2Denitrification Losses and N_2O Emissions from Nitrogen Fertilizer Applied to a Vegetable Field显示文摘A field experiment was conducted on Chinese cabbage (Brassica campestris L. ssp. pekinensis (Lour.) Olsson) in a Nanjing suburb in 2003. The experiment included 4 treatments in a randomized complete block design with 3 replicates: zero chemical fertilizer N (CK); urea at rates of 300 kg N ha-1 (U300) and 600 kg N ha-1 (U600), both as basal and two topdressings; and polymer-coated urea at a rate of 180 kg N ha-1 (PCU180) as a basal application. The acetylene inhibition technique was used to measure denitrification (N2+N2O) from intact soil cores and N2O emissions in the absence of acetylene. Results showed that compared to CK total denitrification losses were significantly greater (P≤0.05) in the PCU180, U300, and U600 treatments, while N2O emissions in the U300 and U600 treatments were significantly higher (P≤0.05) than CK. In the U300 and U600 treatments peaks of denitrification and N2O emission were usually observed after N application. In the polymer-coated urea treatment (PCU180) during the period 20 to 40 days after transplanting, higher denitrification rates and N2O fluxes occurred. Compared with urea, polymer-coated urea did not show any effect on reducing denitrification losses and N2O emissions in terms of percentage of applied N. As temperature gradually decreased from transplanting to harvest, denitrification rates and N2O emissions tended to decrease. 4Cloud point extraction coupled with HPLC-UV for the determination of phthalate esters in environmental water samples显示文摘A method based on cloud point extraction was developed to determine phthalate esters including di-ethyl-phthalate (DEP), di-(2-ethylhexyl)-phthalate (DEHP) and di-cyclohexyl-phthalate (DCP) in environmental water samples using high-performance liquid chromatography separation and ultraviolet detection (HPLC-UV). The non-ionic surfactant Triton X-114 was chosen as extraction solvent. The parameters affecting extraction effciency, such as concentrations of Triton X-114 and Na2SO4, equilibration temperature, equilibration time and centrifugation time were evaluated and optimized. Under the optimum conditions, the method can achieve preconcentration factors of 35, 88, 111 and detection of limits of 2.0, 3.8, 1.0 ngml for DEP, DEHP and DCP in 10-ml water sample, respectively. 8Generation of a monkey with MECP2 mutations by TALEN-based gene targeting显示文摘Gene editing in model organisms has provided critical insights into brain development and diseases. Here, we report the generation of a cynomolgus monkey(Macaca fascicularis) carrying MECP2 mutations using transcription activator-like effector nucleases(TALENs)-mediated gene targeting. After injecting TALENs mRNA into monkey zygotes achieved by in vitro fertilization and embryo transplantation into surrogate monkeys, we obtained one male newborn monkey with an MECP2 deletion caused by frameshifting mutation in various tissues. The monkey carrying the MECP2 mutation failed to survive after birth, due to either the toxicity of TALENs or the critical requirement of MECP2 for neural development. The level of MeCP2 protein was essentially depleted in the monkey's brain. 9Identification of fruit and branch in natural scenes for citrus harvesting robot using machine vision and support vector machine显示文摘With the decrease of agricultural labor and the increase of production cost,the researches on citrus harvesting robot(CHR)have received more and more attention in recent years.For the success of robotic harvesting and the safety of robot,the identification of mature citrus fruit and obstacle is the priority of robotic harvesting.In this work,a machine vision system,which consisted of a color CCD camera and a computer,was developed to achieve these tasks.Images of citrus trees were captured under sunny and cloudy conditions.Due to varying degrees of lightness and position randomness of fruits and branches,red,green,and blue values of objects in these images are changed dramatically.The traditional threshold segmentation is not efficient to solve these problems.Multi-class support vector machine(SVM),which succeeds by morphological operation,was used to simultaneously segment the fruits and branches in this study.The recognition rate of citrus fruit was 92.4%,and the branch of which diameter was more than 5 pixels,could be recognized.The results showed that the algorithm could be used to detect the fruits and branches for CHR.Lü Qiang Cai Jianrong Liu Bin Deng Lie Zhang Yajing 2014International Journal of Agricultural and Biological Engineering2014,7,2:17
