation on the reproductive performance of sows.Methods: A total of 210 sows(parities 3–6) were randomly allocated into six diets(n = 35) throughout gestation to feed graded levels of dietary fiber(DF), including a corn-soybean meal-based control diet with no wheat bran inclusion(CON, 12.5% DF), a wheat bran-rich diet(DF1, 17.4% DF), and another 4 diets(DF2, 17.7% DF;DF3, 18.1%DF;DF4, 18.4% DF;DF5, 18.8% DF) in which wheat bran were equally substituted by 1%, 2%, 3% and 4% purified FIBER MIX(guar gum and cellulose, 1:4). All sows received similar DE and other nutrients throughout gestation.Results: DF treatment during gestation resulted in normal fecal score(1 to 5 with 1 = dry and 5 = watery) in sows compared with those received the CON diet(P < 0.05). The number of total born piglets had a tendency to be affected by dietary treatment(P = 0.07), and correlation analysis revealed a linear response of total born to dietary fiber levels during gestation(P < 0.01). Sows received the DF2, DF3, and DF5 diets during gestation had a greater ADFI during lactation compared with those in the CON group(P < 0.05) without affecting the daily body weight gain of suckling piglets. Gut microbiota compositions were dramatically changed by the gestation stage and some of those were changed by DF inclusion. 10Neonatal vitamin A injection promotes cattle muscle growth and increases oxidative muscle fibers显示文摘Background: Vitamin A and its metabolite,retinoic acid(RA),are important regulators of cell differentiation and organ morphogenesis.Its impact on beef cattle muscle growth remains undefined.Method: Angus steer calves were administrated with 0(control) or 150,000 IU vitamin A(retinyl palmitate in glycerol,i.m.) per calf at birth and 1 month of age.At 2 months of age,a biopsy of the Biceps femoris muscle was obtained to analyze the immediate effects of vitamin A injection on myogenic capacity of muscle cells.The resulting steers were harvested at 14 months of age.Results: Vitamin A administration increased cattle growth at 2 months.At 2 months of age,Vitamin A increased PAX7 positive satellite cells and the expression of myogenic marker genes including PAX7,MYF5,MYOD and MYOG.Muscle derived mononuclear cells were further isolated and induced myogenesis in vitro.More myotubes and a higher degree of myogenesis was observed in vitamin A groups.Consistently,vitamin A increased Latissimus dorsi(LD) muscle fiber size at harvest.In addition,vitamin A increased the ratio of oxidative type I and type IIA fibers and reduced the glycolic type IIX fibers.Furthermore,we found that RA,a key bioactive metabolite of vitamin A,activated PPARGC1 A promoter,which explains the upregulated expression of PPARGC1 A in skeletal muscle.Conclusion: Vitamin A administration to neonatal calves enhanced postnatal muscle growth by promoting myogenesis and increasing satellite cell density,accompanied with a shift to oxidative muscle fibers.Bo Wang Wei Nie Xing Fu Jeanene M de Avila Yannan Ma Mei-Jun Zhu Martin Maquivar Steven M Parish Jan R Busboom Mark L Nelson Min Du 2019Journal of Animal Science and Biotechnology2019,10,1:3

12VSITA, an Improved Approach of Target Amplification in the Identification of Viral Pathogens显示文摘Objective Unbiased next generation sequencing(NGS) is susceptible to interference from host or environmental sequences. Consequently, background depletion and virome enrichment techniques are usually needed for clinical samples where viral load is much lower than background sequences. Methods A viral Sequence Independent Targeted Amplification(VSITA) approach using a set of non-ribosomal and virus-enriched octamers(V8) was developed and compared with traditionally used random hexamers(N6). Forty-five archived clinical samples of different types were used in parallel to compare the V8 and N6 enrichment performance of viral sequences and removal performance of ribosomal sequences in the step of reverse transcription followed by quantitative PCR(qP CR). Ten sera samples from patients with fever of unknown origin and 10 feces samples from patients with diarrhea of unknown origin were used in comparison of V8 and N6 enrichment performance following NGS analysis. Results A minimum 30 hexamers matching to viral reference sequences(sense and antisense) were selected from a dataset of random 4,096(4~6) hexamers(N6). Two random nucleotides were added to the 5' end of the selected hexamers, and 480(30 × 4~2) octamers(V8) were obtained. In general, VSITA approach showed higher enrichment of virus-targeted c DNA and enhanced ability to remove unwanted ribosomal sequences in the majorities of 45 predefined clinical samples. Moreover, VSITA combined with NGS enabled to detect not only more viruses but also achieve more viral reads hit and higher viral genome coverage in 20 clinical samples with diarrhea or fever of unknown origin. 13Synthesis of N-methylene phosphonic chitosan(NMPCS)and its potential as gene carder显示文摘N-Methylene phosphoulc chitosan (NMPCS),an amphiphilic macromolecule with powerful chelating ability of Ca^(2+) ions,was synthesized and characterized.The physicochemical properties of NMPCS and the interactions between NMPCS and plasmid DNA were investigated by FTIR,^(13)C NMR,X-ray,agarose gel electrophorcsis retardation assay,atomic force microscopy (AFM) and circular dichroism (CD).The results suggest that at charge ratio 2:1 or above,DNA could be completely entrapped and spherical complexes with mean size of 80-210 nm were formed.Taking HeLa as host cell,luciferase expression mediated by NMPCS improved about 100 times compared to the expression mediated by chitosan.Dun Wan Zhu Jin Gen Bo Hai Ling Zhang Wen Guang Liu Xi Gang Leng Cun Xian Song Yu Ji Yin Li Ping Song Lan Xia Liu Lin Mei Xiu Lan Li Yang Zhang Kang De Yao 2007Chinese Chemical Letters2007,18,11:3

15Red Electroluminescence and Photoluminescence from Novel Binuclear Europium Complex with Squaric Acid Ligand显示文摘A novel binuclear europium b-diketone complex with squaric acid ligand was synthesized for the first time. Its structure was elucidated by IR, UV, and Elemental Analysis. Red light emitting diode (LED) was fabricated by using the novel europium complex as an emitting layer, tris(8-quinolinolate) aluminum (III) (Alq3) as an electron-transporting layer, N, N’-diphenyl-N, N’-(3-methylphenyl)-1,1’-biphenyl-4,4’-diamine (TPD) as a hole-transporting layer. A cell structure of indium-tin-oxide/TPD/Eu-complex/Alq3/Mg: Ag was employed. Red electroluminescence was observed at room temperature with dc bias voltage of 2 V in this cell. Red emission peaks at about 613 nm with maximum luminance of over 106 cd/m2.