10Tectonic Evolution of the Meso-Tethys in the Western Segment of Bangonghu-Nujiang Suture Zone: Insights from Geochemistry and Geochronology of the Lagkor Tso Ophiolite显示文摘The subduction of the Bangonghu-Nujiang Meso-Tethys and the collision between the Lhasa and Qiangtang blocks were important events in the growth of the Tibetan crust. However,the timing of collision initiation and closure timing,as well as nature and structure of the Bangonghu ocean basin,are still poorly constrained. The Lagkor Tso ophiolite,located in the south of Gerze County,Tibet,is one of the most completed ophiolites preserved in the southern side of the BangonghuNujiang suture zone. This study discussed the tectonic evolution of the Bangonghu-Nujiang suture zone as revealed by the Lagkor Tso ophiolite investigated by field investigations,petrology,geochemistry,geochronology and tectonic analysis methods. We present new LA-ICP-MS zircon U-Pb and 39Ar/40 Ar ages for the Lagkor Tso ophiolite,in addition to geochemical and platinum-group element(PGE) data presented for the Lagkor Tso ophiolite in Tibet. It is suggested that the ancient Lagkor Tso oceanic basin split in Middle Jurassic(161.2 ± 2.7 Ma – 165.4 ± 3.5 Ma),and experienced a second tectonic emplacement during the Early Cretaceous(137.90 ± 6.39 Ma). The Lagkor Tso ophiolite likely developed in an independent suture zone. The Bangonghu-Nujiang ocean subducted southwards,and the dehydration of the subducting oceanic crust materials caused partial melting of the continental mantle wedge,which formed the second-order expanding center of the obduction dish. This led to inter-arc expansion,followed by the formation of inter-arc and back-arc basins with island arc features,which are represented by ophiolites around the Shiquanhe-Lagkor Tso-Yongzhu region. 11Isoliquiritigenin inhibits TGF-β1-induced fibrogenesis through activating autophagy via PI3K/AKT/mTOR pathway in MRC-5 cells显示文摘Isoliquiritigenin(ISL),a natural flavonoid derived from the root of liquorice,has been reported to possess anti-inflammatory and antioxidant activities.Previous studies have found that ISL plays a crucial role in anti-fibrosis of adipose tissue and renal tissue;however,its effect on pulmonary fibrogenesis has not been demonstrated.In this study,we aimed to explore the roles and the underlying mechanisms of ISL in TGF-β1-induced fibrogenesis using human lung fibroblast-derived MRC-5 cells.Cell proliferation and migration were determined by MTT and wound healing assay,respectively.The expression levels of alpha-smooth muscle actin(α-SMA),collagen type I alpha 1(COLIA1)and fibronectin(FN),microtubule-associated protein light chain 3(LC3)and related signaling molecules were detected by quantitative real-time PCR,western blot and immunofluorescence assay,correspondingly.EGFP-LC3 transfection was used for autophagy analysis.The results showed that ISL inhibited the TGF-β1-induced proliferation and migration,and down-regulated the expressions ofα-SMA,COLIA1 and FN.ISL treatment led to up-regulation of LC3 in TGF-β1-treated MRC-5 cells,accompanied by significant decrease in the phosphorylation levels of phosphatidylinositol 3-kinase(PI3K),protein kinase B(AKT),and mammalian target of rapamycin(mTOR).In addition,the inhibitory effects of ISL on TGF-β1-induced fibrogenic features in MRC-5 cells were enhanced by pretreatment with autophagy activator Rapmycin and PI3K/AKT inhibitor LY294002 and reversed by autophagy inhibitor 3-methyladenine and PI3K/AKT activator IGF-1.Taken together,our results demonstrated that ISL could attenuate the fibrogenesis of TGF-β1-treated MRC-5 cells by activating autophagy via suppressing the PI3K/AKT/mTOR pathway.Therefore,ISL holds a great potential to be developed as a novel therapeutic agent for the treatment of pulmonary fibrosis.Jinjuan He Hao Peng Meifang Wang Ying Liu Xingrong Guo Bin Wang Longjun Dai Xueqin Cheng Zhongji Meng Leyong Yuan Fenglin Cai Yijun Tang 2020Acta Biochimica et Biophysica Sinica2020,52,8:16
12Gene Structure and Expression of the High-affinity Nitrate Transport System in Rice Roots显示文摘Rice has a preference for uptake of ammonium over nitrate and can use ammonium-N efficiently. Consequently, transporters mediating ammonium uptake have been extensively studied, but nitrate transporters have been largely ignored. Recently, some reports have shown that rice also has high capacity to acquire nitrate from growth medium, so understanding the nitrate transport system in rice roots is very important for improving N use efficiency in rice. The present study identified four putative NRT2 and two putative NAR2 genes that encode components of the high-affinity nitrate transport system (HATS) in the rice (Oryza sativa L. subsp. japonica cv. Nipponbare) genome. OsNRT2.1 and OsNRT2.2 share an identical coding region sequence, and their deduced proteins are closely related to those from mono-cotyledonous plants. The two NAR2 proteins are closely related to those from mono-cotyledonous plants as well. However, OsNRT2.3 and OsNRT2.4 are more closely related to Arabidopsis NRT2 proteins. Relative quantitative reverse transcription-polymerase chain reaction analysis showed that all of the six genes were rapidly upregulated and then downregulated in the roots of N-starved rice plants after they were re-supplied with 0.2 mM nitrate, but the response to nitrate differed among gene members. The results from phylogenetic tree, gene structure and expression analysis implied the divergent roles for the individual members of the rice NRT2 and NAR2 families. High-affinity nitrate influx rates associated with nitrate induction in rice roots were investigated and were found to be regulated by external pH. 13Small molecule inhibitors reveal allosteric regulation of USP14 via steric blockade显示文摘The ubiquitin system is important for drug discovery,and the discovery of selective small-molecule inhibitors of deubiquitinating enzymes (DUBs) remains an active yet extremely challenging task. With a few exceptions,previously developed inhibitors have been found to bind the evolutionarily conserved catalytic centers of DUBs,resulting in poor selectivity. The small molecule IU1 was the first-ever specific inhibitor identified and exhibited surprisingly excellent selectivity for USP14 over other DUBs. However,the molecular mechanism for this selectivity was elusive. Herein,we report the high-resolution co-crystal structures of the catalytic domain of USP14 bound to IU1 and three IU1 derivatives. All the structures of these complexes indicate that IU1 and its analogs bind to a previously unknown steric binding site in USP14,thus blocking the access of the C-terminus of ubiquitin to the active site of USP14 and abrogating USP14 activity. Importantly,this steric site in USP14 is very unique,as suggested by structural alignments of USP14 with several known DUB X-ray structures. These results,in conjunction with biochemical characterization,Indicate a coherent steric blockade mechanism for U5P14 inhibition by compounds of the IU series. 14Chemical profiling of Qixue Shuangbu Tincture by ultra-performance liquid chromatography with electrospray ionization quadrupole-time-of-flight high-definition mass spectrometry (UPLC-QTOF/MS)显示文摘The present study was designed to develop and validate a sensitive and reliable ultra high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UPLC-QTOF/MS) method to separate and identify the chemical constituents of Qixue Shuangbu Tincture(QXSBT), a classic traditional Chinese medicine(TCM) prescription. Under the optimized UPLC and QTOF/MS conditions, 56 components in QXSBT, including chalcones, triterpenoids, protopanaxatriol, flavones and flavanones were identified and tentatively characterized within a running time of 42 min. The components were identified by comparing the retention times, accurate mass, and mass spectrometric fragmentation characteristic ions, and matching empirical molecular formula with that of the published compounds. 18Colorectal cancer incidence and mortality:the current status,temporal trends and their attributable risk factors in 60 countries in 2000–2019显示文摘Background:Globally,colorectal cancer(CRC)imposes a substantial burden on healthcare systems and confers considerable medical expenditures.We aimed to evaluate the global and regional burden in epidemiological trends and factors associated with the incidence and mortality of CRC.Methods:We used data from the GLOBOCAN database to estimate CRC incidence and mortality worldwide in 2020 and their association with the human development index(HDI).Trends of age-standardized rates of incidence and mortality in 60 countries(2000–2019)were evaluated by Joinpoint regression analysis using data of Global Burden of Disease 2019.The association between exposure to country-level lifestyle,metabolic and socioeconomic factors obtained from the World Health Organization Global Health Observatory and World Bank DataBank data and CRC incidence and mortality was determined by multivariable linear regression.Results:CRC incidence and mortality varied greatly in the 60 selected countries,and much higher incidence and mortality were observed in countries with higher HDIs,and vice versa.From 2000 to 2019,significant increases of incidence and mortality were observed for 33 countries(average annual percent changes[AAPCs],0.24–3.82)and 18 countries(AAPCs,0.41–2.22),respectively.A stronger increase in incidence was observed among males(AAPCs,0.36–4.54)and individuals<50 years(AAPCs,0.56–3.86).Notably,15 countries showed significant decreases in both incidence(AAPCs,0.24 to2.19)and mortality(AAPCs,0.84 to2.74).A significant increase of incidence among individuals<50 years was observed in 30 countries(AAPCs,0.28–3.62).Countries with higher incidence were more likely to have a higher prevalence of alcohol drinking,higher level of cholesterol level,higher level of unemployment,and a poorer healthcare system.Conclusions:Some high-HDI countries showed decreasing trends in CRC incidence and mortality,whereas developing countries that previously had low disease burden showed significantly increased incidence and mortality trends,especially in males and populations≥50 years,which require targeted preventive health programs.Bin Lu Na Li Chen-Yu Luo Jie Cai Ming Lu Yu-Han Zhang Hong-Da Chen Min Dai 2021Chinese Medical Journal2021,,16:13
19Sorafenib after resection improves the outcome of BCLC stage C hepatocellular carcinoma显示文摘AIM: To evaluate whether sorafenib use after resection impacts tumor relapse and survival in Barcelona Clinic Liver Cancer(BCLC) stage C hepatocellular carcinoma(HCC).METHODS: This retrospective study enrolled 36 male BCLC stage C HCC patients with portal vein thrombus and Child-Pugh class A liver function. Twentyfour patients received only surgical resection(SR), and 12 patients received oral sorafenib within 30 d after surgery. The primary outcomes were time to progression(TTP)(the time from surgical resection until HCC recurrence or extrahepatic metastases) and overall survival(OS). The secondary outcome was the rate of postoperative recurrence or metastasis. TTP and OS were analyzed using Kaplan Meier curves.RESULTS: There were no significant differences between the two groups in the serum levels of alpha-fetoprotein, copies of hepatitis B virus-DNA, preoperative laboratory results, degree of hepatic fibrosis, types of portal vein tumor thrombus, number of satellite lesions, tumor diameter, pathological results, volume of blood loss, volume of blood transfusion, or surgery time(all P > 0.05). Patients in the SR + sorafenib group had a significantly longer TTP(29 mo vs 22 mo, P = 0.041) and a significantly longer medianOS(37 mo vs 30 mo, P = 0.01) compared to patients in the SR group. The SR group had 18 cases(75%) of recurrence/metastasis while the SR + sorafenib group had six cases(50%) of recurrence/metastasis. A total of 19 patients died after surgery(five in the SR + sorafenib group and 14 in the SR group). The most common sorafenib-related adverse events were skin reactions, diarrhea, and hypertension, all of which were resolved with treatment.CONCLUSION: Sorafenib after SR was well-tolerated. 20Mechanism of aqueous fructus aurantii immaturus extracts in neuroplexus of cathartic colons显示文摘AIM: To examine the effect of aqueous fructus aurantii immaturus(FAI) extracts on the intestinal plexus of cathartic colons.METHODS: Cathartic colons were induced in rats with dahuang,a laxative used in traditional Chinese medicine. Once the model was established(after approximately 12 wk),rats were administered mosapride(1.54 mg/kg) or various doses of aqueous FAI extracts(1-4 g/kg) for 14 d. Transit function was assessed using an ink propulsion test. Rats were then sacrificed,and the ultramicrostructure of colonic tissue was examined using transmission electron microscopy. The expression of the 5-hydroxytryptamine receptor 4(5-HTR4) and neurofilament-H was assessed in colon tissues using real-time PCR,Western blot,and immunohistochemistry.RESULTS: Mosapride and high dose(4 g/kg) of aqueous FAI extracts significantly improved the bowel movement in cathartic colons compared to untreated model colons as measured by the intestinal transit rate(70.06 ± 7.25 and 72.02 ± 8.74,respectively,vs 64.12 ± 5.19; P < 0.05 for both). Compared to controls,the ultramicrostructure of cathartic colons showed signsof neural degeneration. Treatment with mosapride and aqueous FAI extracts resulted in recovery of ultrastructural pathology. Treatment with mosapride alone upregulated the gene and protein expression of 5-HTR4 compared to untreated controls(P < 0.05 for both). Treatment with aqueous FAI extracts(≥ 2 g/kg) increased 5-HTR4 m RNA levels(P < 0.05),but no change in protein level was observed by Western blot or immunohistochemistry. The m RNA and protein levels of neurofilament-H were significantly increased with mosapride and ≥ 2 g/kg aqueous FAI extracts compared to controls(P < 0.05 for all).CONCLUSION: Aqueous FAI extracts and mosapride strengthen bowel movement in cathartic colons via increasing the expression of 5-HTR4 and neurofilament-H.Shi-Yi Wang Yan-Ping Liu Yi-Hong Fan Lu Zhang Li-Jun Cai Bin Lv 2015World Journal of Gastroenterology2015,21,31:11
